Claims (21)

  1. Conjugués polysaccharide-protéine stables, couplés de manière covalente comprenant des polysaccharides bactériens neutres et des protéines immunogéniques couplés par l'intermédiaire d'agents d'intercalation bigénériques, contenant des liaisons thioéther, dans lesquels lesdits agents d'intercalation bigénériques peuvent être représentés par la formule A-E-S-B, dans laquelle E est où R est H ou CH₃ ; A est où m vaut de 0 à 4, n vaut de 0 à 3, W est O ou NH et Y est CH₂, O, S, NR' ou CHCO₂H, où R' est H ou un groupe alkyle en C₁ ou C₂, de telle sorte que si Y est CH₂ ou NH, alors m et n ne sont pas tous les deux égaux à zéro, et si Y est O ou S, alors m est supérieur à 1 et n est supérieur à 1 ; et B est où p vaut 1 - 3, q vaut 0 - 2, Z est NH₂, CO₂H ou H, et D est où R' est tel que défini ci-dessus.
  2. Conjugués polysaccharide-protéine stables, couplés de manière covalente, selon la revendication 1, dans lesquels les agents d'intercalation bigénériques peuvent être représentés par la formule
  3. Conjugués polysaccharide-protéine selon la revendication 1, dans lesquels le polysaccharide capsulaire bactérien neutre est choisi dans le groupe comprenant les polysaccharides de Streptococcus pneumoniae de types 7F, 14 et 37.
  4. Conjugués polysaccharide-protéine selon la revendication 1, dans lesquels la protéine immunogénique est une protéine de membrane externe du sérotype méningococcique B ou une protéine édestine.
  5. Conjugués polysaccharide-protéine selon la revendication 1, dans lesquels le polysaccharide capsulaire bactérien neutre est un polysaccharide de Streptococcus pneumoniae de type 14, la protéine immunogénique est une protéine de membrane externe du sérotype ménincoccique B et l'agent d'intercalation bigénérique peut être représenté par la formule,
  6. Procédé pour solubiliser des polysaccharides bactériens, neutres, non polyanioniques, qui comprend le chauffage desdits polysaccharides dans de l'eau distillée, l'élimination de l'eau résiduelle et la dissolution du polysaccharide dans un solvant aprotique, polaire, non aqueux.
  7. Procédé selon la revendication 6, dans lequel on ajoute de l'hydrazine aqueuse à 5 - 15 % à l'eau distillée.
  8. Procédé selon la revendication 6, dans lequel on chauffe les polysaccharides pendant de 30 secondes à 10 minutes à 70° - 100°C, on élimine l'eau par lyophilisation et séchage avec P₂O₅ sous vide et dans lequel le solvant aprotique, polaire, non aqueux est le diméthylformamide, le diméthylsulfoxyde, le diméthylacétamide, le formamide ou la N,N'-diméthylimidazolidinone.
  9. Procédé pour modifier de manière covalente un polysaccharide bactérien neutre qui comprend
    (a) la fragmentation du polysaccharide et la solubilisation du polysaccharide fragmenté dans un solvant aprotique, polaire, non aqueux ;
    (b) l'activation du polysaccharide avec un réactif bifonctionnel ; puis
    (c) la réaction de ce polysaccharide activé avec un réactif bis-nucléophile.
  10. Procédé selon la revendication 9, qui comprend également la réaction du polysaccharide activé que l'on a fait réagir avec un réactif bis-nucléophile, avec un réactif produisant des sites électrophiles accrochés.
  11. Procédé selon la revendication 9, dans lequel le polysaccharide est fragmenté par chauffage dudit polysaccharide dans de l'eau distillée puis élimination de l'eau par lyophilisation et séchage avec P₂O₅ sous vide ; le solvant aprotique, polaire, non aqueux est le diméthylformamide, le diméthylsulfoxyde, le diméthylacétamide, le formamide ou la N,N'-diméthylimidazolidinone ; le réactif bifonctionnel est choisi dans le groupe comprenant les dérivés de l'acide carbonique, où R² et R³ sont séparément un groupe azolyle ; des halogénures ; ou des esters phényliques ; et le réactif bis-nucléophile est une diamine de formule, H₂N(CH₂)mY(CH₂)nNH₂, dans laquelle m vaut de 0 à 4, n vaut de 0 à 3, et Y est CH₂, O, S, NR', CHCO₂H, ou R' est H ou un groupe alkyle en C₁ ou C₂, de telle sorte que si Y est CH₂ ou NH, alors m et n ne peuvent pas tous les deux être égaux à 0, et si Y est O ou S, alors m est supérieur à 1 et n est supérieur à 1.
  12. Procédé selon la revendication 11, dans lequel le solvant aprotique, polaire, non aqueux est le diméthylformamide, le réactif bifonctionnel est le carbonyldiimidazole et le réactif bis-nucléophile est la 1,4-butanediamine.
  13. Procédé selon la revendication 10, dans lequel le réactif engendrant des sites électrophiles est où X' est un groupe nitrophénoxy, dinitrophénoxy, pentachlorophénoxy, pentafluorophénoxy, halogénure, O-(N-hydroxysuccinimidyle) ou azido, R est H ou CH₃ et X est Cl, Br ou I ; ou un acide maléimido activé, dans lequel p vaut de 1 à 3 et X' est tel que défini ci-dessus.
  14. Procédé selon la revendication 13, dans lequel le réactif engendrant des sites électrophiles et le bromoacétate de p-nitrophényle.
  15. Procédé de préparation de conjugués polysaccharide-protéine comprenant des polysaccharides capsulaires bactériens neutres et des protéines immunogènes couplés par l'intermédiaire d'agents d'intercalation bigénériques contenant des liaisons thioéther, qui comprend
    (a) la fragmentation du polysaccharide et la solubilisation du polysaccharide dépolymérisé dans un solvant aprotique, polaire, non aqueux ;
    (b) l'activation du polysaccharide avec un réactif bifonctionnel ;
    (c) la réaction de ce polysaccharide activé avec un réactif bis-nucléophile ;
    (d) la réaction de ce polysaccharide activé que l'on a fait réagir avec un réactif bis-nucléophile, avec un réactif engendrant des sites électrophiles, pour former ainsi un polysaccharide avec des sites électrophiles accrochés ;
    (e) la réaction, de manière indépendante, de la protéine immunogénique avec un réactif engendrant des groupes thiol, pour former une protéine avec des groupes thiol accrochés ; puis
    (f) la réaction du polysaccharide avec des sites électrophiles accrochés avec la protéine avec des groupes thiol accrochés pour former un conjugué polysaccharide-protéine qui est couplé par l'intermédiaire d'une liaison thioéther covalente ; puis
    (g) la centrifugation du mélange résultant pour éliminer les polysaccharides et les protéines non liés de manière covalente.
  16. Composition comprenant une quantité immunologiquement efficace, pour la protection soit active soit passive des espèces mammifères vis-à-vis des bactériémies provoquées par l'organisme apparenté, des conjugués polysaccharide-protéine stables, couplés de manière covalente, selon la revendication 1, des antisérums provenant desdits conjugués, ou une gamma-globuline ou d'autres fractions contenant l'anticorps desdits antisérums, et un véhicule pharmaceutiquement acceptable.
  17. Composition selon la revendication 16, qui comprend en outre un adjuvant.
  18. Composition selon la revendication 16, dans laquelle les conjugués polysaccharide-protéine comprennent un polysaccharide pneumococcique de type 14 couplé par l'intermédiaire d'un agent d'intercalation bigénérique, de formule, à une protéine de membrane externe de sérotype méningococcique B, une quantité immunologiquement efficace est une quantité de chacun des conjugués dans la composition telle que chaque conjugué contienne de 2 à 50 µg du polysaccharide sous la forme conjuguée, et l'espèce mammifère est l'homme.
  19. Composition pour son utilisation dans un procédé de traitement thérapeutique pour les espèces mammifères vis-à-vis des bactériémies des organismes apparentés, ladite composition comprenant un ou plusieurs types de conjugués polysaccharide-protéine comprenant des polysaccharides capsulaires bactériens neutres couplés, par l'intermédiaire d'agents d'intercalation bigénériques contenant des liaisons thioéther, à des protéines immunogéniques, et un représentant du groupe comprenant un véhicule pharmaceutiquement acceptable, un adjuvant et un adjuvant et véhicule pharmaceutiquement acceptable.
  20. Composition selon la revendication 19, dans laquelle lesdits conjugués polysaccharide-protéine comprennent un polysaccharide pneumococcique de type 14 couplé, par l'intermédiaire d'un agent d'intercalation bigénérique, de formule, à une protéine de membrane externe de sérotype méningococcique B, l'espèce à traiter est un nouveau-né humain, et la quantité efficace de la composition dans une dose unique est une quantité correspondant à 25 µg du polysaccharide dans la forme conjuguée pour les conjugués des polysaccharides pneumococciques.
  21. Composition selon la revendication 19, dans laquelle une ou deux compositions supplémentaires de rappel d'une quantité d'un conjugué polysaccharide-protéine comprenant un conjugué polysaccharide-protéine comprenant des polysaccharides capsulaires bactériens neutres couplés par l'intermédiaire d'agents d'intercalation bigénériques à des protéines immunogéniques, correspondant à 25 µg de polysaccharide sous la forme conjuguée, peuvent être administrées à des nouveau-nés humains.
