Description

【発明の詳細な説明】 1 産業上の利用分野 本発明はキラルな分析試薬に関するものであ
る。 2 従来技術 不斉な化合物の光学純度を知るための直接的か
つ便利な方法の1つとして、キラルなシフト試薬
を用いたNMR(核磁気共鳴)分析による方法が
ある。例えば、ユーロピウムキレート等のシフト
試薬では、リガンド(配位子)であるβ−ジケト
ンがその性能を決定する重要な化合物であり、一
般にフツ素原子を多く含むものが特に有効である
とされている。また、このβ−ジケトンはフツ素
原子を含むことによつて、撥水撥油剤、界面活性
剤、樹脂添加剤、溶剤等として効果を発揮するこ
とも期待される。 3 発明の目的 本発明は、フツ素原子を数多く含み、しかも適
度な立体的かさ高さを有するキラルな分析試薬を
提供することを目的とするものである。 4 発明の構成 即ち、本発明は、一般式: 〔但、Rfは【式】(但、 C〓*は不斉炭素である。で表わされるフルオロ−
β−ジケトンからなることを特徴とするキラルな
分析試薬に係るものである。 5 発明の効果 本発明による上記フルオロ−β−ジケトンはキ
ラルな例えばユーロピウムキレート等のシフト試
薬を形成するのに有効である。即ち、フツ素原子
によつて、1)電子求引性が高まつてキレートが
活性化され、シフト試薬としてのルイス酸性度が
増大するので、低濃度でもシフト効果に優れたも
のとなる。2)基質とオーバーラツプする水素原
子が少ないため、基質のシグナルとのオーバーラ
ツプが少なくなる。3)有機溶媒に対する溶解度
が増す。4)上記一般式中の一対のRfが共に例
えば【式】で ある(但、n=0)ため、分子構造的にみて対称
的な構造になるから立体効果によつてシフト試薬
が安定化し、かつ不斉中心(炭素)を2個もたせ
ることができる。 また、このフルオロ−β−ジケトン類は、フツ
素の存在によつてフツ素化合物としての既述した
如き作用も示すものと期待できる。 更に、構造的にみて、本発明による上記フルオ
ロ−β−ジケトンはRfが
【式】(但、Rf′は− CF2CF2CF3からなる低級ペルフルオロアルキル
基、n=0)で表わされる基であるから、エーテ
ル結合がその基の構造を安定化するのに寄与して
いる。つまり、そうしたエーテル結合がなく、フ
ルオロアルキル基が上述の【式】に直接結合してい る場合には、Rfが擬似ハロゲンとして働き、
【式】への求核試薬の作用で容易に離脱してしまう が、上記エーテル結合によつてそのような変化が
防止され、上記基を分子内に安定に保持できる。 6 実施例 次に、本発明によるフルオロ−β−ジケトン類
からなる分析試薬をその製造プロセスに従つて具
体的に例示する。 ペルフルオロカルボン酸、例えばペルフルオロ
−2−プロポキシプロピオン酸フツ化物
【式】をヘキサフルオロ −1,2−エポキシプロパンの二量化反応によつ
て得る。このペルフルオロカルボン酸フツ化物1
は次式により(−)−α−フエニルエチルアミン
と縮合させることによりジアステレオマー混合物
2を生成し、これをカラムクロマトグラフイーで
分離して得られた生成物を加水分解し、光学活性
なペルフルオロカルボン酸1′に光学分割する。 なお、本発明者の検討によれば、このペルフル
オロカルボン酸1′において上記Rfとしてペルフ
ルオロイソプロピル基を適用した場合には混合物
2から非常に良好に1′を光学分割できることが
分つている。このうちペルフルオロ−2−イソプ
ロポキシプロピオン酸については、本出願人が先
に特願昭56−155440号による次の反応に従つて合
成されたフツ化物3を経て得るのがよい。 一方、次式の如く酢酸tert−ブチル4から得ら
れるエノレートアニオン5はカルボン酸塩化物か
ら相当するβ−ジケトンを得る有用な試薬である
ことが知られている。 そこで、本発明者は、光学分割された上記ペル
フルオロカルボン酸のうち例えば(+)−ペルフ
ルオロ−2−プロポキシプロピオン酸1′を五塩
化リンによつてその酸塩化物:
【式】6に導き、こ れを次式に従つて反応させ、目的とするキラルな
β−ジケトン−ジ(ペルフルオロ−2−プロポキ
シプロピオニル)−メタン7を合成した。このジ
ケトン7は好収率(例えば90%)で得ることがで
きる。 こうして得られた(+)−β−ジケトン−ジ
(ペルフルオロ−2−プロポキシプロピオニル)−
メタン7は、更に常法に従つて相当する下記の
(+)−ユーロピウムキレート8に導いた。 このユーロピウムキレート8を加えて、幾つか
の基質の1HNMRを調べた結果を下記表−1に
示す。 【表】 この結果によれば、シフト試薬としてのユーロ
ピウムキレート8が、非常に低濃度であるにも拘
らず、満足すべき△△δ値(エナンチオマー間の
シフト差の大きさ)が得られ、優れたシフト効果
を示すことが分る。これに反し、公知のトリス−
(3−ヘプタフルオロブチフル−d−カンフアラ
ト)ユーロピウムをシフト試薬として加えた場
合には、α−フエニルエチルアミンを基質とした
とき、シフト試薬/基質のモル比を0.40〜0.60と
高濃度にしてはじめて△△δ=0.17ppmが得られ
る。このことは、本発明のフルオロ−β−ジケト
ンを用いたシフト試薬は、公知のものに比べて1/
4〜1/6の濃度で済むことを意味し、本発明の優位
性を充二分に示している。 また、本発明によるシフト試薬8は、基質とオ
ーバーラツプする水素原子を有していないため
に、非常にきれいな1HNMRスペクトラムを得る
ことができることも分つた。即ち、シフト試薬8
は、基質が示す1HNMRスペクトルと重なるよう
な1HNMRスペクトルを示さないため、シフト試
薬8を加えて得られた1HNMRスペクトルはシフ
ト試薬8の水素原子の影響を受けずに非常にきれ
いに得られ、シフト効果の判別が容易となる。し
かもこのシフト試薬は、四塩化炭素等の非極性有
機溶媒によく溶け、特に1,1,2−トリクロロ
トリフルオロエタン等のフツ素系溶媒に対しては
無尽蔵に溶解し、溶液中で安定に保持できる。従
つて、NMR分析を行なう場合には、上記シフト
試薬を1,1,2−トリクロロトリフルオロエタ
ン溶液としてマイクロシリンジ等で加えることが
できる。 なお、上記ユーロピウムキレート8は、上記し
た如き顕著な利点を有している他、その構造につ
いても不斉炭素を夫々有する一対のペルフルオロ
プロポキシエチル基【式】が互いに対 称的になつているので、その立体効果によつて分
子構造が安定化しており、また不斉中心が2つあ
るために優れたシフト効果を発揮する。このユー
ロピウムキレートは、NMR分析時のシフト試薬
としてスペクトラムを低磁場側へシフトさせる働
きがあるが、逆に高磁場側へシフトさせるために
上記のEuに代えてPr(プラセオジム)をキレート
の中心原子としたプラセオジムキレートを形成す
ることができる。 次に、本発明を実施例について更に詳細に説明
するが、以下の実施例は本発明の技術的思想に基
づいて種々変形が可能であることが理解されよ
う。 実施例 1 光学活性なペルフルオロ−2−プロポキシプ
ロピオン酸の合成: ペルフルオロ−2−プロポキシプロピオン酸フ
ツ化物13.28g(40mmol)を水浴中で、(−)−
α−フエニルエチルアミン4.84g(40mmol)お
よびトリエチルアミン4.04g(40mmol)を含む
ジクロルメタン溶液(50ml)に滴下した。滴下
後、混合物を室温でさらに30分間撹拌した。反応
混合物を水および1規定塩酸で洗浄し、乾燥後に
溶媒を除去して、油状のジアステレオマー混合物
((+)−(−)および(−)−(−))を得た。 この混合物をヘキサン−ベンゼン混合溶媒
(3:1)を用いてシリカゲルカラムにより分離
した。第一のフラクシヨンとして(−)−(−)ア
ミドを6.23g(72%)得た。このアミドはm.
p.55.5〜56.5℃、〔α〕20 D−82.0゜(C1.00,C6H6)で
あつた。また、(+)−(−)アミドを第二のフラ
クシヨンとして6.06g(70%)得た。これはm.
p.83〜83.5℃、〔α〕20 D−88.0゜(C1.00,C6H6)であ
つた。 両者はIRスペクトルにおいてはともに、3300
cm-1および1700cm-1にそれぞれNHおよびC=0
の特性吸収を示した。1HNMR(in CCl4)におい
ても、両者は全く同様なパターンを示した
(δ1.53(Me),5.12(CH)、6.72(NH),7.27
(Ph))。しかしながら、19FNMRでは、ペルフル
オロプロポキシ基の末端トリフルオロメチル基の
ケミカルシフトにおいて、(−)−(−)アミドの
方が(+)−(−)アミドよりも9Hz低磁場にあら
われることがわかつた。両者はまだガスクロマト
グラフイーにおいても、その保持時間に差がある
ことから、19FNMRおよびガスクロマトグラフイ
ーにより純度を決定することができる。 純粋な(+)−(−)アミド4.33g(10mmol)
を、10%の水酸化ナトリウムを含むエタノール−
水混合溶媒(40ml)中に加え、16時間加熱還流さ
せた。反応液を塩酸で中和し、生成したカルボン
酸をエーテル抽出した。抽出液を硫酸マグネシウ
ムで乾燥したのち、エーテルを除去し、残留物を
減圧蒸留して、(+)−ペルフルオロ−2−プロポ
キシプロピオン酸2.28g(69%)を得た。(α28 D+
26.50゜(neat,l=1))。 同様にして(−)−(−)アミドから、(−)−ペ
ルフルオロ−2−プロポキシプロピオン酸2.38g
(72%)を得た。(α28 D−26.30゜(neat,l=1))

オン酸3.30g(10mmol)を氷浴中で五塩化リン
2.50g(12mmol)に滴下した。加え終わつたの
ち、室温で10分間撹拌し、注意深く蒸留すること
により、酸塩化物を3.24g(93%)で得た。この

プロピオニル(メタン)の合成: イソプロピルマグネシウムクロリド(40m

た。これに酢酸tert−ブチル4.64g(40mmol)
を室温で適下した。反応は発熱的であり、すみや
かにプロパンガスが発生した。エーテルが自己還
流するように滴下速度を調節した。滴下終了後、
さらに1時間撹拌したのち、(+)−ペルフルオロ
−2−プロポキシプロピオン酸塩化物6.54g(18
mmol)を滴下した。滴下後、15分間室温で撹拌
したのち、希塩酸でクエンチした。有機層を分離
したのち、硫酸マグネシウムで乾燥した。エーテ
ルおよび過剰の試薬を留去したのち、少量のp−
トルエンスルホン酸を加え、混合物を120℃で15
分間加熱した。残留物を蒸留して目的物を得た。
この生成物の物性値、スペクトルは次の通りであ

ペンタン抽出液を水で洗い、硫酸マグネシウムで
乾燥した。溶媒を減圧留去し、さらに90℃(0.8
mmHg)で数時間放置したところ、黄色い油状物
を得た。次のIRおよびNMRスペクトルより、ほ
ぼ純粋な目的物(ユーロピウムキレート)である

を幾つかの基質の1HNMRのシフト試薬として常
法に従つて適用し、その結果を下記表−2に示し

ピウムキレートが、非常に低濃度であるにも拘ら
ず、満足すべき△△δ値(エナンチオマー間のシ
フト差の大きさ)が得られ、優れたシフト効果を
示すことが分る。これに反し、公知のトリス−
(3−ヘプタフルオロブチリル−d−カンフアラ
ト)ユーロピウムをシフト試薬として加えた場
合には、α−フエニルエチルアミンを基質とした
とき、シフト試薬/基質のモル比を0.40〜0.60と
高濃度にしてはじめて△△δ=0.17ppmが得られ
た。このことは、本発明のフルオロ−β−ジケト
ンを用いたシフト試薬は、公知のものに比べて1/
4〜1/6の濃度で済むことを意味し、本発明の優位

バーラツプする水素原子を有していないために、

リンジ等で加えることができた。
