Description
以下の手順により、図1A、図1Bに示す構造のセンサー素子を作製した。
1. まず、基板1としてBK7ガラスを用い、BK7ガラス上に厚さ1nmのチタン膜、その上に厚さ45nmの金膜からなる薄膜2を基板中心部にスパッタ法で形成した。チタンは金薄膜の基板への接着性を向上させるために使用したものである。これにより、センサー素子の領域Aに金薄膜が形成されたことになる。
2. アセトンを主成分とし、アルコール類を含む混合溶媒に混合ニトロセルロース(アドバンテック社製)を溶解した溶液を用いスピンコート法で、金薄膜つきガラス基板の全面に、中心部の厚さが4μmの多孔質膜からなるサンプル親和性膜を形成した。
3. 金薄膜が存在し、SPR検出が行える領域Aを含むように、サンプル親和性膜に抗ヒトIgG(Sigma社製)をスポット法で固定した。さらに、スーパーブロックPBS(ピアス社製)を用いて領域Aの部分にブロッキングを施した。
4. 図1Aに示すように、抗体を固定した部分と重ならないように、サンプルパッド(ミリポア社製 AP22素材)をサンプル親和性膜に固定した。
5. 図1Aに示すように、吸収パッド(ミリポア社製 AP22素材)を領域Cに設置した。
実施例1と同様の作製手順で図4A、図4Bに示すようなセンサー素子を作製した。この素子ではサンプル親和性膜の一部を図5Aに示すように除去し、サンプル親和性膜の、サンプルが流れる領域を制限した。この狭くなった2箇所に2種類の抗体、抗ヒトIgG(Sigma社製)と抗ヤギIgG(Sigma社製)をそれぞれ固定化した。
Claims (8)
- 透明基板と、該透明基板上の少なくとも一部に形成された金薄膜と、その少なくとも一部の上に金薄膜が形成された透明基板上に設けられたサンプル親和性薄膜とを有し、前記金薄膜と前記サンプル親和性薄膜がその上に設けられた透明基板は測定領域(領域A)と、該領域Aを間に介するように設けられた試料導入部(領域B)と試料吸収部(領域C)を有し、前記領域A,B,Cのいずれにもそれらの少なくとも一部表面にサンプル親和性薄膜が設けられており、前記領域Aのサンプル親和性薄膜には前記金薄膜に接する面に分子認識膜を含有するサンプル測定部が設けられており、領域Bのサンプル親和性膜上には導入ポートが形成され、領域Cのサンプル親和性膜上には該領域C全体を覆うように吸収パッドが設けられ、サンプル親和性薄膜上で、領域B内の点と領域A内の点と領域C内の点が1つの直線にのるように存在することを特徴とする表面プラズモン共鳴(SPR)効果を利用するSPR測定用センサー素子。
- 前記領域Aに複数のサンプル測定部が設けられていることを特徴とする請求項1記載のSPR測定用センサー素子。
- 前記分子認識膜が、複合体を形成する分子対の一方の分子を固定してなるものであることを特徴とする請求項1または2記載のSPR測定用センサー素子。
- 前記複合体を形成する分子対の一方の分子が抗体であることを特徴とする請求項3記載のSPR測定用センサー素子。
- 前記領域Aのサンプル親和性薄膜の表面がサンプルに対して非親和性の膜で覆われていることを特徴とする請求項1から4のいずれか1項に記載のSPR測定用センサー素子。
- 前記測定領域Aが、サンプル非親和性膜で分割される、又は、サンプル親和性膜が取り除かれることにより分割されることを特徴とする請求項1に記載のSPR測定用センサー素子。
- 前記測定領域Aにおいて、前記領域Aに接続された前記領域B又は前記領域Cよりも、分割された前記領域Aが狭いことを特徴とする請求項1に記載のSPR測定用センサー素子。
- 請求項1から7記載のSPR測定用センサー素子であって、サンプル親和性膜の厚さが最大で10μmであることを特徴とするSPR測定用センサー素子。
