Abstract

Un método para aislar una línea celular de una planta, en el que la línea celular aislada de la planta se caracteriza por que (i) procede de un cambium de la planta, (ii) es homogénea, (iii) no ha experimentado desdiferenciación para formar el callo, comprendiendo el método: (a) recoger un tejido que contiene el cambium de la planta; (b) cultivar el tejido que contiene el cambium, induciendo de esta forma una capa proliferada del cambium a partir del cambium; y (c) recoger las células que tienen continuidad meristemática del meristemo primario sin pasar por la desdiferenciación para obtener el callo aislando la capa de cambium.

y se extrajeron varias veces usando 4 ml de cloruro de metilo. La capa de disolvente orgánico separada se secó a vacío y el residuo se disolvió en 1 ml de metanol. El extracto disuelto se agitó igualmente un limpiador ultrasónico. Después, tras la centrifugación, se retiró el aglomerado (8.000 g X 5 min).

HPLC (cromatografía líquida de alto rendimiento, Shiseido, Japón) se usó para el análisis del contenido, y los
10 productos Sigma se usaron como sustancias patrón del paclitaxel. El envase Capcell (C18, MGII, 5 um, 3,0 mm x 250 mm, Shiseido, Japón) se mantuvo a 40 °C usando el horno, y agua y acetonitrilo (Burdick & Jackson, EE.UU.

También permitió una productividad estable debido a menos cambios en el crecimiento celular y en el patrón de crecimiento durante el cultivo a largo plazo, y fue posible el escalado debido a la menor agregación y múltiples vacuolas de las líneas celulares. Estas líneas celulares permitieron la recuperación tras la crioconservación sin ninguna variación genética.
