Abstract
Die Erfindung betrifft deuterierte substituierte Dihydrofuranone sowie diese Verbindungen enthaltende Arzneimittel.Weiterhin betrifft die Erfindung die Verwendung deuterierter substituierter Dihydrofuranone zur Behandlung von Symptomen bei Reizzuständen degenerativer Gelenkerkrankungen, von akuten Schmerzen und primärer Dysmenorrhoe.Außerdem offenbart die Erfindung pharmazeutische Zusammensetzungen, welche deuterierte substituierte Dihydrofuranone sowie deren physiologisch verträgliche Salze, neben pharmazeutisch verträglichen Hilfs- und/oder Zusatzstoffen, zur Behandlung von Symptomen bei Reizzuständen degenerativer Gelenkerkrankungen, von akuten Schmerzen und primärer Dysmenorrhoe, enthalten.
Description
Deuterierte substituierte Dihydrofuranone sowie diese Verbindungen enthaltende Arzneimittel
Die Erfindung betrifft deuterierte substituierte
Dihydrofuranone sowie diese Verbindungen enthaltende Arzneimittel .
Ein bekannter Vertreter der substituierten Dihydrofuranone ist Rofecoxib (EP 705254, US 5474995), das als selektiver COX-2-Inhibitor unter anderem zur Behandlung von Symptomen bei Reizzuständen degenerativer Gelenkerkrankungen, akuten Schmerzen bei Erwachsenen und primärer Dysmenorrhoe eingesetzt wird.
Aufgabe der vorliegenden Erfindung ist es, substituierte Dihydrofuranone bereitzustellen, die gegenüber den bereits bekannten Verbindungen verbesserte pharmakokinetische und/oder pharmakodynamische Eigenschaften aufweisen.
Überraschenderweise wurde nun gefunden, dass die erfindungsgemäßen deuterierten substituierten Dihydrofuranone wesentlich bessere pharmakokinetische und/oder pharmakodynamische Eigenschaften aufweisen als die undeuterierten Verbindungen.
Erfindungsgemäß wird die Aufgabe also gelöst durch die Bereitstellung deuterierter substituierter Dihydrofuranone der allgemeinen Formel I:

worin R1 Methyl oder teilweise oder vollständig deuteriertes Methyl ist, R2 unabhängig voneinander H oder D bedeutet, R3 unabhängig voneinander H oder D ist, R4 unabhängig voneinander H oder D bedeutet und wobei mindestens einer der Reste R1 bis R4 D ist oder D enthält.
Bevorzugt sind deuterierte substituierte Dihydrofuranone gemäß der allgemeinen Formel I, worin R1 teilweise oder vollständig deuteriertes Methyl ist, R2 unabhängig voneinander H oder D bedeutet, R3 unabhängig voneinander H oder D ist und R4 unabhängig voneinander H oder D bedeutet.
Besonders bevorzugt sind deuterierte substituierte Dihydrofuranone gemäß der allgemeinen Formel I, worin R1 Methyl oder teilweise oder vollständig deuteriertes Methyl ist, R2 D bedeutet, R3 unabhängig voneinander H
oder D ist und R4 unabhängig voneinander H oder D bedeutet.

Weiterhin bevorzugt ist die Verwendung der deuterierten substituierten Dihydrofuranone gemäß der allgemeinen Formel I sowie deren physiologisch verträglicher Salze, zur Behandlung von Symptomen bei Reizzuständen degenerativer Gelenkerkrankungen, von akuten Schmerzen und primärer Dysmenorrhoe.
Besonders bevorzugt ist die Verwendung der deuterierten substituierten Dihydrofuranone gemäß der allgemeinen
Formel I sowie deren physiologisch verträglicher Salze, zur Herstellung von Arzneimitteln zur Behandlung von Symptomen bei Reizzuständen degenerativer Gelenkerkrankungen, von akuten Schmerzen und primärer Dysmenorrhoe.
Insbesondere bevorzugt sind pharmazeutische Zusammensetzungen, welche deuterierte substituierte Dihydrofuranone gemäß der allgemeinen Formel I sowie deren physiologisch verträgliche Salze zur Behandlung von
Symptomen bei Reizzustanden degenerativer Gelenkerkrankungen, von akuten Schmerzen und primärer Dysmenorrhoe, neben pharmazeutisch vertraglichen Hilfs- und/oder Zusatzstoffen, enthalten.
Die Herstellung der erfindungsgemaßen deuterierten substituierten Dihydrofuranone erfolgt zum Teil nach bekannten Herstellungsverfahren für deuterierte
Verbindungen sowie nach Verfahren zur Herstellung der undeuterierten Analoga unter Verwendung von deuterierten
Edukten mit einem Deuterierungsgrad über 98%.
So wird durch Reaktion eines gegebenenfalls deuterierten α-Brom-4- (methylsulfonyl) acetophenons, das durch Bromierung von gegebenenfalls deuterierten 4-
(Methylsulfonyl) acetophenon zugänglich ist (analog US 2763692 oder EP 705254), mit gegebenenfalls deutenerter Phenylessigsaure zur Reaktion gebracht. In Abhängigkeit der verwendeten Edukte sind die erfindungsgemaßen Dihydrofuranone in den Positionen R1 und/oder R2 und/oder R3 und/oder R4 deuteπert.
Zur Synthese von deuteπertem α-Brom-4- (methylsulfonyl) acetophenon stehen verschiedene Reaktionswege zur Verfugung.
So kann nach Miura et al. (Bulletin of the Chemical Society of Japan, Vol. 50, Nr. 5, 1142-1146, 1977) ausgehend von 2, 3, 4, 5, 6-d5-Anιlm das 2, 3, 4, 5, 6-d5- Thiophenol hergestellt werden, das dann mit Methanol oder deuteriertem Methanol analog DE 2649590 oder
Dimethylsulfat oder deuteriertem Dimethylsulfat analog EP 206677 S-methyliert wird. Das deuterierte Methylsulfid wird dann durch Friedel-Crafts-Acylierung, zum Beispiel analog Cutler et al. (Journal of the American Chemical Society, Vol. 74, S. 5475-5481, 1952), unter Verwendung
von gegebenenfalls deuteriertem Acetylchlorid zu deuteriertem 4- (Methylthio) acetophenon umgesetzt. Aus diesem erhält man durch Oxidation, z.B. analog EP 705254, deuteriertes 4- (Methylsulfonyl) acetophenon.
Ein weiterer Syntheseweg für deuteriertes 4- (Methylsulfonyl) acetophenon führt über deuteriertes 4- Chloracetophenon, das durch Friedel-Crafts-Acylierung aus deuteriertem Chlorbenzol zugänglich ist. Dieses Ethanon wird mit gegebenenfalls deuteriertem
Natriummethanthiolat, beispielsweise analog JP 8143534, zum deuterierten 4- (Methylthio) acetophenon umgesetzt. Das deuterierte Natriummethanthiolat kann analog DE 1804266 aus deuteriertem Methylsulfid durch Umsetzung mit NaOH erhalten werden.
Die Oxidation zum deuterierten 4- (Methylsulfonyl) - acetophenon erfolgt dann wie im ersten Reaktionsweg beschrieben.
Die zur Herstellung der erfindungsgemäßen deuterierten
Dihydrofuranone verwendete deuterierte Phenylessigsäure ist kommerziell erhältlich sowie analog bekannter
Herstellungsverfahren für die nicht-deuterierte
Verbindung zugänglich.
Zur Herstellung physiologisch verträglicher Salze der erfindungsgemäßen deuterierten substituierten
Dihydrofuranone können übliche physiologisch verträgliche anorganische und organische Säuren verwendet werden. Das sind beispielsweise Salzsäure, Bromwasserstoffsäure, Phosphorsäure, Schwefelsäure, Oxalsäure, Maleinsäure, Fumarsäure, Milchsäure, Weinsäure, Apfelsäure, Citronensäure, Salicylsäure, Adipinsäure und Benzoesäure. Weitere verwendbare Säuren sind beispielweise in Fortschritte der Arzneimittelforschung, Bd. 10, Seiten
224-225, Birkhäuser Verlag, Basel und Stuttgart, 1966, und Journal of Pharmaceutical Sciences, Bd. 66, Seiten 1- 5 (1977) beschrieben.
Die Säureadditionssalze werden in der Regel in an sich bekannter Weise durch Mischen der freien Base oder deren Lösungen mit der entsprechenden Säure oder deren Lösungen in einem organischen Lösungsmittel, beispielsweise einem niederen Alkohol wie Methanol, Ethanol, n-Propanol oder Isopropanol oder einem niederen Keton wie Aceton, Methyl- ethylketon oder Methyl-isobutylketon oder einem Ether wie Diethylether, Tetrahydrofuran oder Dioxan, erhalten. Zur besseren Kristallabscheidung können auch Mischungen der genannten Lösungsmittel verwendet werden. Darüber hinaus können physiologisch verträgliche wässrige Lösungen von Säureadditionssalzen der erfindungsgemäß verwendeten Verbindungen in einer wässrigen Säurelösung hergestellt werden.
Die Säureadditionssalze der erfindungsgemäßen
Verbindungen können in an sich bekannter Weise, z. B. mit Alkalien oder Ionenaustauschern, in die freie Base überführt werden. Von der freien Base lassen sich durch Umsetzung mit anorganischen oder organischen Säuren, insbesondere solchen, die zur Bildung von therapeutisch verwendbaren Salzen geeignet sind, weitere Salze gewinnen. Diese oder auch andere Salze der neuen Verbindung, wie z. B. das Pikrat, können auch zur Reinigung der freien Base dienen, indem man die freie Base in ein Salz überführt, dieses abtrennt und aus dem Salz wiederum die Base freisetzt.
Gegenstand der vorliegenden Erfindung sind auch Arzneimittel zur oralen, rektalen, topischen (percutan, transdermal, lokal) , subcutanen, intravenösen oder intramuskulären Applikation, die neben üblichen Träger- und Verdünnungsmitteln eine Verbindung der allgemeinen Formel I oder deren Säureadditionssalz als Wirkstoff enthalten.
Die Arzneimittel der Erfindung werden mit den üblichen festen oder flüssigen Trägerstoffen oder Verdünnungsmitteln und den üblicherweise verwendeten pharmazeutischtechnischen Hilfsstoffen entsprechend der gewünschten Applikationsart mit einer geeigneten Dosierung in bekannter Weise hergestellt. Die bevorzugten Zubereitungen bestehen in einer Darreichungsform, die zur oralen Applikation geeignet ist. Solche Darreichungsformen sind beispielsweise Tabletten, Filmtabletten, Dragees, Kapseln, Pillen, Pulver, Lösungen oder Suspensionen oder Depotformen.
Die topische Anwendung kann beispielsweise in der Form von Salben, Cremes, Gelen, Lösungen oder durch Pflaster erfolgen.
Selbstverständlich kommen auch parenterale Zubereitungen wie Injektionslösungen in Betracht. Weiterhin seien als Zubereitungen beispielsweise auch Suppositorien genannt. Entsprechende Tabletten können beispielsweise durch Mischen des Wirkstoffs mit bekannten Hilfsstoffen, bei- spielsweise inerten Verdünnungsmitteln wie Dextrose,
Zucker, Sorbit, Mannit, Polyvinylpyrrolidon, Sprengmitteln wie Maisstärke oder Alginsäure, Bindemitteln wie
Stärke oder Gelantine, Gleitmitteln wie Magnesiumstearat oder Talk und/oder Mitteln zur Erzielung eines Depoteffektes wie Carboxylpolymethylen, Carboxylmethylcellulo- se, Celluloseacetatphthalat oder Polyvinylacetat, erhal- ten werden. Die Tabletten können auch aus mehreren Schichten bestehen.
Entsprechend können Dragees durch Überziehen von analog den Tabletten hergestellten Kernen mit üblicherweise in Drageeüberzügen verwendeten Mitteln, beispielsweise
Polyvinylpyrrolidon oder Schellack, Gummiarabicum, Talk, Titandioxid oder Zucker, hergestellt werden. Dabei kann auch die Drageehülle aus mehreren Schichten bestehen, wobei die oben bei den Tabletten erwähnten Hilfsstoffe verwendet werden können.
Lösungen oder Suspensionen mit dem erfindungsgemäß verwendeten Wirkstoff können zusätzlich geschmacksverbessernde Mittel wie Saccharin, Cyclamat oder Zucker sowie z. B. Aromastoffe wie Vanillin oder Orangenextrakt enthalten. Sie können außerdem Suspendierhilfsstoffe wie Natriumcarboxymethylcellulose oder Konservierungsstoffe wie p-Hydroxybenzoate enthalten. Wirkstoffe enthaltende Kapseln können beispielsweise hergestellt werden, indem man den Wirkstoff mit einem inerten Träger wie Milchzucker oder Sorbit mischt und in Gelatinekapseln einkapselt. Geeignete Suppositorien lassen sich beispielsweise durch Vermischen mit dafür vorgesehenen Trägermitteln wie Neutralfetten oder Polyäthylenglykol bzw. deren Derivaten herstellen.
Die Herstellung der erfindungsgemäßen Arzneimittel zur topischen Applikation ist dem Fachmann bekannt. Bei der Herstellung der erfindungsgemäßen Arzneimittel zur transdermalen Anwendung werden die an sich bekannten Hilfs- und Enhancerstoffe verwendet.
Die Herstellung der erfindungsgemäßen pharmazeutischen Zubereitungen ist an sich bekannt und in den dem Fachmann bekannten Handbüchern beschrieben, beispielsweise Hager' s Handbuch (5.) 2, 622-1045; List et al.,
Arzneiformenlehre, Stuttgart: Wiss. Verlagsges. 1985; Sucker et al., Pharmazeutische Technologie, Stuttgart: Thieme 1991; Ulimann' s Enzyklopädie (5.) A 19, 241-271; Voigt, Pharmazeutische Technologie, Berlin: Ullstein Mosby 1995.
Die erfindungsgemäßen, gezielt mit Deuterium substituierten Verbindungen weisen gegenüber den im Stand der Technik bekannten Verbindungen, welche Deuterium lediglich in der natürlichen Verteilung enthalten, eine Reihe von Vorteilen auf. Durch die Deuterierung wird zum einen die Metabolisierung im Organismus verlangsamt. Hierdurch ist es möglich, die Dosierung zu verändern und länger wirkende Zubereitungen zu schaffen, welche auch in Form von Depotzubereitungen die Compliance verbessern können.
Daneben ist auch die Pharmakodynamik verändert, da die erfindungsgemäßen deuterierten Verbindungen andere Hydrathüllen ausbilden, so dass sich ihre Verteilung im Organismus von der der undeuterierten Verbindungen unterscheidet .
Damit ist es möglich, neuartige Zubereitungsformen zu entwickeln.
Die folgenden Beispiele erläutern die Erfindung:
Beispiel 1
Herstellung von dl0-4- (Methylsulfonyl) acetophenon
Zu 20,9 g dl0-4- (Methylthio) acetophenon in einem Gemisch aus 70 ml Methanol und 350 ml Dichlormethan werden unter Ruhren innerhalb von 30 Minuten 88,1 g des Magnesiumsalzes der Monoperoxyphthalsaure hinzugefugt. Der Reaktionsansatz wird für 3 Stunden bei Raumtemperatur gerührt, anschließend filtriert und das Filtrat nacheinander mit 200 ml wassriger, gesättigter
Natπumhydrogencarbonatlosung und 100 ml Kochsalzlosung gewaschen. Die wassrige Phase wird mit 200 ml Dichlormethan extrahiert, die organischen Phasen werden kombiniert, getrocknet und das Losemittel entfernt. Man erhalt 23,75 g Produkt als weißen Feststoff. Ausbeute: 96 % Schmelzpunkt: 123-127 °C berechnet : C: 51, 89 %, H: 9, 67 % gefunden:
C: 51, 99 %; H: 9,72 %

Beispiel 2
Herstellung von d9-α-Brom-4- (methylsulfonyl) acetophenon 36,57 g dl0-4- (Methylsulfonyl) acetophenon werden in
500 ml Chloroform gelost und die Losung w rd auf -5 °C
abgekühlt. Es werden nacheinander 4 mg Aluminiumchlorid und 8 ml Brom in 60 ml Chloroform hinzugefugt. Der Reaktionsansatz wird mit 300 ml Wasser versetzt und die organische Phase abgetrennt. Die wassrige Phase wird mit 200 ml Essigsaureethylester extrahiert, die organischen Phasen werden kombiniert und getrocknet. Das Losemittel wird entfernt und das zurückbleibende Rohprodukt aus einem Gemisch aus Essigsaureethylester und Hexan im Verhältnis 1:1 umkπstallisiert . Man isoliert 42,43 g Produkt als weißen Feststoff. Ausbeute: 95 % Schmelzpunkt: 124-126 °C berechnet : C: 37,77 %; H: 6,33 % gefunden:
C: 37, 66 % ; H: 6,20 %

Beispiel 3
Herstellung von dl4-4- [4- (Methylsulfonyl) phenyl] -3- phenyl-2- (5H) -furanon 14,4 g α, α, 2 , 3, 4 , 5, 6-d7-Phenylessιgsaure werden bei
25 °C mit 31 g d9-α-Brom-4- (methylsulfonyl) acetophenon m 315 ml Acetonitril gelost und langsam mit 15,4 ml Tπethylamin versetzt. Das Gemisch wird für 20 Minuten bei Raumtemperatur gerührt und es werden unter Eiskuhlung langsam 30,05 ml 1, 8-Dιazabιcyclo [5.4.0] undec-7-en hinzugefugt. Die Eiskuhlung wird beibehalten und der Reaktionsansatz für 20 Minuten gerührt. Nach dem Ansäuern mit Salzsaure werden 1200 ml eines Eis/Wasser-Gemischs hinzugegeben und nach einigen Minuten kann das ausgefallene Rohprodukt abfiltriert werden. Der Feststoff wird mit Wasser gewaschen und in 375 ml Chloroform
gelöst. Das Lösemittel wird getrocknet und ihm werden anschließend 150 g Kieselgel zugefügt. Das Chloroform wird so weit wie möglich entfernt und das Produkt anschließend mit einem 10%-igen Gemisch aus Essigsäureethylester/Dichlormethan aus dem Kieselgel eluiert. Aus der Lösung werden 18,59 g Produkt als schwachgelber Feststoff gewonnen. Ausbeute: 56 % Schmelzpunkt: 205-207 °C berechnet:
C: 62,17 %; H: 8,58 % gefunden:
C: 62,35 %; H: 8,67 %

11. Deuterierte substituierte Dihydrofuranone gemäß Anspruch 1, wobei
R1 Methyl oder teilweise oder vollständig deuteriertes Methyl ist,
R2 unabhängig voneinander H oder D bedeutet und R3 und R4 D sind.
12. Deuterierte substituierte Dihydrofuranone gemäß Anspruch 1, wobei
R1 teilweise oder vollständig deuteriertes Methyl ist, R2 uns R3 D sind und
R4 unabhängig voneinander H oder D bedeutet.
13. Deuterierte substituierte Dihydrofuranone gemäß Anspruch 1, wobei R1 teilweise oder vollständig deuteriertes Methyl ist,
R2 D bedeutet,
R3 unabhängig voneinander H oder D ist und
R4 D bedeutet.
14. Deuterierte substituierte Dihydrofuranone gemäß Anspruch 1, wobei
R1 teilweise oder vollständig deuteriertes Methyl ist, R2 unabhängig voneinander H oder D bedeutet und R3 und R4 D bedeuten.
15. Deuterierte substituierte Dihydrofuranone gemäß Anspruch 1, wobei R1 Methyl oder teilweise oder vollständig deuteriertes Methyl ist und R2, R3 und R4 D bedeuten.
16. Deuterierte substituierte Dihydrofuranone gemäß Anspruch 1, wobei
R1 teilweise oder vollständig deuteriertes Methyl ist
und
R2, R3 und R4 D bedeuten.

32. Verwendung der deuterierten substituierten
Dihydrofuranone gemäß einem der Ansprüche 1 bis 31 sowie deren physiologisch verträglicher Salze zur Behandlung von Symptomen bei Reizzuständen degenerativer Gelenkerkrankungen, von akuten Schmerzen und primärer Dysmenorrhoe.
33. Verwendung der deuterierten substituierten
Dihydrofuranone gemäß einem der Ansprüche 1 bis 31 sowie deren physiologisch verträglicher Salze zur Herstellung von Arzneimitteln zur Behandlung von Symptomen bei Reizzuständen degenerativer Gelenkerkrankungen, von akuten Schmerzen und primärer Dysmenorrhoe.
34. Pharmazeutische Zusammensetzung, welche deuterierte substituierte Dihydrofuranone gemäß einem der Ansprüche 1 bis 31 sowie deren physiologisch verträgliche Salze zur Behandlung von Symptomen bei Reizzuständen degenerativer Gelenkerkrankungen, von akuten Schmerzen und primärer Dysmenorrhoe, neben
pharmazeutisch verträglichen Hilfs- und/oder Zusatzstoffen, enthält.
