【発明の詳細な説明】
細胞におけるCD95リガンドの発現の検出方法
本発明は、細胞におけるCD95リガンドの発現の検出方法に関する。
アポトーシスとは、プログラムされた細胞死に対する名称である。それは、例
えば、器官形成および変態、組織の萎縮および腫瘍の退行において見出される。
アポトーシスは、細胞質の濃縮、細胞膜の絨毛の損失、核の分割および染色体D
NAの広範な分解と関連している。アポトーシスの妨害は、AIDS、自己免疫
疾患、腫瘍および白血病等の種々の疾患で発生することがわかった。AIDSの
場合、アポトーシスの増加がCD4−T細胞の強力な減少の原因であるように思
われる。
活性化されたT細胞および他の細胞において、CD95およびAPO−1とそ
れぞれ称される細胞表面蛋白質が見出されている。CD95リガンドおよびAP
O−1リガンドとそれぞれ称される可溶性または膜結合蛋白質は、それぞれ、こ
の細胞表面蛋白質に結合して、アポトーシスの誘導を引き起こすことができる。
アポトーシスの程度および経路ならびにアポトーシスの原因となる分子は、こ
れまで、決定され得ていない。しかしながら、これは、前記疾患を患っている患
者における追跡管理や好適な治療措置等を行なうことを可能にするために必要で
ある。
したがって、本発明の目的は、アポトーシスの程度およびその経路それぞれの
決定に役立つ産物を提供することにある。
本発明によれば、これは、請求の範囲に定義された主題により達成される。
したがって、本発明の主題は、以下CD95−Lと称するCD95リガンドが
細胞において定量的に決定されうる方法に関する。
本発明は、アポトーシスは、T細胞において量の増大したCD95−Lに起因
するという出願人の発見に基づく。さらに、出願人は、アポトーシスの割合は、
CD95−Lの量により決定されるということを発見した。
本発明の方法は、細胞におけるCD95−Lの量が確実に決定されうる定量的
「ポリメラーゼ連鎖反応」(PCR)に関する。細胞中に存在するCD95−L
mRNAの量は、RT−PCR反応により本発明の方法で決定される。CD9
5−Lに対する定量的PCRにおいて、CD95−L cDNAの量を、CD9
5−L特異的DNA競合断片をPCR反応に添加することにより滴定する。一定
量の全RNAのcDNAへの完全な逆転写は、この工程より先行する。該競合断
片は、ヌクレオチド配列の中央での追加の塩基対によりCD95−Lの「野生型
」cDNAとは異なるが、同一の隣接した配列を有する。したがって、野生型と
競合断片とは、同じプライマーの組合せにより認識され、PCRにおいて共同し
て増幅されうる。同一のプライマー結合部位と類似のヌクレオチド配列により、
両断片は、同じ増幅速度に供される。しかしながら、サイズにおける微小な相違
は、PCRによるゲル電気泳動により両DNA種の分離を可能にする。アガロー
スゲル上でのDNAバンドの強度は、PCRにおいて使用された最初のcDNA
の量を反映する。ゲル上でのCD95−Lおよび競合DNAバンドの結果が同じ
ならば、競合物とCD95−Lとが同じ初期濃度で存在していたと結論すること
ができる。この方法により滴定されたCD95−L cDNAの量は、逆転写が
完全に先行するので、全RNA中のCD95−L mRNAの量を反映する。定
量的PCR反応の図を、図1に示す。
定量的PCRは、以下の反応原理に基づく。CD95−Lの発現割合を定量化
するために、逆転写された全RNAの一定量のcDNAをPCR反応容器中にピ
ペットで入れる。例えば、5〜10000fgの種々の希釈の競合断片を、陰性
対照(−)を除いて各試料に添加する。PCR反応後、増幅したDNA断片を臭
化エチジウムで処理して、アガロースゲル上で分離する。U.V.光で該ゲルを
照射することにより、DNAを可視化する。CD95−L DNAのバンドと競
合DNAのバンドの両方が等しい強度を有するという観点から、滴定点(図1で
矢印により記す)を決定する。両方のバンドの等しい比率は、等しい量の決定対
象のDNAと競合DNAが増幅されたことを示す。同様に、滴定点により決定さ
れたCD95−L cDNAの量(mRNAの量に匹敵する)は、用いた全RN
Aの量中に存在していた。
PCR用に合成されたプライマーは、刊行されたCD95−Lの配列に基づく
ことが好ましい(Takahashi T ら、Int.Immunol.6,(1984),1547-1567を参照
)。PCRは、当業者が精通している条件下で行なわれる。
例えば、以下のプライマー:

競合断片としてPCRに用いる変異したCD95−L断片は、突然変異誘発、
好ましくはPCR突然変異誘発により調製される。T細胞株CEM−S等に由来
するCD95−L cDNAは、PCR突然変異誘発のためのマトリックスとし
て役立つ。Lup/LdownPCR断片の2つの部分を、PCRにおいてプライマー
対Lup/LupOV(230bpの長さを有する断片を生ずる)、および第2のPC
Rにおいてプライマー対Ldown/downOV(349bPの長さを有する断片を生ず
る)を使用することにより増幅する。プライマーLupOVは、プライマーLdownOV
の30bp長の突出配列に相補的な50bp長の突出配列を含む。精製後、2つ
の断片をそれらの相補的領域を介してハイブリダイズさせ、プライマーLup/Ldown
を用いる別のPCR増幅のためのマトリックスとして使用する。その結果は
、ヌクレオチド配列の中央に50個の追加の塩基を含むCD95−Lの変異した
断片である。競合断片の調製のための図を、図2に示す。
変異した断片と野生型断片との間の競合に基づく本発明の方法は、少量の試料
(1〜5x106細胞)におけるCD95−Lの細胞内レベルの正確な決定を可
能にする。
CD95/CD95−Lにより伝達されたアポトーシスは、AIDS疾患の場
合、T細胞の枯渇に関係があることは明白であるので、CD95−Lの発現の定
量的決定は、AIDS患者の追跡管理のためや治療上の介在の指標として使用さ
れうる。さらに、本発明の方法は、T細胞におけるウイルス誘導性、CD95伝
達性アポトーシスを阻害することが可能な潜在的薬物の「薬物スクリーニング」
に関して、イン・ビトロで適する。同様に、細胞におけるCD95リガンドの定
量的決定は、減少したアポトーシス(白血病等)および増加したCD95伝達性
アポトーシスに伴う疾患の場合、疾患の定義、追跡管理および「薬物のスクリー
ニング」におけるリガンド発現の薬理学的影響に関して使用することができる。
応用の直接の分野としては、現在、自己免疫疾患および移植拒絶の免疫抑制治療
のために使用されているアゾチオプリン、シクロスポリンA、シクロホスファミ
ド、コルチゾンおよびメトトレキセート等の薬物の効果の分析等が代表例である

図3は、リンパ系細胞におけるCD95−L mRNAの構成的、誘導性発現
を示す。
以下の実施例により、本発明を説明する。実施例:リンパ系細胞におけるCD95−Lの決定
リンパ系T細胞J16とCEM−Sをそれぞれ、通常の培地で培養し、PMA
(5ng/ml)およびイオノマイシン(2μg/ml)それぞれで、それぞれ
、4時間および24時間処理した。チョムジンスキー(Chomczynski,P.)とサッ
チ(Sacchi,N.)、Anal.Biochem.,162,(1987),156-159による方法に従って、
該細胞から全RNAを得て、AMV逆転写酵素(Amersham,Braunschweig)およ
びオリゴdT16プライマー(Promega,Heidelberg)により、cDNAに転写し
た。20μlの反応液は、1x AMV緩衝液(50mM Tris−HCl、
pH8.3;8mM MgCl2;50mM NaCl;1mM DTT)、2
50μMの各dNTP(Pharmacia,Freiburg)、0.5μM オリゴ−dT16;
0.6U/μl RNアーゼ阻害剤(Amersham,Braunschweig);0.75U/
μl AMV逆転写酵素;50ng/μl 全RNA(添加前に65℃で5分間
変性)からなるものであった。逆転写は、42℃で45分間行ない、次いで、9
4℃まで5分間加熱することにより、反応を停止した。すべての試薬が過剰に存
在していたので、用いたRNAは、完全にcDNAに転写されたと仮定すること
ができる。
CD95−Lの発現割合を定量するためには、逆転写された全RNAの250
ngのcDNAをそれぞれ、PCR反応容器にピペットで入れた。前記したよう
な競合断片を、10〜10000fgの種々の希釈で、陰性対照(−)を除いて
各試料に添加した。反応混合物は:cDNA(=250ngの逆転写された全R
NA)、1xPCR緩衝液(10mM Tris−HCl、pH8.8;50m
M KCl、0.08% NP40)、1mM MgCl2、200μM dN
TP、0.5μM プライマーおよび1.12U/容器 Taqポリメラーゼ(
MBI/Fermentas)を含む50μlであった。増幅は、前記したように、Lupお
よびLdownプライマーを用いて、ロボサイクラー(Stratagene)またはBiozymサ
ーモサイクラーで行なった。反応条件は、以下のようであった:94℃で35秒
間、56℃で90秒間および72℃で120秒間を36サイクル繰り返す。20
μlのPCR反応液を、2%アガロースゲル上で電気泳動により分離し、臭化エ
チジウム染色後、U.V.光で可視化した。CD95−L DNAのバンドと競
合DNAのバンドの両方が等しい強度を有する点を、ゲル上で決定した。該ゲル
を図3に示す。同量の野生型断片と競合断片を示す等しいバンドの強度を、矢印
で示す。
未処理のJ16細胞は、10fgのCD95−L mRNA/250ngの全
RNAを発現し、未処理のCEM−S細胞は、50fgのCD95−L mRN
A/250ngの全RNAを発現したことが示された。PMA/イオノマイシン
処理は、J16細胞の場合、250ngの全RNA当たり1000fgまでのC
D95−L mRNAの発現を増加させ、CEM−S細胞の場合、250ngの
全RNA当たり5000fgまでの発現を増加させた(表1を参照)。
前記データは、CD95−Lが本発明の方法により定量的に決定されうること
を示す。したがって、特に、AIDS等の疾患の場合に、アポトーシスの程度お
よび/または経路を決定することができる。

を有する)
を含むCD95リガンドの定量的決定方法。
2.該細胞が確立された細胞株または患者に由来するものであることを特徴とす
る、請求項1記載の方法。
3.該細胞がリンパ系T細胞であることを特徴とする、請求項2記載の方法。
4.アポトーシスの程度および/または経路を決定するための、請求項1記載の
方法の使用。
5.該決定がAIDS患者の細胞でなされるものである、請求項4記載の方法の
使用。
Claims (1)
- 【特許請求の範囲】 1.下記工程: (a)細胞から全RNAを単離する工程、 (b)(a)由来の全RNAを逆転写によりcDNAに転写する工程、ならびに (c)PCR反応において、CD95リガンド特異的プライマーにより(b)由 来のcDNAおよびCD95リガンド競合断片を増幅する工程 を含むCD95リガンドの定量的決定方法。 2.該CD95リガンド特異的プライマーが、下記配列: Lup 5’−ATGTTTCAGCTCTTCCACCTACAGA−3’ Ldown 5’−CCAGAGAGAGCTCAGATACGTTGAC−3’ を有することを特徴とする、請求項1記載の方法。 3.該細胞が確立された細胞株または患者に由来するものであることを特徴とす る、請求項1または2記載の方法。 4.該細胞がリンパ系T細胞であることを特徴とする、請求項3記載の方法。 5.アポトーシスの程度および/または経路を決定するための、請求項1記載の 方法の使用。 6.該決定がAIDS患者の細胞でなされるものである、請求項5記載の方法の 使用。
