Claims (1)
- 【特許請求の範囲】 1.αまたはβ立体配置いずれかの炭素−炭素結合を介して糖部位のC−1原 子に連結したピレン、アントラセンまたはフェナントレンを含み、ここに該糖部 位がリボースまたはデオキシリボースであることを特徴とするヌクレオシド誘導 体。 2.α−9−フェナントレニルデオキシヌクレオシド 3.α−1−ピレニルデオキシヌクレオシド 4.β−9−フェナントレニルデオキシヌクレオシド 5.β−1−ピレニルデオキシヌクレオシド 6.請求項1記載のヌクレオシドのホスホルアミダイト誘導体 7.該ピレン、アントラセンまたはフォナントレンが、メトキシ、エトキシ、 ジメチルアミノ、ジエチルアミノ、ニトロ、メチル、シアノ、カルボキシ、クロ ロ、ブロモ、ヨードおよびアミノよりなる群から選択される置換基で、利用可能 な炭素位置にて誘導体化された請求項1記載のヌクレオシド誘導体。 8.請求項1ないし6いずれか1記載の少なくとも1つのヌクレオシド誘導体 を含む核酸。 9.a,ピレン、アントラセンまたはフォナントレンのオルガノカドミウムま たはオルガノ亜鉛をホッファーのα−ククロ糖のC1位にカップリングさせ;次 いで、 b.メタノール性塩基で保護基を除去する; ことを特徴とする請求項1記載のヌクレオシド誘導体の合成方法。 10.a.ピレン、アントラセンまたはフォナントレンのオルガノカドミウム またはオルガノ亜鉛をホッファーのα−ククロ糖のC1位にカップリングさせ; b.メタノール性塩基で保護基を除去し; c.塩基の存在下で、5’−OHをジメトキシトリチルクロライドでトリチル 化し;次いで、 d.3’−OHをホスフィチル化する; ことを特徴とする請求項1記載のヌクレオシド誘導体のホスホルアミダイト誘導 体の合成方法。 11.請求項1ないし6いずれか1記載のヌクレオシド誘導体を、該誘導体を 取り込むのに十分な条件下で、RNAまたはDNA分子に取り込むことを特徴と する蛍光標識核酸の調製方法。 12.当該標識および当該プローブの間のハイブリダイゼーションを可能とす る時間および条件下で、標的核酸を、請求項1ないし6いずれか1記載の少なく とも1の核酸誘導体を含む核酸プローブと接触させ;次いで該ハイブリダイゼー ションを検出することを特徴とする、テストすべき試料中の標的核酸を検出する 方法。
