Claims (1)
- 【特許請求の範囲】 1. 患者サンプルを、約30kDaのRNase Lの存在について分析す ることを含むことを特徴とする慢性疲労症候群を診断する方法。 2. (a)前記患者サンプル中のタンパク質を自然条件下で分子量によって 分画化し; (b)前記分画化したタンパク質を、約30kDaのタンパク質がRNase L活性を有することについて分析する、 ことを含むことを特徴とする請求項1の方法。 3. 前記サンプルは、血液又はその分画を含むことを特徴とする請求項1又 は2の方法。 4. 前記サンプルは、末梢血液単核細胞の細胞質抽出物を含むことを特徴と する請求項3の方法。 5. 前記RNase L活性の分析は、28S及び/又は18SRNAの加 水分解による特異的開裂生成物の形成を検出することを含むことを特徴とする請 求項2〜4のいずれかの項に記載の方法。 6. 前記RNase L活性の分析は、ポリ(U)−3’−pCpの加水分 解を検出することを含むことを特徴とする請求項2〜4のいずれかの項に記載の 方法。 7. 前記ポリ(U)−3’−pCpに含まれる標識は、放射標識を含むこと を特徴とする請求項6の方法。 8. 前記標識化されたポリ(U)−3’−pCpは、ポリ(U)−3’−[32 P]pCpであることを特徴とする請求項7の方法。 9. (a)付加プロテアーゼインヒビターの存在下で調製した患者サンプル と、検出可能な標識を担持する構造式(I)に従う化合物を含むプローブとを、 前記標識化されたプローブ化合物と前記サンプル中の2’,5’−オリゴアデニ レート結合タンパク質との共有結合物を形成するのに十分な条件下で接触させ: 構造式(I) 上記式中、 mは0から3までの整数であり、 nは1から3までの整数であり、及び 各R1及び各R2は、互いに他のR1及びR2とは独立して、水素又はN3であ り、少なくとも1つのR1又はR2がN3であることを条件とし、 或は前記化合物の水溶性塩である; (b)前記共有結合物を含む前記サンプルと、前記サンプル中のRNase L種に結合する抗体とを接触させ; (c)前記サンプル中の前記タンパク質をゲル電気泳動で分離し;そして (d)前記分離したタンパク質を、 (i)前記標識化されたプローブ化合物と共有結合物を形成し、及び (ii)前記抗体によって結合された マーカータンパク質について調べ; ドデシル硫酸ナトリウム−ポリアクリルアミドゲル電気泳動による見かけの分 子量約37kDaのマーカータンパク質の存在が、慢性疲労症候群の徴候を示す ことを特徴とする慢性疲労症候群を診断する方法。 10. 前記共有結合物を形成する前記条件は、前記プローブ化合物を前記サ ンプル中の2',5’−オリゴアデニレート結合タンパク質に共有結合させるた めに前記サンプルを紫外線照射することを含むことを特徴とする請求項9の方法 。 11. 前記プローブ化合物においてnが1であることを特徴とする請求項9 又は10の方法。 12. 前記プローブ化合物においてmが1であることを特徴とする請求項9 〜11のいずれかの項に記載の方法。 13. 前記プローブ化合物の各R1が水素であることを特徴とする請求項9 〜12のいずれかの項に記載の方法。 14. 前記プローブ化合物は、5’−O−ホスホリル−アデニリル−(2' →5')−アデニリル−(2’→5')−8−アジドアデノシン、又はその塩であ ることを特徴とする請求項13の方法。 15. 前記サンプルは、血液又はその分画を含むことを特徴とする請求項9 〜14のいずれかの項に記載の方法。 16. 前記サンプルは、末梢血液単核細胞の細胞質抽出物を含むことを特徴 とする請求項15の方法。 17. 前記プローブ化合物上の前記検出可能な標識は、放射標識であること を特徴とする請求項9〜16のいずれかの項に記載の方法。 18. 前記放射標識は、32P原子であることを特徴とする請求項17の方法 。 19. (a)付加プロテアーゼインヒビターの非存在下で調製した患者サン プルと、検出可能な標識を担持する構造式(I)に従う化合物を含むプローブと を、前記標識化されたプローブ化合物と前記サンプル中の2',5’−オリゴア デニレート結合タンパク質との共有結合物を形成するのに十分な条件下で接触さ せ; 構造式(I) 上記式中 mは0から3までの整数であり、 nは1から3までの整数であり、及び 各R1及びR2は、互いに他のR1及びR2とは独立して、水素又はN3であり 、少なくとも1つのR1又はR2はN3であることを条件とし、 或は前記化合物の水溶性塩である; (b)前記共有結合物を含む前記サンプルと、前記サンプル中のRNase L種に結合する抗体とを接触させ; (c)前記サンプル中の前記タンパク質をゲル電気泳動で分離し;そして (d)前記分離したタンパク質を、 (i)前記標識化されたプローブ化合物と共有結合物を形成し、及び (ii)前記抗体によって結合された マーカータンパク質について調べ; ドデシル硫酸ナトリウム−ポリアクリルアミドゲル電気泳動による見かけの分 子量約37kDaのマーカータンパク質の存在、及びドデシル硫酸ナトリウム− ポリアクリルアミドゲル電気泳動による見かけの分子量約80kDaのマーカー タンパク質の非存在が、重度の慢性疲労症候群の徴候を示すことを特徴とする重 度の慢性疲労症候群を検出する方法。 20. 前記共有結合物を形成する前記条件は、前記プローブ化合物を前記サ ンプル中の2',5’−オリゴアデニレート結合タンパク質に共有結合させるた めに前記サンプルを紫外線照射することを含むことを特徴とする請求項19の方 法。 21. 前記プローブ化合物においてnが1であることを特徴とする請求項1 9又は20の方法。 22. 前記プローブ化合物においてmが1であることを特徴とする請求項1 9〜21のいずれかの項に記載の方法。 23. 前記プローブ化合物の各R1が水素であることを特徴とする請求項1 9〜22のいずれかの項に記載の方法。 24. 前記プローブ化合物は、5’−O−ホスホリル−アデニリル−(2’ →5')−アデニリル−(2’→5’)−8−アジドアデノシン、又はその塩で あることを特徴とする請求項23の方法。 25. 自然条件下において患者サンプルを、約30kDa及び約80kDa の分子量を有するRNase L分子の存在について分析することを含み、それ によって、(i)約30kDaのRNase L分子の存在及び約80kDaの RNase L分子の非存在は重度の慢性疲労症候群を示し、(ii)両方の分 子量のRNase L分子の存在は比較的軽度の慢性疲労症候群を示す、 慢性疲労症候群の相対的重度を評価する方法。 26. (a)前記患者サンプル中の前記タンパク質を非変性条件下で分子量 によって分画化し; (b)前記分画化したタンパク質を、約30kDa及び約80kDaの分子が RNase L活性を有することについて分析する、 ことを含むことを特徴とする請求項25の方法。 27. 前記サンプルは、血液又はその分画を含むことを特徴とする請求項2 5又は26の方法。 28. 前記サンプルは、末梢血液単核細胞の細胞質抽出物を含むことを特徴 とする請求項27の方法。 29. 前記RNase L活性の分析は、ポリ(U)−3’−pCpの加水 分解を検出することを含むことを特徴とする請求項26〜28のいずれかの項に 記載の方法。 30. 前記RNase L活性の分析は、28S及び/又は18SRNAの 加水分解による特異的開裂生成物の形成を検出することを含むことを特徴とする 請求項26〜28のいずれかの項に記載の方法。
