Abstract
Procedimiento para la preparación de plantas transgénicas resistentes al ataque de fitopatógenos como Spodoptera littoralis y/o Meloidogyne incognita por interferencia del ARN que comprende las etapas siguientes: a) aislar la secuencia de nucleótidos de ADNc que codifica un receptor acoplado a la proteína G (RAPG), o una porción del mismo, cuya función es vital para el fitopatógeno, en el que dicha secuencia de nucleótidos del ADNc codifica un receptor RAPG seleccionado de entre AlstCR de SEC ID nº 2, DHR de SEC ID nº 3 u Oct/TyrR de SEC ID nº 6 cuando dicho insecto es Spodoptera littoralis y en el que dicha secuencia de nucleótidos de ADNc codifica un receptor RAPG seleccionado de entre el receptor de tipo serotonina de SEC ID nº 9 o el receptor de neuropéptidos de SEC ID nº 11 cuando dicho fitopatógeno es Meloidogyne incognita; b) construir un vector de expresión que comprende la secuencia de nucleótidos de ADNc que codifica un receptor RAPG o una porción del mismo según se determina en la etapa a), flanqueado por dos sitios de recombinación específicos; c) reaccionar por recombinación el vector de expresión de la etapa b) con un vector binario que comprende un activador constitutivo o específico para el tejido, los mismos sitios de recombinación específicos y una secuencia intrónica para obtener un plásmido recombinante; d) transferir dicho plásmido recombinante en células competentes de Agrobacterium tumefaciens y transformar la planta; e) cultivar la planta y obtener la expresión constitutiva o específica para el tejido de ARNbc en dicha planta.
