Abstract

51−14485号)により得られている。 しかし、これら二種の合成法のいずれによっても、その
反応混合物中には、目的とするSEの他に、未反応の糖
、未反応の脂肪酸メチルエステル、残留触媒、石鹸、M
離脂肪酸、揮発分等の夾雑物を含んでおり、これらの夾
雑物のうち含量が規定量を越す不純分は、製品と成る以
前に除去されなければならない、特に、上記夾雑物のう
ち、前者の溶媒法に伴う残留溶媒(揮発分)の除去は、
近来規制が厳しくなって来ている・1)だけに極めて重

し、たとえ低温(0〜5℃)で行っても分解を避けるこ
とができない、このため未反応糖の回収、再利用が困難
となる。 また、方法(2)のように、食塩や芒硝などの中性塩の
水溶液を反応混合物中に加えてもSEは直ちに沈殿する
。この場合、未反応勧の分解は起こらないが、SE中の
有用な成分であるモノエステルが水相側に溶解してしま
うため、大きなロスを生じるのみでなく、特に近来需要

及び製品SEのロスを生じない精製方法を開発すること

に押えることができ、たとえ本反応混合物中に含まれる

の原因として、SEのミセル集合体の形成度合と、塩の
奏する、所謂塩析効果がmHに絡み合っていることは確
かであろう。 以上の塩析作用の程度は、換言すればφの値は、5E(
7)エステル分布及びSE中の脂肪酸残基の種類によっ
て幾分変動するが、合計基量が6%(水84%)を越え

8)や、添加された中性塩や揮発分、例えばジメチルス
ルホキシド(分子量78)は、問題なく、逆浸透膜の微
細孔を通過するであろう。この推定に基づき多くの実験
を重ねた結果、前段の塩析処理を経たショ糖、触媒から
の副生塩、塩析時添加された中性塩及び揮発分並びに、
場合により少贋〜微量のSEを含む水溶液は、温度40
〜60°Cで、分画分子量150〜200近辺の逆浸透
膜に対し、駆動源として限外濾過時より大きな圧力を付
かされつつ接触せしめられたとき、触媒からの副生塩、
加えられた中性塩及び揮発分の王者は、水と共に逆浸透
膜の微細孔を容易に通過することが二・)った、この逆
浸透操作によって、不純なショ糖水溶液(場合により少
量のSEを含む)は、水、触媒からの副生塩及び塩析に
際し加えられた中性塩及び揮発分等の低分子量の物質か
ら分離され、濃縮された粗糖水溶液の形となる。そして
ここに得られた粗糖水溶液を再び新鮮な水に溶解させ、
再度(又は再三)同様の逆浸透処理に付すことにより、

pH8,2〜8.2の領域内がショ糖の品質に影響する
恐れが小さい点で好ましい。 (g−2逆浸透膜) 工業的な逆浸透膜は、近年進歩したものが各社から多数
−L市されている。これら市1′&中の膜の中、耐久性
、耐熱性、耐酸、耐アルカリ性、耐菌性及び耐圧性に優
れたものの例として、架橋ポリアミド系の逆浸透膜があ
る。そしてこの種の膜の例として、例えば、東しエンジ
ニアリング■阪売に係る逆浸透膜、商品名<(SU−2
00>)等は、前述の分画分子量200近辺の値を持ち
、本発明目的によく合致する。 大体、分画分子量が200近辺の逆浸透膜の場合、供給
される水溶液中の溶質濃度は、上限値として20%、望
ましくは、溶質濃度の上限値として15%程度に押える
ことによって、工業的な処理漬方を発揮させることがで
きる。 濃度が20%を越える溶質濃度の場合、逆浸透膜の微細
孔内を水、触媒からの副生塩及び揮発分が、通過し難く
なり、その分、駆動圧を高めることを余儀なくされるか
ら、結果的に膜面植を広くとらざるを得す、かつまた、
大動力を必要とすることになるので甚だ不経済である。 これに対し、8〜15%程度の溶質濃度であれば、工業
的なショ糖の分離は充分に可能である0例えば、下表−
2の組成の水溶液の場合、ショ糖の分離速度は、p)1

糖水溶液を得るのは、技術的に困難となる以外に、経済
性も低下してくる。従って、上記以上の糖濃度が望まれ
るならば、通常の濃縮装置1例えば、多重真空効用缶等
を用いて希望の濃度、例えば50%以上にまで濃縮する
ことができる。 (h 限外濾過) 前記塩析工程により、中性塩の添加により反応混合物水
溶液中から多くの割合で沈殿せしめられたSEは、含水
状態、即ち、泥漿(スラリー)状のものである。このも
のは、比較的少量ではあるが、なお揮発分、塩類、ショ
糖などの夾雑物を含む。発明者はこの不純泥漿の精製法
につき鋭意研究した結果、これを限外濾過することによ
って良好な結果が子IJられることを見出した。  S
Eが水溶液中で一定の条件下で相互に合一して高分子礒
のミセル構造の集合体を作ることは、公知(前掲占10
2頁参照)である。 ところで、SEの種類であるが、シ:!糖の分子の3個
の第一級水酸基の酸素原子のいずれかに、夫々1〜3個
の脂肪酸残基が結合したものを夫々モノエステル、ジエ
ステル及びトリエステルと称している。そして周知の如
く、モノエステルは、親木性がジエステルやトリエステ
ルに比較して大きい代りに、水中におけるミセル形成の
度合いが小さいので、比較的低分子量の(分子の直径の
小さい)SEミセル集合体を形成する。逆に、ジエステ
ルやトリエステルは、親水性が比較的小さい代りにミセ
ル形成能が極めて大きいので、水中では、極めて大きな
分子量の(即ち、分子径の大きい)SEミセル集合体を
形成する。市販のSEでは、モノエステル単品として製
造されることは積であって、通常はモノエステルの含量
が、例えば70%、50%、30%中・・といった混合
組成物として製造されている。 本発明者らは、例えば、モノエステルの含量が70%ト
多いSEは、モノエステル含量が50%と少ないSEに
比べて、より低分子量のSE集合体を作るので、その分
、集合体の微視的径が小さいこと、従って、一定の孔径
を有する限外濾過膜に対してモノエステル含有量50%
のSEよりも通過し易く、このため、未反応の糖や触媒
からの副生塩(触媒を酸で中和して塩としたもの)、揮
発分等と一諸に膜を通過してしまい易いという望ましく
ない傾向を有することを知った。そこで本発明者らは、
これに対する対策として、モノエステル含量の高い不純
SEから未反応の糖、触媒由来の塩、揮発分等を除去し
たい場合は、分画分子量の小さい(即ち、孔径の小さい
)濾過膜を選定するのがよいこと、及び逆にモノエステ
ル含量の低いSHの場合には、分画分子量の大きい(即
ち、孔径の大きい)濾過膜を選定するのが処理速度を速
めるのに好都合であることを見出した。 なお、発明者らは1反応混合物に含まれている物質のう
ち、未反応の脂肪酸メチルエステル、石鹸及び脂肪酸の
王者は、SEのミセル構造集合体中に内包された状態で
存在するため、SEとそれらの王者を濾過手段により分
離するのは事実上不可能であることも、多くの実験結果
から確認した。 そして多くの実験から、結論として言えることは、圧力
を駆動源として限外濾過膜(適当な分画分子量を持つ)
を水と共に通過できる不純物質は、未反応の糖、触媒由
来の塩、添加された中性塩類、及び揮発分(ジメチルス
ルホキシドやジメチルホルムアミド等、SE合成に際し
溶媒とじて用いられた、極性が強く、水溶性が大で、か
つショ糖と親和性の大きい物質)の囲者であり、方、高
分子量のミセル集合体中に取り込まれて濾過膜を通過で
きない物質は、SE、未反応の脂肪酸メチルエステル、
石鹸及び遊離脂肪酸等である。 本工程は、これらの事
実を巧妙に利用すると共に、適当な分画分子量を持つ限
外濾過膜の選定によって2塩析沈殿に夾雑する未反応の
糖、触媒由来の塩及び揮発分の王者をSE、未反応の脂
肪酸メチルエステル、石鹸及び脂肪酸の囲者から分離、
除去しようとするものである。 (濾過対象物質の分子量) 適当な分画分子量を持つ限外濾過膜を選定するためには

触媒(K2CO2)から生じた塩及び揮発分の除去を狙
っても、その限外濾過膜で、分離され得るのは、限外濾
過膜の万両分子1200よりも低い分子量を持つ水、触
媒(K2 CO3)から生じた塩及び揮発分のみである
0分画分子量200より大きい分子量342のショ糖は
、全く限外濾過膜を透過しないから、未反応糖はSEか
ら分離、除去できない。 次に1分画分子量が5,000の限外濾過膜の場合は、
ショ糖、触媒からの塩及び揮発分は、夫々の分子量が5
,000より小さいので、限外濾過膜の微孔を容易に通
過できる。SEは、前述の通すミセル集合体を構成し、
ミセル会合数を例えば10個と仮定すると、そのSEミ
セル集合体の分子量はe 、ooo以上と推定されるの
で、濾過膜の分画分子量が5,000より大きいと該ミ
セル集合体が微孔を通過できないものと推定されるが、
この推定は実験的に確認された。 別に、分画分子量1,000の濾過膜の場合についても
検討したが、結果は予想の通りであった。 このように、限外濾過膜の分画分子量を適当に選定する
ことによって、不純なSEから未反応糖を含む不純物の
除去が可能となる。 (限外濾過膜の具備すべき条件) SE反応混合物に含まれる未反応糖と、触媒(K2 C
O:l )から副生じた塩と、揮発分との王者をSE、
石帥、未反応の脂肪酸メチルエステル及び脂肪酸の囲者
より分離しようとする場合、限外濾過膜のA備すべき条
件は、該膜が適当な分画分子量を有する場合、 ■ 物理的な外力に対し、抵抗力があること。 ■ 耐熱性を有し、微生物によって分解されないこと。 ■ 適当な分画分子量を持ち、処理能力の大きいこと。 ■ 耐用年数が長いこと。 ■ 経済的な価格で入手できること。 等である。 近年の限外濾過膜の製造における技術の進歩には著しい
ものがあるから、市販のものでも上の条件を満たしてい
るものが見出される。 (限外濾過の実際) 本工程の実施に際しては、E述の塩析工程で生成した沈
殿に、水、望ましくは脱イオン水を水/沈殿=5〜40
(重量比) になるように、さらに好ましくは、 水/沈殿=20(重量比) となるように加えて溶解させた後、限外濾過膜と接触さ
せる。このとき、水溶液のpHは、最初の中和工程で略
々中性領域内にある筈であるが、若し何等かの理由でp
)I 8.2〜8.2の範囲内にないならば、適宜酸又
はアルカリを用いて当該中性領域内に調整しておく(p
)16°、2未満ではSEのミセル集合体が低分子化し
て漏れによる損失が増加し、またpH6.2超過では、
SE自体の加水分解が進み易くなる。)。 癌過時の水溶液の温度は、脂肪酸メチルエステルの種類
とは無関係に80℃以下の温度が好ましく、同温度を超
えるとSEが分解する懸念がある0発明渚らは、該温度
が、特に40〜60℃の温度範囲内に在るとき、最大の
濾過速度が得られることを見出した。即ち、濾過温度を
40〜60°C1好ましくは約50°Cに調節すると、
後述の理由で、未反応糖を含むショ糖、触媒(K2 C
O3)由来の副生塩及び添加中性塩並びに揮発分(ジメ
チルスルホキシドやジメチルホルムアミド)の囲者は、
水と共に最も効率良く濾過膜を通過する。この理由とし
ては40〜60℃の温度領域に於てSEのミセル集合体
の分子が巨大化する結果、ミセル集合体の総数が減少し
、未反応糖等の元来ミセル集合体の形成に関与しない物
質がSHの抵抗を受は難くなり、その分、未反応糖等が
通過し易くなることに因るものと推測される。因に、公
知の如く、SE水溶液は一般に40〜80゛Cの間で最
大の粘度を示す(上掲書+03頁参照)が、これは、そ
の温度範囲内でミセル集合体が最大の分子量を持ち得る
ことを示唆するものであり、この事実からも、40〜8
0°Cの範囲で未反応糖等が最大の通過速度を示す理由
を説明することが可能である。 かくして、40〜60℃に維持されたSEを含む反応混
合物水溶液を、ポンプにより1〜20 Kg/c+s2
Gまで加圧して駆動源としての圧力をかけ、 pH8,
2〜8.2の水素イオン濃度領域で限外濾過膜に接触さ
せる。ここに濾過膜として、セルロース系のものは物理
的に弱いだけでなく、かつ微生物にも侵され易いので、
実用上余り望ましくない、実用的に好適であるのは、支
持層で補強されたポリスルホン製もしくはポリ弗化ビニ
リデン製の膜である。これら両種の濾過膜は、現在市販
されており1本膜は、耐熱性、耐酸性及び耐アルカリ性
に優れるのみでなく、物理的外力にも強く、しかも微生
物が膜面で増殖することもない。 前述の通り、濾過膜の分画分子−1の決定に際しては、
SEの洩れなく行なわれ、かつ濾過速度も大である範囲
のものを選定することが重要である。発明基らは、検討
の結果、SHの洩れがなく、未反応糖、副生Jfl及び
揮発分の分離性が損なわれず、かつ濾過速度が大である
という希望条件を満たす膜の分画分子量として、1,0
00−1oo、000の範囲内のものが好適であること
、及び、とりわけSEの洩れがなく、しかも工業的な規
模での処理に適したものとして、分画分子量5,000
の濾過j模が最も好ましいことを発見した。 5,00
0超過の分画分子量のものでは、僅かではあるがSHの
洩れが発生し、逆にs、ooo未膚の分画分子量の膜で
は、濾過速度が減少する。しかしいずれの場合でも、工
業的に採算に乗らない程の不利益をもたらすものではな
い、 現在市販の濾過膜のうちで、発明目的に適うもの
としては、例えば東しエンジニアリング■の販売に係る
限界濾過膜のうち、商品名((TERP−E−5>)

業的に充分な分離速度であり、かつ触媒から副生ずる塩
及び揮発分の分離速度も充分であった。因に、未反応側
、触媒からの塩及び揮発分の除去率は、濾過膜への通液

、限外濾過膜の利用により、SE反応混合物の塩析沈殿
から、工業的に容易に、未反応糖、触媒(K2 C03
)からの副生塩及び揮発分の王者を一括して水と共に除
去することが可能となり、かくして、水のみで、溶媒を
一切使用せずに、未反応糖と触媒(K2 CO3)から
の副生塩及び揮発分の王者を除こうとする目的が達成さ
れる。 以上の限外濾過処理により、塩析沈殿に含まれる揮発分
、シヨ糖分、塩類等の夾雑物を除去された高純度SEは
1通常、固形分濃度1〜10%の水溶液の形で回収され
ることができるが、固形分が7%を超えると限外濾過膜
を透過する水及び不純物の丑が減少してくるので余り好
ましくなく、実用的には、固形濃度4〜6%の範囲が工
業的な操業上望ましい。 かくして精製された含水状態のSEは、略々泥漿(スラ
リー)状を呈し、必要に応じ、例えば、真空C縮機を用
