Abstract
Description
〔産業上の利用分野〕
本発明は、新規なピラゾールアミド誘導体、その製法お
よび該誘導体を有効成分として含有する農園芸用殺菌剤
に関する。
〔従来の技術及び問題点〕
農業生産上、特に果樹、硫菜類の栽培上重要な問題の一
つとして、そう画調に属する植物病原菌に依る病害(例
えば、べと病、疫病など)がある。
これらの菌に依る病害は、その菌の生理的・生態的特殊
性より防除困難な病害の一つとなっており、優れた防除
薬剤の開発が望まれている。べと病、疫病による被害は
多くの作物にわたり、その被害は極めて大きい。
現在、そう囲網に属する植物病属国による病害に対して
は、キャブタン(一般名)、キャプタホル(−船名)、
ジチオカーバメート系殺菌剤〔例えば、ジネブ(一般名
)など〕、クロロタロニル(一般名)などがひろく使用
されている。しかし、これらの殺菌剤は予防的効果が主
であり、治病的効果は殆んど期待できないため、病害の
発生が認められた時に使用したのでは、充分な効果が期
待できないという大きな欠点をもっている。最近開発さ
れ、実用に供されているアシルアラニン系化合物の殺菌
剤、たとえばメタラキシル(−船名)などは、予防、治
病の両効果を有しているが、しかし、すでにその耐性菌
が発生しており、その防除効果は、かなり低下してきて
いる。
従来、本発明化合物に化学構造上類偵のアミド置換アセ
トニトリル誘導体については、特開昭57−16797
8号公報、特開昭58−69866号公報および特開昭
60−255759号公報等に報告されているが、この
先行技術では、実用的な農園芸用殺菌剤としては十分と
はいえず、特に有用作物に対する薬剤による薬害があり
、これが大きな未解決の問題となっている。
〔発明が解決しようとする問題点〕
本発明は、そう画調に属する植物病原菌による病害(例
えば、べと病、疫病など)に対して予防効果および治病
効果を同時に有しながらかつ、有用作物への薬害のない
殺菌剤を提供することを目的とする。
〔問題点を解決するための手段および作用〕前記の問題
点を解決するため前記のピラゾールアミド誘導体につい
て鋭意研究した結果、以下に示す一般式(1)の化合物
群が各種植物の疫病、べと病等に対して予防的、治病的
効果の両方を合わせもち、かつ作物に対する薬害が小さ
いという特徴を示すことを見出し、本発明を完成させた
。
本発明化合物は文献未記載の新規化合物である。

Bは、フリル基、チエニル基、ピラゾリル基、フェニル
基、炭素数2〜8のアルケニル基、炭素数2〜8のアル
キニル基、炭素数2〜10のアルコキシ基、炭素数2〜
8のチオアルコキシ基または炭素数3〜8のシクロアル
キル基を表す。
Dは、シアノ基またはチオカルバモイル基を表す。

原料のアルデヒド〔3〕とアンモニア水及び又は塩化ア
ンモニウムとシアン化ナトリウム、シアン化カリウム等
のシアン化アルカリとをエチルエーテル、テトラヒドロ
フラン等のエーテル類やベンゼン、トルエン等の芳香族
炭化水素類と水との二元溶媒系で反応を行わせるもので
ある。反応温度は通常0〜100°C付近が好ましい。
a−2法は、出発原料のアルデヒド〔3〕とトリメチル
シリルニトリルに代表されるトリアルキルシリルニトリ
ルを必要に応じ沃化亜鉛などのルイス酸の触媒量の存在
下反応させて中間体〔4〕を得る。続いてメタノールや
エタノール等の溶媒に溶解させたアンモニアと反応させ
てアミノアセトニトリル誘導体〔5〕を得る方法である
。
得られたアミノアセトニトリル誘導体〔5]は、ジエチ
ルエーテル等のエーテル系溶媒中、塩化水素などのハロ
ゲン化水素との塩を形成させるなどして、精製し単離す
ることもできる。
次に第2工程は、第1工程で得られたアミノアセトニト
リル誘導体〔5〕と、一般式〔6〕で表される酸ハライ
ドを酸受容体の存在下反応させる。
酸受容体としては例えばトリエチルアミン、ジメチルア
ニリン、ピリジン等に代表される有機塩基、アンモニア
、炭酸カリウム、炭酸ナトリウム、炭酸水素アンモニウ
ム、水酸化ナトリウム、炭酸アンモニウム等に代表され
る無機塩基を挙げることができる。
又、この反応は、溶媒の存在下行うのが好ましく、例え
ばジエチルエーテル、テトラヒドロフラン、ジイソプロ
ピルエーテル等に代表されるエーテル類、酢酸メチル、
酢酸エチル等に代表されるエステルL塩化メチレン、ク
ロロホルム、l、2−ジクロロエタン等に代表されるハ
ロゲン化炭化水素類、アセトニトリル等が使用できる。
反応温度は、発熱反応であるため冷却下に行うことが望
ましく、−30°C付近から50°C付近が好ましく、
実用的には一20〜30°C付近が望ましい。この様に
して、ニトリル誘導体〔7〕を得る。

たアミノアセトニトリル誘導体〔5〕とを、脱水剤の存
在下、脱水縮合しアミド置換アセトニトリル誘導体を得
る方法である。
脱水剤としては、ジシクロへキシルカルボジイミド(D
CC) 、1−エチル−3−(3−ジメチルアミノプロ
ピル)カルボジイミド等のカルボジイミド類、四塩化ケ
イ素等の無機系脱水剤等が特に好ましい。

と室温以下から室温付近で反応させてクロル体を得る。
反応後過剰量の塩化チオニルを留去した後、溶媒中一般
弐BZ(12]で表される化合物と、必要に応じて塩基
の存在下で反応させることによりエステル体〔13〕が
得られる。
ここで用いられる溶媒としては、酢酸エチル、酢酸イソ
ブチル等のエステル系溶媒およびエーテルやアセトニト
リル等も用いられる。又、塩基としてはトリエチルアミ
ン、ピリジン等の有機塩基、炭酸ナトリウム、炭酸アン
モニウム等の無機塩基が使用できる。
ついで、このエステル体(13)をアンモニアと反応さ
せることによりアミド体とする。この反応は、メタノー
ル、エタノール等のアルコール類や、ジエチルエーテル
、テトラヒドロフラン等のエーテル類の溶媒を用い、−

させることにより、ニトリル誘導体〔7〕が得られる。

アミノアセトニトリルと酸クロライド〔6〕との反応は
、通常用いられている縮合反応の条件下で、容易にアシ
ルアミノアセトニトリル誘導体〔14]を得ることがで
きる。本反応では、一般に塩基の存在が好ましく、炭酸
ナトリウム、炭酸アンモニウム等の無機塩基、トリエチ
ルアミン、ピリジン等の有機塩基等が好ましい。
アシルアミノアセトニトリル誘導体〔14〕は、ハロゲ
ン化剤によって、ハロゲン化物(15)に変換できる。
ここで用いられるハロゲン化剤としては、塩素、臭素、
N−クロロサクシイミド、N−ブロモサクシイミド等が
好ましい。このハロゲン化中間体は、熱安定性がよくな
いため、単離精製することなく、そのまま次の行程に導
く。すなわち、ハロゲン化の後、このハロゲン化中間体
〔15〕は溶媒中、必要に応じて塩基の存在下でBZ(
12)と反応させることにより、ニトリル誘導体[7]
が得られる。
この反応条件は(製法3〕の場合と同様である。

(a)〔製法1〕で得られたアミノアセトニトリル誘導
体〔5〕と硫化水素から、目的とするチオアミド誘導体
は容易に得られる。使用する塩基としては、例えばトリ
エチルアミン、ピリジン、ジメチルアニリンなどの有機
塩基が好適で、その添加量としては、原料〔5〕1モル
に対し、0.01〜1モルの範囲で反応は充分進行する
。
溶媒は、使用しなくても反応が進行する場合があるが、
通常は使用した方が好ましく、その種類としては特に限
定されるものではないが例えば、ベンゼン、トルエンな
どの炭化水素類、テトラヒドロフラン、ジオキサンなど
のエーテル類、ピリジン、ジメチルアニリンなどの芳香
族アミン類すどが好ましいものとして挙げられる。また
、これらの2種以上の混合溶媒として使用してもよい。
原料のひとつの硫化水素は通常、市販のガス状の硫化水
素をそのまま用いることができ、その送入量は、原料〔
5〕に対して当モル量または多少過剰量を送入し反応さ
せることができる0反応温度としでは、−20〜100
℃が望ましく、特に、0〜50°Cが好ましい。
反応時間は、室温で反応させた場合は通常5〜6時間で
反応が終了する。
このようにして得られた中間体〔2]は、再結晶によっ
て、容易に単離、精製することができる。
(b) (a)で得られた中間体〔2〕と酸ハライド
〔6〕との反応は、〔製法1〕℃)と同様な条件で行う
ことができる。

ニトリル誘導体〔7〕と硫化水素から、目的とするチオ
アミド誘導体 〔16〕を得る反応は、〔製法5 )
(a)と同様な条件で行うことができる。
かくして得られたピラゾールアミド誘導体は、再結晶、
カラムクロマトグラフィーなどの常法の精製法により純
品とすることができる。
次に本発明化合物の製造方法を具体例をあげて説明する
が、本発明は、これらのみに限定されるものではない。
実施例1
(化合物No、117の合成)
α−(3−フリル)アミノアセトチオアミド(3,1g
)(0,02モル)を、テトラヒドロフラン(THF)
50 mj!に溶かし、続いてトリエチルアミン(2,
4g ) (0,024モル)を加える。
この混合液を水冷下で撹拌しながら、1−シクロブチル
−3−メチル−IH−ピラゾール−5−カルボン酸クロ
ライド(4,0g) (0,02モル)を、テトラヒ
ドロフラン(THF)6.2 mlに溶かした溶液を滴
下した。
その後、室温で5時間撹拌を続けて反応終了にした。続
いて反応液を濃縮後、残渣を水洗した。
得られた結晶を更にトルエン−酢酸エチルから再結晶を
行うことにより、(4,5g) (液体クロマトグラ
フィー純度99%)の結晶をえた。
この結晶は、質量分析、’H−NMRがら目的トスるN
−(3−フリル(チオカルバモイル)メチルツー1−シ
クロブチル−3−メチル−IH−ピラゾール−5−カル
ボキサミドであることが判明した。

α−(3−フリル)アミノアセトニトリル(2,5g)
とトリエチルアミン(2,1g)をア七ト二トリル50
mle溶かし、水冷下、l−シクロブチル−3−メチ
ル−LH−ピラゾール−5−カルボン酸クロライド(2
,0g )を加え、1時間撹拌後室部で5時間撹拌した
。生じた固形物を濾別後、溶媒を留去し、得られた油状
物をカラムクロマドグラフィーにより精製し、N−(シ
アノ−3−フラニルメチル)−1−シクロブチル−3−
メチル−IH−ピラゾール−5−カルボキサミド(3,
42g)の結晶を得た。

本発明は、さらに本発明に係る前記一般式(1)で表さ
れるピラゾールアミド誘導体およびその光学活性体を有
効成分として含有することを特徴とする農園芸用殺菌剤
を提供するものである。
本発明の農園芸用殺菌剤は、各種作物のべと病、疫病に
対して有効であるばかりではなく、他の病害にも有効で
ある。主な病害としては、キユウリベト病、ブドウペと
病、レタスペと病、ハクサイへと病、ホップペと病、ジ
ャガイモ疫病、トマト疫病、キュウリ灰色疫病、ピーマ
ン疫病、ビシニウム菌によってひき起こされるトマト、
キュウリ及びイネの苗立枯病、アファノミセス菌によっ
てひきおこされるビートの苗立枯病などが挙げられる。
本発明の農園芸用殺菌剤の施用方法としては、種子処理
、茎葉散布、土壌施用などが挙げられる。
施用1!および施用濃度は、対象作物、対象病害、病害
の発生程度、施用方法などにより差異はあるが、散布に
当たっては、有効成分量として、1ヘクタール当たり2
〜2000gで適用可能であり、10〜1000gが望
ましい。
散布濃度としては、1〜11000ppで適用可能で、
5〜500pp−が望ましい。
又、本発明の農園芸用殺菌剤は、予防、治病の両効果を
所持していることから、予防的散布あるいは、発病後の
散布の両方が可能で、その散布時期としては、幅広い適
期を持っている。
本発明の農園芸用殺菌剤は、必要に応じて、他の生物活
性を有する化合物、たとえば類似の又は補完的な殺菌剤
、もしくは殺虫剤、除草剤、植物生育調節剤などの農薬
、また肥効性物質、土壌改良剤などとの混合使用はもち
ろんのこと、これらとの混合製剤も可能である。
本発明の農園芸用殺菌剤は、適当な担体、例えばクレー
、タルク、ベントナンド、珪藻土等の固体担体あるいは
水、アルコール類(メタノール、エタノール等)、芳香
族炭化水素類(ベンゼン、トルエン、キシレン等)、塩
素化炭化水素類、エーテル類、ケトン類、エステルM(
酢酸エチル等)。
酸アミド類(ジメチルホルムアミド等)などの液体担体
と混用して適用することができ、所望により乳化剤1分
散剤、懸濁剤、浸透剤、展着剤、安定剤などを添加し、
乳剤、油剤、水和剤、粉剤。
粒剤、フロワブル剤等任意の剤型にて実用に供すること
ができる。
次に、これらの組成物の配合例の各成分の範囲および各
成分の種類を示すが、本発明の農園芸用殺菌剤はこれら
のみに限定されるものではない。
なお、以下の配合例において「部」は重量部を意味する
