ジアゾニウムイオウを経由して変換出来るニトロ
基でもよい。 更にフリーデルクラフト反応は上に述べたアロ
イルハライドのかわりに遊離酸又はその対応する
酸無水物に対して本質的に同じ反応条件を使用し
て行なうことが出来る。これらの代替反応はオラ
ー(Olah)の「フリーデルクラフト及び関連反

適当なイミダゾール−2−オンを、約1当量乃至
約10当量の、好ましくは約2当量のリユイス酸触

化炭化水素;1,2,4−トリクロロベンゼン又

量の適当なアロイル化合物を好ましくは滴下によ

は反応体、溶媒及び約−78゜乃至約150℃好ましく
は約0乃至約100℃、最も好ましくは約60℃の温
度に依存する。 生じるアロイルイミダゾール−2−オンは反応
混合物から任意の適当な技術で知られた手順によ
つて、好ましくは反応混合物を氷水で冷却し、続
いて生成物をろ過で除くか又は抽出及び溶媒除去
により単離する。 ここに記されるフリーデルクラフト反応に使用
するのに有用なリユイス酸触媒は例えば金属例え
ばアルミニウム、セシウム、銅、鉄、モリブデン
タングステン、又は亜鉛;燐酸、硫酸、スルホン
酸、又はハロゲン化水素酸、例えば塩酸、臭化水
素酸などのブロンステツド(Bronstead)酸、ク
ロロ酢酸又はトリフルオロ酢酸などのハロゲン置
換酢酸;又は金属ハロゲン化物例えばハロゲン化
ホウ素、塩化亜鉛、臭化亜鉛、塩化ベリル

ペリジノ、モルホリノ、N′−アルキル−ピペラ
ジノ及びNR3R4である式1の化合物は上述の様
に製造されるか又は式6のフルオロベンゾイルイ
ミダゾール−2−オンの適当なものから製造され
る。 式中R及びR1は式1に定義の通りであり、フ
ツ素原子はオルソ又はパラ位にある。 式6の適当な化合物は約1乃至約10当量のピロ
リジン、ピペリジン、モルホリン、又はN′−ア
ルキル−ピペラジンと適宜反応させられる。この
反応は溶媒と共に又は溶媒なしで行なうことが出
来、好ましくはアミンが溶媒でしかも反応体であ
ることである。もし望むならこの反応に適当であ
る溶媒は例えばジメチルホルムアミド、ジメチル
スルホキシド、石油エーテル、クロロホルム、塩
化メチレン、又は四塩化炭素などの塩素化炭化水
素、二硫化炭素、ジエチルエーテル、テトラヒド
ロフラン、又はp−ジオキサンなどのエーテル性
溶媒、ベンゼン、トルエン又はキシレンなどの芳
香族溶媒、エタノールなどのアルコール溶媒であ

7の化合物は先行技術で知られているか又は式4
のイミダゾール−2−オンをニトロ置換ハロゲン
化ベンゾイル、好ましくはニトロ置換塩化ベンゾ
イルで、上に概略を述べたと類似の手順でフリー
デルクラフトアシル化することにより製造するこ
とが出来る。ニトロ基は任意の適当な技術で知ら
れた手順によつて未置換アミノ基に還元され、そ
れに続いて望むならば末置換アミノは任意の適当
な技術で知られた方法でアルキル化出来る。 ニトロベンゾイルイミダゾール−2−オンは対
応するアミノベンゾイルイミダゾール−2−オン
に、すず、亜鉛、鉄又は他の適当な活性金属で濃
塩酸中溶液で還元することにより適宜変換でき
る。約1当量を約10当量の金属に対して使用し、
反応を約1/2〜約10時間、好ましくは2又は3時
間、反応体及び約25乃至約150℃、好ましくは約
100℃であり得る温度に依存して進行させる。代
わりの方法としてニトロベンゾイルイミダゾール
−2−オンはニツケル、プラチナ、パラジウム又
は他の同様な適当な金属、及び分子状水素で接触
的に還元出来る。その様な反応は典型的にはアル
コール溶媒、好ましくはエタノール中で行なわれ
るが任意の非反応性溶媒を使用出来、金属触媒量
は約0.001当量乃至約0.1当量まで変わり得る。反
応体、溶媒、及び約0〜約100℃、好ましくは約
25℃であり得る温度に依存して反応は約1分乃至
約1時間好ましくは約10分間進行させられる。別
の方法としてニトロベンゾイルイミダゾール−2
−オンはアンモニア水溶液中でアンモニウムビサ
ルフアイト(NH4 SH)で還元することが出来
る。約1乃至約10当量の好ましくは約3当量のビ
サルフアイトを、反応体及び約0〜約150℃、好
ましくは約50℃の反応温度にもよるが約1/2時間
〜約10時間好ましくは約2時間反応させる。最後
にニトロベンゾイルイミダゾール−2−オンは任
意の他の適当な技術で知られた手順によつて対応
アミノ化合物に還元出来る。 未置換のアミノベンゾイルイミダゾール−2−
オンのアルキル化は例えば1又はそれ以上の当量
の式R3X及びR4X(R3とR4は式1に定義した通り
でXはハライドである)の適当なハロゲン化アル
キルとの反応によつて達成することが出来る。典
型的にはこれらの反応は石油エーテル;四塩化炭
素、クロロホルム、又は塩化メチレンなどの塩素
化炭化水素;1,2,4−トリクロロベンゼン、
0−ジクロロベンゼン又はクロロベンゼンなどの
塩素化芳香族;二硫化炭素;ニトロベンゼン;ジ
メチルホルムアミド;ジメチルスルホキシド、ジ
エチルエーテル、テトラヒドロフラン、又はp−
ジオキサンなどのエーテル性溶媒;ベンゼン、ト
ルエン、キシレンなどの芳香族溶媒;メタノー
ル、エタノール又はプロパノールなどのアルコー
ル;水性エタノールなどの水性アルコールなどの
溶媒中で実施される。これらのアルキル化は好ま
しくは1又はそれ以上の当量のトリエチルアミ
ン、ピリジン、水酸化ナトリウム、水酸化カルシ

せジメチルアミン化合物を生成するなどしてアル
キル化出来る。更にハロゲンやヒドロキシなどの
多種多様の他の置換基を式7のニトロ置換ベンゾ
イルイミダゾール−2−オンからジアゾニウムイ
オンを経由して技術に於て良く知られた手順で製
造出来る。 X1又はX2及びX3がヒドロキシである式1の化
合物は上に記載の通りに製造でき、又は好ましく
は適当なアルコキシ、好ましくはメトキシで置換
のベンゾイルイミダゾール−2−オンであつてア
ルコキシ基が所望ヒドロキシ置換の位置にあるも
のから製造出来る。アルコキシ化合物はアール
エル ブルウエル(R.L.Burwell)「エーテルの
開裂」Chem・Rev・54 615−85(1954)(この内
容を本明細書に参照により特に取り入れる)によ
り教えられる様な適当な技術で知られている任意
の手順で開裂して対応するヒドロキシベンゾイル
イミダゾール−2−オンを生成する。 式5aと5cの反応体の安定性を良くするため、
又は任意の反応性X基のアシル化を同時に起こす
ことなくここに記したイミダゾール−2−オン環
窒素原子のアシル化を可能にするために、X1,
X2及びX3置換基は必要に応じて保護されてよい。
例えばX1,X2又はX3がヒドロキシであるか、−
NHR3又は−SO2NH2のアミノ基であるときは、
保護しないならば反応性であるヒドロキシ又はア
ミノ基を封鎖するのにベンジル基を使用すること
が出来る。ベンジル基は続いて例えばパラジウム
触媒上の水素による水素添加分解又は液体アンモ
ニア中のナトリウムで除去することが出来る。 望むならばイミダゾール−2−オン環の窒素原
子の1個又は両方を任意の技術で知られた手順で
アルキル基で置換することが出来る。その様な方
法は本発明の適当なN−未置換アロイルイミダゾ
ール−2−オンを塩基及びアルキル化剤と非反応
性溶媒の存在下に反応させることを含む。この反
応の適当な塩基は例えば水素化ナトリウム、又は
水素化カルシウムなどの水素化物;炭酸ナトリウ
ム又は重炭酸ナトリウムなどの炭酸塩又は重炭酸
塩;ナトリウムフエノキシドなどのフエノキシ
ド;ナトリウムエトキシドなどのアルコキシド;
又は好ましくは水酸化ナトリウムなどの水酸化物
であり得る。この反応の適当なアルキル化剤は例
えば塩化メチル、臭化メチル、又はヨウ化メチル
などのハロゲン化アルキル;又はジメチルサルフ
エートなどのジアルキルサルフエートである。適
当な反応性でない溶媒は例えば石油エーテル;四
塩化炭素、クロロホルム、又は塩化メチレンなど
の塩素化炭化水素;1,2,4−トリクロロベン
ゼン、0−ジクロロベンゼン又はクロロベンゼン
などの塩素化芳香族類;二硫化炭素;ニトロベン
ゼン;ジエチルエーテル、テトラヒドロフラン、
又はp−ジオキサンなどのエーテル性溶媒;ベン
ゼン、トルエン、キシレンなどの芳香族溶媒;又
は好ましくはジメチルホルムアミド(DMF)又
はジメチルスルホキシド(DMSO)などの極性
中性溶媒である。反応は約1分から約1時間進行
させ、温度は約0℃乃至約100℃、好ましくは約
25℃である。イミダゾール−2−オン窒素原子の
ただ一つのみがアルキル基で置換されることが望
まれる場合は適当なイミダゾール−2−オンを約
1当量乃至約10当量の塩基、好ましくは約1当量
の塩基及び約1当量のアルキル化剤と反応させ
る。この手順を利用して両方の可能なモノアルキ
ル化窒素の異性体が生じる。 これらの異性体は慣用の技術で知られた手順で
分離出来る。例えば分別結晶化、分別蒸留、又は
クロマトグラフイーによる。イミダゾール−2−
オン環の窒素の両方共がアルキル置換されること
が望まれるときは、適当なイミダゾール−2−オ
ンを約2当量乃至約10当量の、好ましくは約2当
量の塩基、及び約2当量から約10当量の、好まし
くは約2当量のアルキル化剤と反応させる。最後
に、もし存在するならアロイル環の任意の反応性
の置換基が同時にアルキル化されるかもしれな
い。即ち次のX基、X=OH、−NHR3、SO2NH2
及び未置換ピペリジノが同一の反応条件下にアル
キル化される。もし望まれるならアロイル環置換
基のアルキル化は技術で良く知られた適当な保護
基の使用によつて避けることが出来例えばX=
OH又は−NHR3はベンジル化されそしてあとで
水素添加分解で脱封鎖され得る。 望まれる場合にはイミダゾール−2−オン環の
窒素原子は任意の適当な技術で知られた手順によ
りアルキルカルボニル基で置換されてよい。その
様な方法には本発明のN−未置換アロイルイミダ
ゾール−2−オンをハロゲン化アシル、好ましく
は塩化アセチル、塩化n−プロパノイル、塩化イ
ソプロパノイル又は塩化ブタノイルなどの塩化ア
シルと反応させることが含まれる。通常アシルハ
ライドを使用するアシル化反応はトリエチルアミ
ン又はピリジンなどの酸スポンジ(酸吸収体)を
生成してくるハロゲン化水素を除くために使用す
る。更に対応する遊離酸又は酸無水物を酸塩化物
の代わりに使用出来る。アシル化反応は一般に溶
媒の添加なしに行なわれるが、任意の反応性でな
い溶媒例えば石油エーテル;クロロホルム、塩化
メチレン、又は四塩化炭素などの塩素化炭化水
素;二硫化炭素;ジエチルエーテル、テトラヒド
ロフラン、又はp−ジオキサンなどのエーテル性
溶媒;ベンゼン、トルエン又はキシレンなどの芳
香族溶媒を使用して行なうことが出来る。反応は
約1分〜約100時間、好ましくは約1時間〜約10
時間進行させ、温度は約−78゜から約150℃、好ま
しくは0゜〜100℃でよい。 最後にもし存在するならアロイル環の任意の反
応性置換基は同時にアシル化されるようになる。
即ち、次のX基、X=OH、−NHR3、−SO2NH2
及び未置換ピペリジノは同じ反応条件でアシル化
される。もし望まれるならベンゾイル環置換基の
アシル化は技術で周知の適当な保護基の使用で避
けることが出来、例えばX=OH又は−NHR3は
ベンジル化して後に水素添加分解で脱封鎖を行な
つてよい。 本発明のアロイルイミダゾール−2−オンのア
ルカリ金属、アルカリ土類金属、遷移金属、主群
の金属、アンモニウム、又は有機アンモニウム塩
は、対応する金属又はアンモニウム塩基塩、例え
ばナトリウムメトキシド又はナトリウムエトキシ
ドなどのアルコキシド、ナトリウムフエノキシド
などのフエノキシド、水酸化ナトリウム又は水酸
化カリウムなどのヒドロキシド、又は炭酸ナトリ
ウム、炭酸カリウム、炭酸亜鉛、炭酸マグネシウ
ム、又は炭酸水素ナトリウムなどの炭酸塩からつ
くられる。これらの反応は溶媒と共に又は溶媒な
しで行ない得る。適当な溶媒は例えばメタノー
ル、エタノール、イソプロパノール、n−プロパ
ノール又はn−ブタノールなどの低級アルコー
ル、ベンゼン、トルエン又はキシレンなどの有機
溶媒、ジエチルエーテル、テトラヒドロフラン又
はp−ジオキサンなどのエーテル系溶媒及びクロ
ロホルム、塩化メチレン又は四塩化炭素などのハ
ロゲン化炭化水素である。アロイルイミダゾール
−2−オン、及び塩基は反応体及び約−78゜〜約
150℃、好ましくは約0゜〜約25℃であることの出
来る温度に依存して約1分乃至約24時間反応させ
られる。 アロイルクロライド及びそれらの対応するカル
ボン酸は、本発明のフリーデルクラフトアシル化
に必要であるが、技術において一般に入手可能で
あるかあるいは同様の手順で製造される。式4の
イミダゾール−2−オン出発物質はアール ダス
チンスキー(R.Duschinsky)及びエル エー
ドラン(L.A.Dolan)、J・Am・Chem・Soc・

又はこれを適合させたものにより製造され得る。 一般式1の化合物は欝血性心不全、後方不全、
前方不全、左心室性不全、又は右心室性不全を含
む心臓機能不全又は強心剤で心機能の強化を必要
とする任意の他の症状の治療に使用出来る。多く
の点でこれらの化合物はデイジタリス様作用を有
する。一般式1の化合物は原発性ないし本態性の
高血圧症、ホルモン誘発の高血圧症、腎性高血圧
症、及び化学的誘発の高血圧症を含む高血圧症の
治療にも使用することが出来る。最後に一般式1
の化合物は抗血栓症剤として使用出来る。これら
は血小板の集合を防げることによつて血液凝固に
影響を与えるが、血小板は初期段階及び栓塞段階
の両方に於いて血栓症症状の主要な役割を果たす
のである。動脈血栓、特に心筋及び脳に供給を行
なう動脈に於ける血栓は死亡及び不具になること
の最大の原因である。 望む効果を達成するために化合物は種々の方法
で投与出来る。化合物は単独で又は製薬学的調合
剤の形で治療されるべき患者に経口的又は非経口
的、即ち静脈内又は筋肉内に投与出来る。投与さ
れる化合物量は高血圧症、心臓機能不全又は血液
凝固のひどさ及び投与形式と共に変化するであろ
う。経口投与には化合物の抗高血圧症有効量は1
日当り約0.1mg/Kg(ミリグラム/キログラム)
乃至約500mg/Kg(患者の体重Kg当たり)で、好
ましくは1日当たり患者体重Kg当たり50mg乃至
150mgである。 非経口投与には、化合物の抗高血圧症有効量は
1日患者体重キログラム当り約0.01mg/Kg乃至
150mg/Kgで、好ましくは約0.1mg/Kg乃至約10.0
mg/Kgである。経口的又は非経口的投与には化合
物の強心剤としての有効量は1日患者の体重キロ
グラム当り約0.1mg乃至約500mgまで、好ましくは
約0.1mg/Kgから約10.0mg/Kgまでである。経口
又は非経口投与のための化合物の抗血栓症有効量
は1日患者の体重キログラム当たり0.1mg/Kg〜
約1000mg/Kgかつ好ましくは約1mg/Kg〜約100
mg/Kgである。 経口投与のために単位投与量は例えば10乃至
100mgの活性成分を含んでよい。非経口投与のた
めには単位投与量は例えば5乃至50mgの活性成分
を含有してよい。化合物の毎日の繰返し投与は望
ましく患者の症状及び投与形式によつて変化する
だろう。 ここで使用の患者という用語は温血動物、例え
ばにわとり及び七面鳥などの鳥類、霊長類、人
類、ひつじ、馬、雄牛及び雌牛、ぶた、犬、猫、
ラツト及びマウスなどの哺乳類を意味するように
解釈される。 経口投与には、化合物は固体又は液体製剤例え
ばカプセル、丸薬、錠剤、トローチ、粉剤、溶
液、懸濁液及び乳化液に処方される。固体の単位
投与形はカプセルであり得、これは例えば潤滑剤
及び乳糖、庶糖及びとうもろこし澱粉などの不活
性充填剤を含む普通のゼラチン型のものであり得
る。他の態様では式1の化合物は乳糖、庶糖、及
びコーンスターチなどの慣用の錠剤基質と共にア
ラビアゴム、コーンスターチ又はゼラチンなどの
結合剤、ばれいしよ澱粉又はアルギン酸などの崩
壊剤及びステアリン酸又はステアリン酸マグネシ
ウムなどの潤滑剤を組み合わせて錠剤にされ得
る。 非経口投与には表面活性剤及び他の製薬上受け
入れられる助剤を添加した又は添加しない水及び
油などの滅菌液体であり得る、製薬担体を有する
生理学的に受け入れられる希釈剤中の化合物の溶
液又は懸濁液の注射用投与物として投与すること
が出来る。これらの調製剤中で使用出来る油の例
は石油、動物、植物又は合成起源のもの、例えば
ビーナツツ油、大豆油、及び鉱油である。一般に
水、塩水、水性デキストロース及び関連する糖溶
液、エタノール、及びプロピレングリコール又は
ポリエチレングリコールなどのグリコール類は好

ール(20mg/Kg)又はプロプラナロール塩酸塩
(3mg/Kg)を投与することによつて犬に心臓機
能不全を誘導する。これらの心機能抑制剤のいず
れかの投与に続いて、右心房の圧力が劇的に増加
し、心臓血液搏出量がひどく降下した。表題の化
合物(1mg/Kg)の投与は右心房の圧力と心臓血
液搏出量の処理前の水準近くへの逆転によつて示
される様に機能不全を元に戻す。 実施例C 抗血栓症剤としての1,3−ジヒドロ

−2H−イミダゾール−2−オンの用途 アデノシン二燐酸をクエン酸塩加(特に抗凝固

むこと)の血小板の豊富な人間の血漿に加えた時
血小板の典型的な凝集が起る。しかしながら3,
10,30,及び100μg/mlの濃度でクエン酸塩加血
小板の豊富な人間の血漿に表題の化合物を加え、
続いてアデノシン二燐酸を加えると血小板の凝集

48時間約0゜〜約150℃で処理し、 b X1が式NR3R4(R3及びR4の各々は前に定義
の通り)のアミノ基となるべき場合に於いて
は、 式 〔R,R1、は前に定義の通りである〕の化合
物を任意の適当な方法で還元して対応するアミノ
ベンゾイルイミダゾール−2−オンを製造し、こ
れを任意の適当な方法で次にアルキル化し、 かつ製薬学的に受け入れられる塩を望む場合に
は、このようにいずれかにより生成したアロイル
イミダゾール−2−オンを適当な金属又はアンモ
ニウム塩基塩と反応させることからなる 式 〔式中Arは2−フリル、2−チエニル、オル
ソ、メタ又はパラ位置でX1でモノ置換されたフ
エニル、又はパラ位置でX2で置換され、オルソ
又はメタ位置でX3で置換されたジ置換フエニル
であり;R,R1,X1,X2及びX3は前に定義の通
り〕のアロイルイミダゾール−2−オン又は製薬
学的に受け入れられるその塩の製造方法。

A compound according to clause 6, wherein X 1 is methylthio and R 1 is methyl. A compound according to clause 3, wherein 9 Ar is di-substituted phenyl substituted in the para position by X 2 and in the ortho or meta position by X 3 . A compound according to clause 9, in which 10 X 3 is in the meta position. 11. A compound according to claim 10, wherein X 2 and X 3 are linear or branched lower alkoxy of 1 to 4 carbon atoms or together are methylenedioxy. 12 A compound according to clause 11, wherein R 1 is methyl and X 2 and X 3 are methoxy. 13 A compound according to clause 11, wherein R 1 is methyl and X 2 and X 3 are both methylenedioxy. 14 formula The compound [wherein R 1 is hydrogen or a straight or branched chain lower alkyl group of 1 to 4 carbon atoms] is added to about 1 to 10 equivalents of 2-furoyl halide, 2-
