- 【特許請求の範囲】 1.複製欠損性レトロウイルスベクターを含有する複製欠損性レトロウイルス ベクター粒子を産生しうるプロデューサー細胞へと患者の細胞を変換するための 該複製欠損性レトロウイルスベクターをコードするDNA配列であって、該レト ロウイルスベクターが少なくとも1つの異種遺伝子を含んでなり、該ベクターが 機能的なenvまたはgag-pol遺伝子のいずれをも含有しておらず、該DNA配列が 非レトロウイルス手段により患者に投与するのに適した形態であり該細胞に取込 まれうることを特徴とするDNA配列。 2.複製欠損性レトロウイルスベクター粒子を産生しうるプロデューサー細胞 へと患者の細胞を変換するための一組のDNA配列であって、該配列の組が、請 求項1に記載のDNA配列と、該プロデューサー細胞による感染性レトロウイル スベクター粒子の産生のためのパッケージング成分Env および Gag-Polをコ ードするDNA配列とを含んでなり、該DNA配列の組が、非レトロウイルス手 段により患者に投与するのに適した形態であり該細胞に取込まれうることを特徴 とする一組のDNA配列。 3.前記の少なくとも1つの異種遺伝子が、少なくとも1つの治療的に活性な 遺伝子を含む、請求項1または請求項2に記載のDNA配列。 4.変換される患者の細胞が、治療的に活性な遺伝子を受容させようとする標 的細胞型の細胞である、請求項1〜3のいずれか1項に記載のDNA配列。 5.1以上のプラスミド中に存在する、請求項1〜4のいずれか1項に記載の DNA配列。 6.感染性レトロウイルスベクター粒子中に複製欠損性レトロウイルスベクタ ーを産生しうるプロデューサー細胞であって、該プロデューサー細胞が、該複製 欠損性レトロウイルスベクターをコードするDNA配列を含んでなり、該ベクタ ーが、少なくとも1つの異種遺伝子を含んでなるが機能的なenvまたはgag-pol遺 伝子のいずれをも含有しておらず、該プロデューサー細胞が、患者内に導入し遺 伝子治療で使用するのに適した新鮮な細胞であることを特徴とするプロデューサ ー細胞。 7.前記べクター中の少なくとも1つの異種遺伝子が、少なくとも1つの治療 的に活性な遺伝子を含む、請求項6に記載のプロデューサー細胞。 8.前記細胞が、治療的に活性な遺伝子を受容させようとする標的細胞型の細 胞である、請求項7に記載のプロデューサー細胞。 9.前記細胞が、治療的に活性な遺伝子を受容させようとする標的細胞へ前記 べクターを送達する能力を有する免疫系細胞である、請求項7に記載のプロデュ ーサー細胞。 10.前記プロデューサー細胞が由来する患者内へ再移殖するための、請求項6 〜9のいずれか1項に記載のプロデューサー細胞。 11.複製欠損性レトロウイルスベクターを産生しうるプロデューサー細胞の製 造法であって、該べクターが、少なくとも1つの治療的に活性な遺伝子を含んで なるが機能的なenvまたはgag-pol遺伝子のいずれをも含有しておらず、該製造法 が、該複製欠損性レトロウイルスベクターをコードするDNA配列を新鮮な哺乳 動物細胞内に導入して、遺伝子治療で使用するのに適したプロデューサー細胞を 得ることを含むことを特徴とする製造法。 12.請求項11に記載のDNA配列とパッケージング成分EnvおよびGag-Pol をコードするDNA配列とを含む一組のDNA配列を、前記細胞内へ導入する、 請求項11に記載の製造法。 13.前記プロデューサー細胞が、治療的に活性な遺伝子を受容させようとする 標的細胞型の細胞である、請求項11または請求項12に記載の製造法。 14.前記プロデューサー細胞が、治療的に活性な遺伝子を受容させようとする 標的細胞へ前記べクターを送達する能力を有する免疫系細胞である、請求項11ま たは請求項12に記載の製造法。 15.前記の新鮮な細胞が患者の細胞であり、該細胞を患者内でin vivoでプロ デューサー細胞へ変換する、請求項11〜13のいずれか1項に記載の製造法。 16.前記のDNA配列または一組のDNA配列を、前記の新鮮な細胞内へinvi troで導入する、請求項11〜14のいずれか1項に記載の製造法。 17.前記の新鮮な細胞が患者由来であり、得られたプロデューサー細胞を次に 該患者内へ再移植する、請求項16に記載の製造法。 18.請求項6〜10のいずれか1項に記載のプロデューサー細胞を患者内へ導入 することを含んでなる、該患者において遺伝子治療を実施する方法。 19.請求項1〜5のいずれか1項に記載のDNA配列または一組の配列を非レ トロウイルス手段により患者内へ導入することを含んでなる、該患者において遺 伝子治療を実施する方法。 20.遺伝子治療に使用する製剤中での、請求項6〜10のいずれか1項に記載の プロデューサー細胞の使用。 21.遺伝子治療に使用する製剤中での、請求項1〜5のいずれか1項に記載の DNA配列または一組のDNA配列の使用。
