本発明は、インビトロ培養代用皮膚(in vitro cultured skin substitute)、特にバリアー機能が改良されたインビトロ培養代用皮膚に関する。いくつかの態様において、バリアー機能の改良は培養条件の改良の結果であり、また別の態様において、バリアー機能の改良はケラチノサイトの遺伝子改変に起因する。本発明はさらに、この種の代用皮膚を刺激性試験に用いることにも関する。
皮膚バリアー機能の影響を予測しうる検査用の製品およびサービスには大きな市場がある。開発初期段階にある化合物を有する企業が、費用のかかる動物およびヒトでのパッチテストに頼らずにバリアー機能の試験を行えるようにする有効なアッセイ法に対しては、特別な需要が存在する。化粧品会社は、この種の試験に毎年約5000万ドル〜1億ドルを費やしている。家庭用品会社および製薬企業も同程度の費用を投じている。

本明細書で用いる「GKLF」という用語は、タンパク質または核酸に言及して用いる場合、配列番号:1および/または配列番号:2との同一性が約50%を上回る上に、シトクロムp450IAIプロモーターの基本転写因子と結合するタンパク質、またはタンパク質をコードする核酸のことを指す。結合活性は、Zhangら、J. Biol. Chem., 273(28): 17917-25 (1998)に記載されたように、オリゴヌクレオチド
を用いる電気泳動ゲルシフトアッセイ法によって首尾良くアッセイしうる。したがって、GKLFという用語は、野生型GKLFと同一なタンパク質、および野生型GKLFに由来するもの(例えば、GKLFのバリアント、またはGKLFコード領域の部分を用いて構築されたキメラ遺伝子)の両方を含む。

本発明の方法は、バリアー機能が改良された代用皮膚を作製するために用いうると考えている。一般に、重層化して扁平上皮となりうる細胞または細胞株の任意の源を本発明に用いることができる。したがって、本発明は、扁平上皮へと分化しうる細胞の何らかの特定の源の使用には限定されない。実際には、本発明は、初代ケラチノサイトおよび不死化ケラチノサイトの両方を含む、扁平上皮へと分化しうるさまざまな細胞株および源を用いることを考えている。細胞の源には、ヒトおよび死体ドナーから生検により採取したケラチノサイトおよび真皮線維芽細胞(Augerら、In Vitro Cell. Dev. Biol.−Animal 36: 96-103;米国特許第5,968,546号および第5,693,332号、これらはそれぞれ参照として本明細書に組み入れられる)、新生児包皮(Asbillら、Pharm. 17(9): 1092-97 (2000);Meanaら、Burns 24: 621-30 (1998);米国特許第4,485,096号;第6,039,760号;および第5,536,656号、これらはそれぞれ参照として本明細書に組み入れられる)、およびNM1細胞などの不死化ケラチノサイト細胞株(Baden、In Vitro Cell. Dev. Biol. 23(3): 205-213 (1987))、HaCaT細胞(Boucampら、J. cell. Boil. 106: 761-771 (1988));およびNIKS細胞(細胞株BC-1-Ep/SL;米国特許第5,989,837号、これは参照として本明細書に組み入れられる;ATCC CRL-12191)が含まれる。これらの細胞株はそれぞれ、結果として得られる皮膚同等物のバリアー機能を改良する目的で、以下に説明するように培養または遺伝的改変を行うことができる。
本発明のいくつかの態様においては、皮膚同等物を培養する方法であって、従来の方法によって培養した皮膚同等物と比べてバリアー機能の向上をもたらす方法を提供する。三次元器官培養法を用いることにより、正常ケラチノサイトの完全な重層化および組織学的分化を実現することができる(Bellら、Proc. Nat. Acad. Sci. USA, 76: 1274-1278 (1979);Fusenig, 「The Keratinocyte Handbook」中の「Epithelial mesenchymal interactions regulate keratinocyte growth and differentiation in vitro」I.M. Leigh, Lane, E.B.およびF.M. Watt編、1994, University Press: Cambridge (1994);Parenteauら、Cytotechnology, 9: 163-171 (1992))。真皮線維芽細胞を含むコラーゲンゲルの表面で増殖した正常ケラチノサイトは、重層扁平上皮の正常組織構造の特徴である基底膜およびヘミデスモソームなどの特殊化した構造を生じることができる。正常ケラチノサイトのための器官培養法のおかげで、皮膚の薬剤毒性学的試験のためのインビトロモデルが最近開発された。これは動物試験に代わる重要な選択肢となっている。
本発明は、より優れた特性を備えた皮膚同等物を作製するための、改良された器官培養法を提供する。いくつかの態様においては、真皮同等物を調製する。好ましい態様において、真皮同等物はコラーゲンおよびヒト線維芽細胞を含む。本発明は何らかの特定の種類のコラーゲンの使用には限定されない。実際には、ラット尾腱コラーゲン(I型)を非制限的に含む、さまざまな種類のコラーゲンを用いることを考えている。同様に、本発明は何らかの特定の種類の線維芽細胞には限定されない。実際には、NHDF(正常ヒト真皮線維芽細胞)細胞、およびドナーまたは患者から採取した細胞を非制限的に含む、さまざまな異なる種類の線維芽細胞を用いることを考えている。
本発明は、より優れた特性を備えた皮膚同等物を作製するための、改良された器官培養液を提供する。いくつかの態様において、本発明の培地は、ハムF12培地およびダルベッコ変法イーグル培地(DMEM)である。いくつかの態様においては、ハムF12およびDMEMに、以下の補充成分のうち少なくとも1つを加える:ウシ血清アルブミン、イソプロテレノール、カルニチン、セリン、オレイン酸、リノール酸、アラキドン酸、α-トコフェロール、アスコルビン酸および上皮増殖因子(EGF)。また別の態様においては、ウシ血清アルブミン、イソプロテレノール、カルニチン、セリン、オレイン酸、リノール酸、アラキドン酸、トコフェロール、アスコルビン酸およびEGFのうち少なくとも1つが、インビトロ培養皮膚同等物におけるバリアー機能を改良するのに十分な濃度で提供される。さらに他の態様において、培地は補充用血清、好ましくはFetal Clone IIを含む。いくつかの態様において、培地はさらに、以下の補充成分のうち少なくとも1つを含む:ヒドロコルチゾン、コレラ毒素、インスリン、アデニンおよびCaCl2。好ましい態様において、ウシ血清アルブミンは約0. 1mg/ml〜20mg/ml、好ましくは約1mg/mlの濃度で提供され、イソプロテレノールは約0.1〜10.0μMの濃度で提供され、カルニチンは約1.0〜100.0μMの濃度で提供され、セリンは約1.0〜100.0μMの濃度で提供され、オレイン酸は約1.0〜100.0μMの濃度で提供され、リノール酸は約1.0〜100.0μMの濃度で提供され、アラキドン酸は約1.0〜100.0μMの濃度で提供され、α-トコフェロールは約0.1〜10.0μMの濃度で提供され、アスコルビン酸は約0.005〜5.0mg/mlの濃度で提供され、上皮増殖因子は約0.1〜10.0ng/mlの濃度で提供される。
本発明はまた、セクションAに記した細胞において異種GKLFを発現させることによって、バリアー機能を改良しうることも考えている。異種GKLFの発現を、セクションBに記した改良された培養条件と組み合わせて用いてもよい。上皮分化の最終段階に先だって、角質層を生じさせる生化学的修飾のために必要な酵素をコードする数多くの遺伝子の発現が増大する。さらに、ケラチノサイトの分化を誘発する培養条件または皮膚バリアー機能の実験的破壊によっても、細胞外脂質合成および代謝にかかわる酵素の発現が増大する。これらの遺伝子発現の変化は、1つまたは複数の調節性転写因子が、分化中のケラチノサイトの遺伝子発現プロファイルを上皮バリアーの発生を促進するように変化させる原因であることを示している。発生過程における皮膚バリアー機能の時期が厳密であることは、発生スイッチによる明確な時間的制御が行われている可能性を示唆する。複雑な発生プログラムを1つまたは少数の重要な調節性転写因子の作用によって誘発させることが可能であり、これは時にマスター調節因子または選択遺伝子と呼ばれる。転写因子であるクルッペル様因子4(Klf4)におけるノックアウト変異に関する最近の研究により、皮膚におけるバリアー機能の重要な調節因子と考えられるものが同定されている(Segreら、Nat. Genet., 22(4): 356-60 (1999))。
本発明の代用皮膚にはさまざまな用途があると考えられる。これらの用途には、化合物(例えば、刺激物)のスクリーニングのための使用、腫瘍および病原体(例えば、ヒトパピローマウイルス)を培養するための基質、ならびに創傷閉鎖および熱傷治療のための使用が非制限的に含まれる。これらの用途について、以下にさらに詳細に説明する。
本発明の皮膚同等物は、さまざまなインビトロ試験に用いうる。特に、本皮膚同等物は以下の評価のために有用である:スキンケア用製品、薬物代謝、試験化合物に対する細胞応答、創傷治癒、光毒性、皮膚刺激、皮膚炎症、皮膚腐食性および細胞障害。本皮膚同等物は、6ウェル、24ウェルおよび96ウェルのプレートを非制限的に含む、試験用のさまざまな形式として提供される。さらに、本皮膚同等物を標準的な切離法によって分割した後に検査することもできる。本発明の皮膚同等物は、分化した角質層を有する上皮層、および真皮線維芽細胞を含む真皮層の両方を含んでいる。上記の通り、特に好ましい態様において、上皮層は不死化NIKS細胞に由来する。NIKS細胞を含む他の好ましい細胞株は、i)不死化していること;ii)腫瘍形成性がないこと;iii)分化を誘導すると角化膜を形成すること;iv)器官培養下で正常な扁平分化を生じること;およびv)細胞種特異的な増殖基準を液内培養下で保つことを特徴とし、この際、前記の細胞種特異的な増殖基準には、1)標準的なケラチノサイト増殖培地中でマイトマイシンCで処理した3T3フィーダー細胞の存在下で培養した際に、正常ヒトケラチノサイトの形態的特徴を示すこと;2)連続培養のためには上皮増殖因子に依存すること;および3)トランスフォーミング増殖因子1による増殖阻害、が含まれる。
いくつかの態様においては、本発明の皮膚同等物を、化合物を皮膚刺激活性に関して検査するためのレポーター遺伝子アッセイに用いる。また別の態様において、本発明は、既知の皮膚刺激物に対する応答性がある新規遺伝子を同定するために皮膚同等物を用いる方法を提供する。本発明の方法を用いて同定された新規遺伝子は、さらに、皮膚刺激物の候補のスクリーニング用のほかのレポーター遺伝子構築物を作製するのに有用である。
いくつかの態様において、本発明は、皮膚刺激物に応答するレポーター遺伝子構築物を含む、皮膚同等物(例えば、NIKS)細胞を提供する。適したレポーター遺伝子構築物には、上記のものが非制限的に含まれる。いくつかの態様において、本細胞はさらに、バリアー機能が改良されたものである。
他の態様において、本発明は、新規な刺激物応答性遺伝子を同定するための方法および組成物を提供する。いくつかの態様においては、一般集団における刺激物曝露とよく相関するデータを得るために、複数のプールドナーからの初代ヒトケラチノサイトの器官培養物を既知の皮膚刺激物に曝露させたものにおける全体的な遺伝子変化をアッセイする。本方法によって同定された新規遺伝子は、皮膚同等物(例えば、NIKS)由来の遺伝子レポーター細胞株を用いる刺激性試験アッセイ法、および刺激物予測用遺伝子発現アレイに用いることができる。
本発明の皮膚同等物は、皮膚に天然に生じる腫瘍の培養および試験のため、ならびに皮膚に影響を及ぼす病原体の培養および試験のためにも有用である。したがって、いくつかの態様においては、本発明の皮膚同等物に悪性細胞を播種することを考えている。非制限的な例としては、ヒト扁平上皮癌のモデルを得るために、皮膚同等物に対して、米国特許第5,989,837号(これは参照として本明細書に組み入れられる)に記載された悪性SCC13y細胞を播種する。続いて、これらの播種された皮膚同等物を用いて、化合物または他の治療戦略(例えば、放射線療法またはトモセラピー(tomotherapy))を、自然環境にある腫瘍に対する有効性に関してスクリーニングすることができる。すなわち、本発明のいくつかの態様は、悪性細胞または腫瘍、および少なくとも1つの試験化合物を含む皮膚同等物を提供すること、皮膚同等物を化合物で処置すること、ならびに悪性細胞または腫瘍に対する処置の効果をアッセイすることを含む方法を提供する。本発明のまた別の態様においては、悪性細胞または腫瘍、および少なくとも1つの被験治療法(例えば、放射線療法または光線療法)を含む皮膚同等物を提供すること、皮膚同等物を治療法によって処置すること、ならびに悪性細胞または腫瘍に対する治療法の効果をアッセイすることを含む方法を提供する。
本発明の皮膚同等物は、創傷閉鎖および熱傷治療の用途に有用である。熱傷治療および創傷閉鎖のための自己移植片および同種移植片の使用は、Myersら、A. J. Surg. 170(1): 75-83 (1995)および米国特許第5,693,332号;第5,658,331号;および第6,039,760号に記載されており、これらはそれぞれ参照として本明細書に組み入れられる。いくつかの態様において、皮膚同等物をDERMAGRAFTなどの代用真皮とともに用いてもよい。また別の態様においては、皮膚同等物を、標準的なケラチノサイト源(例えば、NIKS細胞)および移植片を移植される予定の患者からのケラチノサイトの両方から作製する。このため、皮膚同等物は2つの異なる異なる源に由来するケラチノサイトを含む。さらに別の態様において、皮膚同等物はヒト単離組織に由来するケラチノサイトを含む。したがって、本発明は、熱傷によって生じた創傷を含む、創傷閉鎖のための方法であって、本発明によるバリアー機能が改良された皮膚同等物および創傷に罹患した患者を提供すること、ならびに創傷が閉鎖される条件下で患者を皮膚同等物によって治療することを含む。
さらに別の態様においては、対象(subject)に対する治療薬を得るために皮膚同等物を操作する。本発明は何らかの特定の治療薬の送達には限定されない。実際には、酵素、ペプチド、ペプチドホルモン、他のタンパク質、リボソームRNA、リボザイムおよびアンチセンスRNAを非制限的に含む、さまざまな治療薬を対象に送達しうると考えている。これらの治療薬は、遺伝的障害の是正を非制限的に含む、さまざまな目的で送達することができる。いくつかの特に好ましい態様においては、治療薬を、移植片が野生型組織として役立つような遺伝性先天性代謝異常(例えば、アミノ酸尿)の患者を解毒する目的で送達する。治療薬の送達によって障害が是正されると考えられる。いくつかの態様においては、皮膚同等物の形成に用いるケラチノサイトに、治療薬(例えば、インスリン、凝固因子IX、エリスロポエチンなど)をコードするDNA構築物をトランスフェクトし、皮膚同等物を対象に移植する。続いて、治療薬または移植片由来の他の組織を患者の血流内に送達させる。好ましい態様においては、治療薬をコードする核酸を適したプロモーターと機能的に結合させる。本発明は何らかの特定のプロモーターの使用には限定されない。実際には、誘導性、構成性、組織特異的およびケラチノサイト特異的なプロモーターを非制限的に含む、さまざまなプロモーターを用いることを考えている。いくつかの態様においては、治療薬をコードする核酸をケラチノサイト内に直接導入する(すなわち、リン酸カルシウム共沈、またはリポソームを介したトランスフェクションによる)。また別の好ましい態様においては、治療薬をコードする核酸をベクターとして用意し、このベクターを当技術分野で知られた方法によってケラチノサイト内に導入する。いくつかの態様において、ベクターはプラスミドなどのエピソーム性ベクターである。また別の態様において、ベクターはケラチノサイトのゲノム中に組み込まれる。組込みベクターの例には、レトロウイルスベクター、アデノ随伴ウイルスベクターおよびトランスポゾンベクターが非制限的に含まれる。
以下の実施例は、本発明のいくつかの好ましい態様および面を示すとともに、さらに例示するために提供するものであり、その範囲を制限するものとみなされるべきではない。
NIKS細胞の上皮バリアー機能に対する培養条件の効果
さまざまな培養条件によってバリアー機能の種々の面が向上することが示されているが(表1参照、前記)、これらの条件の相乗効果を検討した体系的な取り組みはこれまでなかった。本実施例では、アスコルビン酸、PPAR活性化物質(リノール酸)、FXAR活性化物質(ファルネソール)および相対湿度の低下が、NIKS器官培養物の上皮バリアー機能に及ぼす複合的効果の評価について提示する。処理した培養物のバリアー特性の評価は、表面静電容量(SEC)の測定(Boyceら、J. Invest. Dermatol., 107(1):P. 82-7 (1996))、細胞外脂質組成の分析、および電子顕微鏡による組織切片の超微細構造の観察によって行う。
NIKS細胞における外因性Klf4の発現
本実施例では、NIKS細胞における外因性Klf4の発現について述べる。転写因子クルッペル様因子4(Klf4)は、終末分化の過程にある上皮、特に皮膚および腸管上皮において高レベルで発現されるジンクフィンガータンパク質である。皮膚では、これは分裂活性のない表皮の基底上層に豊富に存在する。Klf4は、NIH 3T3細胞cDNAライブラリーにあるジンクフィンガードメインに対するプローブを用いた低ストリンジェンシーハイブリダイゼーションによって同定された(Shieldsら、J. Biol. Chem, 271(33): 20009-17 (1996))。その3つのC2H2ジンクフィンガーのためにこれはジンクフィンガー転写因子のファミリーと結びつけられており、このファミリーには組織特異的分化のために重要な因子であるEKLFおよびLKLFも含まれる。これは増殖停止細胞で最も高いレベルで発現され、増殖中の細胞におけるレベルは検出不能である。Klf4の構成性発現によってCOS-1細胞のDNA合成は阻害される。これは、シトクロムP450遺伝子CYP1A1の調節に重要な特定のDNA配列と結合する(Zhangら、J. Biol. Chem., 273(28): 17917-25 (1998))。Klf4がCYP1A1内の結合部位と結合するとCYP1A1の発現が阻害されるが、これはDNA結合に関するSP1との競合およびSP1との直接的なタンパク質間相互作用によると考えられている。最近の研究では、Klf4が自らの発現も調節すること、および重要な結合相互作用がp300/CBPとのものであることが報告されている(Geimanら、Nucleic Acids Res., 28(5): 1106-1113 (2000);Mahatanら、Nucleic Acids Res., 27(23): 4562-9 (1999))。他の重要な転写因子もそうであるように、Klf4は遺伝子によっては強力な活性化因子であるが、遺伝子によってはリプレッサーとして作用しうる。
皮膚刺激物に応答したIL-1αの分泌
本実施例では、NIKS細胞が皮膚刺激物に応答してIL-1αを分泌することを示す。NIKS細胞の器官培養物が初代ケラチノサイトに典型的な刺激物応答性を示すか否かを明らかにするために、NIKS培養皮膚同等物を、インビボおよびインビトロで皮膚刺激物応答を誘発することが知られた濃度のSDSに曝露させた。空気曝露させたNIKS器官培養物にSDS溶液(0.1%、0.2%および0.5%)を局所適用し、37℃で24時間インキュベートした。24時間後に、培養物の下方の培地を、Quantikine IL-1α ELISAキット(R&D Systems, Minneapolis, MN)を用いてIL-1αタンパク質に関してアッセイし、細胞生存度を決定するために培養物をMTS試薬とともにインキュベートした(Promega Corp., Madison, WI)。対照およびSDS処理した培養物からの培地におけるIL-1αの濃度を図3に示した。非処理培養物からの培地は25pg/mlのIL-1αを含んでおり、一方、0.1%、0.2%または0.5%SDSで処理した培養物におけるIL-1αの濃度はそれぞれ290、375および275pg/mlであった。
レポーター遺伝子構築物の作製
本実施例には、IL-8プロモーター断片を含むレポーター遺伝子構築物を含むNIKS細胞を作製するための方法を述べる。既知の配列に対するプライマーを用いるPCRにより、IL-8プロモーターおよび隣接配列を含む1.5kbのDNA断片を増幅する。増幅されたDNAを配列解析により確認した後に、このDNA断片を、SEAPのコード領域は含むもののその発現を導くための調節因子を欠いている発現ベクターpSEAP(Tropix, Bedford, MA)中にクローニングする。レポーター構築物の完全性は制限分析およびDNA配列の決定によって確認する。
刺激物応答性遺伝子の同定
本実施例では、刺激物曝露によって発現が誘導される新規遺伝子の同定のために有用な実験について述べる。Genome Systems cDNAマイクロアレイ(GEMマイクロアレイ;Incyte Genomics, Palo Alto, California)を遺伝子発現の分析に用いる。操作はすべてIncyte Genomics社が行う。Incyte社が配列を確認したcDNAクローンから作製された、8514種の一意的なヒトcDNAを含むUniGEM V 2.0マイクロアレイを用いる。より大規模なアレイも、それらが入手可能になれば用いる。成人プールドナー由来の培養ヒトケラチノサイト(Clonetics, Walkersville, MDから入手可能)からインビトロ培養皮膚同等物を調製する。収縮したコラーゲンマトリックス中に真皮線維芽細胞が包埋されたものからなる真皮同等物上に培養ケラチノサイトを播種し、その結果生じた器官培養物を気相界面に挙上して、ケラチノサイト層の分化および重層化を誘導する。14日後に器官培養物に対して、既知の刺激物(例えば、0.2%SDS、0.005%カルシポトリオール、0.025%trans-レチノイン酸)または非刺激物対照(例えば、ミネラルオイル、ワセリン)による局所処理を24時間行う。これらの化合物はいずれも、刺激物で処理した皮膚同等物において炎症誘発性サイトカインIL-1αおよびIL-8の発現および分泌を誘導する(de Brugerolle de Fraissinetteら、前記)。
皮膚同等物の脂質含有量
本実施例では、最適な無血清培地を用いる皮膚同等物の調製、および準最適培地を用いて調製した第2のセットの皮膚同等物について述べる。続いて、その結果得られた培養物の脂質含有量を決定した。
NIKS細胞におけるGKLFの発現
本実施例では、マウスにおけるバリアー機能の発生を媒介すると考えられているタンパク質であるGKLFのNIKS細胞における発現について述べる。
培養方法
本実施例には、以下の実施例に共通する培養方法を述べる。
器官培養の工程には、以下の6種の異なる培地を用いる:3T3フィーダー細胞培地(TM);線維芽細胞増殖培地(FM);NIKS培地(NM);プレーティング培地(PM);重層化培地A(SMA);および重層化培地B(SMB)。TMは、単層培養においてNIKS細胞のフィーダー細胞として作用する3T3細胞を増殖させるために用いられる。TMは、10%仔ウシ血清(Hyclone)を加えたダルベッコ変法イーグル培地(DMB、GibcoBRL)の混合物である。FMはハムF-12培地(GibcoBRL)および10%Fetal Clone II(Hyclone)血清の混合物である。NMはNIKSケラチノサイトを増殖させるために用いられる。NMはハムF-12培地(GibcoBRL)およびDMBの3:1混合物に、2.5%Fetal Clone II(Hyclone)、0.4μg/mlヒドロコルチゾン(Calbiochem)、8.4ng/mlコレラ毒素(ICN)、5μg/mlインスリン(GibcoBRL)、24μg/mlアデニン(Sigma)および10ng/ml上皮増殖因子(EGF、R&D systems)を加えたものである。PMは、NIKS細胞を真皮同等物上に播種する際に用いる培地である。PMは、EGFが存在せず、CaCl2(Sigma)を最終カルシウム濃度1.88mMとなるように添加し、Fetal Clone II血清を0.2%のみ添加する点を除き、NMと同じである。SMAは、PMに1mg/mlウシ血清アルブミン(BSA)、1μMイソプロテレノール、10μMカルニチン、10μMセリン、25μMオレイン酸、15μMリノール酸、7μMアラキドン酸、1μM α-トコフェロール、0.05mg/mlアスコルビン酸(すべてSigma社)および1ng/ml EGFを添加下ものである。SMBは、STRATATEST皮膚同等物の上皮重層化段階およびSTRATAGRAFT皮膚同等物の増殖時に用いられる。SMBは、Fetal Clone II血清補充成分が存在することを除き、SMAと同じである。
STRATATEST皮膚同等物またはSTRATAGRAFT皮膚同等物の器官培養物を開始する前に、3T3フィーダー細胞を調製し、続いてそのまま用いるか、後に用いるために凍結する。3T3細胞を集密に達するまで増殖させ、マイトマイシン-C(TM中に4ug/mlのマイトマイシン-C、Roche)で2時間処理する。続いて細胞を洗浄し、再懸濁した上で、NIKSの増殖を補助する目的で、1.25×106個/100mm組織培養皿の密度でプレーティングする。凍結したフィーダーを用いる場合には、2.5×106個の入った1mlを含む1本の凍結アンプルを解凍し、新鮮なTMで希釈した上で、1つまたは複数の100mm組織培養皿にプレーティングする。これは、NIKS細胞をプレーティングする1日前にNIKS細胞の増殖に必要と考えられるだけの数の培養皿に対して行う。
凍結NHDF細胞を解凍してプレーティングを行う。細胞には翌日に、残留性の凍結保護剤を除去し、その後の細胞増殖を維持させるためにFMを与える。集密に達する前のNHDF細胞を、真皮同等物に用いるために採集する。真皮同等物を調製するには、ラット尾腱コラーゲン(I型、Becton Dickinson)をまず0.03N酢酸中に3mg/mlに希釈して、氷上で冷却する。濃縮ハムF12培地の混合物(通常の8.7倍の強度、HEPESでpH 7.5に緩衝)をFetal Clone IIと混合する。この2つの溶液は溶液の最終容積のそれぞれ11.5および10%である。1N NaOHを培地混合物に添加する(最終溶液の2.5%)。続いて、希釈コラーゲン(74%)を混合物に添加する。2%容積の懸濁線維芽細胞(STRATATEST真皮同等物の場合は2.5×106個/ml、STRATAGRAFT真皮同等物の場合は1×106個)を混合物に添加する。この溶液をゆっくりと十分に混合する。100μlずつに分割して組織培養インサート(Millipore Corp.のMILLICELL)に入れるが、STRATATESTの場合は100mm組織培養皿に25個が入る。STRATAGRAFT皮膚同等物にはCorning社のTRANSWELLインサートを用いる。13mlの真皮同等物を各インサートに注ぎ入れ、厚さがSTRATATEST真皮同等物の概ね3倍になるようにする。ゲル形成のために30分間おいた後、STRATATEST真皮同等物を含む培養皿に20mlのFMを満たす。各STRATATEST真皮同等物の表面に1滴または2滴のFMを滴下する。STRATAGRAFT真皮同等物については、80mlのFMを150mm組織培養皿内のTRANSWELLインサートの周囲に注ぎ、10mlを真皮同等物の上に載せる。インサートは使用時まで37℃、5%CO2、相対湿度90%のインキュベーター内に保つ。真皮同等物にNIKS細胞を播種する1日前に、それらを、組織培養インサートの底面から培地を供給するための2つの吸い上げ用パッドを備えた滅菌ステンレス製メッシュ(S&S Biopath)の上に載せることによって気相界面に挙上する。
NIKSのプレーティングの1日前に、フィーダーを凍結しないまま調製するか解凍した上でTM中にプレーティングする。NIKS細胞をフィーダーの上に約3×105個/100mm培養皿の密度でプレーティングする。NIKS細胞を新たに解凍する場合には、残留性の凍結保護剤を除去するためにプレーティングの1日後に新鮮なNMを与える。NIKS細胞には必要に応じて増殖を維持するためのNMを与える。細胞が集密に達した時点で、NIKS細胞を採集し、算定した上でPM中に再懸濁する。4.65×105個のNIKS細胞/cm2を、気相界面に1日間挙上しておいたMIILLICELLまたはTRANSWELL真皮同等物の表面に播く。培養皿に金属製リフトの下方までPMを満たし、再びインキュベーターに戻す。2日後に培養物にSMAを与える。さらに2日後に培養物にSMBを与えて、湿度75%のインキュベーターに移し、成熟するまでSMBを追加しながらそこに保つ。
本実施例では、1mg/mlコラーゲンを用いる真皮同等物の調製について述べる。簡潔に述べると、10倍濃度に調製したハムF12培地24mlを、50mlコニカルチューブ内で、4.8mlの滅菌水、2.4mlペニシリン/ストレプトマイシン混合物および24mlのFetal Clone IIと混合した。4.11mg/mlのラット尾腱I型コラーゲン(1.46ml)を1.882mlの滅菌水および2.658mlの0.05%酢酸で希釈した。正常ヒト真皮線維芽細胞を培養物から採集し、106個および104個/mlの細胞密度で再懸濁した。0.815mlの培地を含む混合物を、2.619mlの希釈コラーゲンおよび104個/mlの線維芽細胞34μlと合わせた。この混合物を116.5μlずつ組織培養インサート(ペトリ皿1枚につき25個)に入れ、37℃で15分おいてゲル化させた。0.815mlの培地を含む別の混合物を、2.619mlの希釈コラーゲンおよび106個/mlの線維芽細胞137μlと合わせた。 この混合物の116.5μlずつを組織培養インサート内の前記のゲルの上に載せ、30分おいてゲル化させた。
本実施例では、3mg/mlコラーゲンを用いる真皮同等物の調製について述べる。10倍濃度に調製したハムF12培地4.785mlを、50mlコニカルチューブ内で、0.946mlの滅菌水、0.473mlのペニシリン/ストレプトマイシン混合液および4.785mlのFetal Clone IIと混合した。この培地混合物の4.6mlを0.242mlの滅菌水および0.289mlの1N NaOHと混合した。0.92mlの混合物を、3.11mg/mlのラット尾腱I型コラーゲン3mlと混合した。これに対して、106個/mlのヒト真皮線維芽細胞懸濁液186μlを添加した。この混合物の100μlをMILLICELLインサート(直径1cm)に入れ、30分おいてゲル化させた。続いてペトリ皿に20mlのFM培地を満たしてインキュベートした。5日後にFMを除去し、真皮同等物の表面から液体を吸引した。標準的な手順を用いてNIKS(商標)細胞を採集し、プレーティング培地(PM)中に2.345×106個/mlの密度で再懸濁した。この懸濁液150μlを各真皮の表面に加えて2時間インキュベートした。続いて、播種された真皮同等物に20mlのPMを与えた。さらに2日後に培地をペトリ皿(培養物の表面を含む)から除去し、培養物を気相界面に挙上して、約30mlの重層化培地を2日毎に与えた。播種14日後に培養物を分析した。培養増殖の終了時点で、すべての培養物において真皮同等物が上皮で完全に覆われており、その外観は平滑で乾燥していた。このため、これらは商業的用途に非常によく適合していた。
本実施例では、ケラチノサイトを播種する前に真皮同等物をあらかじめ挙上することの有益な効果を示す。10倍濃度に調製した1.31mlのハムF12培地を、50mlコニカルチューブ内で、0.328mlの滅菌水、0.148mlのペニシリン/ストレプトマイシン混合液および1.472mlのFetal Clone IIと混合し、1.63ml(ほぼ半分)を2のチューブに分け入れた。2.92mlのラット尾腱コラーゲン(4.11mg/ml)3.764mlの滅菌水および5.316mlの0.05%酢酸と混合し、0.05%酢酸中にある1mg/mlコラーゲンを得た。5.24mlの希釈コラーゲンを1.63mlの培地混合物に加えた。標準的なプロトコールにより採集した細胞密度104個/mlヒト真皮線維芽細胞74μlを添加し、ゆっくりと混合した。この混合物の116.5μlずつを組織培養インサートに入れ(ペトリ皿1枚につき25個)、37℃で15分おいてゲル化させた。別のコラーゲン5.24mlを、106個/mlの線維芽細胞274μlとともに第2の培地混合物1.63mlに添加した。116.5μlを第1のゲル化したコラーゲン層の上の各インサートに添加した。これを37℃で30分おいてゲル化させた。続いて、真皮同等物が培地中に浸漬された状態で成熟しうるように、ペトリ皿に20mlのFMを満たした。4日後に培地をペトリ皿から除去し(培養物の表面からも含む)、培養物を気相界面まで挙上した上で約30mlのFMを与えた。培養物をこの状態でインキュベーター内に一晩おいた。続いてそれらに、標準的なプロトコールを用いて単層培養物から採集した細胞密度2.345×106個/mlのNIKSケラチノサイト150μlを播種した。播種の後に培養物にPMを与えて、インキュベーターに戻した。2日後に培養物にSMAを与え、その後は2日毎に培養物にSMBを合計8回与えた。培養増殖の終了時点で、すべての培養物において真皮同等物が上皮で完全に覆われており、その外観は平滑で乾燥していた。組織検査により、あらかじめ挙上した試料の真皮および表皮の厚さはほぼ均等であり、表皮にはすべて重層化した層が存在することが判明した。
本実施例では、あらかじめ挙上することが真皮同等物の寿命に及ぼす影響について述べる。培養物を、浸漬させなかった点を除き、上に挙げた奏功した実験とまったく同じように調製した。ゲルに気相界面まで挙上させたMILLICELLインサートを加え、その後のすべての播種および培地供給を培養物を挙上した状態で行った。培養増殖の終了時点で、培養物10個中1個では真皮同等物が表皮で完全に覆われていた。この影響は一部には、ほとんどの試料では真皮同等物が壁面から離されていたことから、真皮同等物のMILLICELLインサートの底面に対する付着が乏しかったためであると考えられる。組織検査により、真皮および上皮層の厚さに大きなばらつきがあることが示された。同様に、上皮の重層化も、十分に分化したものから単層を覆うに過ぎないものまでさまざまであり、商業的用途には不適合であった。
本実施例では、器官培養下でのケラチノサイトの完全な重層化を支援するとともに、バリアー機能が改良された皮膚同等物をも生じさせる、無血清培地の最適化について述べる。
Claims (5)
- 10秒間隔の読み取り値の差異として測定された表面静電容量が40〜240pFである移植前ヒト皮膚同等物を含む組成物であり、前記皮膚同等物が二倍体に近い不死化ケラチノサイト細胞を含む組成物。
- 移植前ヒト皮膚同等物の、10秒間隔の読み取り値の差異として測定された表面静電容量が80〜120pFである、請求項1記載の組成物。
- 移植前皮膚同等物におけるセラミド5、6および7の総合含有量がセラミド総含有量の20〜50%である、請求項1記載の組成物。
- 移植前皮膚同等物におけるセラミド2の含有量がセラミド総含有量の10〜40%である、請求項1記載の組成物。
- 2種の異なる源に由来するケラチノサイトをさらに含む、請求項1記載の組成物。
