Claims (1)
- 【特許請求の範囲】 1.配列からなる組換え核酸分子、好ましくは組換えDNA分子であって、該配列 または遺伝子が誘導可能なリプレッサーおよび誘導可能なトランスアクチベータ ーの組合せの制御下である遺伝子の組換え核酸分子。 2.遺伝子産物がその産物を発現する細胞に対して有毒である請求の範囲第1項 記載の組換えDNA分子。 3.少なくとも1つのトランスアクチベーター配列がテトラサイクリンリプレッ サーの結合部位である請求の範囲第1項または第2項記載の組換えDNA分子。 4.結合部位がtn10由来である請求の範囲第3項記載の組換えDNA分子。 5.少なくとも1つの誘導可能なリプレッサー配列がYY1リプレッサーの結合部 位である請求の範囲第1項〜第4項記載のいずれかに記載の組換えDNA分子。 6.少なくとも1つの誘導可能なリプレッサーが後期主要転写因子(MTLF)の結合 部位である請求の範囲第1項〜第4項のいずれかに記載の組換えDNA分子。 7.少なくとも1つの誘導可能なリプレッサーがアデノ随伴ウイルスp5プロモー ター由来である請求の範囲第1項〜第4項のいずれかに記載の組換えDNA分子。 8.遺伝子がパルボウイルスタンパク質の少なくとも特定の一部分をコードする 前記請求の範囲のいずれかに記載の組換えDNA分子。 9.遺伝子がパルボウイルス非構造遺伝子である前記請求の範囲のいずれかに記 載の組換えDNA分子。 10.遺伝子がアデノ随伴ウイルスrep遺伝子である請求 の範囲第9項記載の組換えDNA分子。 11.前記請求の範囲のいずれかに記載の組換えDNA分子からなる細胞。 12.不死化されている請求の範囲第11項記載の細胞。 13.哺乳類起源である請求の範囲第11項または第12項記載の細胞。 14.組換えDNA分子が、発現を誘導することができ、かつ抑制を誘導することが できる遺伝子からなる請求の範囲第11項、第12項または第13項記載の細胞。 15.アデノ随伴ウイルスのRep78/Rep68、Rep52/Rep40および/またはVP1/VP2 /VP3の発現および抑制の誘導を可能とする請求の範囲第14項記載の細胞。 16.請求の範囲第11項〜第15項のいずれかに記載の細胞および/または請求の 範囲第1項〜第10項のいずれかに記載の組換えDNA分子からなる組換えパルボウ イルス粒子を産生するためのパッケージング細胞系。 17.請求の範囲第16項記載のパッケージング細胞系からなり、rAAVベクターの回 復、複製、パッケージングおよび組換えに必要な配列情報をin cisで含むパルボ ウイルスの逆方向末端反復配列に隣接する興味ある遺伝子からなり、パッケージ ングされる少なくとも1つの追加の組換え核酸分子をパッケージング細胞が伴う 、組換えパルボイウルス粒子を産生するための細胞系。 18.ウイルス粒子の産生に必要な物質がパッケージング細胞の遺伝子物質または 追加の組換え核酸に存在するが両者には存在しない請求の範囲第17項記載の細胞 系。 19.請求の範囲第17項または第18項記載の細胞系を適切 な培養培地で培養することおよび該培養から組換えパルボウイルスを採取するこ とからなる組換えパルボウイルスの産生法。 20.請求の範囲第19項記載の方法によりえられる組換えパルボウイルス。 21.宿主細胞において興味ある遺伝子の発現を誘導可能にする方法であって、該 遺伝子の発現を本質的に完全にスイッチオフとすることができ、該遺伝子が誘導 可能なトランスアクチベーターと誘導可能なリプレッサーの組合せの制御下にあ る方法。
