Claims (1)
- 【特許請求の範囲】 1.試薬と基質との間でアシル化移行機構を含む化学反応を、該基質と変遷複合 体を形成できるイミダゾール系触媒の存在下で行う方法において、触媒性イミダ ゾール機能が、該変遷複合体をアシル基との分子相互作用によって安定化するこ とができる基で片側または両側がフランキンングされた、任意に置換されたイミ ダゾール基を含む化学構造要素によって与えられることを特徴とする方法。 2.前記分子相互作用が、水素結合、静電的相互作用および疎水的相互作用から 選ばれる請求の範囲1に記載の方法。 3.前記フランキンング基が、該変遷複合体と分子相互作用することができる末 端基に結合された、1ないし6個の鎖原子の鎖、好ましくは炭素原子1−4のア ルキル鎖、を含むことを特徴とする請求の範囲1または2に記載の方法。 4.pH条件が、前記アシル移行反応で基質と反応する試薬がかなりの程度プロ トン付加されるように選択される請求の範囲1、2または3に記載の方法。 5.前記フランキンング基が、正に荷電した水素結合ドナーである請求の範囲1 ないし4のいずれか1項に記載の方法。 6.前記化学構造要素が、イミダゾール基とフランキンング基とが幾何学的に相 互に固定された関係にあるように配置されている固定構造を有することを特徴と する請求の範囲1ないし5のいずれか1項に記載の方法。 7.前記化学構造要素がペプチドまたはポリペプチドであることを特徴とする請 求の範囲1ないし6のいずれか1項に記載の方法。 8.前記イミダゾール機能が、任意に置換されたヒスチジン残基によって与えら れることを特徴とする請求の範囲1ないし7のいずれか1項に記載の方法。 9.前記化学構造要素が、例えば、らせん−ループ−らせん型の、設計されたタ ンパク質またはポリペプチドの一部であることを特徴とする請求の範囲7または 8に記載の方法。 10.前記化学構造要素が、α−らせん型であり、そして前記フランキンング基 が、iの位置のイミダゾール機能に対してi+4の位置に配置されていることを 特徴とする請求の範囲7、8または9に記載の方法。 11.前記化学構造要素が、α−らせん型であり、そして前記フランキンング基 が、iの位置のイミダゾール機能に対してi−3の位置に配置されていることを 特徴とする請求の範囲7、8または9に記載の方法。 12.前記フランキンング基が、ヒスチジン、オルニチン、ロイシンおよびアル ギニンから選ばれることを特徴とする請求の範囲7ないし11のいずれか1項に 記載の方法。 13.前記化学構造要素が、官能基を有する大きい構造を構成するかまたはその 一部であり、該官能基は分子内反応において前記変遷複合体を介してアシル移行 によって、基質によって選択的に機能化され得る程近接した位置にあることを特 徴とする請求の範囲1ないし12のいずれか1項に記載の方法。 14.前記官能基が前記フランキンング基または該フランキンング基の一部であ る請求の範囲13に記載の方法。 15.前記官能基が、iの位置のイミダゾール機能に対してi+4の位置にあり 、そしてオルニチン、ロイシンまたはジアミノ酪酸残基であることを特徴とする 請求の範囲14に記載の方法。 16.前記官能基が、iの位置のイミダゾール機能に対してi−3の位置にあり 、そしてロイシン残基であることを特徴とする請求の範囲14または15に記載 の方法。 17.前記官能基が、iの位置のイミダゾール機能に対してi+4およびi−3 の両方の位置にあり、そして例えば、ロイシンであることを特徴とする請求の範 囲15および16に記載の方法。 18.構造要素によって1つより多くのイミダゾール機能が与えられ、そして各 イミダゾール機能が、片側または両側が官能基によりフランキンングされている ことを特徴とする請求の範囲15ないし17のいずれか1項に記載の方法。 19.前記官能基が、i,j、k等の位置のイミダゾール機能に対してi+4、 j+4、k+4等の位置にあることを特徴とする請求の範囲18に記載の方法。 20.前記官能基が、i、j,k等の位置のイミダゾール機能に対してi−3、 j−3、k−3等の位置にあることを特徴とする請求の範囲18に記載の方法。 21.前記官能基が、炭水化物含有残基により機能化されることを特徴とする請 求の範囲12ないし20のいずれか1項に記載の方法。 22.前記基質が固体支持体上に設けられ、それにより前記化学構造要素が該支 持体に固定されることを特徴とする請求の範囲12ないし21のいずれか1項に 記載の方法。 23.請求の範囲1ないし20のいずれか1項に定義された、設計された化学構 造要素。 24.アシル移行機構を含む反応用の化学触媒であって、該触媒が請求の範囲1 ないし20のいずれか1項に定義された、設計された化学構造要素を含むことを 特徴とする化学触媒。 25.請求の範囲7ないし20および22のいずれか1項に定義された、設計さ れた化学構造要素を構成するまたは含むタンパク質またはペプチドの製造法にお いて、宿主生物を、ベクターおよび該構造要素をコードしたDNA配列を含む組 換えDNA構造で形質転換し、該宿主生物を培養して該ペプチド、ポリペプチド またはタンパク質を発現させ、該ペプチド、ポリペプチドまたはタンパク質を培 養物から分離することを特徴とする製造法。 26.ベクターおよび請求の範囲7ないし23のいずれか1項に定義された化学 構造要素をコードしたDNA配列を含む、組換えDNA構造。 27.所望の化学的機能を化学的化合物の特定の部位に導入する方法において、 該化学化合物に請求の範囲1ないし23のいずれか1項に定義された構造要素を 与え、そして該化合物を所望の機能を与えることができる基質と反応させること を特徴とする方法。 28.前記基質が炭水化物残基を含むことを特徴とする、請求の範囲27に記載 の方法。 29.請求の範囲28に記載の方法を、タンパク質分解からタンパク質を保護す るのに使用する方法。 30.請求の範囲28に記載の方法を、タンパク質の折りたたみに影響を及ぼす のに使用する方法。 31.1個より多くの基を選択的に機能化することができることを特徴とする請 求の範囲17ないし20のいずれか1項に記載の方法の使用法。 32.前記官能基が炭水化物含有残基によって選択的に機能化されることを特徴 とする、請求の範囲17ないし20のいずれか1項に記載の方法の使用法。 33.ワクチンの開発に使用する、請求の範囲17ないし22、31および32 のいずれか1項に記載の方法の使用法。 34.免疫系の天然の成分に真似るために使用する、請求の範囲12ないし22 、31および32のいずれか1項に記載の方法の使用法。 35.免疫系の成分のアンタゴニストを構成するために使用する、請求の範囲1 2ないし21、31および32のいずれか1項に記載の方法の使用法。 36.免疫系の成分のアゴニストを構成するために使用する、請求の範囲12な いし21、31および32のいずれか1項に記載の方法の使用法。 37.免疫系アンタゴニストの設計用の構造要素の開発のために使用する、請求 の範囲34に記載の方法の使用法。
