- the pellet is recovered and homogenized using a Polytron TM mill in 15 volumes of double-distilled water at 4 ° C., then it is centrifuged at 8000 g for 20 min. The pellet is removed and the supernatant and the skin layer ("buffy coat") are centrifuged at 40,000 g for 20 min. The pellet is recovered, it is resuspended with 15 volumes of double-distilled water at 4 ° C. and it is centrifuged again at 40,000 g for 20 min before being stored at -80 ° C.
- the tissue is thawed slowly and suspended in 5 volumes of buffer.
- 150 ⁇ l of this membrane suspension are preincubated at 37 ° C. for 30 min, in the dark, in the presence or in the absence of the test compound.
- the membranes are then incubated for 60 min at 37 ° C., in the dark, in the presence of 50 ⁇ l of [ 3 H] ⁇ -bungarotoxin at 1 nM in a final volume of 250 ⁇ l of 20 mM HEPES buffer.
- the reaction is stopped by filtration on Whatman GF / C TM filters previously treated for 3 h with 0.05% polyethyleneimine.
- the filters are rinsed with twice 5 ml of buffer at 4 ° C.
- the non-specific binding is determined in the presence of ⁇ -bungarotoxin at 1 ⁇ M; non-specific binding represents approximately 60% of the total binding recovered on the filter.
- the percentage of inhibition of the specific binding of [ 3 H] ⁇ -bungarotoxin is determined, then the IC 50 , concentration of compound which inhibits 50% of the specific binding, is calculated.
- the IC50 values of the most refined compounds of the invention are are between 0.020 and 0.500 ⁇ M.
- the compounds of the present invention have also been studied as regards their affinity with respect to nicotinic receptors containing the ⁇ 4 ⁇ 2 subunit according to the methods described by Anderson and Arneric in Eur. J. Pharmacol. 1994, 253 , 261 and by Hall et al. in Brain Res. 1993, 600 , 127.
- Male Sprague Dawley rats weighing 150 to 200 g are decapitated and the whole brain is quickly removed, homogenized in 15 volumes of 0.32 M sucrose solution at 4 ° C and then centrifuged at 1000 g for 10 min. The pellet is removed and the supernatant is centrifuged at 20,000 g for 20 min at 4 ° C.

- the tablets can be plain, coated, for example with sucrose, or coated with various polymers or other suitable materials. They can be designed to allow rapid, delayed or prolonged release of the active principle thanks to polymer matrices or to specific polymers used in the coating.
- the active ingredient is mixed with dry pharmaceutical vehicles (simple mixing, dry or wet granulation, or hot fusion), liquids or semi-solids.
- dry pharmaceutical vehicles simple mixing, dry or wet granulation, or hot fusion
- liquids or semi-solids can be hard or soft, film-coated or not, so as to have rapid, prolonged or delayed activity (for example for an enteric form).
- a composition in the form of syrup or elixir or for administration in the form of drops may contain active ingredient together with a sweetener, preferably caloric, methylparaben or propylparaben like antiseptic, flavoring agent and colorant.
- a sweetener preferably caloric, methylparaben or propylparaben like antiseptic, flavoring agent and colorant.
- Water dispersible powders and granules can contain the active ingredient in admixture with dispersion or wetting agents, or agents dispersants like polyvinylpyrrolidone, as well as with sweeteners and flavor correcting agents.
- Suppositories are used for rectal administration prepared with binders melting at rectal temperature, for example cocoa butter or polyethylene glycols.
- aqueous suspensions, isotonic saline solutions or sterile injectable solutions containing pharmacologically dispersing and / or wetting agents compatible for example propylene glycol or butylene glycol.
- the active ingredient can also be formulated in the form of microcapsules, possibly with one or more supports or additives, or with a polymer matrix or with a cyclodextrin (transdermal patches, release forms extended).
- compositions according to the invention comprise a medium compatible with the skin. They can present themselves especially in the form of aqueous, alcoholic or hydroalcoholic, gels, water-in-oil emulsions or oil-in-water having the appearance of a cream or gel, microemulsions, aerosols, or in the form of vesicular dispersions containing ionic lipids and / or non-ionic. These dosage forms are prepared according to the usual methods of the fields considered.
- compositions according to the invention may contain, next to a compound of general formula (I), other active ingredients which may be useful in the treatment of the disorders and diseases indicated above.
