Abstract
L'invention concerne des procédés, des compositions et des kits pour éliminer les faux uraciles positifs en raison de la désamination de cytosines non méthylées dans des dosages à l'aide de cytosine désaminases modifiées pour désaminer des cytosines méthylées. Les procédés, les compositions et les kits utilisent un procédé enzymatique double. Après le traitement de la cytidine désaminase, l'ADN est d'abord incubé avec de l'uracile ADN glycosylase (UDG), qui génère des sites abasiques où des conversions de dC en dU se sont produites. L'ADN est ensuite incubé avec une polymérase à haute fidélité complétée par une désoxycytidyl transférase, telle que Rev1, pour réparer la lésion avec l'installation d'une cytidine.
