本発明は下記式()でしめされる(±)−4
−ヒドロキシ−3−メチル−2−2′−プロピニル
−2−シクロペンテノンの生化学的光学分割法に
関する。さらに詳しくは、アルスロバクター属、
シユードモナス属、アクロモバクター属、アルカ
リゲネス属またはトリコデルマ属に属する微生物
が生産するエステラーゼを(±)−4−ヒドロキ
シ−3−メチル−2−2′−プロピニル−2−シク
ロペンテノンの有機カルボン酸(炭素数1〜18個
の飽和または不飽和のカルボン酸)エステルに作
用させて不斉加水分解して、光学純度の高い光学
活性4−ヒドロキシ−3−メチル−2−2′−プロ
ピニル−2−シクロペンテノンとその対掌体のエ
ステルを得る工業的に有利な4−ヒドロキシ−3
−メチル−2−2′−プロピニル−2−シクロペン
テノンの生化学的光学分割法に関する。
本発明の対象である4−ヒドロキシ−3−メチ
ル−2−2′−プロピニル−2−シクロペンテノン
(以後、これをプロピニルシクロペンテノロンと
略称する。)は、優れた殺虫活性を有するいわゆ
る合成ピレスロイドと呼ばれる一群のエステル化
合物の重要なアルコール成分の1つとして知られ
ている。
例えば該プロピニルシクロペンテノロンの2,
2,3,3−チトラメチルシクロプロパンカルボ
ン酸とのエステルである下記式()で示される
化合物は極めて強いノツクダウン効力および致死
効力を有する優れた殺虫剤である(特公昭50−
15843号公報)。
プロピニルシクロペンテノロンは4位に不斉炭
素を有するために2種の光学異性体が存在し通常
これらのエステルとしての活性は大きく異なる。
例えば上記式()で示されるエステルにおいて
は(+)−プロピニルシクロペンテノロンのエス
テルは対応する(−)−プロピニルシクロペンテ
ノロンのエステルに比し、その殺虫効力は数倍優
れている。従つて通常の製造法により得られる
(±)−プロピニルシクロペンテノロンを工業的に
も有利に光学分割する技術が望まれている。
光学活性プロピニルシクロペンテノロンを製造
する方法としては、(±)−プロピニルシクロペン
テノロンをフタル酸の半エステルとし、次いでこ
れを光学活性アミンを反応させ、光学活性プロピ
ニルシクロペンテノロンのジアステレオマー塩を
生成させ、これを分離し、使用したアミンと半エ
ステルとして回収し、得られた半エステルを加水
分解することにより光学活性プロピニルシクロペ
ンテノロンを得る方法が知られている(特開昭56
−2929号公報)。しかし、この方法は通算収率が
低いこと、複雑な工程を必要とすること、および
高価な光学活性試薬を必要とすることなどの点
で、必ずしも充分な方法とは言い難い。
本発明者らはこれらの諸問題点を克服し工業的
にもより有利な(±)−プロピニルシクロペンテ
ノロンの光学分割法を確立すべく研究を重ねた結
果、アルスロバクター属、シユードモナス属、ア
クロモバクター属、アルカリゲネス属またはトリ
コデルマ属に属する微生物が生産するエステラー
ゼを(±)−プロピニルシクロペンテノロンの有
機カルボン酸(炭素数1〜18個の飽和または不飽
和の有機カルボン酸)エステルに作用させること
により光学純度の高い光学活性プロピニルシクロ
ペンテノロンとその対掌体のエステルが得られる
ことを見い出し、これに種々の検討を加え本発明
を完成するに至つた。次に本発明を詳細に説明す
る。本発明方法において原料として使用される
(±)−プロピニルシクロペンテノロンの有機カル
ボン酸(炭素数1〜18個の飽和の有機カルボン
酸)エステルの製造は、エステル製造の常法、例
えば(±)−プロピニルシクロペンテノロンに有
機カルボン酸の無水物を反応させる方法あるいは
有機カルボン酸クロライドを有機塩基の存在下で
反応させることなどにより容易に製造することが
できる。
本発明で使用されるエステラーゼを生産する微
生物としては、アルスロバクター属、シユードモ
ナス属、アクロモバクター属、アルカリゲネス属
またはトリコデルマ属に属する微生物であつてリ
パーゼを含む広義のエステラーゼを生産する微生
物である。
これらの各属に属する代表的な菌株名を下記に
例示するが、本発明の微生物はこれらの例示限定
されるものではない。

これらの菌株はいずれもAmerican Type
Culture Collection(ATCC)または大阪市の財
団法人醗酵研究所(IFO)に保存され、これらの
保存機関より入手することができる。
上記微生物の培養は、通常常法に従つて液体培
養を行なうことにより培養液を得る。例えば滅菌
した液体培地〔かび類、酵母類用には麦芽エキ
ス・酵母エキス培地(水1にペプトン5.0g、
グルコース10.0g、麦芽エキス3.0g、酵母エキ
ス3.0gを溶解し、PH6.5とする)、細菌類用には
加糖ブイヨン培地(水1にグルコース10.0g、
ペプトン5.0g、肉エキス5.0g、NaCl3.0gを溶
解しPH7.2とする)〕に微生物を接種し、通常20〜
40℃で1〜3日間往復振盪培養を行なう。また必
要に応じて固体培養を行つてもよい。
また、これらの微生物起源のエステラーゼのな
かには市販されているものがあり、容易に入手す
ることができる。市販エステラーゼの具体例とし
てはシユードモナス属のリパーゼ(天野製薬製)、
アルカリゲネス属のリパーゼ(リパーゼPL(各糖
産業製))、アクロモバクター属のリパーゼ(リパ
ーゼAL(各糖産業製))、アルスロバクター属のリ
パーゼ(リパーゼ合同BSL(合同酒精製))など
が挙げられる。
本発明方法を実施するに際し、(±)−プロピニ
ルシクロペンテノロンの有機カルボン酸(炭素数
1〜18個の飽和または不飽和のカルボン酸)エス
テルの不斉加水分解は、上記微生物を培養した培
養液、培養液から分離した菌体、エステラーゼを
含有する培養濾液、あるいは各種酵素分離法によ
つて菌体または培養濾液から分離した粗製エステ
ラーゼ、精製エステラーゼおよびエステラーゼ含
有抽出液または濃縮液と(±)−プロピニルシク
ロペンテノロンの有機カルボン酸エステルを混合
し、撹拌または振盪することにより行われる。ま
た、固定化菌体あるいは固定化エステラーゼも使
用することもできる。
(±)−プロピニルシクロペンテノロンの有機
カルボン酸エステルの不斉加水分解を行なう条件
としては、反応温度は10〜70℃が適当であり、好
熱菌の培養液または好熱菌の培養により得られた
耐熱性エステラーゼでは50〜65℃中温菌の培養液
または特に耐熱性を有しないエステラーゼでは20
〜50℃が好ましい。
反応時間は通常3〜48時間であるが、反応温度
を高めたり酵素量を増加させるなどにより反応時
間の短縮も可能である。
反応中のPHは好アルカリ性菌の培養液のアルカ
リ性エステラーゼではPH8〜11、好アルカリ性で
ない微生物の培養液や耐アルカリ性を有しないエ
ステラーゼではPH5〜8が好ましい。また、加水
分解によつて生成する有機カルボン酸を中和し、
反応中のPHを一定に保つために緩衝液の使用が好
ましく、リン酸ナトリウム、リン酸カリウムなど
の無機酸塩の緩衝液、酢酸ナトリウム、クエン酸
ナトリウムなどの有機酸塩の緩衝液を使用するこ
とができる。
基質である(±)−プロピニルシクロペンテノ
ロンの有機カルボン酸エステルの使用濃度は反応
液に対し1〜50wt%であり、好ましくは5〜
25wt%である。
次に、このようにして不斉加水分解反応を行つ
た後、遊離した光学活性プロピニルシクロペンテ
ノロンと未反応の対掌体エステルを分離回収す
る。この分離回収に際しては水蒸気蒸留、溶媒抽
出、分別蒸留、カラムクロマトグラフイーなどの
操作を適宜採用することができる。例えば反応液
を水蒸気蒸留し留生物をエーテル抽出するかある
いは直接反応液をエーテル、酢酸エチル、ベンゼ
ンなどの有機溶媒で抽出し、この抽出物を分別蒸
留し光学活性プロピニルシクロペンテノロンとそ
の対掌体のエステルを分離取得するか、または抽
出物をシリカゲルのカラムクロマトグラフイーに
かけ、例えばトルエン−酢酸エチル(5:1)溶
液で溶出することにより、先ず光学活性プロピニ
ルシクロペンテノロンの有機カルボン酸エステル
が分離され、次いでトルエン−酢酸エチル(2:
1)溶液で溶出を行なうことによりその対掌体の
遊離のプロピニルシクロペンテノロンが分離され
る。
また、以上のようにして分離された光学活性プ
ロピニルシクロペンテノロンのエステルは、さら
に脱アシル化することにより容易に光学活性プロ
ピニルシクロペンテノロンに導くことができる。
脱アシル化の方法としては例えばプロピニルシク
ロペンテノロンの有機カルボン酸エステルに水お
よび当量の炭酸水素ナトリウムを加え、次いで10
%HClを加え、PH5に調製した後、8時間攪拌下
に還流を行なうと容易にプロピニルシクロペンテ
ノロンが分離される。
以上、詳細に説明したように本発明方法による
(±)−プロピニルシクロペンテノロンの光学分割
は従来知られている有機合成化学的光学分割法に
比べ工程が簡略であり、また高価な光学活性試薬
を必要としないので経済的に有利である。
さらに得られる光学活性プロピニルシクロペン
テノロンの収率、光学純度が高く工業的に極めて
有利である。
次に、本発明を実施例によつてさらに詳細に説
明するが、本発明はこれによつて限定されるもの
ではない。
実施例 1〜4
(±)−プロピニルシクロペンテノロンの酢酸
エステル1.0gと表1に記載した各エステラーゼ
20mgを緩衝液(PH7.0、McIlvaine緩衝液またはPH
9.5、0.2M濃度のNa2CO3−NaHCO3緩衝液)10
mlに加え、30℃で攪拌子を用いて激しく撹拌しつ
つ反応させた。24時間反応を行なつた後、反応物
を酢酸エチルで抽出した。抽出液をガスクロマト
グラフイー(5%DEGS、1.1m、180℃)で分析
し、プロピニルシクロペンテノロンの酢酸エステ
ルとプロピニルシクロペンテノロンのピーク面積
比より加水分解率を算出し下記の結果を得た。抽
出液を濃縮し、シリカゲルカラムクロマトグラフ
イーにかけ、トルエン−酢酸エチル(5:1)溶
液で溶出を行ない未反応のプロピニルシクロペン
テノロンの酢酸エステルを分離取得し、さらにト
ルエン−酢酸エチル(2:1)溶液で溶出し、遊
離プロピニルシクロペンテノロンを取得した。
ここで得られた遊離プロピニルシクロペンテノ
ロンのうち10mgをトルエン1mlに溶解し、ピリジ
ン0.2ml、(−)−α−メトキシ−α−トリフルオ
ロメチルフエニル酢酸クロライド((−)−
MTPAクロライド)45mgを加え、一時間加熱還
留しプロピニルシクロペンテノロンの(−)−
MTPAジアステレオマーとし、ガスクロマトグ
ラフイー(シリコンDCQF−1、30mキヤピラリ
ーカラム、180℃)で光学異性体分析を行ない、
(+)−プロピニルシクロペンテノロンのジアステ
レオマーと(−)−プロピニルシクロペンテノロ
ンのジアステレオマーのピーク面積比より遊離プ
ロピニルシクロペンテノロンの光学異性体型およ
び光学純度を求めた。
一方、カラムクロマトグラフイーの操作により
得られた未反応エステルに水5ml、エステルと当
量の炭酸水素ナトリウムを加え、さらに10%HCl
水を加えPH5に調整した後、8時間攪拌下に還留
し脱アシル化を行なつた。反応液を冷却後酢酸エ
チル、水および食塩を加え抽出し、溶媒を留去後
得られた油状物をシリカゲルカラムクロマトグラ
フイーにかけトルエン−酢酸エチル(2:1)溶
液で溶出し、光学活性プロピニルシクロペンテノ
ロンを分離取得した。
このプロピニルシクロペンテノロンを上記と同
様にしてガスクロマトグラフイーにて光学異性体
比を分析し未反応エステルの光学異性体型および
光学純度を求めた。結果を表1に示す。

芽エキス3.0g、酵母エキス3.0gを溶解し、PH6.5
とする。)細菌類用(実施例8)には加糖ブイヨ
ン培地(水1にグルコース10.0g、ペプトン
5.0g、肉エキス5.0g、NaCl3.0gを溶解し、PH
7.2とする。)〕100mlを入れて殺菌した後、表4に
記載した各微生物を斜面培養から2白金耳接種
し、30℃で48時間往復振盪培養した。次いでこの
培養液に(±)−プロピニルシクロペンテノロン
の酢酸エステル7gを加え、30℃で24時間往復振
盪した後、反応液を水蒸気蒸留し、留出物をエー
テル抽出した。抽出物を濃縮し、実施例1〜4と
同様のカラムクロマトグラフイー操作により遊離
プロピニルシクロペンテノロンを取得した。この
遊離プロピニルシクロペンテノロンを実施例1〜
4と同様の操作により光学異性体分析した。その
