Description
(イ) 本発明はビフエニル代謝機能を有するシユ
ウドモナス・シユウドアルカリゲネス
(Pseudomonas Pseudoalcaligenes)由来の新規
な外来遺伝子pph A−C及びこれを保有する新
規な微生物シユウドモナス・シユウドアルカリゲ
ネスKF707株に関するものである。
本遺伝子は、例えば増殖能の高いシユウドモナ
ス・プチダ(Pseudomonas putida)やシユウド
モナス・エルギノーサ(Pseudomonas
aeruginosa)等に組換え形質転換させた微生物
を用いて化合物2−ハイドロキシ−6−オキソ
−6−フエニルヘキサ−2,4−ジエノイツクア
シツド及びその誘導体を製造せしめ、病源菌とし
て知られている例えばスタフイロコツカス属など
のグラム陽性菌及び大腸菌などのグラム陰菌に対
する殺菌剤や生理活性物質の中間体としての生産
に寄与できることが期待されるものである。
(ロ) 従来の技術
従来、ビフエニルを資化する細菌については数
種類分離され、その代謝様式も検討されている
が、遺伝子単位で検討された例は知られていな
い。
また、ビフエニルの特定の中間代謝物について
は代謝様式を含めて遺伝子単位で検討された例は
ない。
そこで、本発明者らはビフエニル代謝機構に関
連する遺伝子について研究を進めた結果、シユウ
ドモナス・シユウドアルカリゲネス由来の遺伝子
bph A−Cをとり出すことに成功し、本発明を
完成するに至つた。
(ハ) 発明の構成
外来遺伝子bph A−Cの給源としては、ビフ
エニル代謝に関与する遺伝子を体内に保有する微
生物で、本発明者が自然界より分離したシユウド
モナス・シユウドアルカリゲネスKF707
(Pseudomonas PseudoalcaligenesKF707)
(FERM P−8297)を例示できる。
なお、本菌株の菌学的性質は以下のとおりであ
る。

以上の菌学的性質からバージイーズ、マニユア
ル オブ システイマテイク オブ バクテリオ
ロジー第9版に基づき検索した結果、シユウドモ
ナス・シユウドアルカリゲネス(Pseudomonas
pseudoalcaligenes)と認められた。
なお、本菌株は、新規な外来遺伝子bph A−
Cを保有する点で新規な微生物である。
本菌株からの該遺伝子の切出しは、前記菌株を
例えばL倍地(バクトリプトン10g、イーストエ
キス5g、食塩5g、蒸溜水1)に一晩増殖さ
せ、リゾチーム−SDS法により溶菌した染色体
DNAを調製し、次いで、染色体DNA(1μg)を
制限酵素Xhoで切断することにより、本遺伝子
を得ることができる。
本遺伝子の各種制限酵素による切断数は第1表
に示すとおりである。

次に、本遺伝子のビフエニル代謝様式と遺伝子
群との関係は次に示すとおりである。
上記反応式において、A、B、Cは反応を司る
酵素を示し、Aはビフエニル オキシゲナーゼ、
Bはジヒドロキシジオール デヒドロゲナーゼ、
Cはフエニルカテコール オキシゲナーゼを各々
示している。
また、これらの酵素に対応する遺伝子として、
bphA、bphB、bphCが存在しこれらの遺伝子は
オペロンを形成している。またPはプロモーター
を示す。
本遺伝子の利用にあたつては、例えば、大腸菌
エシエリヒア・コリKF637(Escherichia coli
KF637)(FERM P−8296)由来のpKF330を常
法により該菌株より取り出した後、制限酵素Xho
で切断後T4リガーゼで結合させ組換えプラス
ミドpMFB1を構築する(第3図)。
なお、pKF330は第2図に示すようにカナマイ
シン耐性とストレプトマイシン耐性を有する
12.6kbのプラスミドである。カナマイシン耐性遺
伝子部位にはXhoなどの挿入失活部位を有して
おり、またカナマイシンのプロモーターを利用で
きる。
次に宿主株として増殖能の高いシユウドモナ
ス・プチダKF107(Pseudomonasputida KF107)
(FERM P−8294)やシユウドモナス・エネギ
ノーサKF204(Pseudomonas aeruginosa
KF204)などを例示できる。形質転換方法及び形
質転換体の選択にあたつては、該菌株等を対数増
殖期(5×108 細胞/ml)まで培養し、集菌、
洗浄後、冷バツフアー(10mM MOPS−10m
M RbCl−100mM MgCl2、PH7.0)に懸濁し、
次いで遠心後、冷バツフアー(100mM
MOPS−10mM RbCl−100mM CaCl2、PH
6.5)に再懸濁し0℃、30分インキユベートする。
次に遠心後、菌株を1/10量の冷バツフアーに懸
濁する。この0.2mlコンピテントセルに組換えプ
ラスミド(0.5μg)を加え0℃にて1時間インキ
ユベート後42℃で2分間ヒートシヨンクする。3
mlのL倍地を加え、30℃で3時間インキユベート
した後ストレプトマイシン(200μg/ml)を含
むL−寒天培地上でpKF330及び組換えプラスミ
ドを保有する形質転換体を一次スクリーニングす
る。ビフエニル遺伝子を保有する目的とする形質
転換体(組換え微生物)は2,3−ジヒドロキシ
ビフエニル溶液(1mg/ml)を一次スクリーニン
グで生じたコロニーに噴霧することにより黄変す
るコロニーを選択する。ビフエニル代謝遺伝子を
含むクローンはビフエニル及びビフエニル関連化
合物より黄色物質を蓄積させ、黄色物質を酸性下
(PH1〜2)で酢酸エチルで抽出後、トリチルシ
リル化してGC−MSによる分析を行いこれが化
合物(図4)及びその誘導体であることを確認
する。
(ニ) 実施例
実施例 1
ビフエニル資化性菌シユウドモナス・シユウド
アルカリゲネスKF707株(FERM P−8297)を
L培地で一晩培養し、集菌、洗浄後、0.1Mトリ
ス(PH7.9)、1mM EDTA バツフアーに懸濁
する。次にリゾチームを最終濃度2μg/mlにな
るように加え、室温で5分間インキユベートし、
10%SDSを50μ/mlになるように加え溶菌し
た。
次いでプロナーゼ、RNase処理をしたのちフ
エノール抽出を行い、エーテルでフエノールを除
去した。このようにして調製したDNAは10mM
トリス、1mM EDTA バツフアーに透析し
た。
一方、プラスミドpKF330を有するエシエリヒ
ア・コリKF637(FERM P−8296)をL培地で
一晩培養し、アルカリ−SDS法によりpKF330を
調製した。染色体DNA及びプラスミドpKF330
を制限酵素XhoIで切断後、T4−リガーゼで連結
した。次いで組換えプラスミドを宿主株であるシ
ユウドモナス・エルギノーサKF204(FERM P
−8295)に導入した。すなわち、対数増殖期(約
4×108セル/ml)のKF204株を集菌し、等量の
冷バツフアー(10mM MOPS、10mM RbCl、
100mM MgCl2、PH7.0)で洗浄後、冷バツフア
ー(100mM MOPS、10mM RbCl、100m
M CaCl2、PH6.5)に再懸濁し、0℃にて30分間
放置した。次に遠心後、1/10量の冷バツフアー
に再懸濁し、その0.2ml細胞懸濁液と精製した
pMFB1(0.5μg)と0℃、1時間インキユベート
した。42℃で2分間、ヒートシヨツクした後、3
mlのL培地を加え30℃で3時間培養した。ビフエ
ニル代謝遺伝子群(bph A−C)の組込まれた
pMFB1を保有する形質転換体はストレプトマイ
シン(200μg/ml)を含むL−寒天培地で2,
3−ジヒドロキシビフエニル溶液(1mg/ml)を
噴霧して黄色となるコロニーとして選択した。次
いで形質転換体をストレプトマイシン200μg/
mlを含むL培地で一晩培養後、常法によりプラス
ミドを調製した。次いで調製したパラスミド
pMFB1をXhoで切断すると7.9kbのbph A−
C遺伝子が切り出された。本遺伝子は第1図に示
す制限酵素切断点を有していた。
実施例 2
実施例1で調製した組換えプラスミドpMFB1
を用いて、シユウドモナス・プチダKF107
(FERM P−8294)を実施例1の方法によつて
形質転換した。その結果、1×103セル/μg
DNAの頻度でpMFB1を保有する形質転換体シユ
ウドモナス・プチダKF138が得られた。
実施例 3
実施例1により調製したpMFB1を保有するシ
ユウドモナス・エルギノーサKF257を炭素源とし
てこはく酸(1mg/ml)を含むBSM寒天培地に
塗布(摂取)しビフエニル粉末をペトリ皿のふた
においてビニールテープでシールした。KF257株
の増殖とともにビフエニル蒸気をとり込んだ菌体
は、ビフエニルを化合物に変化せしめ、培地は
鮮やかに黄変した。
上記の反応はpMFB1を保有しない宿主株シユ
ウドモナス・エルギノーサKF204(FERM P−
8295)では全く認められなかつた。以上の反応は
pMFB1を保有するシユウドモナス・プチダ
KF138においても同様に認められた。
なお、宿主株及びpMFB1を保有するKF257株
について

以上の性質等によりKF138株はシユウドモナ
ス・プチダであることを確認した。
(ホ) 発明の効果
ビフエニル代謝機能を有する外来遺伝子bph
A−Cを増殖効率の優れた微生物に組換え、形質
転換微生物を用いて、化合物 2−ハイドロキ
シ−6−オキソ−6−フエニルヘキサ−2,4−
ジエノイツクアシツド及びその誘導体を安価に製
