MOG誘発マウス実験的自己免疫性脳脊髄炎モデルの神経症状スコアの上昇に対する、シクロヘキサン誘導体(I)又はその薬理学的に許容される塩の作用を評価した。マウス実験的自己免疫性脳脊髄炎モデルは、Journal of Neuroscience Research、2006年、84巻、p.1225−1234に記載の方法を一部改変して作製した。
MOG誘発マウス実験的自己免疫性脳脊髄炎モデルの神経症状スコアの上昇に対する、シクロヘキサン誘導体(I)又はその薬理学的に許容される塩の作用を評価した。
PLP誘発マウス実験的自己免疫性脳脊髄炎モデルの神経症状スコアの上昇に対する、シクロヘキサン誘導体(I)又はその薬理学的に許容される塩の作用を評価した。マウス実験的自己免疫性脳脊髄炎モデルは、International Immunology、1997年、第9巻、p.1243−1251に記載の方法を一部改変して作製した。
トロンビン活性に対する、シクロヘキサン誘導体(I)又はその薬理学的に許容される塩の作用を、蛍光共鳴エネルギー移動(FRET)を利用したAnaspec社のSensoLyte(登録商標) 520トロンビン活性アッセイキットを用いて評価した。
トロンビン活性阻害率(%)=(1−((トロンビン添加かつ被験化合物添加時の蛍光値)−(トロンビン非添加時かつ被験化合物非添加の蛍光値))/((トロンビン添加かつ被験化合物非添加時の蛍光値)−(トロンビン非添加時かつ被験化合物非添加の蛍光値)))×100 ・・・式1
Claims (6)
- 一般式(I)
[式中、Aは、一般式(IIa)で表される置換基であり、
R1及びR2は、それぞれ独立して、トリフルオロメチル基、メチル基又はメトキシ基であり、
R3は、水素原子であり、R4は、ヒドロキシメチル基又はヒドロキシル基であり、
R5及びR6は、それぞれ独立して、水素原子、フッ素原子、トリフルオロメチル基、カルボキシル基、メトキシ基、ヒドロキシル基又はアセチルオキシ基であり、
R7及びR8は、それぞれ独立して、水素原子又はフッ素原子であり、
Zは、窒素原子又はメチン基である。]
で示されるシクロヘキサン誘導体又はその薬理学的に許容される塩を有効成分として含有する、多発性硬化症の治療剤又は予防剤。 - R 7 及びR 8 は、それぞれ独立して水素原子である、請求項1記載の多発性硬化症の治療剤又は予防剤。
- R 1 及びR 2 は、それぞれ独立して、メチル基又はメトキシ基である請求項1又は2記載の多発性硬化症の治療剤又は予防剤。
- R 5 及びR 6 は、それぞれ独立して、水素原子、メトキシ基又はヒドロキシル基である請求項1〜3のいずれか一項記載の多発性硬化症の治療剤又は予防剤。
- Zは、メチン基である請求項1〜4のいずれか一項記載の多発性硬化症の治療剤又は予防剤。
- 一般式(I)が下記化学式である請求項1〜5のいずれか一項記載の多発性硬化症の治療剤又は予防剤。
