Abstract
本発明は、加齢とともに減少するテストステロンの産生を、体内の経路を用いて促進するテストステロン産生促進剤を提供することにある。
【解決手段】
プロペニルシステイン又はグルタミル−プロペニルシステインからなる群から選択される少なくとも1種を含有することを特徴とするテストステロン産生促進剤であり、好ましくは、前記プロペニルシステイン及びグルタミル−プロペニルシステインがアリウム属植物由来であり、さらに好ましくは、前記アリウム属植物がニンニクであることを特徴とするテストステロン産生促進剤。

Description
(1)プロペニルシステイン又はグルタミル−プロペニルシステインからなる群から選択される少なくとも1種を含有することを特徴とするテストステロン産生促進剤。
(2)前記プロペニルシステイン及びグルタミル−プロペニルシステインがアリウム属植物由来であることを特徴とする(1)記載のテストステロン産生促進剤。
(3)前記アリウム属植物がニンニクであることを特徴とする(2)記載のテストステロン産生促進剤。
(4)(1)〜(3)いずれかに記載のテストステロン産生促進剤を含むことを特徴とするテストステロン産生促進用食品用添加剤。

本発明のプロペニルシステインは、S−1−プロペニルシステイン、及びS−2−プロペニルシステイン(S−アリルシステイン)などを挙げることができる。なお、S−1−プロペニルシステインの天然物(化1)及びS−2−プロペニルシステイン(S−アリルシステイン)の天然物(化2)は、一般に下記の一般式で示される構造を有する。
本発明において好適に用いられる天然素材原料であるアリウム属の植物にはアリイナーゼと呼ばれる酵素が内在し、アリウム属に含まれる含硫アミノ酸誘導体であるアリインを臭気成分アリシンへと変換することが知られている。アリインは本発明の原料となり得るため、また発生する臭気を抑えるためにもアリシンの合成を抑制、すなわちアリイナーゼの作用を抑制することが好ましい。アリインを含む素材の内在するアリイナーゼの失活処理方法は、上記の酵素を失活させることができれば特に限定されないが、例えば、当該素材の内部温度を60℃以上に高められる方法が挙げられる。失活処理方法の具体例としては、熱水加熱処理、蒸気加熱処理、マイクロ波加熱処理などが挙げられる。また、酸処理やアルカリ処理により内在するアリイナーゼを失活処理する方法も挙げられる。
アリイナーゼを熱失活させた天然素材原料からテストステロン産生促進剤を得る方法の一つに、グルタチオンとの反応が挙げられる。グルタチオンは、グルタミン酸、システイン、グリシンの3つのアミノ酸からなるトリペプチドであり、食品や生体内に多量に存在し高い抗酸化性を有することが知られている。グルタチオンには還元型と酸化型(還元型グルタチオン2分子がジスルフィド結合したもの)があるが、生体内では専ら還元型として存在し、過酸化物や活性酸素種の還元、細胞の解毒などの役割を担っている。グルタチオンとしては、化学合成されたグルタチオンを原料として用いてもよいし、グルタチオンを含む素材からのグルタチオンの抽出物、精製物などいずれのグルタチオンを原料として用いてもよい。本発明においては、後述するように、グルタチオンを含む天然素材とアリインを含む素材と共存させることによって、グルタチオンとアリインと反応させることにより本発明のテストステロン産生促進剤を得ることができる。原料として用いるグルタチオンは、1種類単独で使用してもよいし、2種類以上を組み合わせて使用してもよい。
アリイナーゼを熱失活させた天然素材原料からテストステロン産生促進剤を得る方法の一つに、システインとの反応が挙げられる。システインはアミノ酸の一種であり、アリインなどと同じく含硫物である。原料となるシステインは、システイン(L体)の他、これの光学異性体(D体)であってもよいし、各光学異性体の混合物であってもよい。システインとしては、化学合成されたシステインを原料として用いてもよいし、システインを含む素材からのシステインの抽出物、精製物などを原料として用いてもよい。本発明においては、システインを含む素材とアリインを含む原料を共存させることによって、当該原料中に含まれるシステインとアリインを反応させることにより本発明のテストステロン産生促進剤を得ることができる。また、システインの2量体であるシスチンと任意の還元剤を共に用いて、反応中にシステインを作り出して、アリインと反応させても良い。
アリイナーゼを熱失活させた天然素材原料からテストステロン産生促進剤を得る方法の一つに、加熱による反応が挙げられる。アリイナーゼを熱失活させた原料を加熱処理工程に付する際の反応温度としては、20〜75℃の温度で行うことが必要であり、好ましくは25〜65℃、より好ましくは25〜55℃、さらに好ましくは30〜45℃が挙げられる。
本発明のテストステロン産生促進剤の適用量については、使用される製品の種類、用途、期待される効果、適用形態等に応じて適宜設定すればよい。例えば、経口適用される場合であれば、プロペニルシステイン又はグルタミル−プロペニルシステインからなる群から選択される少なくとも1種の成人1日あたりの摂取量が、0.000001〜100g、好ましくは0.00001〜100g、更に好ましくは0.001〜100gとなるように設定すればよい。また経皮適用される場合であれば、プロペニルシステイン又はグルタミル−プロペニルシステインからなる群から選択される少なくとも1種の1日当たりの皮膚への適用量が0.000001〜10000μg/cm2、好ましくは0.0001〜10000μg/cm2、更に好ましくは0.001〜1000μg/cm2となるように設定すればよい。
本発明のテストステロン産生促進剤は、生体内でテストステロンの合成にかかわる遺伝子群の作用を活性化する等により、加齢によって低下するテストステロンの産生を促進することが出来る。そのため、壮年期などにおけるテストステロン不足が関与する疾患を予防又は改善することが可能になる。具体的には、筋肉増強、血清脂質の改善、骨量増加、精子形成促進、勃起不全改善、性衝動増強、食欲増進、不眠改善、抗肥満、抗糖尿病、血圧正常化作用、美白、脳血管疾患リスク低減、心血管疾患リスク低減、抗動脈硬化作用、抗糖化作用、血小板凝集抑制作用、抗炎症作用、免疫賦活化作用、腸内細菌叢正常化作用、肝臓保護、育毛および造血から選ばれる効能をいずれか一以上を有することができる。
本発明のテストステロン産生促進剤の適用形態については、特に制限されないが、例えば、経口、経皮、経腸、経粘膜、経静脈、経動脈、皮下、筋肉内等の任意の適用形態で使用できるが、テストステロン産生促進作用をより一層有効に発揮させるという観点から、好ましくは、経口適用又は経皮適用、経静脈適用が挙げられる。
ニンニク(品種名:福地ホワイト)1kgの芯を除去した後、2〜3cmの鱗片に分け、マイクロ波加熱装置にて加熱処理(周波数:2.45GHz、出力:2000W、照射時間:2分間)を施し、アリイナーゼを失活させた。その後、純水1Lを添加して石臼式粉砕機(増幸産業製、商品名:スーパーマスコロイダー)にて摩砕し、ニンニクペーストを得た。そのペーストに対して、5規定の水酸化ナトリウムをpH8になるように添加して撹拌したのちに35℃に加熱して48時間反応させた。加熱反応終了後、そのまま凍結乾燥・粉砕して、本発明のテストステロン産生促進剤(粉末)を得た。粉末中のプロペニルシステインの一種であるS−2−プロペニルシステインの含有量をHPLCで分析したところ、1.35g/100gであった。なお、S−2−プロペニルシステインスルホキシドは含まれていなかった。
マウス精巣由来がん細胞I−10(JCRB細胞バンク:JCRB9097)を1.5×104細胞/cm2となるように、24ウェルプレート(IWAKI社製)に播種した。その後、37℃、5%CO2の環境下で、インキュベーター(ESPEC社製)を用いて培養を行った。24時間経過後、培地をS−2−プロペニルシステイン(東京化成社製、以下「SAC」と略する場合がある。)含有培地と交換することで、細胞にSACを作用させた。24時間作用後、培養上清を採取して、テストステロン測定キット(Cayman社製)によって定量を行った。また比較例1としてS−2−プロペニルシステインスルホキシド(シグマアルドリッチ社製、以下「アリイン」と略する場合がある。)を同じ濃度で細胞へと付加し、テストステロン合成能を比較した。このときのテストステロン産生関連遺伝子に対する影響を、CTL、SAC1μM、10μM作用細胞よりRNA抽出試薬Isogen(日本ジーン社製)を用いて、抽出を行い、逆転写酵素(タカラバイオ社製)を用いて合成を行ったcDNAを基質としてReal−timePCR(酵素:SYBRPremixEXTagII/タカラバイオ社製、検出機:StepOne/AppliedBiosystems社製)法にて測定した。
6週齢の早期老化モデルマウスであるSenescence−accelerated
mouse Prone8(SAMP8)マウス(日本SLC社製)を2週間予備飼育行った後、体重に差が無いように群分けを実施した。群分け後マウスの粉末飼料に、S−2−プロペニルシステインの終濃度が0.05%となるように実施例1で得られたテストステロン産生促進剤を添加し混合することで試験食1を得た。また試験食2は試験食1に含まれる量と同じ量の試薬SAC(東京化成社製)を含むように調整を行い、試験食3には試験食1にSACと当モル量のグルタミル−プロペニルシステインの1つであるγ‐グルタミル−S−2−プロペニル−L−システイン、通称γ−グルタミル−S−アリルシステイン(GSAC、日本健康・栄養食品協会製)をさらに添加してマウスへと摂餌させた。6週間マウスに対して投与を行った。試験のコントロールは非老化モデルであるSAMR1(日本SLC社製)を用い、SAMP8と同じタイムスケジュールで、通常食のみで飼育を継続した。試験期間終了後マウスをネンブタール麻酔下で解体し、中心静脈より採血を実施、血中のテストステロン量をテストステロン測定キット(Cayman社製)にて測定した。また、血中のγ−GTP量をLタイプワコーγ−GT・J(和光純薬工業社製)にて測定した。さらに同じく解体時にマウスの肝臓をサンプリングし、重量より肝臓の肥大等の所見の有無を確かめた。予備試験又は本試験を通じてマウスは、餌・水共に自由摂取下で飼育を行った。

- 請求項1〜3いずれか1項記載のテストステロン産生促進剤を含むことを特徴とするテストステロン産生促進用食品用添加剤。
