Abstract

Un método para producir tejido cartilaginoso in vitro, comprendiendo el método: a) aislar una población de condrocitos de tejido cartilaginoso donante aislado; b) expandir la población de condrocitos cartilaginosos en un medio de expansión y en un sustrato en condiciones eficaces para impedir la adhesión al sustrato de una mayoría de los condrocitos, en el que las condiciones comprenden expandir la población de células usando condiciones que comprenden: (i) un sustrato que tiene un recubrimiento de ácido hialurónico; o (ii) un sustrato no modificado que usa un medio de expansión que comprende ácido hialurónico en solución; c) sembrar una pluralidad de condrocitos de la población expandida de condrocitos cartilaginosos en un medio de producción tisular que comprenda TGF-β, en un sustrato de policarbonato que tenga una pluralidad de poros a través del mismo, teniendo los poros un diámetro interno de al menos 1 micra a aproximadamente 12 micras; y d) cultivar los condrocitos sembrados en el medio de producción tisular en el sustrato de policarbonato poroso durante un periodo de tiempo suficiente para que los condrocitos produzcan matriz extracelular, produciendo de este modo in vivo un tejido cartilaginoso cultivado caracterizado por una organización de células al azar.
