en cualquier cepa del virus de inmunodeficiencia (por ejemplo, VIH). Por lo tanto, las secuencias con cierta homología con las secuencias de consenso están también dentro del alcance de la invención. Dicha homología permite la sustitución de, por ejemplo, de hasta 3 aminoácidos en un epítopo 8-mer (62.5% de homología) o en un epítopo 9-mer, 10-mer, o 11-mer. Se prefiere que no más de 10 de dichas sustituciones sean identificables en una secuencia de la invención que corresponde a las secuencias completas de la SEQ ID 1-4 (66.6% de homología para un 30-mer). Dichas sustituciones son preferiblemente sustituciones conservadoras de acuerdo con los esquemas de sustitución conocidos.
Un aspecto adicional de la invención proporciona un polipéptido de múltiples epítopos, cuyo polipéptido de múltiples epítopos comprende dos o más unidades en donde cada unidad de las dos o más unidades consiste de la SEQ ID NO: 2 o una secuencia que tiene por lo menos 85% de homología con la SEQ ID NO: 2:

en donde, el polipéptido de múltiples epítopos inmunogénico es inmunogénico a una cepa de VIH en un vertebrado que expresa un alelo complejo de histocompatibilidad principal (MHC).
La numeración de secuencia mencionada en la presente invención se define de acuerdo con principios bien reconocidos. Por lo tanto, la numeración comienza en 1 desde el codón de iniciación de traducción reconocido (ATG). Esto corresponde a una Metionina (M), para el segmento del genoma del virus de VIH que codifica la proteína de interés. En otras palabras, comienza en 1 con respecto a la Metionina mostrada como el primer aminoácido en la secuencia de proteína de interés como se utiliza y define por las bases de datos en las que se han establecido las secuencias (es decir, GenBank, SwissProt, etc.).

La Figura 1 muestra la producción de IFN-γ mediante cultivos de esplenocitos primarios de ratones vacunados con VIHv y NRP estimulados con Con A (10 μg/ml), Lisozima soluble (5 μg/ml), polipéptidos solubles purificados (P1, P2, P3 y P4; 5 μg/ml) y células humanas HLA emparejada T1 (T1) y JURKAT emparejada erróneamente (Ju) transfectadas con ya sea Lisozima, P1, P2, P3 o P4 de acuerdo con el protocolo descrito en el texto (la relación de esplenocitos con células transfectadas es 10: 1); la producción de IFN-γ se representa como el diferencial entre el nivel de producción en respuesta al antígeno considerado menos los IFN-γ producidos en respuesta a ya sea Lisozima soluble o la célula correspondiente transfectada con Lisozima. La Figura 6 muestra la producción de IFN-γ mediante cultivos de esplenocitos primarios de ratones vacunados con VIHv y NRP-v estimulados con cualquiera de Ionomicina (0.78 μg/ml) y PMA (0.05 μg/ml), Con A (5 μg/ml) o células humanas CEM HLA emparejadas infectadas con aislados de VIH-1 de Clado D (UG21-R5) o Clado A (UG-29-X4) de acuerdo con el protocolo descrito en los ejemplos (la relación de esplenocitos con células infectadas es 10:1). La producción de IFN-γ se representa como el número de células que producen IFN-γ en el grupo de VIH-v menos el número de células que producen IFN-γ en el grupo de NRP-v. El número de de valores de células que producen IFN-γ para los esplenocitos VIH-v y NRP-v estimulados con Con A o Ionomicina y PMA está sobre 4900 y sobre 8000 respectivamente. La significación estadística se estableció mediante análisis no paramétrico de Mann-Whitney de los valores de IFN-γ establecidos para los grupos y VIH-v y NRP-v.

La homología mencionada anteriormente con respecto a estas secuencias es preferiblemente 75%, 85%, 95% o sustancialmente 100%.
En la presente invención, la cepa del virus de inmunodeficiencia no está especialmente limitada, y los polipéptidos pueden ser inmunogénicos contra, y/o derivados de cualquiera de las cepas del virus de VIH conocidas. Las cepas futuras que han mutado a partir de cualquiera de estas cepas existentes también pueden ser aquellas contra las que los polipéptidos son inmunogénicos, o de las que los polipéptidos se derivan.
Las secuencias que definen los polipéptidos de la presente invención se situán dentro de las proteínas VPR, VIF, REV y NEF de cualquier cepa del virus VIH (las secuencias consenso de las que todas las secuencias se analizan, o alternativamente las posiciones de las cuales dentro de la proteína, se describieron anteriormente). Las siguientes secuencias específicas se analizaron por los inventores, y preferiblemente las secuencias de VIH a las que se hace referencia en la invención se seleccionan de estas secuencias específicas, o mutaciones de estas secuencias. Por lo tanto, las secuencias específicas homólogas a la SEQ ID 1 a 4 descritas anteriormente son preferiblemente las que están en las posiciones apropiadas dentro de las siguientes proteínas. Del mismo modo, las secuencias de la presente invención definidas por las posiciones de los residuos dentro de las proteínas de cualquier cepa de virus, es decir, residuos 51 a 80 de la proteína VPR, residuos 142-181 de la proteína VIF, residuos 69-95 de la proteína REV, y residuos 81-123 de la proteína NEF. En el día 14 después de inmunización, todos los animales recibieron una inmunización de refuerzo utilizando las mismas dosis y ruta de suministro como se utilizó originalmente. Finalmente, en el día 21 o 22, todos los animales fueron sacrificados y se recolectaron sus bazos.
El protocolo de inmunización se puede representar esquemáticamente como sigue:
Esquema 1 – inoculación
Ratón inmunizado con VIH-v
Esplenocitos recolectados después de 21 o 22 días -cualquier reacción inmunitaria provocará amplificación en las céluas T de ratón reactivas a epítopos presentados correctamente dentro de P1 a P4, que se activarán y/o proliferarán. Si los péptidos son presentados correctamente por ambos MHC de ratón y HLA humano, se pueden presentar (las células T reactivas a epítopos presentados en cada uno de estos contextos (las células que presentan antígenos, APC, en el ratón transgénico expresarán ambos MHC de ratón y HLA humano). Los esplenocitos recolectados pueden comprender cantidades significativas de células T activadas/proliferadas, junto con pequeñas cantidades de otras células, tales como células que presentan antígenos (APC)

ELISA de Citoquinas
Se combinaron los bazos de ratón que pertenecen al mismo grupo experimental, se presionaron suavemente a través de coladores celulares y las células rojas de la sangre se eliminan por tratamiento con regulador de lisis de glóbulos rojos (nueve partes NH4Cl 0.16 M y una parte de Tris 0.17 M, pH 7,2). Las suspensiones de esplenocitos de cada grupo experimental se sembraron en placas de 24 pozos a una densidad de 4x106 células/pozo que contenía una serie de antígenos de polipéptido (5 μg/ml) o, alternativamente, estirpes celulares tratadas con MMC (relación de esplenocitos a células (S:C) 10:1) transfectadas con los antígenos de polipéptido como se describió anteriormente.
Después de 4 días de incubación a 37°C, se recogió el sobrenadante y se analizó para IFN-γ e IL-4 por un ELISA de citoquina intercalado de acuerdo con el protocolo del fabricante (Pharmingen). Los límites de detección más bajos para el ensayo fueron 9.77 pg/ml para IL-4 y 39.06 pg/ml para IFN-γ.
Después de 4 días de incubación a 37°C, se recogió el sobrenadante y se analizó para IFN-γ e IL-4 mediante ELISA de citoquina intercalado de acuerdo con el protocolo del fabricante (Pharmingen). Los límites de detección más bajos para el ensayo fueron 9.77 pg/ml para IL-4 y 39.06 pg/ml para IFN-γ.
Resultados
Reactividad del péptido 1
Cada uno de los polipéptidos descritos en esta solicitud de patente (que incluyen P1, P2, P3 y P4 probados en este ejemplo) contiene varios epítopos de células T CD8+ de los cuales varios son específicos para HLA-A*0201. Luego el procesamiento interno del polipéptido por las células que presentan antígeno (APC) de un receptor que lleva HLAA*0201, estos epítopos específicos de células T CD8+ se presentan en la superficie de la APC, donde proceden a activar células T CD8+ no tratadas anteriormente e inducir una respuesta inmunitaria similar a Th1 específica para el péptido que se prueba, en este caso, P1, P2, P3 y P4.
Para confirmar esto, las estirpes celulares humanas que llevan HLA-A*0201 (T1) y no llevan (Jurkat) se cargaron intracelularmente con P1 por medio de un vehículo de lípido (Lipofectina, Invitrogen). Se encontró que los esplenocitos

Nota: “Lys” significa el fondo de control negativo sobre el que se calculan todos los valores. “Sol” significa péptido soluble presentado a la población de esplenocitos primaria. “Pro” significa que el péptido se presenta en complejo con las moléculas de células HLA luego del procesamiento interno y la carga de los epítopos resultantes sobre las moléculas de MHC. Experimento 2 -Inmunización de VIH-v induce una respuesta específica de antígeno contra las células humanas infectadas con aislados de campo de VIH
El propósito de este estudio es evaluar si se induce la inmunización con el VIH identificado con péptidos de poliepítopos de célula T conservados de VIH (VIH-v) y la respuesta específica de antígeno frente a células humanas infectadas con aislados de campo de VIH.
Material y métodos,
Péptidos, virus y estirpe celular
La vacuna candidato (VIH-v) utilizada en este estudio se compone de varios polipéptidos (es decir, P1: VPR aa 51 a 80 (SEQ ID 1); P2: VIF: aa 142 a 181 (SEQ ID 2); P3 : REV aa 69 a 95 (SEQ ID 3); P4: NEF: aa 81 a 123 (SEQ ID 4)), que todo se sintetizaron mediante química de Fmoc y se resuspendieron en DMSO en PBS (la concentración de DMSO en la preparación final fue menor del 10%). La lisozima (Sigma) desnaturalizada por ebullición se utilizó como la preparación no relevante de control (NRP-v).
Las cepas infecciosas del VIH-1 se obtuvieron de NIBSC y los aislados de Uganda representados de Clado D (UG21-R5) y Clado A (UG-29-X4) de VIH-1. La CEM es una estirpe de células T CD4+ linfoblastoides susceptibles a infección por los aislados de VIH mencionados anteriormente. Esta estirpe celular se mantuvo en medio RPMI-1640 (Sigma) suplementado con 50 IU/50 mg/ml de penicilina/estreptomicina (Sigma) y, como medio completo, FCS al 10%. Los cultivos de células se mantuvieron a 37°C en una atmósfera humidificada de CO2 al 5%.
Los esplenocitos primarios se mantuvieron en medio IMDM (Invitrogen) suplementado con β-mercaptoetanol 0.02 mM (Sigma), 50 IU/50 mg/ml de penicilina/estreptomicina (Sigma) y FCS al 10% (Sigma) a 37°C en una atmósfera humidificada de CO2 al 5%.
Inmunizaciones
En el día 1, ratones C57BL/6-Tg(HLA-A2.1)1Enge/J de siete a diez semanas de edad (HLA-A*0201 transgénico en un fondo C57/BL6, Jackson Labs) se inmunizaron por vía subcutánea en la base de la cola con una dosis de 200 μl de la preparación de antígeno emulsionada en IFA (Sigma). En el grupo de prueba (n = 4), cada dosis de la preparación de antígeno contenía 40 nmol de una mezcla equimolar de los cuatro péptidos (10 nmol de cada uno), mientras que en el grupo de control (n = 4), cada dosis de la preparación de antígeno contenía 40 nmol del polipéptido no relevante.
En el día 15 todos los animales recibieron una inmunización de refuerzo utilizando las mismas dosis y vía de administración como se utilizó originalmente.
En el día 21 todos los animales se eligieron y se recogieron sus bazos.
Tinción de citoquinas intracelular
Se combinaron bazos de ratón que pertenecen al mismo grupo experimental, se presionan suavemente a través de coladores celulares y se eliminan las células rojas de sangre por tratamiento con regulador de lisis de glóbulos rojos (nueve partes de NH4Cl 0.16 M y una parte de Tris 0.17 M, pH 7.2). Las suspensiones de esplenocitos de cada grupo experimental se sembraron en placas de 96 pozos (Falcon, BD discovery labware) a una densidad de 2x105 células/pozo que contienen ya sea Ionomicina (0.78 μg/ml) y PMA (0.05 μg/ml), Concanavalina A en 5 μg/ml o 2x104 células CEM irradiadas con UV infectadas por VIH y no infectadas (relación de esplenocitos a célula (S:C) relación de 10:1).
Después de 3 horas de incubación a 37°C con CO2 al 5%, se agregó Brefeldina A a todos los pozos a 10 μg/ml de concentración final. Las placas luego se incubaron durante 3 horas adicionales en placas a 37°C y luego se transfirieron las placas a un refrigerador a + 4°C para almacenamiento durante la noche. Después de centrifugación (200 g, 5 min), el sobrenadante se eliminó de los pozos y las células se contratiñeron con anticuerpo anti-CD3-FITC durante 15-20 minutos a temperatura ambiente en la oscuridad. Las células luego se lavaron con PBS mediante centrifugación (200 g, 5 min), se fijaron durante 20 min a temperatura ambiente (reactivo de fijación A, Caltag) y se lavaron de nuevo. Después de permeabilizar las células durante 20 min (reactivo de permeabilización B, Caltag) a temperatura ambiente, las células se lavaron y se agregaron los anticuerpos anti-IFN-γ-PE y anti-IL4-APC. Después de 30 min de incubación, las células se lavaron dos veces y se resuspendieron en 40% (p/v) de formaldehído en PBS antes de ser analizadas en un citómetro de flujo.
