Description
当該方法が、
(a)前記オイル産生生物のバイオマスの細胞を溶解する工程と、
(b)溶解されたバイオマスからオイルを機械的に分離し、粗製オイルを回収する工程と
を含み、
前記細胞を溶解する工程(a)が、下記2つのパート:
(a1)第1の温度で実行される第1のパート、次いで
(a2)前記第1の温度よりも低い第2の温度で溶解を実行し続ける第2のパート
を含むことを特徴とする、方法に関する。
培養は、専用の自動操作、およびコンピュータステーションによる監視を備えた1~5Lの有用な容積を持つ発酵槽(バイオリアクター)中で行った。培養は、Aurantiochytrium mangroveiの2株を用い、2つの異なる培養プロトコルで行った。系は、塩基(実施例b1およびb2についてはNH4OH、実施例aについてはNaOH)の添加によりpH5に調整し、窒素供給を行いながら(実施例b1およびb2について)、培養の全期間にわたってpH調整を行った。培養温度は、30℃、その後22℃、最終的に培養の終了時に18℃とした。
総バイオマス濃度は、乾燥質量を測定することによってモニターした(測定前に、Whatman GF/Fフィルタで濾過し、その後105℃で、最小で24時間オーブン乾燥させた)。脂肪酸分析は、バイオマスについては、ISO12966-2規格に準拠した方法に従って、またオイルについては、欧州薬局方9.0(2.4.29.)に従って行った。
10m3(実施例d)または180m3(実施例e)のタンクなどの工業用サイズの発酵槽中で、培地b、および炭素:窒素:リン(CNP)のモル比率が533:0.4:1である強化溶液の形態でのグルコース添加により、同様の条件で増殖させたとき、高DHA含有Aurantiochytrium mangrovei株は、同様の脂肪酸組成を有するバイオマスを生産する。
実施例2の条件下で生産され、60%超のDHAを含有する、同一のバイオマスからオイルを抽出した(表4)。
以下の手順に従って、実施例2で生産されたバイオマスからオイルを抽出した:
(a)65℃の温度で4時間の、酵素的手段(例えば、Novozymes社製のAlcalase 2.5LまたはAlcalase 2.4LまたはNovozym 37071)による細胞溶解、
(b)5から40℃に温度を下げながら、30分から30時間、引き続き溶解、
(c)遠心プレートセパレータによるオイルの機械的分離。

Claims (12)
- オイルを含むオイル産生生物のバイオマスから、PUFAに富むオイルを抽出する方法であって、
当該方法が、
(a)前記オイル産生生物のバイオマスの細胞を酵素的に溶解する工程と、
(b)溶解されたバイオマスからオイルを機械的に分離し、粗製オイルを回収する工程と
を含み、
前記細胞を溶解する工程(a)が、下記2つのパート:
(a1)50℃と95℃の間である第1の温度で実行される第1のパート、次いで
(a2)40℃以下である第2の温度で溶解を実行し続ける第2のパートを含み、
パート(a1)およびパート(a2)が、細胞の溶解が実施される媒体に塩、酸または塩基を追加することなく実施され、
前記第2の温度が、前記第1の温度よりも少なくとも10℃低いことを特徴とする、方法。 - 前記オイル産生生物のバイオマスが、PUFA産生微生物のバイオマスであることを特徴とする、請求項1に記載の方法。
- 前記バイオマスが、前記微生物の懸濁液であることを特徴とする、請求項2に記載の方法。
- 前記オイル産生生物によって産生されるオイルが50%超のPUFAを含むことを特徴とする、請求項1~3のいずれか一項に記載の方法。
- 前記PUFAが、ドコサヘキサエン酸(DHA、C22:6n3)、ドコサペンタエン酸(DPA、C22:5n6)、アラキドン酸(ARA、C20:4n6)、エイコサペンタエン酸(EPA、C20:5n3)およびそれらの混合物から選択されるものであることを特徴とする、請求項1~4のいずれか一項に記載の方法。
