- 【特許請求の範囲】 1.連鎖球菌コリン結合タンパク質であって、そのタンパク質が連鎖球菌により 発現され、かつ下記の特徴: a)コリン結合活性、及び b)少なくとも約10%のコリンの存在下のクロマトグラフィーカラムからの溶離 を有し、但し、連鎖球菌コリン結合タンパク質がPspAまたは自己溶菌酵素(LytA) ではないことを条件とすることを特徴とする連鎖球菌コリン結合タンパク質。 2.c)宿主細胞へのバクテリアの付着を抑制すること、 d)感染された患者またはバクテリアによる感染から回復している患者からの血 清と反応性であること、 e)少なくとも約10%のコリン中の溶離によりコリンアフィニティーカラムから 単離された肺炎球菌タンパク質に対し産生されたウサギ抗血清と反応性であるこ と、及び f)連鎖球菌溶解を必要としないで(即ち、無傷バクテリア中で)フルオレセイ ンイソチオシアネート(FITC)により標識されること からなる群から選ばれた特徴の一つ以上を有する請求の範囲第1項に記載の連鎖 球菌コリン結合タンパク質。 3.ストレプトコッカス・ニゥモニエから単離される請求の範囲第1項に記載の 連鎖球菌コリン結合タンパク質。 4.112kDa、90kDa、84kDa、70kDa、及び50kDaからなる群から選ばれた少なくと も約10%のSDS-PAGEによる見掛分子量を有する請求の範囲第3項に記載の連鎖球 菌コリン結合タンパク質。 5.配列番号1−10、配列番号19、配列番号21及び配列番号25からなる群から選 ばれた部分アミノ酸配列を有する請求の範囲第1項に記載の連鎖球菌コリン結合 タンパク質。 6.検出可能な標識で標識された請求の範囲第1項に記載の連鎖球菌コリン結合 タンパク質。 7.配列番号19のアミノ酸配列を有する連鎖球菌コリン結合タンパク質。 8.配列番号25のアミノ酸配列を有する連鎖球菌コリン結合タンパク質。 9.請求の範囲第1項〜第8項のいずれかに記載の連鎖球菌コリン結合タンパク 質及び医薬上許されるアジュバントを含むことを特徴とするワクチン。 10.a)異なる連鎖球菌コリン結合タンパク質、 b)PspA、 c)自己溶菌酵素(LytA)、及び d)以上の一種以上のあらゆる組み合わせ からなる群から選ばれた抗原を更に含む請求の範囲第9項に記載のワクチン。 11.請求の範囲第1項〜第8項のいずれかに記載の連鎖球菌コリン結合タンパク 質及び医薬上許されるキャリヤーを含むことを特徴とする医薬組成物。 12.a)PspAまたは自己溶菌酵素(LytA)、 b)抗生物質、 c)コリン結合タンパク質が下記の特徴: i)コリン結合活性、及び ii)少なくとも約10%のコリンの存在下のクロマトグラフィーカラムから の溶離を有し、但し、連鎖球菌コリン結合タンパク質がPspAまたは自己溶菌酵素 (LytA)ではないことを条件とする、抗連鎖球菌コリン結合タンパク質ワクチン、 d)ステロイド、及び e)抗連鎖球菌ワクチン からなる群から選ばれた活性成分を更に含む請求の範囲第11項に記載の医薬組成 物。 13.コリン結合タンパク質が下記の特徴: i)コリン結合活性、及び ii)少なくとも約10%のコリンの存在下のクロマトグラフィーカラムから の溶離を有し、但し、連鎖球菌コリン結合タンパク質がPspAまたは自己溶菌酵素 (LytA)ではないことを条件とすることを特徴とする連鎖球菌コリン結合タンパク 質の精製抗体。 14.請求の範囲第1項〜第8項のいずれかに記載の連鎖球菌コリン結合タンパク 質のモノクローナル抗体。 15.請求の範囲第14項に記載のモノクローナル抗体を産生する不死化細胞系。 16.検出可能な標識で標識された請求の範囲第13項または第14項に記載の抗体。 17.標識が酵素、蛍光を発する薬品、及び放射性元素からなる群から選ばれる請 求の範囲第16項に記載の抗体。 18.請求の範囲第1項〜第8項のいずれかに記載のコリン結合タンパク質の抗体 及び医薬上許されるキャリヤーを含むことを特徴とする医薬組成物。 19.請求の範囲第1項〜第8項のいずれかに記載の連鎖球菌コリン結合タンパク 質、またはその少なくとも15ヌクレオチドのフラグメントをコードする精製核酸 。 20.a)配列番号1−10、または19もしくは21もしくは25に示された配列を有する ポリペプチドをコードするDNA配列、 b)高度にストリンジェントなハイブリダイゼーション条件下で(a)のDNA 配列にハイブリッドを形成するDNA配列、及び c)(A)または(B)の以上のDNA配列によりコードされたアミノ酸配列をコー ドするDNA配列 からなる群から選ばれたヌクレオチド配列を有するDNA分子である請求の範囲 第19項に記載の核酸。 21.請求の範囲第20項に記載の組換えDNA分子。 22.ヌクレオチド1から終止コドンTAAまで配列番号18に示されたヌクレオチド 配列を有する請求の範囲第21項に記載の組換えDNA分子。 23.配列番号24のコーディング領域に示されたヌクレオチド配列を有する請求の 範囲第21項に記載の組換えDNA分子。 24.DNA分子が発現調節配列に機能可能なように結合される請求の範囲第21項 に記載の組換えDNA分子。 25.請求の範囲第19項に記載の核酸から調製された別の種中の連鎖球菌コリン結 合タンパク質をコードする核酸についてスクリーニングすることができるオリゴ ヌクレオチド。 26.請求の範囲第24項に記載の組換えDNA分子で形質転換された単細胞宿主。 27.請求の範囲第24項に記載の組換えDNA分子を含むことを特徴とする核酸ワ クチン。 28.請求の範囲第1項に記載の連鎖球菌コリン結合タンパク質の存在の検出方法 であって、その連鎖球菌コリン結合タンパク質が a)連鎖球菌コリン結合タンパク質の存在または活性が疑われているサンプルを 結合パートナーへの連鎖球菌コリン結合タンパク質の結合が起こることを可能に する条件下で連鎖球菌コリン結合タンパク質の抗体と接触させ、そして b)結合がサンプルからの連鎖球菌コリン結合タンパク質と抗体の間で起こった か否かを検出することにより測定され、結合の検出がサンプル中の連鎖球菌コリ ン結合タンパク質のその存在または活性を示すことを特徴とする検出方法。 29.請求の範囲第1項に記載の連鎖球菌コリン結合タンパク質の遺伝子を有する バクテリアの存在の検出方法であって、 a)バクテリアの存在または活性が疑われているサンプルをその遺伝子へのオリ ゴヌクレオチドの特異的ハイブリダイゼーションが起こることを可能にする条件 下で連鎖球菌コリン結合タンパク質遺伝子にハイブリッドを形成するオリゴヌク レオチドと接触させ、そして b)ハイブリダイゼーションがオリゴヌクレオチドと遺伝子の間で起こったか否 かを検出し、ハイブリダイゼーションの検出がサンプル中のバクテリアのその存 在または活性を示すことを特徴とする検出方法。 30.免疫有効投薬量の請求の範囲第9項に記載のワクチンを被験者に投与するこ とを特徴とする連鎖球菌コリン結合タンパク質を発現するバクテリアによる感染 症の防止方法。 31.免疫有効投薬量の請求の範囲第27項に記載のワクチンを被験者に投与するこ とを特徴とする連鎖球菌コリン結合タンパク質を発現するバクテリアによる感染 症の防止方法。 32.治療有効投薬量の請求の範囲第11項に記載の医薬組成物を被験者に投与する ことを特徴とする連鎖球菌コリン結合タンパク質を発現するバクテリアによる感 染症の治療方法。 33.治療有効投薬量の請求の範囲第18項に記載の医薬組成物を被験者に投与する ことを特徴とする連鎖球菌コリン結合タンパク質を発現するバクテリアによる感 染症の治療方法。 34.アミノ酸配列WQPPRARI(配列番号11)を有する15以下のアミノ酸残基のペプ チド、エノラーゼ、及びアミノ酸配列WQPPRARIに特異性の抗体からなる群から選 ばれたフィブロネクチンへの連鎖球菌付着のインヒビターを含むことを特徴とす る医薬組成物。 35.治療有効投薬量の請求の範囲第34項に記載の医薬組成物を被験者に投与する ことを特徴とする連鎖球菌コリン結合タンパク質を発現するバクテリアによる感 染症の治療方法。 36.フィブロネクチンへの連鎖球菌付着を抑制するヒンダード陽イオン小分子を 投与することを特徴とする連鎖球菌コリン結合タンパク質を発現するバクテリア による感染症の治療方法。 37.ヒンダード陽イオン小分子がリシン、コリン、及びアルギニンからなる群か ら選ばれる請求の範囲第36項に記載の方法。 38.ヒンダード陽イオン小分子がフィブロネクチンへのエノラーゼの結合を抑制 する請求の範囲第36項に記載の方法。 39.下記の特徴: a)コリン結合活性、及び b)少なくとも約10%のコリンの存在下のクロマトグラフィーカラムからの溶離 を有し、但し、連鎖球菌コリン結合タンパク質がPspAまたは自己溶菌酵素(LytA) ではないことを条件とするコリン結合タンパク質、コリン結合タンパク質の抗体 、エノラーゼ、ヒンダード陽イオン小分子、ペプチドWQPPRARI(配列番号11)、 及びアミノ酸配列WQPPRARI(配列番号11)を有するエピトープに特異性の抗体か らなる群から選ばれた付着抑制剤を肺に投与することを特徴とする連鎖球菌コリ ン結合タンパク質を発現するバクテリアによる感染症の治療方法。 40.ヒンダード陽イオン小分子がリシン、コリン、及びアルギニンからなる群か ら選ばれる請求の範囲第39項に記載の方法。
