るに当り、触媒としてヨウ素または無機ヨウ化物
の存在下に反応させることを特徴としている。
一般式又はで示されるフエノキシカルボン
酸誘導体は医薬品の製造に極めて有用な化合物で
あり就中、カルコン誘導体及びジヒドロカルコン
誘導体は強い抗消化性潰瘍作用を示し(Chem
Pharm Bull.,27,2943(1979))非常に有用な化
合物である。すなわち本発明は、抗消化性潰瘍剤
として有用なカルコン誘導体、ジヒドロカルコン
誘導体あるいはそれらの中間体を主として製造す
るための方法に関する。
一般式あるいはで示される化合物のうちフ
エノキシ酢酸誘導体を製造するに当つて、一般式
で示されるフエノール誘導体とモノブロム酢酸
誘導体を原料とする場合には概して高収率で反応
が進行することが知られている。たとえば後述の
参考例1の如く2―ハイドロキシ―4―プレノキ
シアセトフエノンを炭酸カリウムの存在下、モノ
ブロモ酢酸エステルと反応させると2―エトキシ
カルボニルメチルオキシ―4―プレノキシアセト
フエノンが79.6mol%の高収率で得られる。
しかしながら、原料のモノブロム酢酸誘導体は
モノクロル酢酸誘導体に比して著しく高価であ
り、経済的に問題があるのみならず、催涙性が大
で製造操作上困難な点が多い。このためモノブロ
ム酢酸誘導体をモノクロル酢酸誘導体に代えるこ
とも考えられるが(特開昭53−116355号)、単に
クロル化物に変更しただけでは後述の比較例1お
よび2に示す如く目的物の収率が夫々8.9mol%
および27.7mol%と著しく低く、工業的に有効な
方法とはなり得ない。
本発明者らは、かかる実情に鑑みクロル化物を
利用した効果的な合成法につき鋭意研究を行つた
結果、モノクロル酢酸誘導体を用いる場合に、触
媒としてヨウ素または無機ヨウ化物を存在させる
ことの有効性を見出し、更に検討を進めたところ
酢酸誘導体に限られないことを知り本発明に到達
した。即ち、本発明は、ヨウ素または無機ヨウ化
物を触媒として反応系に存在させることにより、
ブロム化物に比してより安価でかつ取扱いの容易
なモノクロルカルボン酸誘導体を一方の原料とし
て、短かい反応時間にも拘らず高い収率で目的物
を得ることを可能とするものであり、フエノキシ
カルボン酸誘導体の経済的な製造法をもたらすも
のである。
本発明方法で用いられる無機ヨウ化物とは、ヨ
ウ化アンモニウムおよび元素の周期律表の族a
およびb、族aおよびb、族aおよびb、
族aおよびb、V族のおよびb、族aおよび
b、族b、族に属する各種元素とヨウ素との
化合物を指称するものであり、たとえばヨウ化リ
チウム、ヨウ化ナトリウム、ヨウ化カリウム、ヨ
ウ化ルビジウム、ヨウ化銅、ヨウ化カルシウム、
ヨウ化セシウム、ヨウ化ストロンチウム、ヨウ化
マンガン、ヨウ化コバルト、ヨウ化クロム、ヨウ
化セリウム、ヨウ化鉄、ヨウ化バリウム、ヨウ化
スズ、ヨウ化亜鉛、ヨウ化ニツケル、ヨウ化マグ
ネシウム、ヨウ化ビスマス、ヨウ化銀、ヨウ化イ
ツトリウム、ヨウ化アルミニウム、ヨウ化ホウ素
等が挙げられるが、ここに記載の化合物に限定さ
れるものではなく、またそれらの化合物は単用に
限られず、2種以上を併用して有益な場合もあ
る。
本発明に於いてフエノール誘導体とは、一般式

尚、以上の如きフエノール誘導体において芳香
環の3位3′位等が既述のR1によつて置換されたフ
エノール誘導体も同様に採用可能である。
又、モノクロルカルボン酸誘導体は例えば一般
式ClCH2COOR4(但し、R4は前述の定義のとうり
であるものとする)で表わされるモノクロル酢酸
誘導体、これを具体的に例示するならば、モノク
ロル酢酸、モノクロル酢酸メチル、モノクロル酢
酸エチル、モノクロル酢酸―n―プロピル、モノ
クロル酢酸―iso―プロピル、モノクロル酢酸―
n―ブチル、モノクロル酢酸―iso―ブチル、モ
ノクロル酢酸―n―ヘキシル等や、または3―ク
ロルプロピオン酸メチル、4―クロル―n―酪酸
エチル、6―クロル―n―カプロン酸メチル等が
挙げられるが、これらの化合物に限定されるもの
ではない。
又、本発明の反応に於いて使用される塩基とし
ては、アルカリ金属及びアルカリ土類金属の水酸
化物、炭酸塩、炭酸水素塩、水素化物及びアルコ
ラートが挙げられる。これらを具体的に例示する
ならば、水酸化ナトリウム、水酸化カリウム、水
酸化マグネシウム、水酸化カルシウム、炭酸ナト
リウム、炭酸カリウム、炭酸バリウム、炭酸カル
シウム、炭酸水素ナトリウム、炭酸水素カリウ
ム、炭酸水素マグネシウムカリウム、水素化ナト
リウム、水素化カルシウム、ナトリウムエチラー
ト、カリウムエチラートなどであるが、これらの
化合物に限定されるものではない。更にまた、そ
れらの化合物は単用に限られず、2種以上を併用
し得ることは勿論である。
更に本発明の反応に於いては適当なる溶媒を希
釈剤として用いてもよく、アセトン、メチルエチ
ルケトン、メチル―iso―ブチルケトンの如きケ
トン類、ジエチルエーテル、テトラヒドロフラ
ン、1,2―ジメトキシエタンの如きエーテル
類、ベンゼン、トルエンの如き芳香族炭化水素、
n―ヘキサンの如き脂肪族炭化水素、N,N―ジ
メチルホルムアミド、ジメチルスルフオキシド、
ヘキサメチルホスホリツクトリアミドの如き極性
非プロトン溶媒などが利用し得るものの例であ
る。
更にまた、本反応を窒素、アルゴン等の不活性
ガス雰囲気下に於いて行なう時は、空気と接触し
て起こる副生成物の生成を抑制し、目的物たるフ
エノキシカルボン酸誘導体の品質を向上せしめ得
るが、このことは決して空気雰囲気下での反応を
妨げることを意味するものではない。
また、この場合、必要によりアスコルビン酸、
エリソルビン酸、ハイドロキノンモノメチルエー
テル、ビタミンE、ジブチルヒドロキシトルエ
ン、ブチルヒドロキシアニソール、p―tert―ブ
チルカテコール、ギ酸、亜硫酸塩、酸性亜硫酸
塩、チオ硫酸塩、次亜硫酸塩等の酸化防止剤の1
種以上を適宜に併用してもよい事は勿論である。
本発明方法を更に詳細に説明すれば、フエノー
ル誘導体1モルに対しモノクロルカルボン酸誘導
体1〜5モル、塩基1〜5モル、ヨウ素あるいは
無機ヨウ化物0.0001〜0.5モル、溶媒0〜50モル
を加え、室温〜140℃好ましくは室温〜100℃の温
度に於いて5〜20時間、常圧あるいは加圧下に反
応を行なうものである。尚、原料の仕込割合は反
応方法、反応温度、反応時間等上記範囲内に限定
されるものではない。
また、本発明の実施に当り、予めフエノール誘
導体と塩基とを反応せしめ、フエノール誘導体の
アルカリ塩もしくはアルカリ土類塩を調製し、こ
れを単離したあと或いは単離せずにそのまま、こ
の塩にモノクロルカルボン酸誘導体を仕込んで反
応を行うものであつても良い。この場合に触媒
は、反応の前段、後段のいずれの時期に添加して
も構わない。
更にまた、本反応を水の存在下またはアルカリ
金属水酸化物もしくはアルカリ土類金属水酸化物
の存在下に行う場合においては、モノクロルカル
ボン酸エステルを原料とするのであつても、生成
するフエノキシカルボン酸誘導体が遊離のカルボ
ン酸として得られることもある。そして本反応を
アルコール類の存在下に行う場合においては、モ
ノクロルカルボン酸を原料とするのであつても、
生成するフエノキシカルボン酸誘導体が一部エス
テルとして得られることもある。
又、本発明に於いて水の存在は必要ないが、存
在してもさしつかえない。以上、詳述した如く、
本発明の方法は、安価でかつ取扱いの容易なモノ
クロルカルボン酸誘導体を用いてフエノキシカル
ボン酸誘導体を高収率で製造できるので工業的に
有利な方法である。
以下に本発明を実施例により更に具体的に説明
し、本発明の効果をより明らかにするが、これら
の実施例によつて本発明の範囲が限定されるもの
ではない。
実施例 1
還流冷却器、温度計、窒素ガス導入管及び撹拌
装置を付した100mlの四ツ口フラスコに2―ハイ
ドロキシ―4―プレノキシアセトフエノン2.20g
(10mmol)、モノクロル酢酸メチル1.68g、無水
炭酸カリウム2.76g、ヨウ化ナトリウム0.16g及
びアセトン20gを仕込み窒素雰囲気下に於いて60
℃の温度に10時間撹拌を続けながら反応を行なつ
た。
反応終了後、無機物を別し、ケーキをアセト
ンにて洗浄を行ない、液と合併した後、アセト
ンを減圧下に留去した。残液をn―ヘキサン―ト
ルエン混液より再結晶し、2―メトキシカルボニ
ルメトキシ―4―プレノキシアセトフエノン2.31
g(収率79.0mol%)を得た。NMR(CDCl3),

参考例 1
還流冷却器、温度計、窒素ガス導入管及び撹拌
装置を付した100mlの四ツ口フラスコに2―ハイ
ドロキシ―4―プレノキシアセトフエノン2.20g
(10mmol)、モノブロム酢酸エチル2.00g
(12.0mmol)、無水炭酸カリウム1.38g
(10mmol)及びアセトン20gを仕込み窒素雰囲
気下に於いて、60℃の温度に10時間撹拌を続けな
がら反応を行なつた。
反応終了後無機物を別し、ケーキをアセトン
にて洗浄を行ない、液と合併した後アセトンを
減圧下に留去した。残液ををn―ヘキサン―トル
エン混液より再結晶し、2―エトキシカルボニル
メトキシ―4―プレノキシアセトフエノン2.44g
(収率79.6mol%)を得た。
実施例 2
実施例1に於いて無水炭酸カリウム2.76gを50
%水酸化ナトリウム水溶液2.5gに代え、同様に
反応を行なつた。

実施例1に於いてヨウ化ナトリウムをヨウ化銅
()に代え、同様に処理して2―メトキシカル
ボニルメトキシ―4―プレノキシアセトフエノン
2.25g(収率77.0mol%)を得た。
比較例 1
還流冷却器、温度計、窒素ガス導入管、滴下ロ
ート及び撹拌装置を付した100mlの四ツ口フラス
コに2―ハイドロキシ―4―プレノキシアセトフ
エノン1.1g(5mmol)、水酸化カリウム(微粉
末)0.3gおよびアセトン20gを仕込み、撹拌し
ながらモノクロル酢酸メチル1.0gを滴下し、滴
下終了後室温で2時間撹拌しながら反応を行つ
た。
反応終了後無機物を別し、ケーキをアセトン
にて洗浄を行い、液と合併した後内部標準法を
用いてガスクロマトグラフイーにて分析を行つ
た。その結果、得られた目的物である2―メトキ
シカルボニルメトキシ―4―プレノキシアセトフ
エノンは0.13g(収率8.9mol%)であつた。また
原料である2―ハイドロキシ―4―プレノキシア
セトフエノンは1.00g(回収率90.9mol%)回収
された。
参考例 2
実施例1に於いて、ヨウ化ナトリウムを用いず
に同様の処理を行なつた処、得られた2―メトキ
シカルボニルメトキシ―4―プレノキシアセトフ
エノンは0.81g(収率27.7mol%)であつた。
実施例 4
実施例1と同様の四ツ口フラスコに2′―ハイド
ロキシ―4,4′―ジプレノキシカルコン1.96g
(5.0mmol)、モノクロル酢酸0.71g、無水炭酸ナ
トリウム1.06g、ヨウ化バリウム―2水化物0.21
g及びテトラヒドロフラン10gを仕込み、窒素雰
囲気下に於いて還流下、10時間撹拌を続けながら
反応を行なつた。
反応終了後無機物を別し、ケーキをテトラヒ
ドロフランにて洗浄を行ない、液と合併した後
テトラヒドロフランを減圧下に留去した。残査を
エタノールより再結晶し、淡黄色結晶の2′―カル
ボキシメトキシ―4,4′―ジプレノキシカルコン
1.23g(収率54.6mol%)。

実施例 5
実施例1と同様のフラスコに水素化ナトリウム
(60%)0.2gをn―ヘキサン5gに懸濁させて仕
込み窒素ガスを通しながら氷冷下に、2―ハイド
ロキシ―4―ゲラノキシアセトフエノン1.44gを
N,N―ジメチルホルムアミド10gに溶解させた
ものを30分を要して滴下した。滴下終了後、室温
で30分撹拌し、次いでヨウ化カリウム0.08g及び
モノクロル酢酸エチル0.92gをN,N―ジメチル
ホルムアミド10gに溶解させたものを30分を要し
て滴下し、更に室温で5時間反応させた。
反応終了後希塩酸を加えて酸性とし、エーテル
抽出を行ないエーテル層を水洗した後無水硫酸ナ
トリウムで乾燥した。
エーテル留去後の残査をエーテル―n―ヘキサ
ン混液より再結晶した処、2―エトキシカルボニ
ルメトキシ―4―ゲラノキシアセトフエノン1.33
gを得た。
一方、ヨウ化カリウムを用いない場合、得られ
た2―エトキシカルボニルメトキシ―4―ゲラノ
キシアセトフエノンは0.24gであつた。
実施例6〜21、比較例2〜3
種々のフエノール誘導体、触媒、溶媒及び塩基
を用いて実施例1と同様に反応を行なつた。
その結果を第1表に示す。尚、モノクロル酢酸
誘導体としてはモノクロル酢酸メチルを7.5mmol
用いた。分析は内部標準を用い、高速液体クロマ
