Definitions
- the present invention relates to a method for producing natural or synthetic nucleosides, nucleoside analogs or oligonucleotides with at least two identical or different such nucleoside monomers.
- oligonucleotides with a defined sequence are synthesized using the phosphite triester method.
- an oligonucleotide is successively synthesized from its 3'-terminus in the 5'-direction.
- the synthesis begins with the binding of the 3'-terminal nucleoside, which carries a protective group on its 5'-hydroxyl group and, if necessary, on its nucleoside base, to a chemically inert solid phase.
- the 5'-hydroxy protecting group is then split off.
- the internucleotide bond is formed by reacting with a 3'-phosphoramidite of an adequately protected nucleoside.
- the resulting phosphite triester is then oxidized to the desired internucleotide bond.
- the next synthesis cycle can be initiated with the cleavage of the 5'-terminal protective group of the dinucleotide immobilized on the solid phase.
- the synthesis cycles are repeated until one arrives at the finished oligonucleotide bound to the solid phase.
- the synthesis ends with the elimination of the protective groups on the nucleoside bases, hydroxyl groups and the internucleotide bonds (Caruthers, MH, Science 230; 281-285, 1985).
- the nucleoside or oligonucleotide used contains, for example, the groups of the formulas
- the problem with this synthetic process is the condensation reaction between the phosphoramidite and the free hydroxyl group of the nucleosides or oligonucleotides immobilized on the solid phase. During this reaction step, the most embarrassing exclusion of water must be observed. Even through the use of absolute solvents, the absence of water is not completely guaranteed. Therefore, in the coupling step of solid phase oligonucleotide synthesis, considerable amounts of nucleoside phosphoramidite are converted into nucleoside phosphoramidite hydrolyzate, for example the nucleoside cyanoethyl hydrogen phosphonate.
- Bschreibinger (Bauchinger, M.P., thesis, Eidgenössische Technische Hochhoff, ETH No. 9377, 1991) causes the hydrolyzate of a methyl phosphoramidite of a nucleoside analogue to react with sodium hydroxide solution to form the nucleoside analogue with a free hydroxy group. Under the hydrolysis conditions he uses, however, the protective groups of the nucleoside bases are split off.
- the predvoditelev et al. (Predvoditelev, D.A., Tyuganova, M.A., Nifant'ev, E.E., Rogovin, Z.A., Zh. Prikl. Khim. 40: 171-177, 1967), Nifnt'ev et al. (Nifnt'ev, EE, Predvoditelev, DA, Fursenko, LV., Zh. Obsh. Khim. 51: 2435-2441, 1981), Troev and Simeonov (Troev, K., Simeonov, M., Phosphorus and Sulfur 19: 363-367, 1984) and Westheimer et al.
- Another process is the Lewis acid-catalyzed transesterification with terakis-benzyloxy-titanium, which is carried out at 40 ° C. in moderate yields within 72 hours and is particularly disadvantageous to the hydrolysis sensitivity of the titanate (Froneman, M., Modro, TA, Synthesis 201-204, 1991).
- the present invention now provides a process for converting nucleoside hydrogen phosphonates or hydrogen phosphinates into nucleosides which have a free hydroxyl group at the position at which the hydrogen phosphonate or phosphinate function was located.
- the selectivity of the cleavage can in some cases be so great that even after several days of exposure under the given reaction conditions, the protective groups of the nucleoside bases are not cleaved off. This reaction also gives high yields.
- the method according to the invention enables for the first time the recycling of the nucleoside excesses that occur during nucleotide coupling according to the usual phosphite triester method during the routine continuous operation of manual or automated oligonucleotide synthesis.
- the present invention relates in particular to a process for the production of natural or synthetic nucleosides, nucleoside analogs or oligonucleotides from at least two such nucleosides and / or nucleoside analogs, the basic structure of which is an optionally substituted residue of a nucleic base B and a protected hydroxyl group of the formula R 1 -O-, wherein R 1 stands for a protective group, contains, or which are bonded to a solid support material directly or via a bridging group with the oxygen atom of a hydroxyl group, from nucleoside monomers or from oligonucleotides, on the backbone of which, in addition to said radicals, a group of the formulas I, Ia or Ib is bound by converting the group of the formulas I, la or Ib into a hydroxyl group -OH, by reacting the nucleosides or oligonucleotides, optionally in an inert organic solvent, with an excess of an ali
- the groups of the formulas I, Ia and Ib can be bound in the 3'- or 5'-positions of the nucleosides or oligonucleotides, the group R 1 O- or the solid support material in the 5'- or 3'-positions is bound.

Abstract
Description
- Auch die von Predvoditelev et al. (Predvoditelev, D.A., Tyuganova, M.A., Nifant'ev, E.E., Rogovin, Z.A., Zh. Prikl. Khim. 40:171-177, 1967), Nifnt'ev et al. (Nifnt'ev, E.E., Predvoditelev, D.A., Fursenko, LV., Zh. Obsh. Khim. 51:2435-2441, 1981), Troev und Simeonov (Troev, K., Simeonov, M., Phosphorus and Sulfur 19:363-367, 1984) und Westheimer et al. (Westheimer, F.H., Huang, S., Covitz, F., J. Am. Chem.
