Description
【発明の詳細な説明】
本発明は、新規なピラノキノリン誘導体および
その薬理上許容されうる塩に関する。さらに詳し
くは、抗アレルギー作用を有する6−アルキル−
5,6−ジヒドロ−4,5−ジオキソ−4H−ピ
ラノ〔3,2−c〕キノリン−2−カルボン酸誘
導体およびその薬理上許容される塩に関する。
発明の背景
本発明化合物に類似した構造を有するピラノキ
ノリン−4−オン−2−カルボン酸誘導体とし
て、5,6−ジヒドロ−4,5−ジオキソ−4H
−ピラノ〔3,2−c〕キノリン−2−カルボン
酸誘導体(特開昭52−17498、および同52−
109000号公報)、およびそのエステル誘導体(特
開昭54−160398号公報)がすでに本発明者らによ
つて提供されている。
これらの化合物は、コツクスらによりアレルギ
−性喘息に有効であると報告されたクロモグリク
酸ナトリウムと同様の作用メカニズム(Adv.in
Drug Res.,5,115(1970))により抗アレルギ
ー作用を示すことが見出されている。すなわち、
上記化合物は、IgE様抗体によりひき起こされる
抗原抗体反応の結果生じる肥満細胞からのケミカ
ルメジエーターの遊離を抑制することによつて抗
アレルギー作用を示しうると考えられる。
発明の概要
本発明者らは、前記従来化合物と同等あるいは
それ以上の薬理活性を有する化合物を鋭意研究し
た結果、新規な6−アルキル−5,6−ジヒドロ
−4,5−ジオキソ−4H−ピラノ〔3,2−c〕
キノリン−2−カルボン酸誘導体およびその薬理
上許容される塩が優れた薬理活性を示すことを見
出して本発明を完成するに至つた。
本発明化合物の従来化合物との構造的差異は、
6位の窒素原子に炭素数1〜5のアルキル基が置
換したことである。従来の5,6−ジヒドロ−
4,5−ジオキソ−4H−ピラノ〔3,2−c〕
キノリン−2−カルボン酸誘導体が下記の5−オ
キソ型(a)、5−ハイドロキシ型(b)の互変異性を示
すことと比較し、本発明化合物は、6位にアルキ
ル置換基を有するため、上記の互変異性を示さず
5−オキソ型に固定されており、このため、有機
溶媒に対する溶解性が向上し、各製造工程および
最終生成物における精製が容易になつた。
本発明化合物は、動物実験においてクロモグリ
ク酸ナトリウムと同様な抗アレルギー作用を示
す。この事実はこの化合物が喘息、アレルギー性
鼻炎、アトピー性皮膚炎等のアレルギー症に対す
る治療薬として有用である可能性を示唆するもの
である。
発明の具体的説明
1 本発明化合物
本発明によれば、下記一般式〔〕で表わされ
る新規な化合物である6−アルキル−5,6−ジ
ヒドロ−4,5−ジオキソ−4H−ピラノ〔3,
2−c〕キノリン−2−カルボン酸誘導体および
その薬理上許容される塩が提供される。
(式中、Rは水素または炭素数1〜8のアルキ
ル基であり、R′は炭素数1〜5のアルキル基で
あり、そしてR1,R2、およびR3はそれぞれ水素、
炭素数1〜4のアルキル基、炭素数1〜4のアル
コキシ基およびハロゲン原子である。)
R,R′,R1,R2およびR3としては、具体例に
以下の基が挙げられる。

−1−ブチルエステル、3,3−ジメチル−2−
ブチルエステル、2−メチル−3−ペンチルエス
テル、n−ヘプチルエステル、2−ヘプチルエス

また、本発明化合物のうち、遊離カルボン酸誘
導体は薬理上許容される塩基の塩として使用する
事ができる。
また上記カルボン酸化合物の薬理上許容される
塩も、本発明化合物に包含される。具体的には、
ナトリウム、カリウム等のアルカリ金属塩、マグ
ネシウム、カルシウム等のアルカリ土類金属塩、
アルミニウム塩、アンモニア塩、トリス(ヒドロ
キシメチル)アミノメタン塩、N,N−ビス(ヒ
ドロキシエチル)ピペラジン塩、2−アミノ−2
−メチル−1−プロパノール塩、エタノールアミ
ン塩、N−メチルグルカミン塩、およびL−グル
カミン塩等が挙げられる。
本発明化合物のうち有用性の点から特にR1,
R2,R3が無置換およびR3がメトキシ基、および
メチル基の化合物が好ましく、またR′としては
炭素数2〜4の場合が好ましい。
具体的な化合物としては前述の(2),(3),(5),
(7),(15)およびそのエステルが好ましい。
2 化合物の製造法
一般的〔−A〕または〔−B〕で示される
本発明による化合物は、合目的的な任意の方法で
製造することができるが、下記の図に示す方法に
より、具体的に製造することができる。
(R′,R1,R2およびR3は前記と同様であり、
R″は炭素数1〜8のアルキル基であり、Xは塩
素、臭素またはヨウ素である)
(1) 遊離カルボン酸誘導体〔−A〕の製造法
この製造法では、出発物質である式〔〕で表
わされる3−アセチル−1−アルキル−4−ヒド
ロキシ−2−キノロン誘導体を溶媒中、もしくは
無溶媒下に塩基の存在下で式〔〕で表わされる
シユウ酸ジエステルと反応させて中間体〔〕を
製造し(工程A)、更にこの中間体〔〕を溶媒
中あるいは無溶媒下で酸により閉環ならびにエス
テルを加水分解反応させることにより、目的とす
る遊離カルボン酸誘導体〔−A〕を合成する
(工程B)。
〔工程A〕
出発物質 〔〕
出発物質である3−アセチル−1−アルキル−
4−ヒドロキシ−2−キノロンは、任意適当な方
法で合成することができるが、具体的には、例え
ば、J.Am.Chem. Soc., 68,324(1946)、に記載
された方法に従つてアルキルアニリンとジエチル
アセチルマロネートとを反応させるか、あるいは
薬学雑誌、71、1100(1951).に記載された方法に
従つて、1−アルキル−4−ヒドロキシ−2−キ
ノロンのフリーデルクラフツアシル化反応により
製造される。
シユウ酸ジエステル 〔〕
式〔〕で示されるシユウ酸ジエステルとして
は、前記Rの具体例で示したアルコール残基のジ
アルキルエステルが使用できる。使用量は化合物
〔〕に対しモル比で0.8〜20程度、好ましくは2
〜10である。
塩 基
工程Aを進行させるに際して用いられる塩基と
しては、例えば、RONa又はROK(式中R
は炭素数1〜5のアルキル基である)のアルコ
ラート、NaNH2,NaH,NaOH,KOH、ナト
リウム、カリウム等があげられ、使用量は化合物
〔〕に対しモル比で0.8〜20程度、好ましくは1
〜10である。
好適反応条件
温度 0〜200℃、好ましくは60〜150℃である。
時間 20分〜1日、好ましくは30分〜5時間であ
る。
溶媒 エチルエーテル、テトラヒドロフラン、ジ
オキサン等のエーテル類、クロロホルム、
ジクロルメタン、トリクレン等のハロゲン
化アルキル、メタノール、エタノール、プ
ロパノール、ブタノール、オクタノール等
のアルコール類、ベンゼン、トルエン、キ
シレン等の芳香族炭化水素類、ジメチルス
ルホキシド、リン酸ヘキサメチルトリアミ
ド(HMPA)等が用いられる。使用量は
化合物〔〕に対し重量比で1〜200であ
る。
反応終了後、後処理せずにそのまま工程Bに進
むことも可能であるが、中間体〔〕を単離する
場合は、必要に応じて溶媒を留去し、大過剰の水
を加え、酢酸等の酸にて中和し、析出した沈殿を
取するかあるいはクロロホルム、酢酸エチル等
の通常の溶媒で抽出することにより中間体〔〕
を得る。
〔工程B〕
酸
〔工程B〕を進行させるに際して用いられる酸
としては、例えば塩酸、硫酸、臭化水素酸等の鉱
酸、酢酸、パラトルエンスルホン酸、ベンゼンス
ルホン酸等の有機酸、および酸性樹脂等があげら
れ、使用量は化合物〔〕に対しモル比で1〜
100程度、好ましくは2〜50である。
好適反応条件
温度 0〜200℃、好ましくは50〜100℃である。
時間 5分〜10時間、好ましくは30分〜3時間で
ある。
溶媒 溶媒としては、メタノール、エタノール、
プロパノール、ブタノール、オクタノール
等のアルコール類、エチルエーテル、テト
ラヒドロフラン、ジオキサン等のエーテル
類、クロロホルム、ジクロルメタン、トリ
クレン等のハロゲン化アルキル、ベンゼ
ン、トルエン、キシレン等の芳香族炭化水
素、酢酸、プロピオン酸等の有機酸、ジメ
チルスルホキシド、HMPA等が用いられ
る。使用量は化合物〔〕に対し重量比で
1〜200である。
反応終了後、必要に応じて溶媒を留去し、大過
剰の水を加え、析出した沈殿を取するか、ある
いはクロロホルムや酢酸エチル等の通常の溶媒で
抽出した後、再結晶やカラムクロマト等の方法で
精製することにより遊離カルボン酸誘導体〔−
A〕を得る。
(2) カルボン酸エステル誘導体〔−B〕の製造
法
カルボン酸エステル誘導体〔−B〕は、大き
く分類して、(イ)前述の工程Aおよび工程Bにより
得られる遊離カルボン酸誘導体〔−A〕を、酸
ハライド法、酸触媒法、縮合剤法、酸無水物法、
活性エステル法、ハロゲン化アルキル法などの通
常のエステル化法によりエステル誘導体に変換す
るか、(ロ)前述のごとく製造した中間体〔〕を加
水分解することなく閉環するか、あるいは(ハ)特開
昭54−160398号公報に開示したようにして製造さ
れたピラノキノリン誘導体の6位をアルキル化す
ることにより製造される。
製造法の具体例のいくつかを示せば、下記のと
おりである。
イ) 遊離カルボン酸誘導体〔−A〕のエステ
ル化
a) 酸ハライド法
前述の一般式〔−A〕で表わされる遊離カル
ボン酸誘導体あるいはその塩を出発物質とし、溶
媒中あるいは無溶媒下で、所望により塩基の存在
下、酸ハライド化試薬により酸ハライド誘導体
〔〕とし(工程C)、更にこの誘導体〔〕を、
溶媒中あるいは無溶媒下で所望により塩基の存在
下、一般式〔〕で表わされるアルコールと反応
させて目的とするカルボン酸エステル〔−B〕
を合成する(工程D)。
〔工程C〕
酸ハライド化試薬
工程Cを進行させるに際して用いられる酸ハラ
イド試薬としては、具体的にチオニルクロライ
ド、三塩化リン、五塩化リン、オキシ塩化リン、
三臭化リン、三沃化リン等が挙げられる。使用量
は、出発物質〔−A〕に対しモル比で0.5〜50
程度、好ましくは1〜20である。
塩 基
工程Cを進行させるに際して、必要に応じて用
いられる塩基としては、具体的にジメチルアニリ
ン、ピリジン、トリエチルアミン等の有機塩基が
挙げられる。使用量は、出発物質〔−A〕に対
しモル比で0.01〜5である。
好適反応条件
温度 −20〜200℃程度、好ましくは0〜150℃で
ある。
時間 10分〜2日程度、好ましくは30分〜10時間
である。
溶媒 溶媒を使用する場合は、例えばジクロルメ
タン、クロロホルム、トリクレン等のハロ
ゲン化アルキル、ベンゼン、トルエン、キ
シレン等の芳香族炭化水素、エーテル、テ
トラヒドロフラン、ジオキサン等のエーテ
ル類、ジメチルホルムアミド等が用いら
れ、使用量は、出発物質〔−A〕に対し
重量比で1〜100程度、好ましくは5〜50
である。
反応終了後、後処理せずにそのまま(工程D)
に進むことも可能であるが、カルボン酸ハライド
〔〕を単離する場合は、必要に応じて酸ハライ
ド化試薬及び溶媒を留去した後、水を加え、析出
した結晶を取するか、あるいはクロロホルム、
酢酸エチル等の通常の溶媒で抽出する。チオニル
クロライドを使用した場合は濃縮乾固するだけで
次の工程に使用することができる。
〔工程D〕
ROH 〔〕
工程Dを進行するに際して炭素数1〜8のアル
キルアルコールが用いられ、具体的には以下の化
合物が挙げられる。
メタノール、エタノール、n−プロパノール、
i−プロパノール、n−ブタノール、i−ブタノ
ール、t−ブタノール、n−ペンタノール、2−
ペンタノール、3−ペンタノール、3−メチル−
1−ブタノール、2−メチル−1−ブタノール、
2,2−ジメチル−1−プロパノール、3−メチ
ル−2−ブタノール、n−ヘキサノール、2−ヘ
キサノール、3−ヘキサノール、4−メチル−1
−ペンタノール、2−メチル−1−ペンタノー
ル、3−メチル−1−ペンタノール、4−メチル
−2−ペンタノール、3−メチル−2−ペンタノ
ール、3,3−ジメチル−1−ブタノール、3,
3−ジメチル−2−ブタノール、2−メチル−3
−ペンタノール、n−ヘプタノール、2−ヘプタ
ノール、3−ヘプタノール、n−オクタノール、
2−オクタノール、3−オクタノール、4−オク
タノール等。使用量は、カルボン酸クロリド
〔〕に対しモル比で1〜100程度、好ましくは
1.2〜50である。
塩 基
工程Dを進行させるに際して必要に応じて塩基
を用いることができ、具体的にトリエチルアミ
ン、ジメチルアニリン、ピリジン、トリトンB、
ジアザビシクロウンデセン(DBU)等の有機塩
基、NaOH、KOH、K2CO3、NaCO3、NaNH2
等のアルカリ類が用いられる。使用量は、カルボ
ン酸クロリド〔〕に対するモル比で0.5〜10程
度、好ましくは1〜5である。
好適反応条件
温度 −30〜200℃程度、好ましくは−10゜〜150
℃である。
時間 30分〜1日程度、好ましくは1時間〜10時
間である。
溶媒 溶媒を使用する場合は、例えばジクロルメ
タン、クロロホルム、トリクレン等のハロ
ゲン化アルキル、ベンゼン、トルエン、キ
シレン等の芳香族炭化水素、ジオキサン、
テトラヒドロフラン等のエーテル類、アセ
トン、メチルエチルケトン等のケトン類、
ジメチルホルムアミド、ジメチルスルホキ
シド等が用いられ、使用量は、カルボン酸
クロライド〔〕に対する重量比で1〜
100程度、好ましくは5〜50である。
反応終了後は、必要に応じて反応液を濃縮し、
n−ヘキサン等の難溶性溶媒を加えて、析出する
結晶を取するか、あるいは水を加えて、クロロ
ホルム、酢酸エチル等の溶媒にて抽出後、再結晶
やカラムクロマトグラフイー等の通常の方法で精
製することにより、一般式〔−B〕で表わされ
るカルボン酸エステル誘導体を得る。
b) 酸触媒法
遊離カルボン酸〔−A〕あるいはその塩を、
塩化水素、硫酸、パラトルエンスルホン酸等の酸
触媒の存在下に一般式〔〕で示されるアルコー
ルと反応させて、カルボン酸エステル〔−B〕
を製造する。
c) 縮合剤法
遊離カルボン酸〔−A〕あるいはその塩を、
ジシクロカルボジイミド(DCC)、ジエチルフオ
スホリルシアナイド(DEPC)、ジフエニルフオ
スホリルアジド(DPPA)等の縮合試薬の存在下
に一般式〔〕で示されるアルコールと反応させ
て、カルボン酸エステル〔−B〕を製造する。
d) 酸無水物法
遊離カルボン酸〔−A〕をクロル炭酸エチル
等と反応させて混合酸無水物を形成させ、更にア
ルコール〔〕と反応させてカルボン酸エステル
〔−B〕を製造する。
e) 活性エステル法
遊離カルボン酸〔−A〕を、2,4−ジニト
ロフエノール、N−ヒドロキシサクシンイミド等
と反応させて、活性エステル〔〕を形成させ、
更にアルコール〔〕と反応させてカルボン酸エ
ステル〔−B〕を製造する。
f) ハロゲン化アルキル法
遊離カルボン酸〔−A〕のナトリウム、カリ
ウム等の金属塩、あるいはトリエチルアミン等の
有機塩基の塩〔〕をアルコール類、ケトン類お
よびHMPA等の溶媒中、式〔R″−〕で示され
るハロゲン化アルキルと反応させてカルボン酸エ
ステル誘導体〔−B〕を得る。
(R″およびXは前記と同様)
ロ) 中間体〔〕の酸閉環法
前述のようにして製造した中間体〔〕を溶媒
の存在下、あるいは無溶媒下で、塩酸、硫酸、臭
化水素酸等の酸触媒にて閉環しカルボン酸エステ
ル誘導体を得る。
ハ) 化合物〔〕の6位のアルキル化法
特開昭64−160398号公報に記載された方法にて
得られたキノロピラン−4−オン−2−カルボン
酸エステル〔〕を出発物質とし、この化合物
を、溶媒中、NaOH、KOH、NaH、NaNH2等
の塩基の存在下に式〔R′−X〕で示されるハロ
ゲン化アルキルと反応させカルボン酸エステル誘
導体〔−B〕を得る。
また、こうして得られたカルボン酸エステル誘
導体〔−B〕は、酸あるいは塩基の存在下、通
常のエステル加水分解条件にて、遊離カルボン酸
〔−A〕とすることができる。
3 薬効試験
本発明化合物の抗アレルギー作用の評価を、ラ
ツトにおける受動的皮膚過敏症試験(P.C.A)に
より行なつた。
(1) カルボン酸塩
ラツト(SLC系Wister rat)に5回再結晶した
卵白アルブミンおよび百日咳ワクチン(Bod−
etalla pertussis Vaccine)を皮内投与し、13日
後の血清を採取した。得られた血清は、ヒトのレ
アギンに類似した性質を有する抗体を含有し、抗
体価64以上を示した。この血清を12倍に希釈して
ラツトの背部に皮内投与し、48時間後に、表1に
示す各カルボン酸塩誘導体を1mg/Kg蒸留水に溶
解したものを、静脈内投与した。5分後、卵白ア
ルブミンと色素(エバンス・ブルー)の生理食塩
水溶液を静脈内投与し、さらに30分後、背部皮膚
を剥離して、抗原抗体反応の結果漏出した色素量
をNa2SO4とアセトンより抽出し、620mμにて比
色定量した。
得られた値を薬物を投与しない場合と比較して
アレルギー反応の抑制率を算出し、結果を表1に
示した。

(2) カルボン酸エステル誘導体ラツトに抗体投与
48時間後、表−2に示す各カルボン酸エステル
誘導体30mg/Kgを1%トラガント溶液に懸濁さ
せたものを経口投与し、30分後、卵白アルブミ
ンと色素を静脈内投与した以外は(1)と同様にし
て色素を比色定量した。
そしてこうして得られた値を(1)と同様に薬物
を投与しない場合と比較して、アレルギー反応
の抑制率を算出し、結果を表−2に示した。

表1および表2の評価結果から遊離カルボン酸
誘導体塩はいずれも静脈内投与した場合に、また
カルボン酸エステル誘導体はいずれも経口投与し
た場合に抗アレルギー作用を有することがわか
る。
4 製造例
実施例 1
5,6−ジヒドロ−6−メチル−4,5−ジオ
キソ−4H−ピラノ〔3,2−c〕キノリン−
2−カルボン酸
ナトリウム(460mg)無水EtOH(5ml)から
NaOEtを調整し、これに無水ベンゼン(15ml)、
シユウ酸ジエチル(3.5ml)、3−アセチル−4−
ヒドロキシ−1−メチル−2−キノロン(1.09
g)を加え、2時間還流撹拌した。溶媒を減圧留
去後、残渣に水50mlを加え、酢酸にてPH4〜5に
調節し、析出する沈殿を取した。
次いで、この沈殿を濃塩酸(4ml)、酢酸(10
ml)中に加え、40分還流撹拌した。冷後、反応液
を水(50ml)中に注ぎ、析出した結晶を取後乾
燥し、粗カルボン酸950mgを得た。
収率:70%

用して、表4に示される遊離カルボン酸塩を合成

結晶を3−メチル−1−ブタノール(1ml)に加
え80〜90℃で30分撹拌し、冷却後、n−ヘキサン
(約3ml)を加え析出した結晶を取した。クロ
ロホルム−n−ヘキサンより再結晶し淡黄色プリ
ズム晶(110mg)を得た。
収率:65%
