および16の3種の同族体が主体となつていること
が明らかにされた〔R.W.Keenan et al.,
Biochemical Journal,165,405(1977)参照〕。
また、ドリコール類はブタの肝臓のみならず、哺
乳動物体内に広く分布しており、生体の生命維持
の上で極めて重要な機能を果していることが知ら
れている。例えばJ.B.Harfordらは子牛や豚の脳
白髄質を用いる試験管内テストにより、外因性ド
リコールがマンノースなどの糖成分の脂質への取
り込みを促進し、その結果、生体の生命維持の上
で重要な糖蛋白質の形成を増大させる作用をもつ
ことを明らかにしている〔Biochemical and
Biophysical Research Communication,76,
1036(1977)参照〕。ドリコールによるかかる脂質
への糖成分の取り込み促進効果は成長期の生体に
おけるよりも既に成熟している動物において顕著
であることから、老化防止の点でのドリコールの
働きが注目されている。また、R.W.Keenanらは
幼年期などの急速に成長を続けている生体にとつ
ては外からドリコールを摂取し、自己の体内で生
合成して得られるドリコールを補うことが重要で
あると述べている(Archives of Biochemistry
and Biophysics,179,634(1977)参照〕。さら
に、赤松らはラツトの再生肝中のドリコールリン
酸エステルを定量し、その量が通常の肝中よりも
著るしく減少しており、肝組織での糖蛋白の合成
機能が大巾に低下していること、および外因性ド
リコールリン酸エステルを加えると該機能が改善
されることを見い出した〔第54回日本生化学会大
会(1981年)にて発表〕。このようにドリコール
類は生体にとつて極めて重要な物質であり、医薬
品またはその合成中間体、化粧品などにおける用
途開発が強く望まれている。 しかしながら、従来ドリコール類は入手が困難
であるため、充分に研究を行なうことができない
というのが実情である。たとえば豚の肝臓10Kgか
ら複雑な分離操作を経てやつと約0.6gのドリコ
ールが得られるに過ぎない〔J.Burgos etal.,
Biochemical Journal,88,470(1963)参照〕。 一方、ドリコール類を全合成することは、それ
らの複雑で特異な分子構造から明らかなように、
現在の有機合成化学の技術では至難のことであ
る。そこで、合成中間体を天然物に依存し、これ
に簡単な合成化学的処理を加えるのみでドリコー
ル類を得ることができるならば有利であるが、従
来そのような好都合な物質は見出されていない。 従来、種々の植物からポリプレノール化合物を
採取しうることが知られており、下記のポリプレ
ノール類が採取されている。 (1) ソラネソール (2) フイカプレノール類 (3) ベツラプレノール類 ベツラプレノール類はドリコール類と同様にω
−末端イソプレン単位に2個のトランス型イソプ
レン単位が連なつた構造を有するが、これまでに
知られているベツラプレノール類は上記のように
シス型イソプレン単位の数が最大でも6個しかな
く、このものからシス型イソプレン単位の数が14
個、15個および16個のものを主体とするドリコー
ル類を合成するためには、8個以上ものイソプレ
ン単位をシス型に規制して延長することが必要で
あるが、それは現在の有機合成技術ではほとんど
不可能である。 本発明者らは、哺乳類ドリコール類の合成原料
として有用なポリプレニル化合物を植物源に求め
て各種の植物からの抽出物を分析した結果、今
回、驚くべきことに、コウヤマキ科(Family
Sciadopitydaceae、イヌマキ科(Family
Podocarpaceae)、イヌガヤ科(Family
Cephalotaxaceae)またはナンヨウスギ科
(Family Araucariaceae)に属する植物の葉か
ら一般式()においてAが水酸基またはアセト
キシ基でありnが14〜16であるポリプレニル化合
物を比較的高濃度に含有するポリプレニル画分が
得られること、該ポリプレニル画分は所望により
その構成成分である個々のポリプレニル同族体に
比較的容易に分離しうること、したがつて該ポリ
プレニル画分およびそれから分離される各ポリプ
レニル同族体はいずれも哺乳類ドリコール類の合
成中間体として非常に適していることを見出し
た。 しかして、本発明によれば、コウヤマキ科、イ
ヌマキ科、イヌガヤ科またはナンヨウスギ科に属
する植物の葉を有機溶媒で抽出し、得られる抽出
物からポリプレニル化合物またはその混合物を分
離し、必要に応じその分離前または分離後に該ポ
リプレニル化合物またはその混合物を加水分解、
エステル化もしくはエステル変換反応またはそれ
らの2種以上の反応に付することを特徴とする一
般式()で示されるポリプレニル化合物または
その混合物を製造する方法が提供される。コウヤ
マキ科、イヌマキ科、イヌガヤ科またはナンヨウ
スギ科に属する植物の葉を有機溶媒で抽出し、得
られる抽出物を、必要に応じ加水分解したのち、
クロマトグラフイー、分別溶解法、分別冷凍沈殿
法、分子蒸留法またはこれらの方法の2種もしく
はそれ以上の組合わせからなる分離法に付して、
メルク社製薄層クロマト用プレート〔シリカゲル
60F254被覆(pre−coated);層の厚さ0.25mm〕を
用いてn−ヘキサンと酢酸エチルとの容量比で
9:1の混合溶媒を展開溶媒とする薄層クロマト
グラフイー(10cm展開)において標準物質として
のソラネシルアセテートのRf値が0.40〜0.45とな
る条件下に0.18〜0.25および/または0.50〜0.55
の範囲内のRf値を示す画分を単離回収すること
により、Aが水酸基である一般式()の化合物
および/またはAがアセトキシ基である一般式
()の化合物の混合物から本質的になり、そし
てnが14である一般式()の化合物、nが15で
ある一般式()の化合物およびnが16である一
般式()の化合物の少なくとも3種を必須成分
としてそれぞれ実質量含有するポリプレニル組成
物を得ることができる。 本発明において原料として用いるコウヤマキ科
植物、イヌマキ科植物、イヌガヤ科植物またはナ
ンヨウスギ科植物(以下、これらを単に原料植物
と称することがある。)はいずれも種子植物門裸
子植物亜門球果植物綱球果植物目に属する植物で
ある。コウヤマキ科植物コウヤマキ
(Sciadopitys verticillata)の1属1種しか存在
せず主として日本にのみ存在する。イヌマキ科植
物は主として暖帯および南半球温帯に分布し、11
属よりなるが、とくにイヌマキ(Podocarpus
macrophylla)およびナギ(Podocarpus nagi)
が好適に使用される。イヌガヤ科植物は主として
東アジアに分布し1属しか存在しないがハイイヌ
ガヤ(Cephalotaxus Harringtonia subsp.nana)
が好適に使用される。また、ナンヨウスギ科植物
は主として暖帯および南半球温帯に存在し、2属
に分類されるがなかでもブラジルマツ
(Araucaria brasiliana)が好適に使用される。 原料植物の葉は乾燥後に本発明に従う抽出処理
に供してもよく、また未乾燥のまま用いることも
できる。一般には乾燥した葉の方が好ましく、そ
の際の乾燥の程度は乾燥葉の重量基準で含水率が
一般に約30%以下、好ましくは10%以下とするこ
とが有利である。さらに、葉は破砕した後に抽出
することが好ましく、それにより、抽出溶媒との
接触面積が増大し、抽出効率を上げることができ
る。 前記一般式()で示されるポリプレニル同族
体は原料植物の葉の中では一般に遊離のアルコー
ルの形および/または酢酸エステルの形でかなり
高濃度で含まれており、該葉から該ポリプレニル
同族体を効果的に抽出するためには、該ポリプレ
ニル同族体をよく溶解する油溶性の有機溶媒が好
適に使用される。かかる油溶性の有機溶媒として
は、一般に誘電定数εが32.7以下、好ましくは
25.0以下、さらに好ましくは20.7以下のものが好
適であり、具体的には下記に例示する溶媒がそれ
ぞれ単独でまたは2種もしくはそれ以上の混合溶
媒として使用できる。 (a) 炭化水素類:例えば、石油エーテル、ペンタ
ン、ヘキサン、ヘプタン、ベンゼン、トルエ
ン、キシレンなど。 (b) ハロゲン化炭化水素類:例えば、クロロホル
ム、塩化メチレン、四塩化炭素、四塩化エタ
ン、パークロルエチレン、トリクロルエチレン
など。 (c) エステル類:例えば、酢酸メチル、酢酸エチ
ル、プロピオン酸エチルなど。 (d) エーテル類:例えば、ジエチルエーテル、ジ
イソプロピルエーテル、テトラヒドロフラン、
ジオキサンなど。 (e) ケトン類:例えば、アセトン、メチルエチル
ケトン、ジエチルケトン、ジイソプロピルケト
ンなど。 (f) アルコール類:例えば、メチルアルコール、
エチルアルコール、プロピルアルコール、ブチ
ルアルコールなど。 使用する溶媒の選択にあたつては、目的とする
前記一般式()のポリプレニル化合物を選択的
に高効率で抽出し、それ以外の物質はできるだけ
抽出しないものが望ましく、かかる観点からすれ
ば、上記溶媒中、炭化水素類、ハロゲン化炭化水
素類、エステル類、ジエチルエーテル、ジイソプ
ロピルエーテルの如き極性の低いエーテル類およ
びケトン類が特に好適である。 抽出溶媒の使用量は臨界的なものではなく、用
いる溶媒の種類、抽出すべき葉の種類や状態等に
応じて広範に変えることができるが、一般には原
料植物の葉の1重量部(乾燥重量基準)当り約1
〜約100重量部、好ましくは5〜50重量部、さら
に好ましくは10〜30重量部の範囲内で用いること
が有利である。 抽出は上記の溶媒中に原料植物の葉を浸漬し、
必要に応じて連続的または間欠的に撹拌すること
により行なうことができる。抽出時の温度も臨界
的なものではなく、用いた溶媒の種類や量等の条
件に応じて広範に変えることができるが、一般に
は約0℃から溶媒の還流温度までの温度を用いる
ことができ、通常は室温で充分である。かかる条
件下に抽出は普通1〜10日間行なうのが有利であ
る。 抽出処理後の浸漬液は葉その他の固形分を除去
したのち必要に応じて溶媒を除去して濃縮液とす
る。かくして得られる抽出物をついでクロマトグ
ラフイー、分別溶解法、分別冷凍沈殿法、分子蒸
留法またはこれらの方法の2種もしくはそれ以上
の組合わせからなる分離工程に付して、目的とす
るポリプレニル画分を回収することができる。 上記分離工程におけるポリプレニル化合物が含
有されている画分の確認は、メルク社製薄層クロ
マト用プレート(シリカゲル60F254pre−
coated;層厚0.25mm)を用いかつn−ヘキサンと
酢酸エチルとの容量比で9:1の混合溶媒を展開
溶媒とする薄層クロマトグラフイー(10cm展開)
において標準物質としてのソラネシルアセテート
のRf値が0.40〜0.45となる条件下に、0.18〜0.25
〔前記一般式()においてAが水酸基を表わす
場合〕および/または0.50〜0.55〔前記一般式
()においてAがアセトキシ基を表わす場合〕
の範囲内のRf値のところにスポツトが存在する
か否かにより行なうことができる。しかして、以
下の説明において薄層クロマトグラフイーのRf
値を言及する場合には、特にことわらない限り、
上記条件下に測定した値を意味することを了解す
べきである。 上記抽出物の分離工程において使用しうるクロ
マトグラフイー、分別溶解法、分別冷凍沈殿法お
よび分子蒸留法の各方法の操作はそれ自体公知の
ものであり、本発明においても公知の方法に準じ
て行なうことができるので、各方法の詳細につい
ては文献の引用を以つて説明に代え、ここには特
に注意すべき点のみを記載する。 (A) クロマトグラフイー〔例えば、H.Heftman,
“Chromatography”,Reinhold Publish Co.,
New York(1961)参照〕 抽出物が少量の場合には薄層クロマトグラフイ
ーおよび液体クロマトグラフイーが適当である
が、大量の抽出物の処理にはカラムクロマトグラ
フイーが好適である。使用しうるクロマトグラフ
イー用担体としては、シリカゲル、アルミナ、フ
ロリジル、セライト、活性炭、セルロースなどが
挙げられ、中でもシリカゲルが好適である。 シリカゲルカラムを使用して分離操作を行なう
場合の展開溶媒としては、例えば、ヘキサン/酢
酸エチル(容量比95:5〜80:20)、ヘキサン/
ジイソプロピルエーテル(容量比95:5〜80:
20)、石油エーテル/酢酸メチル(容量比95:5
〜80:20)、石油エーテル/イソプロピルアルコ
ール(容量比99:1〜90:10)、ベンゼン/ジエ
チルエーテル(容量比95:5〜80:20)、ベンゼ
ン/酢酸エチル(容量比98:2〜80:20)などの
混合溶媒系あるいはクロロホルム、メチレンクロ
リドなどが挙げられる。 (B) 分別溶解法〔例えば、L.C.Craig,
“Technique of Organic Chemistry”,Vol,
13,Interscience,(1951)参照〕 前記一般式()のポリプレニル化合物はペン
タン、ヘキサンのような非極性溶媒に易溶であ
り、一方、メタノールや水などの極性溶媒には難
溶であるので、この溶解性の差を利用して分別溶
解法により精製できる。例えば抽出液濃縮物のよ
うな粗精製物を上記非極性溶媒に溶解し、ついで
該非極性溶媒と非混和性の極性溶媒で洗浄するこ
とによつて、極性溶媒に易溶な不純物を大幅に除
去することができる。本方法で好適に使用される
非極性溶媒としては、例えば、石油エーテル、ペ
ンタン、ヘキサン、ヘプタン、ベンゼン、トルエ
ンなどの炭化水素系溶媒およびメチレンクロリ
ド、クロロホルムなどのハロゲン化炭化水素系溶
媒が好適である。また、かかる非極性溶媒と非混
和性の極性溶媒としては例えば水あるいはメタノ
ールが好適である。 (C) 分別冷凍沈殿法〔例えば、E.W.Berg,
“Physical and Chemical Methods of
Separation”Chapter 14,15,McGraw−
Hill,New York(1963)参照〕 前記一般式()のポリプレニル化合物は約−
10℃以下で固化する。従つて、抽出物を−10℃以
下、好ましくは約−15〜約−30℃に冷却下に放置
し、目的物を固化させたのち固−液分離すること
によつて、かかる温度で固化しない不純物から精
製することができる。しかしながら、該ポリプレ
ニル化合物はあまり優れた結晶性を有さず、ワツ
クス状固体となるため、本方法により完全に精製
することは難しいので、他の精製方法と組合せて
実施することが好ましい。なお、逆にポリプレノ
ール中に存在する結晶性の不純物を優先的に析出
させて除去することも可能である。 (D) 分子蒸留法〔例えば、G.Burrows,
“Molecular Distillation”,Clarendon Press,
Oxford(1960)参照〕 前記一般式()の化合物は分子量が大きいた
め、分子蒸留法を用いることによつて低分子量の
不純物を除去することができる。例えば10-3〜
10-5mmHgの真空度において100〜250℃の加熱条
件下に分子蒸留して、低分子留分と高分子留分と
に分割される。このとき、高分子留分に目的物質
は保持され、低分子量不純物を大幅に除去するこ
とができる。 上記の各分離法によつて充分に純度の高いポリ
プレニル画分が得られない場合には、これら分離
法の2種もしくはそれ以上の組合わせを用いるこ
ともできる。例えば、クロマトグラフイーと分別
溶解法;クロマトグラフイーと分別冷凍沈殿法と
分別溶解法;クロマトグラフイーと分別冷凍沈殿
法と分別溶解法と分子蒸留法;クロマトグラフイ
ーと分子蒸留法と分別溶解法;クロマトグラフイ
ーと分子蒸留法;分子蒸留法と分別溶解法;分子
蒸留法と分別溶解法と分別冷凍沈殿法などの組合
わせを用いることができる。 かくして、薄層クロマトグラフイーにおける
Rf値が0.18〜0.25および/または0.50〜0.55の画
分が単離回収される。Rf値が0.18〜0.25の画分
は、前記一般式()におけるAが水酸基を表わ
す場合の同族体の混合物から実質的に成り、他
方、Rf値が0.50〜0.55の画分は前記一般式()
におけるAがアセトキシ基を表わす場合の同族体
の混合物から実質的に成る。 かくして得られる画分をさらに例えば分配型高
速液体クロマトグラフイーに付することによつ
て、個々の同族体成分を単離することもできる。 なお、上記分離工程において、抽出物を上記の
分離操作に供する前に、該抽出物を加水分解し
て、そこに含まれうる前記一般式()における
Aがアセトキシ基を表わす場合の同族体を対応す
るAが水酸基を表わす場合の同族体に予め転化す
ることが可能である。かくすることにより次いで
行なう分離操作が簡単になることがある。しか
し、かかる加水分解は勿論分離操作が終つた後の
Rf値が0.50〜0.55の成分を含む画分に対して行な
うこともできる。この加水分解は公知の脂肪酸エ
ステル類を加水分解するために知られている通常
の任意の方法を用いて行うことが可能であり、た
とえば含水メタノールまたはエタノール中に水酸
化ナトリウムまたは水酸化カリウムを溶解させた
溶液(アルカリ金属水酸化物濃度は好ましくは約
0.1〜30重量%とすることができる)100重量部に
対して上記の抽出物または画分を約5〜50重量部
の割合で加えて約25〜90℃で約0.5〜5時間反応
させればよい。 以上述べた方法によつて単離回収されるポリプ
レニル画分においてRf値が0.18〜0.25の画分は前
記一般式()におけるAが水酸基を表わす場合
の複数のポリプレノール同族体の混合物から実質
的になり、またRf値が0.50〜0.55の画分は前記一
般式()におけるAがアセトキシ基を表わす場
合の複数のポリプレニルアセテート同族体の混合
物から実質的になるものである。抽出物中におけ
る前者と後者の存在比率(ポリプレノール同族体
混合物:ポリプレニルアセテート同族体混合物)
は大体20:1乃至1:100(重量比)の範囲内にあ
り、また各画分中のポリプレノールまたはポリプ
レニルアセテート同族体の分布状態(分布パター
ン)は原料植物の種類によつて異なり、同一種の
原料植物から得られるものの分布状態は大体同じ
であり、原料として用いた葉の若さや採取時期お
よび地域などの要因に関係なくほぼ一定である。 本発明方法により得られるポリプレニル組成物
を哺乳動物のドリコール類に誘導するに際して該
組成物をそのまま用いてもよく、あるいは必要に
応じて該組成物を構成する各ポリプレニル化合物
を単離したのち反応させることもできる。さら
に、該組成物または単離されたポリプレニル化合
物を、それが遊離のアルコールであるかまたはア
セテートの形であるかに応じて、加水分解、エス
テル化もしくはエステル交換反応またはそれらの
2種以上の反応に付することにより、ポリプレノ
ールをポリプレニルエステルに、またはポリプレ
ニルアセテートを一般式()で示される他のポ
リプレニルエステルもしくはポリプレノールに変
換してもよい。 本発明方法により製造される一般式()で示
されるポリプレニルエステルの例として、一般式
()中のAをRCOO−で表わすとき、該Rが水
素原子、炭素数1〜18のアルキル基(例えば、メ
チル、エチル、n−プロピル、イソプロピル、n
−ブチル、sec−ブチル、イソブチル、tert−ブ
チル、n−ペンチル、イソアミル、n−ヘキシ
ル、n−オクチル、n−デシル、n−ドデシル、
n−ウンデシル、ステアリルなど)、炭素数2〜
18のアルケニル基(例えば、3−ブテニル、3−
ペンテニル、4−ペンテニル、ゲラニル、ゲラニ
ルメチル、フアルネシル、フアルネシルメチルな
ど)、炭素数5〜7のシクロアルキル基(例えば、
シクロペンチル、シクロヘキシル、メチルシクロ
ヘキシル、シクロヘプチルなど)、炭素数6〜10
のアリール基(例えば、フエニル、トリル、キシ
リル、ナフチルなど)、炭素数7〜11のアラルキ
ル基(例えば、ベンジル、フエネチル、メチルベ
ンジル、ジメチルベンジル、α−またはβ−ナフ
チルメチルなど)または1〜3個のフツ素原子も
しくは塩素原子で置換されたメチル基(例えばモ
ノフルオロメチル、トリフルオロメチル、モノク
ロルメチル、ジクロルメチルなど)であるエステ
ル類を挙げることができる。これらのポリプレニ
ルエステルまたはその混合物を得るための一般式
()で示されるポリプレノールまたはその混合
物のエステル化またはエステル交換反応はエステ
ル類を得るための自体公知のエステル化またはエ
ステル交換反応方法および条件を用いて行なうこ
とができる。例えば、エステル化は一般式()
で示されるポリプレノールまたはその混合物をエ
ステル化触媒および溶媒の存在または不存在下に
ギ酸、酢酸、モノフルオロ酢酸、モノクロル酢
酸、トリフルオロ酢酸、プロピオン酸、酪酸、カ
プロン酸、カプリル酸、カプリン酸、ラウリン
酸、パルミチン酸、ステアリン酸、オレイン酸、
リノール酸、リノレン酸、フアルネシル酸、フア
ルネシル酢酸、安息香酸、3,5−ジメチル安息

はない。なお、実施例および参照例中、IR分析
は液膜で測定し、NMRはTMSを内部標準とし
てCDCl3中で測定した。FD−MASS分析(電界

mm)を使用し、10cm展開〕においてRf値0.51およ
び0.20を含む画分を分離して約23gの油状物を得
た。なお、上記薄層クロマトグラフイーにおいて
ソラネシルアセテートはRf値0.40を示した。この
油状物にアセトン約400mlを加えてアセトン可溶
成分を溶解し、得られた混合物を過し、液を
濃縮し、得られた液状物をメタノール400ml、水
40mlおよび水酸化ナトリウム30gと共に2時間65
℃に加熱したのちメタノールを留去し、残留物に
ジエチルエーテル400mlを加えて抽出し、エーテ
ル層を約100mlの飽和食塩水で5回洗浄したあと
無水硫酸ナトリウムで乾燥し、溶媒を留去して
18.2gの液状物を得た。ついで、この液状物を約

化カリウム5gと共に2時間65℃に加熱したのち
メタノールを留去し、残留物中にヘキサン200ml
を加えて抽出し、ヘキサン層を約50mlの飽和食塩
水で5回洗浄したあと無水硫酸マグネシウムで乾
燥し、溶媒を留去して2.7gの液状物を得た。つ
いで、この液状物を約100gのシリカゲルおよび
クロロホルムを用いて分離し、前述の薄層クロマ
トグラフイーにおいてRf=0.20を示す画分として

gの液状物を得た。このものを約1Kgのシリカゲ
ルを用い、メチレンクロリドを展開液とするカラ
ムクロマトグラフイーにより精製して7.2gの液
状物を得た。この液状物は90%以上の純度を有す
るポリプレノールであり、このものについて実施
例1と同じ方法で測定した分子量分布は下記のと

出溶媒を抽去して残留物の重量を測定し、抽出物
総量として表4にまとめた。これらの抽出物をヘ
キサン200mlに溶解し、その溶液をメタノール/
水=9/1(容量比)の混合溶液約100mlで3回洗
浄したのち、無水硫酸マグネシウムで乾燥し、溶
媒を留去して油状物を得た。この油状物をメタノ
ール50ml、水酸化カリウム1gとともに2時間65
℃に加熱したのちメタノールを留去し、残留物に
ジエチルエーテル100mlを加えて抽出し、エーテ
ル層を約50mlの飽和食塩水で3回洗浄したのち無
水硫酸マグネシウムで乾燥し、溶媒を留去して油
状物を得た。この油状物を100gのシリカゲルを
用いn−ヘキサン/酢酸エチル=9/1(容量比)
の混合液で分離し、実施例1におけると同様の薄
層クロマトグラフイーによりRf値0.19を示す画分
(ポリプレノール混合物)を得た。この画分の重
量をポリプレノール含量として表4にまとめて示
した。なお、得られたポリプレノール混合物の組
成は、用いた溶媒の種類に関係なく、実施例2に
おいて得られたポリプレノール混合物のそれと実
質的に一致していた。

ルエーテルを留去し、淡黄色粘性液体を得た。こ
のものをシリカゲルカラムクロマトグラフイー
(ヘキサン/酢酸エチルを展開液として使用)に
より精製し1.01gの微黄色液体を得た。このもの
についてIR分析したところ約3300cm-1の原料ポ
リプレノールのOH基に起因する吸収が消失し、
−OCOCH3に起因する1745cm-1および1255cm-1の
吸収が現われた。また、NMR分析を行なつたと
ころ原料ポリプレノールの−CH 2OHに帰属さ
れるシグナル(doublet,δ=4.08)が消失し、−
CH 2OCOCH3に帰属される新たなシグナル
(doublet,δ=4.55)が観測された。−
CH2OCOCH 3に帰属されるべきシグナルは−C
H2−C=に帰属されれるシグナル(δ=2.04)
と重なつて観測された。また、FD−MASS分析
によりm/e=1216を得た。以上のことからこの
液体がn=14、A=OCOCH3である一般式()
のポリプレニルアセテートであることが確認され
た。nが14以外のポリプレニルアセテートおよび
nが10〜22に任意に分布するポリプレニルアセテ
ート混合物も同様の操作により合成できた。 実施例 28 実施例1と同様にしてコウヤマキから得られた
n=14、A=OHである一般式()のポリプレ
ノール1.17g、オレイン酸メチル0.5gおよび水
素化ナトリウム0.01gをトルエン50ml中に溶解し
110℃で24時間窒素ガス雰囲気下で加熱した。反
応溶液を室温まで冷却したのち飽和食塩水で洗浄
し、無水硫酸マグネシウム上で乾燥したのち溶剤
を留去して黄色液状物を得た。このものをシリカ
ゲルカラムクロマトグラフイー(ヘキサン/酢酸
エチルを展開液として使用)により精製して0.44
gの無色粘性液体を得た。この液体をIR分析し
たところ原料ポリプレノールのOH基に起因する
吸収約3300cm-1が消失していた。また、FD−
MASS分析によりm/e=1438を与えた。以上
のことからこのものがn=14、A=OCOC17H33
である一般式()のポリプレニルオレエートで
あることが確認された。nが14以外のポリプレニ
ルオレエートおよびnが10〜22に任意に分布する
ポリプレニルオレエート混合物も同様の操作によ
り合成できた。 実施例 29 実施例2と同様にしてハイイヌガヤから得られ
たn=16、A=OHである一般式()のポリプ
レノール1.31gとピリジン10mlとの混合物中に室
温でベンゾイルクロリド0.28gを加え、一夜室温
で撹拌した。ついで反応混合物を約150mlの水中
に注ぎ、ジエチルエーテルで抽出し、得られたエ
ーテル層を飽和食塩水、希塩酸水、飽和炭酸水素
ナトリウム水、そして再び飽和食塩水で洗浄した
のち、無水硫酸マグネシウム上で乾燥し、ジエチ
ルエーテルを留去して黄色液体を得た。これをシ
リカゲルカラムクロマトグラフイー(ヘキサン/
酢酸エチルを展開液として使用)により精製し
0.79gの微黄色液体を得た。このものについてIR
分析したところ原料ポリプレノールのOH基に起
因する吸収が消失し、1715cm-1及び1270cm-1にエ
ステル結合に起因する吸収が現われた。またFD
−MASS分析したところm/e=1414を与えた。
以上のことからこの液状物がn=16、A=
OCOC6H5である一般式()のポリプレニルベ
ンゾエートであることが確認された。nが16以外
のポリプレニルベンゾエート及びnが12〜20に任
意に分布するポリプレニルベンゾエート混合物も
同様の操作により合成できた。 参考例 アルゴン置換した3つ口フラスコに、マグネシ
ウム細片0.316g(13mmol)と無水テトラヒド
ロフラン0.5mlおよび1,2−ジブロモエタン
0.08mlを入れ、これをドライヤーで激しく泡立つ
まで加熱した。次に2−〔4−ブロモ−3−メチ
ルブトキシ〕−テトラヒドロ−2H−ピラン2.51g
(10mmol)の無水テトラヒドロフラン(3.0ml)
溶液を、この活性化されたマグネシウムに溶媒が
丁度沸騰するような速度で滴下した。滴下終了後
この混合物を70℃にて15分間撹拌した。これに無
水テトラヒドロフラン60mlを加えてグリニヤール
溶液とした。 別にアルゴン置換した3つ口フラスコに一般式
()においてn=15、A=−OCOCH3であるポ
リプレニルアセテート6.42g(5mmol)の無水
テトラヒドロフラン(15ml)溶液とLi2CuCl4の無
水テトラヒドロフラン溶液(0.1モル溶液、2.0
ml)を入れた。これに先に調製したグリニヤール
溶液を0℃で1時間かけて滴下し、さらに0℃で
2時間撹拌を続けた。そののち、この反応混合物
に飽和塩化アンモニウム水を加えて加水分解し、
エーテル抽出した。エーテル層を飽和食塩水で洗
浄し、無水硫酸マグネシウム上で乾燥したのち回
転蒸発器を用いて溶媒留去して7.69gの淡黄色液
状物を得た。このものはシリカゲル薄層クロマト
グラフイー(ヘキサン/酢酸エチル=97/3を展
開液として使用)によりRf=0.35に主スポツトを
有していた。また、この淡黄色液状物をFD−
MASS分析したところ原料ポリプレニルアセテ
ートの存在を示すm/e=1284は全く検出され
ず、一般式()においてn=15、Z=テトラヒ

前または分離後に該ポリプレニル化合物またはそ
の混合物を加水分解、エステル化もしくはエステ
ル変換反応またはそれらの2種以上の反応に付す
ることを特徴とする一般式 (式中、Aは水酸基またはアシルオキシ基を表
わし、【式】はトランス型イソ プレン単位を表わし、【式】は シス型イソプレン単位を表わし、nは10から22ま
での整数である。) で示されるポリプレニル化合物またはその混合物

件下に0.18〜0.25および/または0.50〜0.55の範
