Description
本発明は、トコフエロール及び特にd−α−ト
コフエロールの製造方法、並びにその方法の中間
生成物及びその応用に関する。
トコフエロール、特にd,l−α−トコフエロ
ール(ビタミンE)の工業的な全合成は、1938−
1940年に発表されたP.Karrerの研究(例えば、
P.Karrer、H.Fritzsche、B.H.Ringier、H.
Salomon、Helv.Chim.Acta21,520(1938))に
までさかのぼることができ、又その方法は、トリ
メチルヒドロキノンと、フイトール又はイソフイ
トールとの縮合に基づいている。このためには、
ギ酸、塩化亜鉛又は三フツ化ホウ素エーテラート
のような酸性の縮合剤が必要である(ドイツ特許
第713749号1939年;第731972号1938年)。
トリメチルヒドロキノンの代りに、銅/亜鉛粉
末とギ酸との存在の下で、トリメチルベンゾキノ
ンを使用することもできる(M.KaJiwara、D.
Sakamoto、S.Ohta、Heterocycles14,1995
(1980))。
ピロリジンのような塩基性の縮合剤は、2−ア
セチル−3,5,6−トリメチルヒドロキノンと
フアルネシルアセトンとの反応に使用され、その
生成物は次の還元と水素化との段階で同様にd,
l−トコフエロールになる(H.J.Kabbe、H.
Heitzer、synthesis1978,888)。
フイトール又はイソフイトールの代りに、3,
7,11,15−テトラメチル−1,3−ヘキサデカ
ンジオールも、酸性の触媒(ZnCl2、BF3・
OEt2)の存在の下で、トリメチルヒドロキノン
に縮合することができる(L.Bla′ha、J.
Hodrov′a、J.Weichet、Collection Czechosloc.

(1965))。
殆んど専ら出発製品として使用されるトリメチ
ルヒドロキノンの製造には、一連の反応段階
(3,5−ジメチルフエノール又は1,2,4−
トリメチルベンゼン(プソイドクメン)から出発
する)が必要である。文献から公知である全収率
は30%を下廻る。
全収率が60%である比較的新らしいトリメチル
ヒドロキノンの4段階での合成は、出発製品とし
てスルフアニル酸の外に、4−メトキシ−3−ブ
テン−1−インから60乃至70%の収率で得られる
4−メトキシ−3−ブテン−2−オンを必要とす
る。
従来通りの方法を、天然のビタミンEの製造に
変形することはできない。天然に存在するd−α
−トコフエロールの合成は、これまで手に入れる
ことが困難な2R−配置クロマン前駆体(例えば
2−ホルミルクロマン)を経てのみ可能であつ
て、それにイソプレン側鎖を結合する(H.
Mayer、P.Schudel、R.Ru¨egg、O.Isler、Helv.

本発明は、トリアルキルヒドロキノンを使用し
ないで、融通性のある方法で実施することのでき
るトコフエロール及び特に天然のビタミンEを製
造する方法を開発するという課題に基づいてい
る。特にその方法は、天然のd−α−トコフエロ
ールを合成できるものでなくてはならないが、そ
れは天然のトコフエロールはその生物学的活性に
おいて、ラセミ体のd,l−製品よりも35%程度
まで勝つているからである(ビタミン試験方法、
第3版、Interscience社1966年参照)。この課題
のより詳細な様相は、以下の説明から明らかにな
る。
この課題は、始めに上げたような方法を開発す
ることによつて解決されるが、その方法は、アル

反応生成物(10)から金属カルボニル部分を分離し、
金属の入つていない生成物(11)を脱エーテルして、
酸性の触媒の存在の下に希望するトコフエロール
に環化することを特徴とする。
本発明によつて使用されるカルボニル−アルケ
ニルカルベン−金属錯体(1)に包含される金属原子
は、一般には第6亜族の金属を表わすが、この場
合これらのものの中で再びクロムが特に好適であ
る。特に好都合な合成結果は、式(1)、
の化合物を指定する際に得られた。この化合物に
おいては、1つ又は2つ以上のカルボニル基を他
の配位子、例えば水素原子を一般には再びアルキ
ル化又はアリール化したホスフイン配位子、アミ
ン配位子、あるいは、例えばベンゼン、シクロペ
ンタジエニルのようなπ−結合芳香族によつて、
置き換えることができる。そのような配位子や他
の配位子は、カルベン化学から公知である。
カルボニル−アルケニルカルベン−金属錯体と
アルキノールとの反応は、一般には保護ガスの存
在の下で実施される。保護ガスとしては、例えば
窒素、アルゴンなどが上げられる。溶媒として
は、供与体溶媒、例えばジアルキルエーテル、
THFなどが好適に考慮される。反応は、20乃至
80℃の温度範囲において、高い反応速度で行われ
る。
原料のカルベン錯体(1)は、それ自体公知の方法
で製造することができるが、それはクロムペンタ
カルボニル錯体によつて、更に詳細に説明する積
りである。上述の場合には、市販のヘキサカルボ
ニルクロムと1−リチオアルケン並びに求電子性
アルキル化剤(例えばオキソニウム塩、硫酸ジア
ルキル、トリアルキルハロゲンシラン、アシル化
合物)から出発するが、その場合、原料化合物(1)
を約70乃至90%の収率で得ることができる。合成
はC=C−二重結合に対して立体規則的に起る。
1−リチオアルケンは、1−ハロゲンアルケンと
リチウム又はリチウム−アルキル化合物とから製
造される(F.G.Bordwell、Ph.S.Landis,J.
Amer.Chem.Soc.79,1593(1957);D.Seebach、
H.Neumann、Chem.Ber.107,847(1974))。
クロムのカルベン錯体は、一部は0℃以下の融
点を有する赤色の結晶を一般には示し、保護ガス
(窒素、アルゴンなど)の存在の下で冷時長く貯
蔵することができる。
アルキノールの製造は、アルキン−エンから出
発して行われるが、プレニルプロマイドとアルキ
ニル−グリニヤール試薬とからの上述の合成は、
欧州特許明細書第0015436号(Hoff−man−La
Roche)(公開番号第00 100 822.8号)に記載さ
れている。このアルキン−エンは過酸、例えばm
−クロロ過安息香酸によつて酸化されてエポキシ
−アルキンとなり、それは例えばLiAlH4などに
よつて容易に還元されて希望するアルキノールに
なる。
カルボニル−アルケニルカルベン−金属錯体と
アルキノールとの反応は、ほぼ定量的な収率で行
われるが、その際反応生成物はなお金属カルボニ
ル部分を包含している。反応生成物は、原料アル
キノールの種類には無関係に、純粋な鏡像体又は
ラセミ混合物を示す。アルキノールを純粋に鏡像
体の形で添加すると、生成物(10)は同様に純粋な鏡
像体の形で生成する。反応生成物(10)は、生成物(11)
を生成して金属カルボニル部分から解放される。
一般にこの場合加圧した一酸化炭素によつて反応
を行わせるが、ここで生成した金属カルボニル
は、原料のカルボニル−アルケニルカルベン−金
属錯体(1)を生成するために、戻すことができる。
この変形によつて、特に有利にまたは合理的に本
方法を実施することができる。金属の入つていな
いヒドロキノンモノエーテル製品(11)もまたほぼ定
量的な収率で得られる。この金属の入つていない
生成物(11)は、種々な方法で脱エーテルすることが
できる。それは例えばエーテル残基R3の種類に
よつて決めることができるが、加水分解、水素化
分解又はルイス酸を使用して行うことができる。
エーテル型置換基がアシル残基である場合には、
加水分解によつて除去される。R3が例えばSiR3
(R3=低級アルキル)を表わす場合には、この残
基は水素化分解によつて、あるいはR3=低級ア
ルキルである場合には、三臭化ホウ素によつて除
去される。脱エーテルも高い収率で行われ、例え
ば80乃至96%の範囲にある。次いでヒドロキノン
誘導体(12)は、好適には酸性の触媒、例えば三フツ
化ホウ素−エーテラート、p−トルエンスルホン
酸、ギ酸などによつて環化されて、希望するトコ
フエロールになる。
本発明による方法の過程は、次の反応図式1に
示してあるが、ここでは金属カルベン錯体とし
て、ペンタカルボニルクロム−カルベン錯体を使
用している。しかしながらこの特別なカルベンを
指定したことは、この過程を理解させるためだけ
であるので、これによつて本発明による方法を制
限することを何等目的とするものではない。

それ故に、本発明による合成は特に重要である
が、それは、反応が立体異性を選体的に行われ、
それによつて天然のd−α−トコフエロールの合
成が直ちに可能であるからである。このことは次
に示す反応図式2から明らかになる筈である。
注目に値することは、上述の反応図式2におい
て、イノールが縮合段階で(閉環してクロマンに
なる)転化して反応することである。それ故に、
6S−イノールから出発して、縮合段階における
転化で、天然の2R−α−トコフエロール(ビタ
ミンE)を得る新しい方法が可能になる。
鏡像体6S−(あるいは6R−)イノールの製造
は、例えば軸左右対称の金属アルキル化合物によ
つて、ケトンを立体異性選択的にアルキル化して
行われる(D.Seebach,有機合成における有機金
属化合物、オランダ・ワーゲニンゲン シンポジ
ウム、1982年3月18/19日参照)。
対応するエン−イン(2)を立体異性選択的に(8)に
酸化する時に、別の方法で鏡像体6S−乃至は6R
−イノールを得ることもできる。
本発明には、式(10)と(11)との中間生成物も包含さ
れるが、その際(10)は金属トリカルボニル部分をな
お包含している。それ故に、本発明は特に式(10)の
化合物、
を包含する。
同様にして本発明による式(11)のヒドロキノンモ
ノエーテル、
が包含される。
最後に、本発明によつて、トコフエロール、特
にビタミンEの製造へのヒドロキノンモノエーテ
ル(11)並びにその金属トリカルボニル置換誘導体(10)
も、特許請求される。
本発明を、以下に示す実施態様で更に明らかに
する筈であるが、それは本発明を制限するものと
して解すべきものではない。
実施態様
金属−カルベン錯体(1)、例えばペンタカルボニ
ル(E−2−アルケニルカルベン)クロム−錯
体(1)の製造
300mlのテトラヒドロフラン(又はジエチルエ
ーテル)中の6.6g(30mmol)のヘキサカルボ
ニルクロムに、−40℃で、等モル量のZ−2−リ
チオ−2−アルケンの−30℃に冷却したエーテル
溶液を、保護ガスの存在の下で滴下する。その場
合、色は黄橙色に変化する。−40℃で1時間放置
した後、溶媒を除去する。残留物を50mlの水の中
に懸濁し、200mlのペンタンをその上に注ぎ、更
に4.5gのトリメチルオキソニウムテトラフルオ
ロボラートを添加すると、色は直ちに深赤色に変
化する。ペンタン相を硫酸ナトリウムで乾燥し、
蒸発する。冷却すると赤色の結晶が得られる。
アシル化された金属化合物もテトラアルキルア
ンモニウム塩として遊離され、塩化メチレン中の
均一相で、トリエチルオキソニウムテトラフルオ
ロボラート、ハロゲン化アシル(塩化アセチル、
塩化ベンゾイル)又はトリメチルシリルクロライ
ドによつてアルキル化される。

チル−2−ノナデシン(8)の製造
50mlの塩化メチレン中の3.44gの3−クロロ過
安息香酸(77%純度)に、0℃で、20mlの塩化メ
チレンに溶解した4.50gの6,10,14,18−テト
ラメチル−5−ノナデセン−2−インを滴下す
る。2時間かきまぜた後ろ過し、ろ液を10%の亜
硫酸ナトリウム溶液と、5%の炭酸水素ナトリウ
ム溶液とで洗浄し、エーテルで抽出する。濃縮し
た有機相をシリカゲルのクロマトグラフにかけ
る。溶離剤として塩化メチレンを使つて4.25gの
無色の油が得られる(収率91%)。
