Description

【発明の詳細な説明】[Detailed description of the invention]

本発明はピペラジニルアルコキシインダン類及
びその酸付加塩に関するものである。さらに詳し
くは抗不安作用を有し鎮静薬として有効なピペラ
ジニルアルコキシインダン類に関するものであ
る。 本発明化合物は下記一般式() で示されるピペラジニルアルコキシインダン類で
ある。 一般式()中nは3または4の整数を示し、
R1はハロゲン原子、アルキル基、アルコキシ基、
ヒドロキシ基、フエニル基またはニトロ基を示
し、ハロゲン原子としてはフツ素原子、塩素原
子、シユウ素原子等、アルキル基としてはメチル
基、エチル基、プロピル基、イソプロピル基、ブ
チル基等の炭素数1〜5の低級アルキル基が挙げ
られる。アルコキシ基としてはメトキシ基、エト
キシ基、プロポキシ基、ブトキシ基等の炭素数1
〜5の低級アルコキシ基が挙げられる。R2はフ
エニル基を示し、またこのフエニル基はハロゲン
原子またはトリフルオロメチル基で置換されてい
てもよい。ハロゲン原子としては、R1の場合と
同様なものが挙げられる。 インダン環上のR1、ピペラジニルアルコキシ
基の位置およびR2の置換基の位置はとくに限定
されない。 本発明化合物は以下のようにして製造される。
すなわち式() (上記式中nは3または4の整数を示し、R1
は一般式(1)のR1と同義であり、Xはハロゲン原
子を示す。)で示されるハロゲノアルコキシイン
ダン類と式() (上記式中R2は一般式()中のR2と同義で
ある)で表わされるピペラジン類との反応により
得られる。 ハロゲノアルコキシインダン類とピペラジン類
とはそれぞれ1:1で反応するが、通常ピペラジ
ン類を過剰に使用する方が反応が円滑に進行す
る。従つてピペラジン類はハロゲノアルコキシイ
ンダン類1モルに対し1〜10モル使用される。 反応は無溶媒でも十分進行するが、反応を円滑
に進めるために、不活性溶媒を用いてもよい。溶
媒としては水、ジオキサン、テトラヒドロフラ
ン、ジメチルホルムアミド、ジメチルスルホキシ
ド、低級アルコールまたはこれらの二種以上に溶
媒の混合物が用いられる。 反応温度は特に限定されないが通常室温から
150℃である。 反応時間は反応温度及び原料の反応性、溶媒の
種類により異なるが通常10分から20時間の範囲で
ある。 また反応により生ずるハロゲン化水素を捕集し
て反応を促進させるために、塩基類を添加しても
よい。塩基類としては、水酸化カリウム、炭酸カ
リウム、水酸化ナトリウム、炭酸水素ナトリウ
ム、炭酸ナトリウム等の無機塩類、ピリジン、ト
リエチルアミン等の第三級有機アミン類等であ
る。その使用量はピペラジン類1モルに対し通常
1〜5モルである。 望ましい酸付加塩を得るためには、反応終了後
過剰のアミン類及び溶媒を蒸留あるいは水洗によ
り除き、水酸化ナトリウムあるいは水酸化カリウ
ム等の強塩基水溶液を加え、遊離のピペラジニル
アルコキシインダン類とし、その後エーテル、ク
ロロホルム、ベンゼン、トルエン等の溶媒で本化
合物を抽出する。さらに望ましい酸を加えて中和
すると目的とする酸付加塩が得られる。 本発明化合物の具体例を以下に例示する。 6―フルオロ―5―〔3―(4―フエニル―1

ル)―1―ピペラジニル〕プロポキシ}インダン 等である。 本発明化合物はいかなる方法でも投与できる
が、好適には以下のような方法が実施される。 すなわち皮下注射、静脈内注射、筋肉注射、腹
腔内注射等の非経口投与もまた経口投与も可能で
ある。 投与量は患者の年令、健康状態、体重、同時処
理があるならばその種類、処置頻度、所望の効果
の性質等により決定される。 一般的に有効成分の1日投与量は0.5〜50mg/
Kg体重、通常1〜30mg/Kg体重であり、1回ある
いはそれ以上投与される。 本発明化合物を経口投与する場合は錠剤、カプ
セル剤、粉剤、液剤、エリキシル剤等の形体で、
また非経口投与の場合は液体あるいは懸濁等の殺
菌した液状の形体で用いられる。上述の様な形体
で用いられる場合、固体あるいは液体の毒性のな
い製剤的担体が組成に含まれ得る。 固体担体の例としては通常のゼラチンタイプの
カプセルが用いられる。また有効成分を補助薬と
ともにあるいはそれなしに錠剤化、粉末包装され
る。 これらのカプセル、錠剤、粉末は一般的に5〜
95%、好ましくは25〜90%重量の有効成分を含
む。 すなわちこれらの投与形式では5〜500mg、好
ましくは25〜250mgの有効成分を含有するのがよ
い。 液状担体としては水あるいは石油、ピーナツ
油、大豆油、ミネラル油、ゴマ油等の動植物起原
の、または合成の油等が用いられる。 また、一般に生理食塩水、デキストロースある
いは類似のシヨ糖溶液、エチレングリコール、プ
ロピレングリコール、ポリエチレングリコール等
のグリコール類が液状担体として好ましく、とく
に生理食塩水を用いた注射液の場合には通常0.5
〜20%、好ましくは1〜10%重量の有効成分を含
むようにする。 経口投与の液剤の場合、0.5〜10%重量の有効
成分を含む懸濁液あるいはシロツプがよい。 この場合の担体としては香料、シロツプ、製剤
学的ミセル体等の水様賦形剤を用いる。 以上説明したように本発明化合物は抗不安作用
を有する鎮静薬として有効に使用できる。 製造例 5―(3―ブロモプロポキシ)―6―フルオロ
―インダン27.3g(0.1モル)とN―フエニルピ
ペラジン17.8g(0.11モル)とトリエチルアミン
15gとをジメチルホルムアミド300mlにとかし80
℃で2時間加熱する。反応液を1N NaOH水溶液
500mlに加えエチルエーテル400mlで2回抽出す
る。エチルエーテルを飽和食塩水で洗い無水硫酸
ナトリウムで乾燥した後エチルエーテルを留去す
る。残査油状物質に20%HCl―エチルアルコール
溶液を50ml加えると結晶が析出する。放冷取後
エチルアルコールから再結晶する融点212―3℃
の6―フルオロ―5―〔3―(4―フエニル―1
―ピペラジニル)プロポキシ〕インダン2HCl塩
が36.5g(85.6%)得られる。 以下同様な方法で化合物を製造し、結果を上記
化合物と併せ表2に示す。
