<標的物質の測定>
図19の装置において、抗CEA抗体、抗AFP抗体、抗PSA抗体及び抗PAP抗体を固定化させた標的物質検出素子を装置にセットし以下のプロトコールをおこなう。
(1)三方バルブを洗浄液側に切り替え、pHを7.4に調整したバッファー溶液を0.1(ml/min.)の流速で10分間流し、標的物質検出素子をよく洗浄する。
(2)バルブを試料溶液側に切り替え、試料溶液を0.1(ml/min.)の流速で10分間流し抗原抗体反応をおこす。
(3)試料溶液を送液しながら、一定時間ごとに反射スペクトルを測定する。
(4)再び、バルブを洗浄液側に切り替え、バッファー溶液を流速0.1(ml/min.)で10分間流し、非特異的に吸着した抗原を洗い流す。

<標的物質の測定>

(1)バルブ1を洗浄液側に、バルブ2を廃液タンク側に設定し、pHを7.4に調整したバッファー溶液を0.1(ml/min.)の流速で10分間流し、標的物質検出素子をよく洗浄する。
(2)バルブ1を試料溶液側に、バルブ2を循環用チューブ側に切り替え、試料溶液を0.1(ml/min.)の流速で10分間流し抗原抗体反応をおこす。
(3)試料溶液を送液しながら、一定時間ごとに反射スペクトルを測定する。
(4)再び、バルブを洗浄液側に切り替え、バッファー溶液を流速0.1(ml/min.)で10分間流し、非特異的に吸着した抗原を洗い流す。
