Abstract

Una células son genéticamente modificadas de manera que expresen un luminoforo, por ejemplo una proteína fluorescente verde (GFP, EGFP) modificada (F64L, S65T, Y66H) acoplada a un componente de una vía de señalización intracelular, como un factor de transcripción, una proteína quinasa GMPc o AMPc dependiente, una serina/treonina quinasa dependiente de cilina, de calmodulina, o de fosfolípido o activada por mitógeno, una tirosina quinasa, o una proteína fosfatasa (por ejemplo PKA, PKC, Erk, Smad, VASP, actina, p38, Jnk1, PKG, I{ka}B, CDK2, Grk5, Zap70, p85, proteína-tirosina fosfatasa 1C, Stat5, NFAT, NFl{ka}B, RhoA, PKB). Una influencia permite modular la vía de señalización intracelular de tal manera que el luminóforo se redistribuye o transloca con el componente en células vivas de manera determinada experimentalmente, para una correlación con el grado de influencia. Se mide la redistribución registrando la intensidad luminosa, la duración de vida de la fluorescencia, la polarización, eldesplazamiento de longitud de onda, la transferencia de energía de resonancia o de otras propiedades mediante un aparato constituido, por ejemplo, por un microscopio de fluorescencia y una cámara CCD. Unos datos almacenados en forma de imágenes digitales son tratados para obtener números que representan el grado de redistribución. El procedimiento puede utilizarse como un programa de clasificación PATRA identificar un compuesto que modula un componente y es susceptible de tratar una enfermedad asociada a la función del componente.
