Abstract
Método para detectar y/o purificar complejos de biomoléculas y/o proteínas que comprende los pasos: (a) proporcionar un ambiente de expresión que contiene uno o más ácidos nucleicos heterólogos que codifican una o más subunidades de un complejo de biomoléculas, estando las subunidades fusionadas a al menos dos marcadores de afinidad diferentes, uno de los cuales está constituido por uno o más dominios de fijación de IgG de la proteína A de Staphylococcus, (b) mantener el ambiente de expresión en condiciones que facilitan la expresión de la una o más subunidades en una forma nativa como proteínas de fusión con los marcadores de afinidad, y en condiciones que permiten la formación de un complejo entre la una o más subunidades y posiblemente otros componentes capaces de complejarse con la una o más subunidades, (c) detectar y/o purificar la una o más subunidades por una combinación de al menos dos pasos de purificación por afinidad diferentes cada uno de los cuales comprende fijar la una omás subunidades por la vía de un marcador de afinidad a un material soporte capaz de fijar selectivamente uno de los marcadores de afinidad y separar la una o más subunidades del material soporte después que las sustancias no fijadas al material soporte se han eliminado.
