Description

アムロジピン、ブフロメジル、アミルナイトライト、ジルチアゼム、ジピリダモール、ニトログリセリン、イソソルビドジナイトレート、リドフラジン、モルシドミン、ニカルジピン、ニフェジピン、オキシペンチフィリンおよびペンタエリスリトールテトラナイトレートのような血管拡張剤;
それらのN−オキシド、それらの薬学的に許容される酸または塩基付加塩およびそれらの立体化学的異性形体。
ここで用いられる用語「薬学的に許容される界面活性剤」は、薬学的に許容されるイオン性または非イオン性活性剤を指す。界面活性剤の取り入れは、僅水溶性活性成分を含むマトリックスにとって特に好ましい。界面活性剤は剤型から排出される活性成分の瞬間的乳化を達成する可能性があり、および/または消化管の水性液体中での、活性成分の沈降を防止する可能性がある。
ポリオキシエチレンアルキルエーテル、たとえばポリオキシエチレン(3)ラウリルエーテル、ポリオキシエチレン(5)セチルエーテル、ポリオキシエチレン(2)ステアリルエーテル、ポリオキシエチレン(5)ステアリルエーテル;ポリオキシエチレンアルキルアリールエーテル、たとえばポリオキシエチレン(2)ノニルフェニルエーテル、ポリオキシエチレン(3)ノニルフェニルエーテル、ポリオキシエチレン(4)ノニルフェニルエーテルまたはポリオキシエチレン(3)オクチルフェニルエーテル;
ポリエチレングリコール脂肪酸エステル、たとえばPEG−200モノラウレート、PEG−200ジラウレート、PEG−300ジラウレート、PEG−400ジラウレート、PEG−300ジステアレートまたはPEG−300ジオレエート);
アルキレングリコール脂肪酸モノエステル、たとえばプロピレングリコールモノラウレート(Lauroglycol(登録商標));
ショ糖脂肪酸エステル類、たとえば蔗糖モノステアレート、蔗糖ジステアレート、蔗糖モノラウレートまたは蔗糖ジラウレート;
ソルビタンモノラルラウレート(Span(登録商標))20、ソルビタンモノオレエート、ソルビタンモノパルミテート(Span(登録商標)40)、またはソルビタンステアレートのようなソルビタン脂肪酸モノエステル,
ポリオキシエチレンヒマシ油誘導体、たとえばポリオキシル35ヒマシ油(Cremophor(登録商標)EL、BASF社)またはポリエチレングリコール40水素添加ヒマシ油(Cremophor(登録商標)RH40、BASF社)またはポリエチレングリコール60水素添加ヒマシ油(Cremophor(登録商標)RH60、BASF社)、または
エチレンオキシドとプロピレンオキシドのブロックコポリマー(別名ポリオキシエチレンポリオキシプロピレンブロックコポリマーまたはポリオキシエチレンポリプロピレングリコール、例:Poloxamer(登録商標)124、Poloxamer(登録商標)188、Poloxamer(登録商標)237、Poloxamer(登録商標)388またはPoloxamer(登録商標)407(BASF社))、または、
ポリオキシエチレン(20)ソルビタンのモノ脂肪酸エステル、たとえばポリオキシエチレン(20)ソルビタンモノオレエート(Tween(登録商標)80)、ポリオキシエチレン(20)ソルビタンモノステアレート(Tween(登録商標)60)、ポリオキシエチレン(20)ソルビタンモノパルミテート(Tween(登録商標)40)、ポリオキシエチレン(20)ソルビタンモノラウレート(Tween(登録商標)20)、またはそれらの1種以上の混合物。
これらの加熱ゾーンH1、H2およびH3におけるバレル1の温度は、活性成分とマトリックス形成剤の分散体の溶融を調節するために、調節することができる。
Claims (25)
- 生理活性成分と薬学的に許容されるマトリックス形成剤を押出機に供給することおよび均一な押出物を作ることを含む、生理活性成分の固体分散体を生産するための製薬プロセスであって、前記押出機が少なくとも2個の回転シャフト(2)を含み、該シャフト(2)の各々は、複数の処理エレメントを持ち、該処理エレメントは、
(i)供給−搬送セクション(A)、
(ii)少なくとも1個の逆フライトセクション(R、D)、および、
(iii)排出セクション(E)、
と定義され、
前記複数の処理エレメントは、前記シャフト(2)の軸線上に(i)、(ii)、(i
ii)の順に配置され、
前記供給−搬送セクション(A)と定義される前記処理エレメントは、順フライトスクリュー式エレメント(3)を含み、
前記逆フライトセクション(R、D)と定義される前記処理エレメントは、少なくとも2個の逆フライトエレメント(14)、および当該逆フライトエレメント(14)の間に配置された少なくとも1個の順フライトスクリュー式エレメント(3)を含み、
前記排出セクション(E)と定義される前記処理エレメントは、順フライトスクリュー式エレメント(3)を含み、
前記少なくとも2個の逆フライトエレメント(14)は、それぞれ前記押出機の全体的な搬送方向と逆の搬送方向を有するスクリュー式エレメントに基づくものであり、
前記少なくとも2個の逆フライトエレメント(14)のスクリューピッチは、前記少なくとも1個の順フライトスクリュー式エレメント(3)のスクリューピッチの(−0.8)倍〜(−1.2)倍の範囲にあることを特徴とする製薬プロセス。 - 前記逆フライトセクション(R、D)と定義される前記処理エレメントが、少なくとも3個の逆フライトエレメント(14−1〜14−3)を含み、前記少なくとも1個の順フライトスクリュー式エレメント(3)がそれぞれの連続する逆フライトエレメント(14)の間に配置される、請求項1に記載の製薬プロセス。
- 第二と第三の逆フライトエレメント(14−2、14−3)の間に配置される前記順フライトスクリュー式エレメント(3)の長さが、第一と第二の逆フライトエレメント(14−1、14−2)の間に配置される前記順フライトスクリュー式エレメント(3)の長さの1〜15倍の範囲にある、請求項2に記載の製薬プロセス。
- 前記逆フライトセクション(R、D)と定義される前記処理エレメントが、少なくとも4個の逆フライトエレメント(14−1〜14−4)を含み、前記少なくとも1個の順フライトスクリュー式エレメントがそれぞれの連続する逆フライトエレメントの間に配置され、ならびに連続する逆フライトエレメントの間に配置される順フライトスクリュー式エレメント(3)が同じ長さを持つ、請求項1に記載の製薬プロセス。
- 前記処理エレメントが、さらにスクリュー式エレメントに由来する少なくとも1つの混合エレメント(11、12、13)を含み、前記混合エレメント(11、12、13)が前記スクリュー式エレメントのスクリューフライトに形成されたくぼみを有する、請求項1〜4のいずれかに記載の製薬プロセス。
- 少なくとも1個の逆フライトエレメント(14)が前記混合エレメント(11、12、13)の下流に配置される、請求項5に記載の製薬プロセス。
- 少なくとも1つの前記混合エレメント(11、12、13)がスクリュー式エレメントを旋削した溝によって形成された複数の同心性リング部分(16、25)を有し、
前記混合エレメント(11、12、13)は、前記リング部分(16、25)の前記溝によってのみ分断される、連続的なスクリューフライトを有する、請求項5または6に記載の製薬プロセス。 - 少なくとも1つの前記混合エレメント(11、12、13)が全体的な搬送方向に平行で逆向きの法線をもつ平坦な表面を持たない、請求項5〜7のいずれかに記載の製薬プロセス。
- 少なくとも1つの前記混合エレメント(11、12、13)が全体的な搬送方向に対して垂直である面を持たない、請求項5〜8のいずれかに記載の製薬プロセス。
- 少なくとも1つの前記混合エレメント(11、12、13)が全体的な搬送方向に対して垂直である突合せ面を持たない、請求項5〜9のいずれかに記載の製薬プロセス。
- 前記処理エレメントは、
(i)供給−搬送セクション(A)、
(ii)第一混合セクション(B)、
(iii)中間搬送セクション(C)、
(iv)逆フライトセクション(D)、および、
(v)排出セクション(E)、
と定義され、第一混合セクション(B)と定義される処理エレメントは、少なくとも1個の混合エレメント(11、12、13)を含み、逆フライトセクション(D)と定義される処理エレメントは、少なくとも1個の混合エレメント(11、12、13)およびその下流に少なくとも1個の逆フライトエレメント(14)を含むことを特徴とする、請求項5〜10のいずれかに記載の製薬プロセス。 - 前記マトリックス形成剤の少なくとも一部が前記押出機のホッパーに供給され、
(i)前記マトリックス形成剤の残り、
(ii)生理活性成分、
(iii)添加物、および、
(iv)それらの組合せ
から選択される少なくとも1つの成分が、押出機バレル(1)にある開口部(8)を通して混合セクション(B)または逆フライトセクション(R、D)の位置、またはそれらよりも上流の位置で前記押出機に導入される、請求項1〜11のいずれかに記載の製薬プロセス。 - 少なくとも1つの成分が供給−搬送セクションと混合セクション(B)または逆フライトセクション(R、D)の接合点の、またはその近くの、位置で前記押出機に導入される、請求項12に記載の製薬プロセス。
- 少なくとも1つの成分が液体であるか、または液化性である、請求項12または13に記載の製薬プロセス。
- 少なくとも1つの成分が薬学的に許容される界面活性剤を含む、請求項12〜14のいずれかに記載の製薬プロセス。
- 前記生理活性成分が固溶体の状態でポリマー中に分散する、請求項1〜15のいずれかに記載の製薬プロセス。
- 前記マトリックス形成剤がポリマーを含む、請求項1〜16のいずれかに記載の製薬プロセス。
- 前記ポリマーが、
N−ビニルラクタムのホモポリマー、
N−ビニルラクタムと窒素含有モノマーまたは酸素含有モノマーから選択される1つ以上のコモノマーのコポリマー、
セルロースエステルおよびセルロースエーテル、
高分子量ポリアルキレンオキシド、
ポリアクリレートおよびポリメタクリレート、
オリゴ−およびポリ−サッカライド、
ポリ(ヒドロキシ酸)、またはそれらの混合物
からなる群から選択される、請求項17に記載の製薬プロセス。 - 前記マトリックス形成剤がポリオール、ワックスおよび脂質のいずれかから選択される部材を含む、請求項1〜18のいずれかに記載の製薬プロセス。
- さらに、流量調整剤類−潤滑油、充填剤、崩壊剤、可塑剤、スタビライザーまたは防腐剤、からなる群から選択される少なくとも1つの添加剤を押出機中に供給することを含む、請求項1〜19のいずれかに記載の製薬プロセス。
- 押出物が1つの剤型に直接成形される、請求項1〜20のいずれかに記載の製薬プロセス。
- 成形がカレンダー加工、射出成形または異形押出成形によって遂行される、請求項21に記載の製薬プロセス。
- さらに、固化した押出物を磨砕することを含む、請求項1〜22のいずれかに記載の製薬プロセス。
- さらに、前記の固体分散体を錠剤に成形することまたは前記固体分散体をカプセル殻に詰めることを含む、請求項23に記載の製薬プロセス。
- さらに、前記錠剤にフィルムコートを塗布することを含む、請求項24に記載の製薬プロセス。
