Description

【発明の詳細な説明】 本発明はピペラジニルアルコキシインダン類系
抗不安薬に関するものである。さらに詳しくは本
発明は毒性が低く、副作用と考えられる筋弛緩作
用も弱い抗不安薬に関するものである。 本発明の抗不安薬は下記式()で表わされる
化合物を有効成分とするものである。 一般式()中nは3または4の整数を示し、
R1は水素原子、低級アルキル基またはハロゲン
原子を示す。低級アルキル基としてはC1〜C5
アルキル基が挙げられ、特にC1〜C2のアルキル
基が好ましい。ハロゲン原子はフツ素原子、塩素
原子、シユウ素原子等が挙げられ、特にフツ素原
子が好ましい。R2はハロゲン原子を有していて
もよいフエニル基を示し、ハロゲン原子としては
R1と同様なハロゲン原子が挙げられ、中でもフ
ツ素原子が好ましい。 R1の置換位置及びR2上のハロゲン原子の置換
位置は特に限定されない。 一般式()に示されるインダン類は以下のよ
うにして製造される。 すなわち式() (上記式中n及びR1は一般式()のn及びR1
と同義であり、Xはハロゲン原子を示す。)で示
されるハロゲノアルコキシインダン類と式() (上記式中R2は一般式()中のR2と同義であ
る)で表わされるピペラジン類との反応により得
られる。 ハロゲノアルコキシインダン類とピペラジン類
とはそれぞれ1:1で反応するが、通常ピペラジ
ン類を過剰に使用する方が反応が円滑に進行す
る。従つてピペラジン類はハロゲノアルコキシイ
ンダン類1モルに対し1〜10モル使用される。 反応は無溶媒でも十分進行するが、反応を円滑
に進めるために、不活性溶媒を用いてもよい。溶
媒としては水、ジオキサン、テトラヒドロフラ
ン、ジメチルホルムアミド、ジメチルスルホキシ
ド、低級アルコールまたはこれらの二種以上の溶
媒の混合物が用いられる。 反応温度は特に限定されないが通常室温から
150℃である。 反応時間は反応温度及び原料の反応性、溶媒の
種類により異なるが通常10分から20時間の範囲で
ある。 また反応により生ずるハロゲン化水素を捕集し
て反応を促進させるために、塩基類を添加しても
よい。塩基類としては、水酸化カリウム、炭酸カ
リウム、水酸化ナトリウム、炭酸水素ナトリウ
ム、炭酸ナトリウム等の無機塩類、ピリジン、ト
リエチルアミン等の第三級有機アミン類等であ
る。その使用量はピペラジン類1モルに対し通常
1〜5モルである。 望ましい酸付加塩を得るためには、反応終了後
過剰のアミン類及び溶媒を蒸留あるいは水洗によ
り除き、水酸化ナトリウムあるいは水酸化カリウ
ム等の強塩基水溶液を加え、遊離のピペラジニル
アルコキシインダン類とし、その後エーテル、ク
ロロホルム、ベンゼン、トルエン等の溶媒で本化
合物を抽出する。さらに望ましい酸を加えて中和
すると目的とする酸付加塩が得られる。 一般式()で示されるインダン類の具体例を
以下に例示する。 5−〔3−(4−フエニル−1−ピペラジニル)
プロポキシ〕インダン 5−〔4−(4−フエニル−1−ピペラジニル)
ブトキシ〕インダン 5−{3−〔4−(4−フルオロフエニル)−1−
ピペラジニル〕プロポキシ}インダン 5−{4−〔4−(4−フルオロフエニル)−1−
ピペラジニル〕ブトキシ}インダン 6−フルオロ−5−〔3−(4−フエニル−1−
ピペラジニル)プロポキシ〕インダン 6−メチル−5−〔3−(4−フエニル−1−ピ
ペラジニル)プロポキシ〕インダン 4−フルオロ−5−{3−〔4−(4−フルオロ

塩化水素酸、シユウ化水素酸、硫酸、リン酸、硝
酸等の無機酸、酢酸、コハク酸、アジピン酸、プ
ロピオン酸、酒石酸、フマル酸、マレイン酸、シ
ユウ酸、クエン酸、安息香酸、トルエンスルホン
酸、メタンスルホン酸等の有機酸が挙げられる。 以下本発明の抗不安作用について説明する。 一般式()に示される化合物の抗不安効果は
以下の方法で検討した。 抗不安薬あるいはマイナ−トランキライザーと
しての公知のクロロジアゼポキサイド、既知化合
物Aを対照薬として用いた。結果は50%有効量
(ED50、mg/KgPO)として表に示した。 動物はddy系雄性マウス(20−22g)を用い、
抗不安作用の指標として抗−フアイテイング
(fighting)作用及び抗一モルヒネ(morphine)
作用を検討した。即ち抗−fighting作用は

ドを介して足部に与えることにより発現する

表1に示すごとく、化合物1〜7は、対照薬No.
8及びクロロジアゼポキサイドにまさる抗不安効
果が認められたが、なかでもNo.2の化合物はクロ
ロジアゼポキサイドに比し低毒性で、かつ筋弛緩
作用も弱く、安全性の高い薬物である。 本発明の薬物はいかなる方法でも投与できる
が、好適には以下のような方法が実施される。 すなわち皮下注射、静脈内注射、筋肉注射、腹
腔内注射等の非経口投与もまた経口投与も可能で
ある。 投与量は患者の年令、健康状態、体重、同時処
理があるならばその種類、処置頻度、所望の効果
の性質等により決定される。 一般的に有効成分の1日投与量は0.5〜50mg/
Kg体重、通常1〜30mg/Kg体重であり、1回ある
いはそれ以上投与される。 本発明の薬物を経口投与する場合は錠剤、カプ
セル剤、粉剤、液剤、エリキシル剤等の形体で、
また非経口投与の場合は液体あるいは懸濁等の殺
菌した液状の形体で用いられる。上述の様な形体
で用いられる場合、固体あるいは液体の毒性のな
い製剤的担体が組成に含まれ得る。 固体担体の例としては通常のゼラチンタイプの
カプセルが用いられる。また有効成分を補助薬と
ともにあるいはそれなしに錠剤化、粉末包装され
る。 これらのカプセル、錠剤、粉末は一般的に5〜
95%、好ましくは25〜90%重量の有効成分を含
む。 すなわちこれらの投与形式では5〜500mg、好
ましくは25〜250mgの有効成分を含有するのがよ
い。 液状担体としては水あるいは石油、ピーナツ
油、大豆油、ミネラル油、ゴマ油等の動植物起原
の、または合成の油等が用いられる。 また、一般に生理食塩水、デキストロースある
いは類似のシヨ糖溶液、エチレングリコール、プ
ロピレングリコール、ポリエチレングリコール等
のグリコール類が液状担体として好ましく、とく
に生理食塩水を用いた注射液の場合には通常0.5
〜20%、好ましくは1〜10%重量の有効成分を含
むようにする。 経口投与の液剤の場合、0.5〜10%重量の有効
成分を含む懸濁液あるいはシロツプがよい。 この場合の担体としては香料、シロツプ、製剤
学的ミセル体等の水様賦形剤を用いる。 以上説明したように本発明によれば優れた抗不
安薬を提供することができる。 製造例 5−(3−ブロモプロポキシ)インダン25.5g
(0.1モル)とN−フエニルピペラジン17.8g
(0.11モル)とトリエチルアミン15gとをジメチ
ルホルムアミド300mlにとかし80℃で2時間加熱
する。反応液を1NNaOH水溶液500mlに加えエチ
ルエーテル400mlで2回抽出する。エチルエーテ
ルを飽和食塩水で洗い無水硫酸ナトリウムで乾燥
した後エチルエーテルを留去する。残査油状物質
に20%HCl−エチルアルコール溶液を50ml加える
と結晶が析出する。放冷取後エチルアルコール
から再結晶すると融点195−7℃の5−〔3−(4
−フエニル−1−ピペラジニル)プロポキシ〕イ
ンダンが35g(85.6%)得られる。 以下同様な方法で化合物を製造し、結果を上記
