Description
本発明は、2001年8月2日出願の米国特許仮出願60/309,180(そのすべてを出典明示により本明細書の一部とする)の利益を享受する。
本発明は、アシル部分が脂肪族ホモポリマーまたはコポリマー・ポリエステルから生じる生分解性多価アルコールエステルに関する。
最近、星状ポリマーすなわち中心コアに結合する3以上の高分子アームを有する一種の分枝ポリマーに関心が増加している。この種のポリマーは今まで生物医学的な利用がなされていない。
生物医学的な利用にとりわけ有用な生分解性星状ポリマーを、多価アルコールをエステル化して、脂肪族ホモポリマーまたはコポリマー・ポリエステルから生じるアシル部分を得ることに基づき、製造できることを見出した。
重量平均分子量を、ポリスチレン標準を用いるゲル浸透クロマトグラフィーで測定する。
用語「生分解性」は、人体および生物体(例えば、バクテリア)の正常な官能化および/または水環境において、トリプシンやリパーゼなどの酵素またはリソソームにより分解され得ることを意味する。
エステルのアシル部分が脂肪族ホモポリマーまたはコポリマー・ポリエステルから生じ、遊離ヒドロキシルをその末端に含有する生分解性多価アルコールエステルであって、その平均分子量が1,000〜80,000、例えば2,000〜50,000である、第1態様の化合物について説明する。
2‐カルボキシエテニル官能化多価アルコールエステルと多糖の反応産物とを溶液中で光架橋して、生分解性ポリエステル−多糖ヒドロゲルを得る。光架橋のためのホトイニシエーターおよび照射手段は、光架橋についての上記のものと同じである。得られるヒドロゲルは生分解性である(生分解性ヒドロゲルは、水によりおよび/または天然に存する酵素により分解されるポリマーを架橋することで形成されるヒドロゲルである)。
アミノキシル含有ラジカルを含める反応は、カルボキシルのヒドロキシルを、2,2,6,6−テトラメチルピペリジン−1−オキシの4位に結合するイミノで、2,2,5,5−テトラメチルピペリジン−1−オキシの3位に結合するイミノで、または2,2,5,5−テトラメチルピペリジン−1−オキシ−3−カルボニルのカルボニルに結合するイミノで置き換えるのに適するスピン標識の反応により実施する。適当なスピン標識は米国特許5,516,881に記載されている。用語「アミノキシル」は、構造>N−O-の意味で使用する。用語「アミノキシル含有ラジカル」は、構造>N−O-を含有するラジカルの意味で使用する。産物は、腫瘍のアミノキシルラジカル処置、腫瘍の化学療法およびイオン化照射療法についての効力増加、損傷、病変または老化のヒト血管の再構築、自然形成の酸化窒素過剰の制御、中和、減少の手段として有用である。
本発明を下記の実施例でもって説明する。
上記で製造のPGCL−OH−1(0.01モル)およびPGCL−OH−1のヒドロキシルに対する5等量のマレイン酸無水物を三首フラスコにN2気下で入れ、130℃で24時間保持した。ついで、反応混合物を室温に冷やし、クロロホルムに溶解した。得られたクロロホルム溶液を過剰の石油エーテルに注ぎ、産物を沈澱せしめた。沈澱を蒸留水500ml中で4時間攪拌し過剰のマレイン酸無水物を除去した。ろ過後、沈澱を蒸留水で4回洗い、P2O5で減圧室温で一定の重量になるまで乾燥した。産物をPGCL−Ma−1と呼ぶ。
PGCL−Ma−1(1g)をジオキサン(5%wt/vol)に溶解した。ホトイニシエーター、2,2'−ジメトキシ−2−フェニルアセトフェノン(PGCL−Ma−1の4重量%)を溶液に加え、数分間激しく攪拌しながら溶解した。溶液をガラスプレート上に注ぎ、365nm長波UVランプ(Model UVL-18, 8 watt handheld, UVP, Upland, CA, USA) で5時間刺激した。得られた架橋フィルムを減圧室温で乾燥した。産物は乳白色フィルムであり、NPGCL−Ma−1と呼ぶ。
上記で製造のPPCL−OH−1(0.01モル)およびPPCL−OH−1のヒドロキシルに対する5等量のマレイン酸無水物を三首フラスコにN2気下で入れ、熔融物を24時間持続的に攪拌した。24時間後、反応混合物を室温に冷やし、クロロホルムに溶解した。クロロホルム溶液を6倍量の石油エーテルに注ぎ、産物を沈澱せしめた。沈澱を蒸留水500ml中で4時間攪拌し、過剰のマレイン酸無水物を除去した。ろ過後、蒸留水で4回洗い、P2O5で減圧室温で一定の重量になるまで乾燥した。産物をPPCL−Ma−1と呼ぶ。
他の事例において、PGCL−Ma−3を等モル量のPPCL−Ma−3に代える以外は、同じ操作である。
他の事例において、PGCL−Ma−3を等モル量のPPCL−Ma−3に代える以外は、同じ操作である。
他の事例において、上記の調製のPGCL−Ma−3を等モル量のPPCL−Ma−3に代える。
上記の多くの変形は当業者に明らかである。従って、本発明は請求項により定義つけられる。
Claims (4)
- 生分解性ポリエステル−多糖ヒドロゲルであって、多糖のヒドロキシと以下に示す生分解性多価アルコールエステルの2−カルボキシエテニル基との反応により形成された不飽和反応産物の光架橋により形成されるヒドロゲル:
生分解性多価アルコールエステルのアシル部分が脂肪族ホモポリマーまたはコポリマー・ポリエステルから生じ、アシル部分の末端に遊離のヒドロキシルを含有する生分解性多価アルコールエステルから得られ、該生分解性多価アルコールエステルの末端の遊離のヒドロキシルのいくつかまたはすべてが、2−カルボキシエテニル基に変換されており、平均分子量が1,000〜80,000である、末端に2−カルボキシエテニル基を有する生分解性多価アルコールエステル。 - 多糖がデキストランである、請求項1の生分解性ポリエステル−多糖ヒドロゲル。
- 配送すべき薬物または他の生物活性物質を捕捉しているか、またはそれと共有結合している請求項2の生分解性ヒドロゲルを含む薬物配送システム用組成物。
- 請求項3の薬物配送システム用組成物を含む被覆を含有する血管ステント。
