Description
アルクチゲニンの分化細胞(ヒト皮膚繊維芽細胞)への影響を調べるため、ヒト皮膚繊維芽細胞をアルクチゲニンまたはケルセチンで処理したときの細胞生存率を調べた。ケルセチンは、上述した特許文献1において、未分化細胞除去効果を有することが報告されている。
次に、アルクチゲニンの未分化細胞への影響を調べるため、iPS細胞をアルクチゲニンまたはケルセチンで処理したときの細胞生存率を調べた。
アルクチゲニンの分化細胞への影響を調べるため、細胞をアルクチゲニンで処理したときの細胞生存率を調べた。分化細胞には、ヒト皮膚繊維芽細胞(KF-4009、クラボウ)、ヒト心筋細胞(C-12810、タカラバイオ)およびヒトiPS由来心筋細胞(Y50015、タカラバイオ)を用いた。無菌下(クリーンベンチ内)で、細胞を、2×104 cells/well(生細胞数)の密度で96wellプレートに接種し、37℃で24時間培養した。ヒトiPS由来心筋細胞は、細胞培養マトリックス(Fibronectin from human plasma、F0895、SIGMA)にて、あらかじめコートしたウェル(well)にて培養した。
次に、アルクチゲニンの未分化細胞への影響を調べるため、未分化細胞(iPS細胞)をアルクチゲニンで処理したときの細胞生存率を調べた。未分化細胞には、異なる方法で作成された3種類のiPS細胞、すなわちRPChiPS771-2(リプロセル)、201B7(CiRA)および253G1(CiRA)を用いた。無菌下(クリーンベンチ内)で、各細胞を、2×104 cells/well(生細胞数)の密度で96wellプレートに接種し、37℃で24時間培養した。iPS細胞は、細胞培養マトリックス(アイマトリックス-551溶液、892011、ニッピ)であらかじめコートしたウェル(well)にて培養した。培地には、StemFit AK02N(RCAK02N、リプロセル)を用いた。
アルクチゲニンと2-デオキシ-D-グルコース(以下、「2-DG」と表記する。)を併用したときの分化細胞への影響を調べるため、細胞をアルクチゲニンおよび2-DGの両方で処理したときの細胞生存率を調べた。分化細胞には、ヒトiPS由来心筋細胞(Y50015、タカラバイオ)を用いた。無菌下(クリーンベンチ内)で、細胞を、2×104 cells/well(生細胞数)の密度で96wellプレートに接種し、37℃で24時間培養した。ヒトiPS由来心筋細胞は、細胞培養マトリックス(Fibronectin from human plasma、F0895、SIGMA)にて、あらかじめコートしたウェル(well)にて培養した。培地には、MiraCell(商標)CM Culture Medium(Y50013、タカラバイオ)を用いた。
アルクチゲニンと2-DGを併用したときの未分化細胞への影響を調べるため、未分化細胞(iPS細胞)をアルクチゲニンおよび2-DGの両方で処理したときの細胞生存率を調べた。未分化細胞には、201B7(CiRA)を用いた。無菌下(クリーンベンチ内)で、細胞を、2×104 cells/well(生細胞数)の密度で96wellプレートに接種し、37℃で24時間培養した。未分化細胞(iPS細胞)である201B7細胞は、細胞培養マトリックス(アイマトリックス-551溶液、892011、ニッピ)であらかじめコートしたウェル(well)にて培養した。培地には、StemFit AK02N(RCAK02N、リプロセル)を用いた。2-DGとして、2-Deoxy-D-glucose(D0051、東京化成工業)を用いた。
アルクチゲニンとグルコース阻害による未分化細胞への影響を調べるため、未分化細胞(iPS細胞)をグルコース濃度の低い培地(低グルコース培地)においてアルクチゲニンで処理したときの細胞生存率を調べた。未分化細胞には、201B7(CiRA)を用いた。無菌下(クリーンベンチ内)で、細胞を、2×104 cells/well(生細胞数)の密度で96wellプレートに接種し、37℃で24時間培養した。未分化細胞(iPS細胞)である201B7細胞は、細胞培養マトリックス(アイマトリックス-551溶液、892011、ニッピ)であらかじめコートしたウェル(well)にて培養した。培地には、DMEM/Ham's F-12(No Glucose) with L-Glu and Sodium Pyruvate(09893-05、ナカライ)を用いた。グルコースとして、(D-(+)Glucose solution 100 ml 45% in H2O(G8769-100ML、SIGMA)を用いた。
Claims (8)
- アルクチゲニンを有効成分として含有する、iPS細胞除去剤。
- iPS細胞を分化誘導した細胞群に残存する未分化のiPS細胞を除去するための、請求項1に記載のiPS細胞除去剤。
- グルコース代謝拮抗物質をさらに含有する、請求項1または2に記載のiPS細胞除去剤。
- 請求項1〜3のいずれか1項に記載のiPS細胞除去剤と、低グルコース培地とを含む、iPS細胞を除去するためのキット。
- in vitroにおいてアルクチゲニンをiPS細胞に接触させる工程を含む、iPS細胞を除去する方法。
- 前記iPS細胞が、iPS細胞を分化誘導した細胞群に残存する未分化のiPS細胞である、請求項5に記載の方法。
- 前記接触させる工程において、前記アルクチゲニンを低グルコース培地において前記iPS細胞に接触させる、請求項5または6に記載の方法。
- 前記接触させる工程において、グルコース代謝拮抗物質を前記iPS細胞に接触させる、請求項5〜7のいずれか1項に記載の方法。
