PROCEDIMIENTO PARA LA DETECCION SENSIBLE Y ESPECIFICA DE ERWINIA CAROTOVORA SUBSP. ATROSEPTICA MEDIANTE "ELISA-ENRIQUECIMIENTO". EL PROCEDIMIENTO PARTE DE LA INSOLUBILIZACION DE ANTIGENOS DEL AGENTE PATOGENO ERWINIA CAROTOVORA SUBSP. ATROSEPTICA (ECA) EN MICROPLACAS, MEDIANTE ANTICUERPOS MONOCLONALES O POLICLONALES. POSTERIORMENTE, EN LA MISMA MICROPLACA, SE AÑADE LA MUESTRA OBJETO DE ESTUDIO ADICIONADA CON UN MEDIO SELECTIVO PARA ERWINIAS (D-PEM) Y SE INCUBA EN ANAEROBIOSIS A 25 C DURANTE 24-48 HORAS. LA REACCION SE REVELA MEDIANTE ANTICUERPOS MONOCLONALES ESPECIFICOS DE ECA (4G4) BIOTINILADOS Y POSTERIOR ADICION DE ESTREPTOAVIDINA MARCADA CON UNA ENZIMA O DIRECTAMENTE CON ANTICUERPOS 4G4 MARCADOS CON UNA ENZIMA (ELISA-DAS O DAS BIOTINA/ESTREPTOAVIDINA) O CON ANTICUERPOS 4G4 SIN MARCAR Y POSTERIOR ADICION DE UN CONJUGADO ANTI-ESPECIE (ELISA-DASI). LA HIDROLISIS DEL SUBSTRATO ENZIMATICO IMPLICA LA APARICION DE UN PRODUCTO COLOREADO QUE ES VALORADO FOTOCOLORIMETRICAMENTE.
