Abstract

1. procedimiento para la obtención de antigenos superficiales, o asociados a membranas, a partir de células enteras o medios de cultivo en donde se desarrollan, caracterizado porque comprende: a) destruir y fijar dichas células; (b) separar dichas células del medio; (c) extraer antigeno de dichas células o medio de cultivo con una solución alcalina acuosa de ph regulado de sal de metal alcalino que tiene un ph de aproximadamente 6,0 a aproximadamente 11,0 , en donde la concentración de dicho regulador de ph y sal es de por lo menos 0,01 molas; d) concentrar dicho antigeno; (e) precipitar con ácido dichos antigeno concentrado con un ácido acuosos o una solución de regulador de ácido que tiene un ph de aproximadamente 1,0 a aproximadamente 6,0 en donde la concentración de dicho ácido o regulador es de por lo menos 0,01 molar; f) solubilizar dicho antigeno precipitado con ácido con una solución alcalina acuosa de ph regulado uqe tiene un ph de aproximadamente 6,0 a aproximadamente 11,0 en donde la concentración de dicho regulador es de por lo menos 0,01 molar y , si se desea, digerir el antigeno solubilizado con una enzima proteolitica durante aproximadamente 1/4 hr a aproximadamente 4hr entre aproximadamente 0ºC y aproximadamente 45ºC; y detener la digestión con enzima del antigeno.
