Abstract
Un método para obtener una cepa de microorganismos modificados para contener un fragmento de una secuencia específica de desoxiribonucleótidos, cuyo método comprende: (I) purificar un fragmento de una secuencia de desoxiribonucleótidos mediante las siguientes etapas: (a) proveer una población de transcritos de ADNc de los polirribonucleótidos. (b) Someter los transcritos de ADNc a la acción de una preparación de endonucleasa de restricción capaz de catalizar la hidrólisis de los transcritos de ADNc en cada uno de los puntos de restricción. (c) fraccionar los fragmentos producidos por la acción de la endonucleasa de restricción de acuerdo con su longitud. (d) tratar previamente el ADN para eliminar cualquier grupo terminal 5''-fosfato. (e) incubar el ADN con una endonucelasa de restricción capaz de actuar sobre la secuencia específica deseada de nucleótidos para producir dos sub-fragmentos lineales del mismo. (f) fraccionar los sub-fragmentos de acuerdo con su longitud. (g) aislar los dos sub-fragmentos, (h) reunir los sub-fragmentos covalentemente, en una reacción catalizada por ADN ligasa, (i) fraccionar las moléculas de ADN reunidas de acuerdo con su longitud, (II) recombinar el producto procedente de la secuencia anterior con un vector de transferencia de ADN y transferirlo a un microorganismo.
