Abstract

Se revelan métodos nuevos para aumentar la masa muscular mediante la inmunización contra el factor de diferenciación de crecimiento 8 (GDF-8, miostatina). La inmunización se efectua preferentemente mediante la administración de análogos de GDF-8 capaces de inducir la producción de anticuerpos contra GDF-8 homólogos. Se prefiere especialmente como un inmunógeno el GDF-8 homólogo modificado por la introducción de un solo o varios, epitopes de célula T extraños inmunodominantes y promiscuos a la vez que se conserva sustancialmente la estructura terciaria del GDF-8 homólogo. También se presenta la vacunación de ácido nucleico contra GDF-8 y la vacunación usando vacunas vivas, como asi también métodos y medios útiles para la vacunación. Dichos métodos y medios incluyen métodos para la identificación de análogos de GDF-8 inmunogénicos, métodos para la preparación de análogos de GDF-8 inmunogénicos, métodos para la preparación de análogos y formulaciones farmacéuticas como asi también fragmentos de ácido nucleico, vectores, células transformadas polipéptidos y formulaciones farmacéuticas.Novel methods of increasing muscle mass are revealed by immunization against growth differentiation factor 8 (GDF-8, myostatin). Immunization is preferably effected by administration of GDF-8 analogs capable of inducing the production of antibodies against homologous GDF-8. Especially preferred as an immunogen is homologous GDF-8 modified by the introduction of one or more, immunodominant and promiscuous foreign T-cell epitopes while substantially conserving the tertiary structure of the homologous GDF-8. Nucleic acid vaccination against GDF-8 and vaccination using live vaccines are also presented, as well as useful methods and means for vaccination. Such methods and means include methods for the identification of immunogenic GDF-8 analogs, methods for the preparation of immunogenic GDF-8 analogs, methods for the preparation of analogues and pharmaceutical formulations as well as nucleic acid fragments, vectors, transformed cells. polypeptides and pharmaceutical formulations.
