物の存在下に反応を行うことを特徴とする、フエ
ノキシ化合物誘導体を製造する方法に関するもの
である。 一般式の化合物および一般式もしくはの
化合物より製造されるフエノキシ化合物誘導体
は、医薬品の製造に極めて有用な化合物であり、
就中、カルコン誘導体及びジヒドロカルコン誘導
体は強い抗消化性潰瘍作用を示し(Chem
Pharm Bull.、27、2943(1979))非常に有用な化
合物である。すなわち、本発明は主として抗消化
性潰瘍剤として有用なカルコン誘導体、ジヒドロ
カルコン誘導体あるいはそれらの中間体の製造方
法に関する。 従来一般式で示されるある種合の化合物より
フエノキシ化合物誘導体を製造するに当つて、一
般式で示されるある種のフエノール誘導体と臭
化プレニルまたはモノブロム酢酸誘導体を原料と
する場合には、概して高収率で反応が進行するこ
とが知られている。 たとえば、後述の参考例1の如く、2,4−ジ
ハイドロキシアセトフエノンを炭酸カリウムの存
在下、臭化プレニルと反応せしめると、2−ハイ
ドロキシ−4−プレノキシアセトフエノンが
75.8mol%の高収率で得られ、また参考例2の如
く、2−ハイドロキシ−4−プレノキシアセトフ
エノンを炭酸カリウムの存在下、モノブロム酢酸
エチルと反応せしめると、2−エトキシカルボニ
ルメトキシ−4−プレノキシアセトフエノンが
79.6mol%の高収率で得られる。 しかしながら原料の臭素化物は塩素化物に比し
て著しく高価であり、経済的に問題があるのみな
らず、臭化(ポリ)プレニルにおいては塩化(ポ
リ)プレニルに比してその安定性が遥かに劣るた
め貯蔵が困難であつたり、またモノブロムカルボ
ン酸エステルは催涙性が大であるため、製造操作
上不都合な点が多い。 このため原料の臭素化物を塩素化物に代えるこ
とも考えられるが(特開昭53−116355号)、例え
ば塩化プレニルに関して実施例1において比較デ
ータを示した如く、またモノクロル酢酸メチルに
関しては実施例16で比較データを示した如く、単
に塩素化物に変更しただけでは目的物の収率が著
しく低く(例えば前記比較データでは収率が夫々
38.1および27.7mol%)、工業的に有効な方法とは
なり得ない。 本発明者らは、かかる実情に鑑み塩素化物を利
用した効果的な合成法につき鋭意研究を行つた結
果、前記の各塩素化物を用いる場合に触媒として
特定の第四級化合物またはクラウンエーテル化合
物を存在させることの有効性を見出し、更に検討
を進めたところ、それらの例に限られず一般式
またはで表わされる塩素化物全般に有効である
ことを知り本発明に到達した。即ち、本発明は、
反応系に相間移動触媒の機能を有することで知ら
れている第四級アンモニウムブロマイド、第四級
アンモニウムアイオダイド、第四級ホスホニウム
ブロマイド、第四級ホスホニウムアイオダイドお
よびクラウンエーテル化合物より成る群から選ば
れる少くとも1種の化合物を存在させることを特
徴とするものである。そして本発明によれば、有
機反応溶媒中において、相間移動触媒の働きによ
り反応種のイオン化性が高められ、溶媒に固相、
水溶液相等として分散する塩基による脱ハロゲン
水素が効果的に促進されることによつて、一方の
原料として臭素化物に比してより安価でかつ取扱
いの容易な塩素化物を用い、短い反応時間にも拘
らず高収率で目的物が得られ、目的とするフエノ
キシ化合物誘導体の経済的な製造法が確立され
る。 本発明に於いて使用し得る触媒は、相間移動触
媒として作用する次のものである。 (i) 第四級アンモニウムブロマイド:たとえば、
テトラアルキルアンモニウムブロマイド(テト
ラメチルアンモニウムブロマイド、テトラエチ
ルアンモニウムブロマイド、n−オクチルトリ
アルキルアンモニウムブロマイド、n−デシル
トリアルキルアンモニウムブロマイド等)、ベ
ンジルトリアルキルアンモニウムブロマイド
(ベンジルトリエチルアンモニウムブロマイド
等)、フエニルトリアルキルアンモニウムブロ

い。 又、本発明の反応に於いて使用される塩基とし

化物、炭酸塩、炭酸水素塩、酸化物水素化物、ア
ミド及びアルコラートが挙げられる。これらを具
体的に例示するならば、たとえば水酸化ナトリウ
ム、水酸化カリウム、水酸化リチウム、水酸化マ
グネシウム、水酸化カルシウム、炭酸ナトリウ
ム、炭酸カリウム、炭酸バリウム、炭酸カルシウ
ム、炭酸水素ナトリウム、炭酸水素カリウム、酸

種あるいはそれ以上を併用できる。又、ここに記
載の化合物のみに限定されるものではない。 更にまた、必要により、これらの塩基は、溶媒
懸濁あるいは水溶液あるいはアルコール溶液とし
て用いることもできる。 更に、本発明の反応に於いて適当なる溶媒を希
釈剤に用いてもよく、アセトン、メチルエチルケ
トン、メチル−iso−ブチルケトンの如きケトン
類、ジエチルエーテル、テトラヒドロフラン、
1,2−ジメトキシエタンの如きエーテル類、n
−ヘキサン、ベンゼン、トルエンの如き炭化水素
類、水、メタノール、エタノールの如きアルコー
ル類、N,N−ジメチルホルムアミド、ジメチル
スルフオキシド、ヘキサメチルホスホリツクトリ
アミドの如き極性非プロトン溶媒などが利用し得
るものの例である。 更にまた、本反応を窒素、アルゴン等の不活性
ガス雰囲気下に於いて行なう時は、空気と接触し
て起こる副生成物の生成を抑制し、目的物たるフ
エノキシ化合物誘導体の品質を向上せしめ得る
が、このことは決して空気雰囲気下での反応を妨
げることを意味するものではない。また、この場
合、必要により、アスコルビン酸、エリソルビン
酸、ハイドロキノンモノメチルエーテル、ビタミ
ンE、ジブチルヒドロキシトルエン、ブチルヒド
ロキシアニソール、p−tert−ブチルカテコー
ル、ギ酸、亜硫酸塩、酸性亜硫酸塩、チオ硫酸
塩、次亜硫酸塩等の酸化防止剤を併用してもよい
ことは勿論である。 本発明の実施に当つて、撹拌は普通撹拌でよい
が、連続剪断粉砕型の撹拌を行なうのも効率的で
ある。すなわち、反応系が不均一の場合、フエノ
ール誘導体と塩化プレニル、塩化ポリプレニルま
たはモノクロルカルボン酸誘導体の反応で生成し
た塩化物が塩基の表面に付着して塩基の活性を低
下せしめるものであること、つまり、連続剪断粉
砕を行なう事により、常に塩基の活性表面を保持
し得るため、反応時間が短縮されるものである。
しかしながら、このことは決して普通撹拌をさま
たげるものではない。 かかる目的に使用される連続剪断粉砕型撹拌機
として代表的なものを例示するならば、スイス
国、キネマテイカ社(Kinematica G.m.B.H)製
の「ポリトロン(polytron)」の如きホモジナイ
ザーがある。 本発明の方法を更に詳細に説明すれば、フエノ
ール誘導体1モルに対し、塩化プレニル又は塩化
ポリプレニル0.5〜10モル、モノクロルカルボン
酸誘導体では0.5〜5モル、塩基0.5〜10.0モル、
本発明の触媒0.0001〜0.5モル、溶媒0〜50モル
を加え、0℃〜150℃、好ましくは0℃〜100℃の
温度に於いて1〜20時間、常圧あるいは加圧下に
反応を行なうものである。尚、原料の仕込割合、
反応方法、反応温度、反応時間等は上記範囲内に
限定されるものではない。 又、本発明の実施に当たり、あらかじめフエノ
ール誘導体と塩基を反応せしめ、フエノール誘導
体のアルカル塩もしくはアルカリ土類塩を調製
し、これを単離した後あるいは単離せずにそのま
ま塩化プレニル、塩化ポリプレニルまたはモノク
ロルカルボン酸誘導体と反応せしめてもよい。 以上、詳述した如く、本発明の方法は安価で、
かつ取扱いの容易な塩素化合物を用いてフエノキ
シ化合物誘導体を高収率で製造できるので工業的
に有利な方法である。 以下に参考例および実施例を挙げ、本発明を更
に具体的に説明し、本発明の効果をより明らかに
するが、これらの実施例によつて本発明の範囲が
限定されるものではない。 参考例 1 還流冷却器、温度計、窒素ガス導入管及び撹拌
装置を付した100mlの四ツ口フラスコに2,4−
ジハイドロキシアセトフエノン1.52g
(0.01mol)、無水炭酸カリウム1.66g、アセトン
20g及び臭化プレニル1.98gを仕込み、窒素雰囲
気下に於いて60℃の温度に4時間、撹拌を続けな
がら反応を行なつた。 反応終了後、無機物を別し、ケーキをアセト
ンにて洗浄を行ない、液と合併した後、アセト
ンを減圧下に留去した。残渣をn−ヘキサンより
再結晶し、2−ハイドロキシ−4−プレノキシア
セトフエノン1.67g(収率75.8mol%)を得た。 参考例 2 還流冷却器、温度計、窒素ガス導入管及び撹拌
装置を付した100mlの四ツ口フラスコに2−ハイ
ドロキシ−4−プレノキシアセトフエノン2.20g
(10mmol)、モノブロム酢酸エチル2.00g(12.0
mmol)、無水炭酸カリウム1.38g(10mmol)及
びアセトン20gを仕込み、窒素雰囲気下に於いて
60℃の温度に10時間撹拌を続けながら反応を行な
つた。 反応終了後、無機物を別し、ケーキをアセト
ンにて洗浄を行ない、液と合併した後、アセト
ンを減圧下に留去した。残液をn−ヘキサン−ト
ルエン混液より再結晶し、2−エトキシカルボニ
ルメトキシ−4−プレノキシアセトフエノン2.44
g(収率79.6mol%)を得た。 実施例 1 還流冷却器、温度計、窒素ガス導入管及び撹拌
装置を付した100mlの四ツ口フラスコに2,4−
ジハイドロキシアセトフエノン1.52g
(0.01mol)、無水炭酸カリウム1.66g、テトラエ
チルアンモニウムアイオダイド0.13g、アセトン
20g及び塩化プレニル(純度76.5%)1.64gを仕
込み窒素雰囲気下に於いて60℃の温度に3時間撹
拌を続けながら反応を行なつた。 反応終了後、無機物を別し、ケーキをアセト
ンにて洗浄を行ない、液と合併した後、アセト

キシカルボニルメトキシ−4−プレノキシアセト
フエノンは0.13g(収率8.89mol%)であつた。
又、原料である2−ハイドロキシ−4−プレノキ
シアセトフエノンは1.00g(回収率90.9mol%)
回収された。 実施例22〜29および比較例4〜5 種々のフエノール誘導体、触媒、溶媒及び塩基
を用いて実施例16あるいは実施例18と同様に反応
を行なつた。 その結果を第2表に示す。尚、モノクロル酢酸
メチルを7.5mmol用いた。分析は内部標準を用
い、ガスクロマトグラフイーあるいは高速液体ク
ロマトグラフイーにて行なつた。
