Abstract
Una composición de goma de silicona curable antimicrobiana que comprende en al menos una porción de la superficie expuesta de dicha composición una matriz orgánica que contiene partículas de plata metálica homogéneamente dispersas que tienen un tamaño de partícula en el intervalo de 1 a 50 nm (nanopartículas de plata) en una cantidad que proporciona una concentración de plata antimicrobiana pero menos que citotóxica de más de 1 nmol/l a menos de 1 µmol/l sobre al menos una porción de la superficie de dicha composición.

La presente invención se refiere a una
composición de goma de silicona curable antimicrobiana.
Las composiciones de goma de silicona se han
usado ampliamente en la industria para una amplia variedad de
aplicaciones diferentes. Estas aplicaciones incluyen por ejemplo
mangos de sartenes, tapas de lentes de cámaras, manillares de
bicicletas, antideslizantes para espectáculos, varias láminas de
goma y tejidos recubiertos de goma tales como láminas y cortinas
que se usan por ejemplo en hospitales. Las composiciones de goma de
silicona se usan además como tubos y revestimientos para recipientes
de alimentos o varios tipos de objetos higiénicos.
En muchas de estas aplicaciones, existe una
demanda de eficacia antimicrobiana y antifúngica de las
composiciones de goma de silicona. Esta demanda surge del hecho de
que muchos de los entornos de uso son proclives a la contaminación
microbiana o fúngica. Por ejemplo, tales entornos de uso incluyen la
presencia de superficies contaminadas tales como a y/o la presencia
de humedad ambiental en combinación con temperaturas ambiente o
incluso mayores.
En la solicitud de patente japonesa nº
7-65149, presentada el 28 de febrero de 1995 se ha
sugerido incorporar en una composición de goma de silicona un
agente antimicrobiano inorgánico, comprendiendo el agente plata
cargada sobre un material inorgánico tal como zeolita, un fosfato de
circonio o un fosfato de calcio. Tales composiciones de goma de
silicona no obstante son sólo capaces de liberar los átomos de plata
en forma iónica y por lo tanto son citotóxicas.

composición de goma de silicona curable antimicrobiana que comprende
en al menos una porción de la superficie expuesta de dicha
composición una matriz orgánica que contiene partículas
homogéneamente dispersas de plata metálica que tienen un tamaño de
partícula en el intervalo de 1 a 50 nm (nanopartículas de plata) en
una cantidad que proporciona sobre la superficie de dicha
composición una concentración de plata eficaz en sentido
antimicrobiano pero menos que citotóxica de más de 1 nmol/l a menos
de 1 \mumol/l sobre al menos una porción de la superficie de
dicha composición.
La presente invención proporciona además una
composición de goma de silicona según las reivindicaciones 1 y 2,
caracterizada porque dicha matriz orgánica comprende dichas
nanopartículas de plata en una cantidad de 1 a 2000 ppm,
preferiblemente de 5 a 1000 ppm y más preferiblemente de 10 a 250
ppm.
La presente invención proporciona además una
composición de goma de silicona caracterizada porque comprende
nanopartículas de plata que tienen un tamaño de partícula de 2 a 20
nm, preferiblemente de 5 a 10 nm.
La presente invención proporciona además una
composición de goma de silicona caracterizada porque dicha matriz
orgánica fluida comprende un fluido orgánico en el que están
dispersas dichas nanopartículas de plata.
La presente invención proporciona además una
composición de goma de silicona caracterizada porque dicho fluido
orgánico viscoso comprende un hidrocarburo alifático o aromático, un
aceite mineral, vaselina, glicerol, un alcohol graso,
polipropilenglicol, un aceite te o grasa animal y/o vegetal o un
aceite de silicona.
La presente invención proporciona además un
procedimiento para fabricar una composición de goma de silicona
curable antimicrobiana que comprende las etapas de:
- -
- proporcionar una composición de goma de silicona curable
- -
- proporcionar una matriz líquida orgánica que comprende plata metálica que tiene un tamaño de partícula en el intervalo de 1 a 50 nm
- -
- mezclar dicha matriz líquida orgánica en dicha composición de goma de silicona en una cantidad que proporciona una concentración de plata antimicrobiana pero menos que citotóxica de más de 1 nmol/l a menos de 1\muml/l sobre al menos una porción de la superficie de dicha composición.
- -
- curar opcionalmente la mezcla de dicha composición con dicha matriz orgánica.

Se ha descubierto sorprendentemente que la
composición de goma de silicona antimicrobiana de la presente
invención muestra una excelente actividad antimicrobiana y
fungicida no proporcionando citotoxicidad alguna y es activa frente
a una amplia gama de microbios sin indicación de resistencia alguna
a causa de las muy pequeñas cantidades de plata liberadas a partir
de la superficie de dicha composición de goma de silicona a lo largo
de un periodo prolongado. Basándose en esto, el producto para el
cuidado corporal de la presente invención es idealmente adecuado
para aplicaciones en las que dicha composición se usa en condiciones
que permiten la proliferación de microbios y
hongos.

Para el propósito de la presente invención, la
eficacia antimicrobiana, es decir, la actividad frente a una amplia
gama de microbios se puede cuantificar por ejemplo mediante
procedimientos tales como los descritos en "Der Erlanger
Silberkatheter: In-vitro Ergebnisse zur
antimikrobielle Wirksamkeit" en Infection 26 (1998), Supl. 1,
edición alemana, páginas 25 a 31.
Para el propósito de la presente invención, la
citotoxicidad se puede cuantificar por ejemplo mediante
procedimientos tales como los descritos en "Untersuchung der
akuten Zytotoxizität des Erlanger Silberkatheters zur Bestimmung
der Biokompatibilität" en Infection 26 (1998), Supl. 1, edición
alemana, páginas 36 a 39.
Según una forma de realización preferida de la
presente invención, dichas nanopartículas de plata están dispersas
en dicha matriz de goma de silicona en una cantidad tal que se
proporciona una concentración de plata de más de 1 nmol/l a menos
de 1 \mumol/l, específicamente cuando está en contacto con fluidos
acuosos presentes sobre la superficie de la composición de la
presente invención.
Preferiblemente, al menos las superficies
expuestas de la composición de goma de silicona de la presente
invención comprende dichas nanopartículas de plata en una cantidad
de 1 a 2000 ppm, preferiblemente de 5 a 1000 ppm y más
preferiblemente de 10 a 250 ppm.
Las nanopartículas de plata homogéneamente
dispersas en dicha matriz de goma de silicona preferiblemente tienen
un tamaño de partícula de al menos 2 nm, más preferiblemente de al
menos 5 nm. Además, las nanopartículas de plata dispersas tienen
preferiblemente un tamaño de partícula de no más de 20 nm, más
preferiblemente de no más de 10 nm, de la forma más preferible de
no más de 8 nm.
La matriz de goma de silicona en la que dichas
nanopartículas de plata se dispersan después puede ser sólida o
fluida. El término fluida incluye líquida y semisólida y cubre un
intervalo de viscosidad de 5 a 5000 mPa.s, preferiblemente de 5 a
500 mPa.s medido a 60ºC usando un viscosímetro rotatorio (tal como
un viscosímetro de Brookfield) a 60 rpm usando un husillo del
número 2.
Según una forma de realización de la presente
invención, al menos las superficies expuestas de la composición de
goma de silicona de la presente invención comprenden una matriz
fluida orgánica en la que están dispersas homogéneamente dichas
nanopartículas de plata. Dicha matriz orgánica fluida
preferiblemente es un fluido orgánico viscoso que tiene una
viscosidad como se ha mencionado anteriormente y comprende
preferiblemente un hidrocarburo alifático o aromático, un aceite
mineral, vaselina, glicerol, un alcohol graso, polipropilenglicol,
un aceite te o grasa animal y/o vegetal o un aceite de silicona. Se
prefieren específicamente aceites de silicona, es decir,
polisiloxanos tales como polimetilsiloxano
fenil-funcional que tiene una viscosidad a 37ºC que
varía de 5 a 5000 mPa.s, más preferiblemente de 5 a 2000 mPa.s,
según se mide con el viscosímetro mencionado anteriormente a una
temperatura de 37ºC. Un aceite de silicona adecuado está disponible
de Dow Corning Corporation, Michigan, EE.UU., con la designación
DC556 polidimetilsilicona de calidad cosmética (dimeticona).
La incorporación de nanopartículas de plata se
puede hacer con un aparato tal como se describe en la patente
alemana 44 40 521 usando un procedimiento que implica la evaporación
a vacío de plata metálica sobre un líquido en una cámara de vacío
en la que el fluido orgánico fluye a lo largo del exterior de un
cilindro. Esto permite la preparación de una matriz orgánica
líquida que comprende una dispersión homogénea extremadamente fina
de nanopartículas de plata que tienen un tamaño de partícula de
aproximadamente 5 nm. Esta matriz orgánica líquida que contiene
plata se puede usar para recubrir la parte del producto de cuidado
corporal de la presente invención que está en contacto con la piel
y/o para impregnar o para incorporar mediante amasado en el material
proporcionando tal superficie, hecha por ejemplo de
polivinilbenceno, polietileno, polipropileno u oligómeros cremosos,
tales como vaselina
blanca.

Una composición de goma líquida adecuada
comprende 100 partes en peso de goma de organopolisiloxano en la que
la unidad media está representada por
R_{a}SiO_{(4-a)/2}
(en la que R es un radical
hidrocarburo monovalente sustituido o no sustituido y ocho está
entre 1,95 y 2,05), 10-100 partes en peso de sílice
húmeda con un área superficial específica de 50 metros cuadrados por
gramo o más y una cantidad suficiente de un agente
endurecedor.
En principio se pueden hacer antimicrobianas y
antifúngicas todas las composiciones de goma de silicona conocidas
en la técnica con los medios descritos en el presente documento.
Ejemplo
Se produjo una suspensión que contenía
nanopartículas de plata con un intervalo de tamaño individual de 5
a 50 nm mediante evaporación térmica de plata en una base de aceite
de silicona líquido. El aceite de silicona que contenía las
nanopartículas de plata se incorporó entonces en una goma de
silicona obtenida mediante condensación de adición y reticulación
proporcionando un mordedor para niños.
El mordedor para niños o chupete demostró
eficacia antifúngica y antibacteriana cuando contenía 50 ppm de
nanopartículas de plata cuando se añadieron directamente en un
componente antes de la reticulación. Las muestras se colocaron en
agar salino mineral (no nutritivo) y se inocularon con una
suspensión mixta de esporas fúngicas de Aspergillus Níger y
Penicillium oinophilium, Chaetomium globosum, Aureobasidium
pullulans y Gliocadium virens. Después de 28 días de
incubación a 28ºC no se observó crecimiento alguno de ninguno de los
microorganismos.
Claims (6)
1. Una composición de goma de silicona curable
antimicrobiana que comprende en al menos una porción de la
superficie expuesta de dicha composición una matriz orgánica que
contiene partículas de plata metálica homogéneamente dispersas que
tienen un tamaño de partícula en el intervalo de 1 a 50 nm
(nanopartículas de plata) en una cantidad que proporciona una
concentración de plata antimicrobiana pero menos que citotóxica de
más de 1 nmol/l a menos de 1 \mumol/l sobre al menos una porción
de la superficie de dicha composición.
2. La composición de goma de silicona según la
reivindicación 1, caracterizada porque dicha composición
comprende dichas nanopartículas en una cantidad de 1 a 2000 ppm,
preferiblemente de 5 a 1000 ppm, más preferiblemente de 10 a 250
ppm.
3. La composición de goma de silicona según una
cualquiera de las reivindicaciones 1 y 2, caracterizada
porque comprende nanopartículas de plata que tienen un tamaño de
partícula de 2 a 20 nm, preferiblemente de 5 a 10 nm.
4. La composición de goma de silicona según
cualquiera de las reivindicaciones precedentes, caracterizada
porque comprende en dicha matriz orgánica un fluido orgánico en el
que están dispersas dichas nanopartículas de plata.
5. La composición de goma de silicona según la
reivindicación 4, caracterizada porque dicho fluido orgánico
viscoso comprende un hidrocarburo alifático o aromático, un aceite
mineral, vaselina, glicerol, un alcohol graso, polipropilenglicol,
un aceite te o grasa animal y/o vegetal o un aceite de silicona.
6. Un procedimiento para fabricar una
composición de goma de silicona curable antimicrobiana que comprende
las etapas de:
- -
- proporcionar una composición de goma de silicona curable
- -
- proporcionar una matriz líquida orgánica que comprende plata metálica que tiene un tamaño de partícula en el intervalo de 1 a 50 nm
- -
- mezclar dicha matriz líquida orgánica en dicha composición de goma de silicona en una cantidad que proporciona una concentración de plata antimicrobiana pero menos que citotóxica de desde más de 1 nmol/l a menos de 1 \muml/l sobre al menos una porción de la superficie de dicha composición.
- -
- curar opcionalmente la mezcla de dicha composición con dicha matriz orgánica.
