- 【特許請求の範囲】 1.液体サンプル中の複数の異なるポリヌクレオチド分析物を検出または定量 するためのデバイスであって、該デバイスは、 (i)サンプル注入口、(ii)1つ以上の検出チャンバ、および(iii)各該チャンバ と該注入口との間に行き止まり流体連通を提供するチャネル手段を有するサンプ ル分配ネットワークを規定する基板を備え、ここで該検出チャンバの少なくとも 2つは各々、検出可能シグナルを生じる、該サンプル中に存在し得る異なるポリ ヌクレオチド分析物を検出するための検出試薬を含み、そして 該基板は、各チャンバについて、このようなシグナルを検出するための検出手 段を提供し、 それにより、該注入口のサンプルの適用前の該ネットワークの排気は、該チャ ンバの各々に真空によりサンプルを流すのに有効である、 デバイス。 2.前記チャネル手段が、前記検出チャンバが前記流体連通により連結されて いる1つのチャネルを備える、請求項1に記載のデバイス。 3.前記チャネル手段が、第1の群の検出チャンバがこのような行き止まり流 体連通により連結されている第1のチャネル、および第2の群の検出チャンバが このような行き止まり流体連通により連結されている第2のチャネルを備える、 請求項1に記載のデバイス。 4.前記チャネル手段が、前記注入口と各検出チャンバとの間に行き止まり流 体連通を提供するために、各検出チャンバについて個々のチャネルを備える、請 求項1に記載のデバイス。 5.前記サンプル注入口と前記検出チャンバに液体連絡している部位で前記チ ャネル手段に連結された真空ポートをさらに備える、請求項1に記載のデバイス 。 6.前記真空ポートが、前記検出チャンバの下流の部位で前記チャネル手段に 連結されている、請求項5に記載のデバイス。 7.前記チャネル手段と液体連絡した非フロースルー真空リザーバをさらに備 える、請求項1に記載のデバイス。 8.前記検出手段が各検出チャンバと関連した光学透明窓を備え、それを通っ てこのようなシグナルが光学的に検出され得る、請求項1に記載のデバイス。 9.前記分析物特異的検出試薬が、プライマーにより開始されるポリメラーゼ 連鎖反応による選択されたポリヌクレオチド分析物セグメントの増幅のために、 該セグメントに相補鎖の反対末端領域にハイブリダイズするのに有効な配列を有 する第1および第2のオリゴヌクレオチドプライマーを含む、前記請求項のいず れかに記載のデバイス。 10.前記分析物特異的検出試薬が、前記分析物が前記サンプル中に存在する 場合に検出可能な蛍光シグナルを生じるために、前記プライマーの1つの下流の 領域の分析物セグメントにハイブリダイズし得る蛍光剤−消光剤オリゴヌクレオ チドをさらに含む、請求項9に記載のデバイス。 11.前記分析物特異的検出試薬が、選択された分析物のオリゴヌクレオチド 連結アッセイ検出のために、該分析物中の標的配列の隣接した連続的な領域に結 合するのに有効な第1および第2のオリゴヌクレオチドを含む、前記請求項のい ずれかに記載のデバイス。 12.前記分析物特異的検出試薬が、リガーゼ連鎖反応による前記ポリヌクレ オチド分析物の増幅のために、第1のオリゴヌクレオチド対により結合された領 域に相補的な隣接した連続的な領域に結合するのに有効である第2のオリゴヌク レオチド対を含む、請求項11に記載のデバイス。 13.前記分析物特異的検出試薬が、前記ポリヌクレオチド分析物中の選択さ れた配列にハイブリダイズするのに有効な結合ポリマーを含み、そして該結合ポ リマーが、該選択された配列にハイブリダイズする際に検出可能シグナルを生じ る蛍光染料部分を含む、前記請求項のいずれかに記載のデバイス。 14.前記分析物特異的検出試薬が、二本鎖ポリヌクレオチドをインターカレ ートする際に光学検出可能シグナルを生じるインターカレート化合物をさらに含 む、前記請求項のいずれかに記載のデバイス。 15.前記基板が、各検出チャンバの温度を制御するための温度調節手段をさ らに含む、前記請求項のいずれかに記載のデバイス。 16.前記基板が、少なくとも2つのこのようなサンプル分配ネットワークを 規定する、前記請求項のいずれかに記載のデバイス。 17.前記ネットワークの内部が真空下にある、前記請求項のいずれかに記載 のデバイス。 18.液体サンプル中の複数のポリヌクレオチド分析物の1つ以上を検出また は定量するための方法であって、該方法は、 液体サンプルを前記請求項のいずれかに記載のデバイスのサンプル注入口に適 用し、そして該サンプルを真空作用によりネットワークに流させ、該サンプルを 検出チャンバに送達する工程、 該送達されたサンプルを、該選択された分析物が該サンプル中に存在する場合 に各検出チャンバにおいて検出可能シグナルを生じるのに有効な条件下で、各検 出チャンバにおいて分析物特異的試薬と反応させる工程、および 該反応チャンバで生じたシグナルを測定し、どの選択された分析物が該サンプ ル中に存在するかを決定する工程 を包含する、方法。
