Description
パピローマウイルスはタンパク質キヤプシドにより取り囲まれた塩基対約7900の環状ゲノムを有するDNAウイルスである。多数のウシ(BPV)およびヒト(HPV)パピローマウイルスタイプが同定され、かつこれらのゲノム配列が決定されている[Pfister, 1987, The papovaviridae: The Papillomaviruses (Salzmanおよび Howley 著)Plenum Press, New York, 1-38頁]。このものは初期領域と後期領域を含んでいる。後期領域は、このキヤプシドの主成分をコードする二つの読み枠(reading frame )L1 およびL2 を含む。初期領域は少なくとも読み枠E1 、E2 、E4 、E5 、E6 およびE7 を含む。E1およびE2 発現産物はウイルス複製およびウイルス遺伝子の発現を制御するが、一方E5 、E6 およびE7 領域のものは感染細胞の発がん性形質転換の諸工程に関与する。事実、BPV−1 E5 タンパクは in vitro で細胞を形質転換できることが実験的に見い出されている(Schlegelら、1986, Science 233, 464-467)。BPV−1 E6 およびHPV−16 E7 タンパク質は発がん性形質転換の誘発および保存に関係する。E7 の形質転換力はHPV−16およびHPV−18の場合について実証されている(Kanda ら、1988, J. Virol. 62, 610-613; Vousden ら、1988, Oncogene Res. 3, 1-9; Bedellら、1987, J. Virol. 61, 3635-3640 )。E4 の場合にはなんらの機能も実証されていない。
また本発明は、パピローマウイルスからのポリペプチドと免疫刺激分子との組み合わせによる製剤を提供する。かかる組成物の利点の一つは、ウイルス抗原により誘発される特異的免疫性と、特異的応答を強化するように設計された免疫刺激分子により誘発される特異的免疫性とが併合されることにある。
(1)パピローマウイルスの初期領域からの少なくとも一種のポリペプチドおよびパピローマウイルスの後期領域からの少なくとも一種のポリペプチド、
(2)パピローマウイルスの初期領域からの少なくとも一種のポリペプチド、パピローマウイルスの後期領域からの少なくとも一種のポリペプチドおよび免疫刺激活性を有する少なくとも一種のポリペプチド、または
(3)パピローマウイルスの初期または後期領域からの少なくとも一種のポリペプチドおよび免疫刺激活性を有する少なくとも一種のポリペプチド
を含有する。
(1)パピローマウイルスの、E6 領域からのポリペプチド、E7 領域からのポリペプチド、L1 領域からのポリペプチドおよびL2 領域からのポリペプチド、
(2)パピローマウイルスのE6 領域からのポリペプチド、E7 領域からのポチペプチド、およびインターロイキン−2から誘導されるポリペプチド、
(3)パピローマウイルスのE6 領域からのポリペプチド、E7 領域からのポチペプチド、および上記分子B7.1から誘導されるポリペプチド、
(4)パピローマウイルスのE6 領域からのポリペプチド、E7 領域からのポチペプチド、上記分子B7.1から誘導されるポリペプチド、およびインターロイキン−2から誘導されるポリペプチド、
(5)パピローマウイルスの、E6 領域からのポリペプチド、E7 領域からのポチペプチド、L1 領域からのポリペプチド、L2 領域からのポリペプチド、およびインターロイキン−2から誘導されるポリペプチド、
(6)パピローマウイルスのE6 領域からのポリペプチド、E7 領域からのポチペプチド、L1 領域からのポリペプチド、L2 領域からのポリペプチド、および上記分子B7.1から誘導されるポリペプチド、または
(7)パピローマウイルスのE6 領域からのポリペプチド、E7 領域からのポチペプチド、L1 領域からのポリペプチド、L2 領域からのポリペプチド、上記分子B7.1から誘導されるポリペプチド、およびインターロイキン−2から誘導されるポリペプチド
を含んでいる。
(1)パピローマウイルスの初期領域からの少なくとも1種のポリペプチドおよびパピローマウイルスの後期領域からの少なくとも1種のポリペプチド、
(2)パピローマウイルスの初期領域からの少なくとも1種のポリペプチド、パピローマウイルスの後期領域からの少なくとも1種のポリペプチド、および免疫刺激活性を有する少なくとも1種のポリペプチド、または
(3)パピローマウイルスの初期または後期領域からの少なくとも1種のポリペプチド、および免疫刺激活性を有する少なくとも1種のポリペプチド;
をコードするDNAフラグメントが挿入される1種または2種以上の組み換えベクターを含有し、上記DNAフラグメントは宿主細胞または生物体中でのそれら発現に要する諸要素の支配下に置かれる。

図1は、プロモーターp7.5Kの統制下に置かれたCopenhagenワクシニアのTK遺伝子座、HPV−16後期遺伝子L1 およびL2 のTK遺伝子座へのトランスフアーを可能にするベクターpTG 5021を示す略図であり、この2つのカセットは互いに関し反対配向をなしている。
A. HPV16 L1 およびL2 遺伝子の単離
L1 およびL2 タンパク質をコードするフラグントは従来の一般的手法に従ってCaski 細胞(ATCC 1550)のゲノムDNAからPCRにより単離する。このL1 配列を保有する増幅用フラグメントを、ベクターM13TG130(Kieny ら、1983, Gene 26, 91-99)へとサブクローニングすると、構築体M13TG8171を与える。クローン化L1 遺伝子配列は、Genebank(届出番号(accession ) K02718号)中に含まれた配列と比較すると、いくつかの突然変異を現す:位置248でのAに代えてC、位置253でのAに代えてC、位置537でのAに代えてG、位置682でのCに代えてG、位置874でのAに代えてG、位置1393におけるトリプレットACTの挿入、位置1390におけるトリップレットGATの欠失。
イニシエーターATGの上流にBg1 II部位を創ることにより、L1 タンパク質をコードする遺伝子が修飾される。この突然変異遺伝子をBg1 II−SacI消化により先行のベクター(M13TG8185)から切除し、pTG186−ポリのBamHIとSaCI部位の間に挿入する。得られた構築体をpTG4019と呼称する。このトランスフアーベクターpTG186−ポリの詳細はフランス特許第 2,583,429号公報中に記載がある。トランスフアーされる遺伝子およびその発現を制御するプロモーターp7.5Kの挿入に対する制限部位は10箇所ある。
E6 およびE7 遺伝子は欧州特許EP第 0,462,187号の実施例2および3記載のようにCaski細胞ラインから単離する。2つの構築体は、次のクローニグ段階を容易にするように、HPV−16 E6 とE7 遺伝子とを含むクローンM13E7 /E6 から誘導された。第1の命名M13TG8188は、E7 遺伝子の上流と下流それぞれに、部位指向性突然変異誘発によりPstIおよびBamHI部位を導入することにより得られ、第2のM13TG8189はE6 遺伝子の上流でPstI部位を含んでいる。イニシエーターATGの上流および停止コドンの下流への点突然変異の導入は当業者の通常能力以内のものである。 網膜芽腫遺伝子の産物とHPV−16 E7 タンパク質との組み合わせは多くの著者により実証(例えば、Mungerら、1989, EMBO J. 8, 4099-4105 参照)され、その形質転換力と関係ずけられている。明らかな安全性の理由で、形質転換機能に関与する未変性E7 タンパク質のアミノ酸21から26をコードする配列が削除されている非発がん性突然変異体は、オリゴヌクレオチドoTG5118(SEQ ID NO:1)を用いたベクターM13TG8188の部位指向性突然変異誘発により生じる。M13TG9104が得られる。この突然変異体E7 遺伝子を以後E7 * と呼称する。
Copenhagen ワクシニアウイルスゲノム(Guillard ら、1985, J. Virol. 53, 316-318 )の左末端の5.2kb EcoRI Kフラグメントを、EcoRIで線形化したプラスミドpUC8 (Gibco BRL )中にクローン化する。得られたベクターをBg1 IIを用いる制御消化に処し、次いでK1L宿主制限遺伝子(Guillardら、1986, Proc.Natl. Acad. Sci., USA 83, 5573-5577)をコードする855bpフラグメントが削除されるように連結させる。この3.4kb Bg1 IIフラグメントをpUC8Khrと呼称する中間構築体から単離し、次いでプラスミドpUC7 (Gibco BRL)のBamHI部位中にクローン化する。pBAC1が生じ、 この中にXhoIアダプターがユニークBg1 II部位の左側に導入される。XhoIとBg1 IIによる消化後、p7.5Kワクシニアプロモーターを有するSa1 I−Bg1 IIフラグメントを挿入する。この2つのEcoRI部位はEcoRI消化に続くKlenow処理および再連結により除去される。このベクターpTG2147は、Bg1II−BamHIフラグメントの形で単離されたベクター p ポリII(Lathe ら、187, Gene 57, 193-201 )から単離される多重クローニング部位をユニークBg1II部位中に導入して得られる。したがってこのものは、制限部位が続くp7.5Kプロモーターを取り囲むK1 L遺伝子座中への挿入を可能にする組み換えアームを含んでいる。 上記E6 * およびE7 * 遺伝子はベクターM13TG9132中に含まれるワクシニアプロモーターpH5Rの下流にクローン化され、M13TG9138およびM13TG9139をそれぞれ与える。参考までに、ベクターM13TG9132はウイルスゲノムからPCRにより単離されたH5R遺伝子のプロモーターをフアージM13TG6131へ挿入することにより得られる。
ヒトインターロイキン−2をコードするcDNAをPstI消化によりプラスミドpTG36から単離し、かつプラスミドpTG186のPstI部位中に挿入して、pTG188を得る。相同的組み換えにより得られるこのウイルスをVVTG188と呼称する。
B7.1をコードするcDNAは、Daudi細胞ライン(ATCC CCL−213)から得られたmRNA調製物から、プライマーoTG6353およびoTG6352(SEQ ID NO: 3および 4)を用いる逆PCRにより単離される。この遺伝子の配列は届出番号第M27533としてGenebankに開示されている。この増幅フラグメントはM13TG6131のBg1IIとEcoRIサイトとの間にクローン化されてM13TG9149に導かれ、次いでpTG186のBamHIとEcoRIとの間にクローン化される。この構築体をpTG5090と呼称し、またこのウイルスは相同的組み換えVVTG5090により得られる。K1L遺伝子座で組に込まれたHPV−16E6 *およびE7 *遺伝子、ならびにTK遺伝子座に組み込まれたヒトB7.1遺伝子を発現するワキシニアウイルスは、ウイルスVVTG5061および同VVTG5090を用いたチキン胚の共感染により得ることができる。この組み換えウイルスをVVTG5061&5090と呼称する。
このMVAウイルスは、ワクシニアウイルスのAnkara株から導かれる。哺乳類細胞上で感染粒子を発生させることはできないが、チキン胚繊維芽細胞上では満足に発生する。これらの細胞へそれを適応すると、この種タイプの細胞上での発生と感染サイクルにとり必須でない6領域の切除が起きる(ウイルスゲノムの約15%が消失;Meyer ら、1991, J. Gen. Virol. 72, 1031-1038)。外生遺伝子材料はこれら切除帯域のいずれのレベルでも組み込める。本発明の枠組以内で、HindIII制限フラグメントNおよびAそれぞれのレベルに位置する切除IIおよびIIIが使用される(Altenburger ら、1989, Arch. Virol.105,15-27 )。
上記と同じ戦略を用いてベクターpTG6056中にIL−2遺伝子を導入する。組み換えオリゴヌクレオチドoTG10503およびoTG10502(SEQ ID NO:15および16)をクローン化してpTG6076を生産後、XhoIを用いて後者を開裂し、かつ相同的組み換えにより挿入したM13TG9185(pH5R−IL−2)からBg1II−EcoRIフラグメントを調製する。かくして得られるベクターpTG6090は、”不安定”型gusAマーカー遺伝子、HPV−16初期遺伝子の発現用カセット、ならびにヒトIL−2遺伝子カセットを含んでいる。MVAゲノムを用いた相同的組み換えにより得られる組み換えウイルスをMVATG6090と呼称する。この後期遺伝子は上記のようにpTG6018を用いて切除帯域II中に組み込める。
A. 毒性研究
静脈内、筋肉内、皮下または頭蓋内ルートでVVTG5021&5065の
107 pfuまたは野生型ワクシニアウイルスの107 pfuをヌードマウスに投与した。注射ウイルスのタイプ、および投与ルートに応じて5マウスからなる群を形成させ、ワクシニアに関連する病変を有する動物数を注射後26日で評価した。組み換えウイルスを投与したマウスは採用投与ルートに係わらずなんらの病変も示さなかった。一方、野生型ワクシニアを用いて処置した大部分の動物は病変を有し、その頻度および重大さは投与ルートに依存する。その上、静脈内および頭蓋内注射の場合は複数の死亡例が記録される。これらのデータでは、VVTG5021&5065は野生型ウイルスに較べて減衰的であり、かつこのものは頭蓋内注射後でさえも病変を起こさせないことを示す。
C57BL6 マウスをVVTG5021&5065の107 pfuを用いて皮下ルートで3回ワクチン投与した。最終免疫化の3日後、これらの動物に皮下経由で103 E7 W1 細胞を移植して抗原投与する。参考までに、このE7 W1 細胞はHPV−16発がん性E7 遺伝子を発現するベクターを用いてトランスフエクションしたネズミ(murine)リンパ腫ラインから得られる。時間の関数としての動物の生存率%を非組み換えワクシニアウイルスVVTG186(上記TG186ベクターの誘導体)の107 pfuを用いて処置した対照マウスで得られた生存率%と比較する。死亡率のモニタリングによれば、2群間に差異がみられる。対照群では、D36において100%の動物が死滅し、一方、VVTG5021&5065を用いたワクチン処置では40%の動物がD36において依然として生存し、D51では30%を超えて生存していた。

皮下経由(D0 )で移植した103 E7 W1 細胞を用いてC57BL6 マウスに接種する。次いで107 pfuの組み換えウイルスを同じく皮下ルートでD3 、さらにD6 で最終的にはD9 において投与し、動物の生存率%を、非組み換えウイルスを投与した対照動物と比較して決定した。対照動物の100%が死滅したが、一方、VVTG5061&5021およびVVTG188の混合物を用いて注射した動物の生存率は著しい増加が観測される。同様の結果が、VVTG5065&5021とVVTG188の投与後に得られる。
Claims (18)
- パピローマウイルス感染または腫瘍の治療を意図した医薬組成物であって、治療剤として、パピローマウイルスの(i)初期E6 領域からの1種のポリペプチド、(ii)初期E7 領域からの1種のポリペプチド、および(iii)インターロイキン−2、インターロイキン−7、および共付着分子B7.1およびB7.2からなる群から選択される免疫刺激活性を有する少なくとも1種のポリペプチド、を含む医薬組成物。
- パピローマウイルスの初期E6 および/またはE7 領域からのポリペプチドがパピローマウイルスのE6 および/またはE7 タンパク質の非発がん性変異体であることを特徴とする、請求項1記載の医薬組成物。
- 免疫刺激活性を有するポリペプチドが、インターロイキン−2から誘導されることを特徴とする、請求項1〜2のいずれか1項に記載の医薬組成物。
- 免疫刺激活性を有するポリペプチドが、上記分子B7.1から誘導されることを特徴とする、請求項1〜3のいずれか1項に記載の医薬組成物。
- 次の:
(1)パピローマウイルスのE6 領域からのポリペプチド、E7 領域からのポリペプチド、およびインターロイキン−2から誘導されるポリペプチド、
(2)パピローマウイルスのE6 領域からのポリペプチド、E7 領域からのポリペプチド、上記分子B7.1から誘導されるポリペプチド、または
(3)パピローマウイルスのE6 領域からのポリペプチド、E7 領域からのポリペプチド、上記分子B7.1から誘導されるポリペプチド、およびインターロイキン−2から誘導されるポリペプチド、
を含むことを特徴とする、請求項1から4のいずれか1項に記載の医薬組成物。 - パピローマウイルスが、HPV−16、HPV−18、HPV−31および/またはHPV−45の型から選択されることを特徴とする、請求項1から5のいずれか1項に記載の医薬組成物。
- パピローマウイルス感染または腫瘍の治療を意図した医薬組成物であって、治療剤として、パピローマウイルスの(i)初期E6 領域からの1種のポリペプチド、(ii)初期E7 領域からの1種のポリペプチド、および(iii)インターロイキン−2、インターロイキン−7および共付着分子B7.1およびB7.2からなる群から選択される免疫刺激活性を有する1種以上のポリペプチドをコードするDNAフラグメントが挿入される1種の組み換えベクターを含み、上記DNAフラグメントが、宿主細胞または生物中でのそれらの発現に必要な諸要素の統制下に置かれる医薬組成物。
- 上記ポリペプチドが請求項1から6に定義した特性を有することを特徴とする、請求項7記載の医薬組成物。
- 組み換えベクターが、ポックスウイルス、アデノウイルス、レトロウイルス、ヘルペスウイルスおよびアデノウイルス関連ウイルスから選択されるウイルスのゲノムから誘導されるウイルスベクターであることを特徴とする、請求項7または8記載の医薬組成物。
- 組み換えベクターが、ワクシニアウイルス、カナリヤポックスウイルスおよび禽痘ポックスウイルスからなる群から選択されるポックスウイルスから誘導されることを特徴とする、請求項9記載の医薬組成物。
- 組み換えベクターが、Copenhagen、Wyethおよび修飾Ankara(MVA)株から選択されるワクシニアウイルスから誘導されることを特徴とする、請求項10記載の医薬組成物。
- 上記ポリペプチドをコードするDNAフラグメントの発現に必須な諸要素が、チミジンキナーゼ(TK)、7.5K、H5RおよびK1L遺伝子のプロモーターから選択されたワクシニアウイルスの遺伝子のプロモーターを含むことを特徴とする、請求項10または11記載の医薬組成物。
- 組み換えベクターが、Copenhagen株のワクシニアウイルスから誘導され、かつ上記ポリペプチドをコードするDNAフラグメンが上記ワクシニアウイルスのTK遺伝子座および/またはK1L遺伝子座中に挿入されることを特徴とする、請求項11または12記載の医薬組成物。
- 組み換えベクターが、MVA株のワクシニアウイルスから誘導され、かつ上記ポリペプチドをコードするDNAフラグメントが、上記ワクシニアウイルスのI、II、III、IV、VおよびVI切除から選択された切除帯域のいずれかのレベルで挿入されることを特徴とする、請求項11または12記載の医薬組成物。
- 次の:
(1)パピローマウイルスのE6 タンパク質をコードする1種のDNAフラグメント、パピローマウイルスのE7 タンパク質をコードするDNAフラグメント、および上記分子B7.1コードするDNAフラグメント、
(2)パピローマウイルスのE6 タンパク質をコードする1種のDNAフラグメント、パピローマウイルスのE7 タンパク質をコードするDNAフラグメント、インターロイキン−2をコードするDNAフラグメント、または
(3)パピローマウイルスのE6 タンパク質をコードする1種のDNAフラグメント、パピローマウイルスのE7 タンパク質をコードするDNAフラグメント、上記分子B7.1をコードするDNAフラクメント、およびインターロイキン−2をコードするDNAフラグメント、
が挿入されるCopenhagenまたはMVA株のワクシニアウイルスから誘導される1種の組み換えベクターを含むことを特徴とする、パピローマウルス感染または腫瘍の治療を意図した、請求項7から14のいずれか1項に記載の医薬組成物。 - 組み換えベクターが生存または死滅していることを特徴とする、請求項7から15のいずれか1項に記載の医薬組成物。
- ヒトまたは動物中への注射による投与を可能にする、薬剤的に許容される担体を含むことを特徴とする、請求項1から16のいずれか1項に記載の医薬組成物。
- 子宮頚部がん、低い等級の頚部の異形成、およびパピローマウイルス感染の治療のための医薬物としての、請求項1から17のいずれか1項に記載の医薬組成物。
