ら18まで分布し、j=14,15および16の3種の同
族体が主体となつていることが明らかにされた
〔R.W.Keenan et al.,Biochemical Journal,
165,505(1977)参照〕。 ドリコール類はブタの肝臓のみならず、哺乳動
物体内に広く分布しており、生体の生命維持の上
で極めて重要な機能を果していることが知られて
いる。例えば、J.B.Harfordらは子牛やブタの脳
内白髄質を用いるin vitro試験により、外因性ド
リコールがマンノースなどの糖成分の脂質への取
り込みを促進し、その結果、生体の正命維持のう
えで重要な糖蛋白質の形成を増大させる作用を持
つことを明らかにしている〔Biochemical and
Biophysical Research Communication,76,
1036(1977)参照〕。 ドリコール類によるかかる脂質への糖成分の取
り込み促進効果は成長期の生体におけるよりも既
に成熟している動物において顕著であることか
ら、老化防止の点でのドリコール類の働きが注目
されている。また、R.W.Keenanらは幼年期など
の急速に成長を続けている正体にとつては外から
ドリコールを摂取し、自己の体内で生合成して得
られるドリコールを補うことが重要であると述べ
ている〔Archives of Biochemistry and
Biophysics,179,634(1977)参照〕。さらに、
赤松らはラツトの再生肝中のドリコールリン酸エ
ステルを定量し、その量が正常な肝中よりも著し
く減少しており、肝組織での糖蛋白の合成機能が
大巾に低下していることおよび外からドリコール
リン酸エステルを加えると該機能が改善されるこ
とを見出した〔第54回日本生化学会大会(1981
年)において発表〕。 上記のようなドリコール類は生体にとつて極め
て重要な機能を司る物質であり、医薬品またはそ
の中間体として有用であるが、従来その入手は容
易でなく、例えばブタの肝臓10Kgから複雑な分離
操作を経てやつと0.6gのドリコールが得られるに
過ぎない〔F.W.Burgos et al., Biochemical
Journal,88,470(1963)参照〕。ドリコール類を
全合成することは、それらの複雑で特異な分子構
造から明らかなように現在の有機合成の技術では
至難のことである。合成中間体を天然物に依存
し、これに簡単な合成化学的処理を加えるのみで
ドリコール類を得ることができるならば有利であ
るが、従来そのような好都合な物質は見出されて
いない。従来、下記の一般式(B) 〔但し、k=4〜6〕で示されるポリプレノール
類(これらはベツラプレノール類と呼ばれてい
る)がシラカンバ(Betula verrucola)から採
取し得ることは知られているが、これらからシス
型イソプレン単位の数が14,15および16のものを
主体とするドリコール類を合成することは現在の
有機合成技術ではほとんど不可能である。また
K.Hannusらはヨーロツパ赤松(Pinus
sylvestris)の葉から乾燥重量基準で1%の収率
でポリプレニル成分を単離し、この成分がイソプ
レン単位10〜19個を主としてシス配置で有するポ
リプレニルアセテート混合物であることを報告し
ているが〔Phytochemistry,13,2563(1974)参
照〕、彼らの報告には該ポリプレニルアセテート
中のトランスおよびシス配置についての詳細まで
は解明されていない。さらに、D.F.Zinckelらは
ストローブ松(Pinus strobus)の葉の抽出物中
にイソプレン単位数18個またはイソプレン単位数
の平均値が18であるC90のポリプレノールが存在
することを報告しているが〔Phytochemistry,
11,3387(1972)参照〕、この報告では該ポリプレ
ノールのトランス、シス配置について詳細な解析
を行なつていない。 本発明者らの一部とその共同研究者らは、先
に、イチヨウ(Ginkgo biloba)およびヒマラヤ
杉(Cedrus deodara)の葉から有機溶媒で抽出
される抽出物を必要に応じてケン化したのちクロ
マトグラフイー、分別溶解法その他の適当な分離
法によつて処理することにより一般式()にお
いてnが11から19まで分布しているポリプレノー
ルおよび/またはその酢酸エステル同族体混合物
からなるポリプレニル画分が得られること、該ポ
リプレニル画分は哺乳類ドリコール類に比べてα
−末端の飽和イソプレン単位が存在しないだけで
哺乳類ドリコール類におけるポリプレニル同族体
の分布に非常によく以たポリプレニル同族体の分
布を示すこと、該ポリプレニル画分は所望により
その構成成分である個々の(イソプレン単位数が
一様な)ポリプレニル同族体に比較的容易に分離
しうること、従つて該ポリプレニル画分およびそ
れから分離された各ポリプレニル同族体はいずれ
も哺乳類ドリコール類の合成中間体として非常に
適していることを見出した。また本発明者らは、
ヒマラヤ杉と同様にマツ科に属するクロマツ
(Pinus thunbergii)、アカマツ(Pinus
desiflora)などの葉からも一般式()で示さ
れるポリプレノールおよび/またはそのエステル
(一般に酢酸エステル)が得られることを見出し
た。ただし、クロマツおよびアカマツからは、通
常、一般式()においてnが10から18まで分布
しているポリプレニル同族体混合物が得られる。 しかしながらこれらの植物から有機溶媒で抽出
される抽出物中には通常一般にクロロフイル、カ
ロチノイド、ステロイド、グリセリド、スフイン
ゴリピド、フラボノイドなど、上記ポリプレノー
ル類との分離が必ずしも容易でない成分が含まれ
ており、該抽出物をケン化処理したのち分別溶解
法とクロマトグラフイーとの組も合せなどによつ
て分離、精製して得られるポリプレノールにはし
ばしばそれらの不純物が数パーセント混入してく
る。 本発明者らは上記の問題点を解決して一般式
()で示されるポリプレノールおよび/または
そのエステルを含有する植物の抽出物から一般式
()で示されるポリプレノールを容易に高純度
で得るために鋭意研究を重ねた結果、粗ポリプレ
ノール画分をエステルに転化して精製したのちに
ポリプレノールに戻す工程を含む下記方法により
所期の目的が達成されることを見出し、本発明を
完成するに至つた。 すなわち、本発明によれば、一般式()で示
されるポリプレノールの少なくとも1種および/
またはそのエステルを含有する植物を脂溶性の有
機溶媒で抽出して得られる抽出物をケン化処理し
たのち一般式()で示されるポリプレノールを
主成分とする画分を分離し、該ポリプレノールを
その低級脂肪族モノカルボン酸エステルとし、該
ポリプレニルエステルを精製したのちポリプレノ
ールに転化し、生成した一般式()で示される
ポリプレノールを回収することにより、一般式
()で示されるポリプレノールを容易に高純度
で得ることができる。 一般式()で示されるポリプレノールの少な
くとも1種および/またはそのエステルを含有す
る植物から該ポリプレニル成分を抽出するために
使用される脂溶性の有機溶媒としては、一般に誘
電定数εが32.7以下、好ましくは25.0以下、さら
に好ましくは20.7以下のものが好適であり、具体
的には下記に例示する溶媒がそれぞれ単独でまた
は2種もしくはそれ以上の混合溶媒として使用で
きる。 (a)炭化水素類:例えば、石油エーテル、ペンタ
ン、ヘキサン、ヘプタン、ベンゼン、トルエン、
キシレンなど。 (b)ハロゲン化炭化水素類:例えばクロロホルム、
塩化メチレン、四塩化炭素、四塩化エタン、パー
クロルエチレン、トリクロルエチレンなど。 (c)エステル類:例えば、酢酸メチル、酢酸エチ
ル、プロピオン酸エチルなど。 (d)エーテル類:例えば、ジエチルエーテル、ジイ
ソプロピルエーテル、テトラヒドロフラン、ジオ
キサンなど。 (e)ケトン類:例えば、アセトン、メチルエチルケ
トン、ジエチルケトン、ジイソプロピルケトンな
ど。 (f)アルコール類:例えば、メチルアルコール、エ
チルアルコール、プロピルアルコール、ブチルア
ルコールなど。 使用する溶媒の選択にあたつては、一般式
()のポリプレノールおよびそのエステルを選
択的に高効率で抽出し、それ以外の物質はできる
だけ抽出しないものが望ましく、かかる観点から
すれば、上記溶媒中、炭化水素類、ハロゲン化炭
化水素類、エステル類、ジエチルエーテル、ジイ
ソプロピルエーテルの如き極性の低いエーテル類
およびケトン類が特に好適である。 抽出溶媒の使用量は臨界的なものではなく、用
いる溶媒の種類、抽出すべき植物の種類、部位、
状態等に応じて広範に変えることができるが、一
般には抽出に供する植物体1重量部(乾燥重量基
準)当り約1〜約100重量部、、好ましくは5〜50
重量部、さらに好ましくは10〜30重量部の範囲内
で用いることが有利である。 抽出は上記の溶媒中に植物体を浸漬し、必要に
応じて連続的または間欠的に撹拌することにより
行なうことができる。抽出時の温度も臨界的なも
のではなく、用いた溶媒の種類や量等の条件に応
じて広範に変えることができるが、一般には約0
℃から溶媒の還流温度までの温度を用いることが
でき、通常は室温で充分である。かかる条件下に
抽出は普通1〜10日間行なうのが有利である。 抽出処理後の浸漬液は植物体その他の固形分を
除去したのち必要に応じて溶媒を除去して濃縮液
とする。かくして得られる抽出物をそのままケン
化処理に供することができるが、所望により該抽
出物を適宜前処理によりある程度精製したのちケ
ン化処理に供することもできる。この前精製には
後述する分別溶解法および/またはクロマトグラ
フイーがとくに好適に用いられる。この前精製に
よつて炭化水素溶媒(たとえばn−ヘキサン)に
不溶な夾雑物および/またはタール分を除去して
おくと後の精製が容易となる。ケン化処理は、自
体公知の方法および条件を用いて行うことが可能
であり、たとえば含水率約1〜20重量%程度の含
水メタノールまたは含水エタノール中に水酸化ナ
トリウムまたは水酸化ナトリウムを溶解させた溶
液(アルカリ金属水酸化物濃度は好ましくは約
0.1〜30重量%とすることができる)100重量部に
対して上記の抽出物を約5〜50重量部の割合で加
えて約25〜90℃で約0.5〜5時間反応させればよ
い。 ケン化処理後、ケン化反応混合物から一般式
()で示されるポリプレノールを主成分とする
画分(粗ポリプレノール画分)を分離取得する。
この粗ポリプレノール画分の取得は通常好ましく
は次の方法により行われる。すなわち、まずケン
化反応混合物を水の存在下、実質的に水と混和し
ない有機溶媒で抽出し、有機層と水性層とに分離
する。 ここで用いうる有機溶媒としては、例えば石油
エーテル、ペンタン、ヘキサン、ヘプタン、ベン
ゼン、トルエン、キシレンなどの炭化水素類、ク
ロロホルム、塩化メチレン、四塩化炭素、四塩化
エタン、パークロルエチレン、トリクロルエチレ
ンなどのハロゲン化炭化水素類、酢酸イソプロピ
ル、酢酸ブチル、プロピオン酸エチル、プロピオ
ン酸ブチル、ジエチルエーテル、ジイソプロピル
エーテル、ジn−ブチルエーテル、メチルイソブ
チルケトン、ジイソブチルケトンなどを挙げるこ
とができる。これら抽出溶媒の使用量は臨界的で
はないが、一般にケン化反応混合物に対して5倍
(重量)から100(重量)が好適である。この抽出
に際し、水は有機層と水性層とが層分離を生じる
に必要な最少量以上存在すればよいが、通常有機
層と水性層との容量比が1:10〜10:1となるよ
うな割合で用いることが好ましい。こうして得ら
れる有機層を必要に応じ溶媒を留去して濃縮後、
クロマトグラフイー、分別溶解法、分子蒸留法ま
たはこれらの方法の2種もしくはそれ以上の組合
わせからなる分離工程に付して粗ポリプレノール
画分を回収することができる。 上記分離工程におけるポリプレノールが含有さ
れている画分の確認は、メルク社製薄層クロマト
用プレート(シリカゲル60F254被覆;層の厚さ
0.25mm)を用いかつn−ヘキサンと酢酸エチルと
の容量比で9:1の混合溶媒を展開溶媒とする薄
層クロマトグラフイー(10cm展開)において標準
物質としてのソラネシルアセテートのRf値が0.40
〜0.45となる条件下に0.18〜0.25の範囲内のRf値
のところにスポツトが存在するか否かにより行な
うことができる。しかして、以下の説明において
薄層クロマトグラフイーのRf値を言及する場合
には、特にことわらない限り、上記条件下に測定
した値を意味することを了解すべきである。 上記分離工程において使用しうるクロマトグラ
フイー、分別溶解法、分別冷凍沈殿法および分子
蒸留法の各方法の操作はそれ自体公知のものであ
り、本発明においても公知の方法に準じて行なう
こができるので、各方法の詳細については文献の
引用を以つて説明に代え、ここには特に注意すべ
き点のみを記載する。 (A)クロマトグラフイー〔例えば、H.Heftman,
“Chromatography”,Reinhold、Publish Co.,
New York(1961)参照〕 抽出物が少量の場合には薄層クロマトグラフイ
ーおよび液体クロマトグラフイーが適当である
が、大量の抽出物の処理にはカラムクロマトグラ
フイーが好適である。使用するクロマトグラフイ
ー用担体としては、シリカゲル、アルミナ、フロ
リジル、セライト、活性炭、セレロースなどが挙
げられ、中でもシリカゲルが好適である。 シリカゲルカラムを使用して分離操作を行なう
場合の展開溶媒としては、例えば、ヘキサン/酸
エチル(容量比95:5〜80:20)、ヘキサン/ジ
イソプロピルエーテル(容量比95:5〜80:20)、
石油エーテル/酢酸エーテル(容量比95:5〜
80:20)、石油エーテル/イソプロピルアルコー
ル(容量比99:1〜90:10)、ベンゼン/ジエチ
ルエーテル(容量比95:5〜80:20)、ベンゼ
ン/酢酸エチル(容量比98:2〜80:20)などの
混合溶媒系あるいはクロロホルム、メチレンクロ
リドなどが挙げられる。 (B)分別溶解法〔例えば、L.C.Craig,“Technique
of Organic Chemistry”,Vol.13,Interscience,
(1951)参照〕 前記一般式()のポリプレノールはペンタ
ン、ヘキサンのような非極性溶媒に易溶であり、
一方、メタノールや水などの極性溶媒には難溶で
あるので、前記抽出液濃縮物を上記非極性溶媒に
溶解し、ついで該非極性溶媒と非混和性の極性溶
媒で洗浄することによつて、極性溶媒に易溶な不
純物を大幅に除去することができる。本方法で好
適に使用される非極性溶媒としては、例えば、石
油エーテル、ペンタン、ヘキサン、ヘプタン、ベ
ンゼン、トルエンなどの炭化水素系溶媒およびメ
チレンクロリド、クロロホルムなどのハロゲン化
炭化水素系溶媒が好適である。また、かかる非極
性溶媒と非混和性の極性溶媒としては例えば水あ
るいはメタノールが好適である。 (C)分別冷凍沈殿法〔例えば、E.W.Berg,
“Physical and Chemical Methods of
Separation”Chapter 14,15,McGraw−Hill,
New York(1963)参照〕 前記一般式()のポリプレノールは約−10℃
以下で固化する。従つて、抽出物を−10℃以下、
好ましくは約−15〜約−30℃に冷却下に放置し、
目的物を固化させたのち固−液分離することによ
つて、かかる温度で固化しない不純物から精製す
ることができる。しかしながら、該ポリプレノー
ルはあまり優れた結晶性を有さず、ワツクス状固
体となるため、本方法により完全に精製すること
は難しい。なお、逆にポリプレノール中に存在す
る結晶性の不純物を優先的に析出させて除去する
ことも可能である。 (D)分子蒸留法〔例えば、G.Durrows,
“Molecular Distillation”,Clarendon Press,
Oxford(1960)参照〕 前記一般式()のポリプレノールは分子量が
大きいため、分子蒸留法を用いることによつて低
分子量の不純物を除去することができる。例えば
10-3〜10-5mmHgの真空度において100〜200℃の
加熱条件下に分子蒸留して、低分子留分と高分子
留分とに分割される。このとき、高分子留分に目
的物質は保持され、低分子量不純物を大幅に除去
することができる。 これらの分離法の2種もしくはそれ以上の組合
わせを用いることもできる。例えば、クロマトグ
ラフイーと分別溶解法;クロマトグラフイーと分
別冷凍沈殿法と分別溶解法;クロマトグラフイー
と分別冷凍沈殿法と分別溶解法と分子蒸留法;ク
ロマトグラフイーと分子蒸留法と分別溶解法;ク
ロマトグラフイーと分子蒸留法;分子蒸留法と分
別溶解法;分子蒸留法と分別溶解法と分別冷凍沈
殿法などの組合せを用いることができる。 本発明の好ましい実施態様の1つにおいては、
前記ケン化処理後有機溶媒で抽出された抽出物を
カラムクロマトグラフイーを主体とした分離精製
法によつて処理することにより、前記薄層クロマ
トグラフイーにおいてRf値が0.18〜0.25となる画
分(粗ポリプレノール画分)を分離取得する。 かくして得られる粗ポリプレノール画分を一旦
ポリプレニルエステルに転化し、該ポリプレニル
エステルの形で精製すると簡単な操作で容易に高
純度に精製することができる。ポリプレノールを
ポリプレニルエステルに転化するためには、ポリ
プレノールを酢酸、プロピオン酸、酪酸、吉草酸
などの低級脂肪族モノカルボン酸またはそれらの
反応性誘導体例えば無水酢酸のごとき酸無水物ま
たはアセチルクロリドのごとき酸ハライドとエス
テル化反応させ、あるいはポリプレノールを酢酸
メチル、酢酸エチル、プロピオン酸メチルなどの
低級脂肪族モノカルボン酸低級アルキルエステル
とエステル交換反応させればよい。これらエステ
ル化反応およびエステル交換反応は自体公知の方
法および条件を用いて行うことができる。たとえ
ばエステル化反応は次のようにして行うことがで
きる。すなわちポリプレノール画分とこれに含ま
れるポリプレノール1モルあたり約0.8〜10モル、
好ましくは1〜5モルの前記酸無水物または酸ハ
ライドとを好ましくはペンタン、ヘキサン、ベン
ゼン、トルエン、ジエチルエーテル、ジイソプロ
ピルエーテルなどの適当な溶媒の存在下、ピリジ
ン、トリエチルアミンなどの適当な塩基の存在下
に約−30〜+80℃、好ましくは0〜30℃で反応さ
せればよい。また、エステル交換反応はたとえば
次のようにして行うことができる。すなわち前記
のごとき低級脂肪族モノカルボン酸の低級アルキ
ルエステルとポリプレノールとを溶媒の存在下ま
たは無溶媒で、水素化ナトリウム、水素化カリウ
ムなどの適当なエステル交換触媒の存在下に50〜
200℃、好ましくは80〜150℃で反応させればよ
い。 上記のようにして生成させたポリプレニルエス
テルを精製することにより前記ポリプレノール画
分からは容易には除去し難い來雑物とくにクロロ
フイル、カロチノイド、ステロイド、グリセリ
ド、スフインゴリピド、フラボノイドなどを容易
に除去することができる。この精製には先に述べ
たクロマトグラフイー、分別溶解法、分別冷凍沈
殿法、分子蒸留法などを単独でまたは2種以上組
合わせて用いることができるが、クロマトグラフ
イーの使用がとくに好適である。クロマトグラフ
イーの好ましい実施態様はポリプレノール画分を
得るために用いる分離方法に関して前記したもの
と同様である。大量のポリプレニルエステルを精
製するには、たとえばシリカゲルカラムおよび展
開溶媒としてたとえばヘキサン/酢酸エチル(容
量比99:1〜90:10)、ヘキサン/ジイソプロピ
ルエーテル(容量比99:1〜90:10)などを用い
てカラムクロマトグラフイーにより精製するのが
簡便であり、とくに好ましい。 精製されたポリプレニルエステルは好ましくは
加水分解によつて目的とするポリプレノールに転
化される。この加水分解反応は公知の高級アルコ
ールのエステル類を加水分解するために有用であ
ることが知られている種々の方法により行うこと
ができる。好適には、ポリプレニルエステルをメ
タノール、エタノール、イソプロパノールなどの
アルコール中でポリプレニルエステル1モルあた
り約1〜10モルのアルカリ金属水酸化物とくに水
酸化ナトリウムまたは水酸化カリウムと共に加熱
撹拌することによつて実施される。別法としてポ
リプレニルエステルをジエチルエーテル、テトラ
ヒドロフランなどの溶媒中でポリプレニルエステ
ルに対して0.25〜5.0モル当量の水素化アルミニ
ウムリチウムと共に室温またはその近傍の温度で
反応させることによつてもポリプレノールに転化
することができる。 これらの反応後、反応混合物から有機層を分離
し、水洗、乾燥および濃縮などの簡単な後処理を
行うことにより一般式()で示されるポリプレ
ノールを回収することができる。得られたポリプ
レノールがその同族体混合物である場合、所望に
よりさらに例えば分配型高速液体クロマトグラフ
イーによつて個々の同族体成分を分取することも
できる。このようにして得られるポリプレノール
またはそれらの混合物はクロロフイル、カロチノ
イド、ステロイド、グリセリド、スフインゴリピ
ド、フラボノイドなどの着色性不純物をほとんど

去が容易となることが明らかである。 実施例2 5月下旬に採取したヒマラヤ杉の葉10Kg(未乾
燥重量)を約50℃で8時間熱風乾燥したのち室温
(約20℃)でクロロホルム100中に浸漬して1週
間抽出した。この抽出液から溶媒を留去し、黒か
つ色の液状物を得た。このものを約1Kgのシリカ
ゲルを用いたシリカゲルカラムによりn−ヘキサ
ン/酢酸エチル=95/5(容量比)の混合溶媒を
用いて処理することにより、n−ヘキサン/酢酸
エチル=9/1(容量比)の混合溶媒を用いたシ
リカゲル薄層クロマトグラフイー〔メルク社製
TLC Plate(silica gel 60F254 Precoated,層厚
0.25mm)を使用して10cm展開〕においてRf=0.5
付近およびRf=0.2付近の画分を集めて黒かつ色
の液状物58gを得た。この薄層クロマトグラフイ
ーにおいてソラネシルアセテートは0.41のRf値を
示した。 次いでこの黒かつ色の液状物をメタノール400
ml、水40mlおよび水酸化カリウム15gと共に4時
間65℃に加熱撹拌したのち室温に冷却し、ヘキサ
ン約800mlを加えて抽出し、ヘキサン層を約200ml
の飽和食塩水で5回洗浄し、無水硫酸マグネシウ
ムで乾燥し、溶媒を留去して黒かつ色の液状物
49gを得た。この液状物を約1Kgのシリカゲルを
用いたシリカゲルカラムによりn−ヘキサン/酢
酸エチル=97/3(容量比)の混合溶媒を用いて
処理することにより、前記シリカゲル薄層クロマ
トグラフイーにおいてRf=0.21となる画分を分離
して赤かつ色液状物23.2gを得た。 次いでこの液状物をピリジン20g、n−ヘキサ
ン300ml中に溶解し、室温で撹拌しながら無水プ
ロピオン酸25gを滴下し、滴下完了後、一夜撹拌
を継続した。得られた反応液を飽和食塩水約100
mlで5回洗浄したのち無水硫酸マグネシウムで乾
燥し、n−ヘキサンを留去して赤かつ色の液状物
を得た。 この液状物を約1Kgのシリカゲルを用いたシリ
カゲルカラムによりn−ヘキサン/酢酸エチル=
97/3(容量比)の混合溶媒を用いて処理するこ
とにより、前記シリカゲル薄層クロマトグラフイ
ーにおいてRf=0.51となる画分を分離して淡黄色
液状物20.8gを得た。 次いでこの液状物を200mlの無水テトラヒドロ
フランに溶解し、室温で撹拌しながら水素化アル
ミニウムリチウム0.5gを少しづつ加え、添加完了
後室温で2時間撹拌を続けたのち約5mlの水を注
意深く加え、回転蒸発器でテトラヒドロフランを
留去し、約500mlのヘキサンで抽出した。ヘキサ
ン溶液を約100mlの飽和食塩水で5回洗浄したの
ち、無水硫酸マグネシウムで乾燥し、ヘキサンを
留去して淡黄色液状物18.4gを得た。この液状物
について実施例1と同様に高速液体クロマトグラ
フイーによる分析を行なつたところ、一般式
()で示されるポリプレノール以外のピークは
殆んど検出されず、99%以上の純度を有すること
が確認された。また、このポリプレノールについ
て実施例1と同じ方法で測定した分子量分布は下
記のとおりであつた。

℃)で抽出した。この抽出液から溶媒を留去して
黒かつ色の液状物を得た。このものをメタノール
400ml、水40mlおよび水酸化カリウム30gと共に
2時間65℃に加熱、撹拌したのち室温に冷却し、
ジエチルエーテル500mlを加えて抽出し、ジエチ
ルエーテル層を飽和食塩水で5回洗浄後、無水硫
酸ナトリウムで乾燥し、溶媒を留去して黒かつ色
液状物75gを得た。この黒かつ色液状物の各5gを
下記工程に従つて処理し、精ポリプレノールを得
た(実施例3〜5)。 黒かつ色液状物→第1精製→エステル化→第2
精製→ケン化→精ポリプレノールの単離。 また比較のため第1精製後のエステル化および
第2精製後のケン化を行わなかつた以外は実施例
3〜5と同様にして精ポリプレノールを得た(比
較例3〜5)。 結果を表3にまとめて示した。

溶媒を用いて処理することによりn−ヘキサン/
酢酸エチル=9/1(容量比)の混合溶媒を用い
たシリカゲル薄層クロマトグラフイーにおいて
Rf=0.21となる画分を分離して6.9gの赤かつ色液
状物を得た。 次いでこの液状物をピリジン5g、n−ヘキサ
ン100ml中に溶解し、室温で撹拌しながら無水酢
酸5gを滴下し、滴下完了後、一夜室温で撹拌し
た。得られた反応混合物を飽和食塩水約20mlで5
回洗浄したのち無水硫酸マグネシウムで乾燥し、
n−ヘキサンを留去して赤かつ色の液状物を得
た。この液状物を約300gのシリカゲルを用いた
シリカゲルカラムによりn−ヘキサン/酢酸エチ
ル=97/3(容量比)の混合溶媒を用いて処理す
ることにより、前記と同様のシリカゲル薄層クロ
マトグラフイーにおいてRf=0.53となる画分を分
離して黄色液状物5.18gを得た。 次いでこの液状物を100mlの無水テトラヒドロ
フランに溶解し、室温で撹拌しながら水素化アル
ミニウムリチウム0.2gを少しづつを加え、添加完
了後、室温で2時間撹拌を続けたのち約5mlの水
を注意深く加え、回転蒸発器でテトラヒドロフラ
ンを留去し、約100mlのヘキサンで抽出した。 得られたヘキサン層を約50mlの飽和食塩水で5回
洗浄したのち、無水硫酸マグネシウムで乾燥し、
ヘキサンを留去して淡黄色液状物5.02gを得た。 この液状物について実施例1と同様に高速液体
クロマトグラフイーによる分析を行なつたとこ
