Claims (1)
- 【特許請求の範囲】 1.ヌクレオチド標識相互作用領域において少なくとも1つの変異を有するDNA ポリメラーゼであって、ここで該DNAポリメラーゼは、フルオレセイン型色素標 識ヌクレオチドに対する差別が低減した、DNAポリメラーゼ。 2.前記変異が、(i)O-ヘリックス、(ii)Kヘリックス、および(iii)O-Pヘリッ クス間ループからなる群より選択されるヌクレオチド標識相互作用領域の部分に ある、請求項1に記載のDNAポリメラーゼ。 3.前記変異が、E520、A531、L522、R523、E524、A525、H526、P527、I528、V5 29、E530、K531、I532、R536、E537、R573、Q582、N583、V586、R587、P589、Q5 92、R593、R595、D610、T612、Q613、E615、R636、D637、T640、F647、V654、D6 55、P656、L657、R659、R660、T664、E681、L682、A683、I684、P685、E688、F6 92、Q754、H784、L817、E820、L828、K831、およびE832からなる群より選択され るアミノ酸残基に対応する酵素のセグメントである、請求項2に記載のDNAポリ メラーゼ。 4.前記変異が、R595、D655、R660、T664、およびE681からなる群より選択され る位置にある、請求項3に記載のDNAポリメラーゼ。 5.前記DNAポリメラーゼが、Taq DNAポリメラーゼである、請求項4に記載のDN Aポリメラーゼ。 6.前記変異が、R660D、D655L、E618G、およびR595Eからなる群より選択される 、請求項5に記載のDNAポリメラーゼ。 7.(G46D,R660D,F667Y)、(G46D,R595D,R660D,F667Y)、および(G46D,R660D,F667 Y,E681G)、および(G46D,F667Y,E681G)からなる群に属する変異の組を含む、請求 項6に記載のDNAポリメラーゼ。 8.前記DNAポリメラーゼが耐熱性DNAポリメラーゼである、請求項2に記載のDN Aポリメラーゼ。 9.請求項1に記載のDNAポリメラーゼをコードするポリヌクレオチド。 10.プロモーターを有する発現ベクターであって、ここで該ベクターが、該プ ロモーターと機能的に組み合せた請求項9に記載のポリヌクレオチドを含む、ベ クター。 11.請求項10に記載の発現ベクターを含む、宿主細胞。 12.蛍光標識ポリヌクレオチドを合成する方法であって、該方法が、請求項1 に記載のDNAポリメラーゼとプライムされる鋳型とを混合する工程を包含する、 方法。 13.前記プライムされる鋳型が、鎖終結配列決定反応においてプライムされる 鋳型である、請求項12に記載の方法。 14.前記プライムされる鋳型が、ポリメラーゼ連鎖反応におけるプライムされ る鋳型である、請求項12に記載の方法。 15.ポリヌクレオチドを蛍光標識するためのキットであって、該キットが、請 求項1に記載のDNAポリメラーゼおよび蛍光標識ヌクレオチドを含む、キット。
