Description
本発明は12α−ヒドロキシプレグナ−4−エン
−3−オン−20−カルブアルデヒド及びその製造
法に関する。
本発明により提供される12α−ヒドロキシプレ
グナ−4−エン−3−オン−20−カルブアルデヒ
ドは公知文献に未記載の新規化合物であり、優れ
た抗炎症作用を有するプレドニゾン、プレドニゾ
ロンに代表されるコルチコイド系ステロイドに合
成原料となる12α−ヒドロキシプレグナ−4−エ
ン−3,20−ジオンを製造するための中間体とし
て有用である。
従来、プレドニゾンの製造法としては、デオキ
シコール酸を出発原料として20数段階の工程を経
る方法〔エル・エフ・フイーザー、エム・フイー
ザー共著;ステロイド(レインホルド社発行、
1959年)、634〜647頁参照〕が知られているが、
この方法は用いる試薬が高価であるうえに工程が
長く、工業的実施には必ずしも適していない。
本発明者らはプレドニゾンを製造するためにそ
の有用な中間体について鋭意検討した結果、12α
−ヒドロキシプレグナ−4−エン−3−オン−20
−カルブアルデヒドがデオキシコール酸及び/又
はその塩にシユードモナス属に属する特定の細菌
を作用させることによつて容易に得られ、かつプ
レドニゾンの合成原料となる12α−ヒドロキシプ
レグナ−4−エン−3,20−ジオンに簡単に誘導
できることを見出し、本発明に至つた。

本発明によれば、デオキシコール酸及び/又は
その塩を基質として12α−ヒドロキシプレグナ−
4−エン−3−オン−20−カルブアルデヒドを生
産するシユードモナス属に属する細菌を、デオキ
シコール酸及び/又はその塩を含む培地に培養す
ることにより、12α−ヒドロキシプレグナ−4−
エン−3−オン−20カルブアルデヒドを高選択
率、高収率で得ることができる。
上記のシユードモナス属に属する細菌として
は、シユードモナス・プチダD4014−A357
(Pseudomonas putidaD4014−A357)菌株(微
工研条寄第206号)がある。この菌株は土壌中か
ら採取したシユードモナス・プチダD4014
(Pseudomonas putidaD4014)菌株(微工研条寄
第205号)に突然変異処理を施して得られた変異
株である。
本発明者らが得たシユードモナス・プチダ
D4014菌株及びシユードモナス・プチダD4014−
A357菌株の菌学的性質を列挙すると次表のとお
りである。

上記の表に示した菌学的性質に基づき、シユー
ドモナス・プチダD4014菌株及びシユードモナ
ス・プチダD4014−A357菌株の同定を行なつた。
シユードモナス・プチダD4014菌株は、桿菌であ
ること、極鞭毛を有していること、グラム染色が
陰性であることなどの顕微鏡的所見並びにオキシ
ダーゼ反応及びカタラーゼ反応がともに陽性であ
ること、好気性であること、O−Fテストの結果
が酸化的(Oxidative)であることなどの生理学
的性質からバージエイズ・マニユアル・オブ・デ
イターミネイテイブ・バクテリオロジー第7版及
び第8版に基づき、シユードモナス属に属する細
菌であると同定した。さらにシユードモナス・プ
チダD4014菌株は、培養液が螢光色を帯びる点、
ゼラチンを液化しない点、37℃で生育する点、ア
ルギニン・ジヒドロラーゼを生成する点などから
シユードモナス属のプチダ種に属する細菌である
と同定した。また、一般に突然変異株はその親株
と同じ種に属するものと考えられており、シユー
ドモナス・プチダD4014−A357菌株はシユード
モナス属のプチダ種に属する細菌であると判定し
た。
本発明による12α−ヒドロキシプレグナ−4−
エン−3−オン−20−カルブアルデヒドの生産
は、デオキシコール酸及び/又はその塩を基質と
して12α−ヒドロキシプレグナ−4−エン−3−
オン−20−カルブアルデヒドを生産するシユード
モナス属に属する細菌を、デオキシコール酸及
び/又はその塩を含む培地に培養することにより
行なわれる。デオキシコール酸の塩としては具体
的にはデオキシコール酸のナトリウム、カリウム
などのアルカリ金属の塩又はカルシウム、マグネ
シウムなどのアルカリ土類金属の塩が挙げられ
る。デオキシコール酸及び/又はその塩の濃度は
通常約1〜200g/の範囲でよいが、生産され
る12α−ヒドロキシプレグナ−4−エン−3−オ
ン−20−カルブアルデヒドの収量、培養条件及び
操作性などの経済的観点から約2〜50g/の範
囲が好ましい。培養方法は原則的には一般微生物
の好気培養で採用される方法と同じであるが、通
常は液体培地による振盪培養法又は通気撹拌培養
法が用いられる。培地は上記のデオキシコール酸
及び/又はその塩を基質として12α−ヒドロキシ
プレグナ−4−エン−3−オン−20−カルブアル
デヒドを生産するシユードモナス属に属する細菌
が資化利用できる栄養源を含有するものであれば
よい。炭素源としてはデオキシコール酸及び/又
はその塩を単一炭素源としてもよく、或いはデオ
キシコール酸及び/又はその塩にグルコース、グ
リセリン、ペプトン、肉エキス、酵母エキスなど
を併用してもよい。また窒素源としては、例えば
硫酸アンモニウム、塩化アンモニウム、燐酸アン
モニウム、硝酸アンモニウム、硝酸ナトリウム、
硝酸カリウムなどの無機窒素源、又はポリペプト
ン、ペプトン、肉エキスなどの有機窒素源が用い
られる。また、この他に燐酸水素2カリウム、燐
酸2水素カリウム、硫酸マグネシウムなどの無機
塩類が添加される。培養条件に特徴はないが、通
常25〜35℃で10時間〜7日間振盪培養又は通気撹
拌培養を行なう。
このようにして培養液中に蓄積された12α−ヒ
ドロキシプレグナ−4−エン−3−オン−20−カ
ルブアルデヒドは、基質のデオキシコール酸又は
その塩と比較して水に対する溶解度が著しく小さ
く、通常は培養液中に析出沈澱してくる。この
12α−ヒドロキシプレグナ−4−エン−3−オン
−20−カルブアルデヒドを分離採取するには、沈
澱している12α−ヒドロキシプレグナ−4−エン
−3−オン−20−カルブアルデヒドをデカンテー
シヨンにより浮遊している菌体を含む培養液から
分離するか、または浮遊している菌体が沈澱しな
いような回転数で遠心分離を行ない、析出してい
る12α−ヒドロキシプレグナ−4−エン−3−オ
ン−20−カルブアルデヒドを沈澱させたのち上記
のデカンテーシヨンにより12α−ヒドロキシプレ
グナ−4−エン−3−オン−20−カルブアルデヒ
ドを分離する方法が採られる。沈澱した12α−ヒ
ドロキシプレグナ−4−エン−3−オン−20−カ
ルブアルデヒドを除去した培養液に含まれる菌体
その他の不溶成分を濾過又は遠心分離などにより
分離除去して得られた培養濾液又は上清に、水酸
化ナトリウム、水酸化カリウム、水酸化カルシウ
ムなどのアルカリを加えてその培養濾液又は上清
をアルカリ性としたのち、上記のアルデヒドを溶
解しかつ水と相分離する有機溶媒、例えば酢酸エ
チル、クロロホルム、クロロホルムとメタノール
の混合液などを用いて抽出操作を行ない、得られ
た抽出液を集め、これより溶媒を溜去することに
よつて、培養液中に溶解している12α−ヒドロキ
シプレグナ−4−エン−3−オン−20−カルブア
ルデヒドを回収することができる。この有機溶媒
による抽出操作は培養濾液又は上清についてのみ
でなく、培養液そのものについて行なうことがで
きる。上記の方法で得られた沈澱物又は抽出物中
には12α−ヒドロキシプレグナ−4−エン−3−
オン−20−カルブアルデヒドの他には残存基質の
デオキシコール酸及び/又はその塩並びに副生物
はほとんど含まれておらず、例えばメタノール水
溶液からの再結晶により容易に高純度の12α−ヒ
ドロキシプレグナ−4−エン−3−オン−20−カ
ルブアルデヒドを取得することができる。
本発明により得られる12α−ヒドロキシプレグ
ナ−4−エン−3−オン−20−カルブアルデヒド
は、(方法−1)これを炭酸カリウム、酸化バリ
ウムなどの脱水剤の存在下にピペリジンと反応さ
せて12α−ヒドロキシ−22−(N−ピペリジル)
ビスノル−4,20(22)−コラジエン−3−オンと
し、ついでこの生成物をオゾン酸化することによ
り12α−ヒドロキシプレグナ−4−エン−3,20
−ジオンに誘導できる。
また(方法−2)によれば、12α−ヒドロキシ
プレグナ−4−エン−3−オン−20−カルブアル
デヒドと一般式()

(式中、R1はアルキル基を表わす。)で示され
るカルボン酸又はその反応性誘導体、例えば酸ハ
ライド、酸無水物などとを常法により反応させる
ことにより一般式()で示される12α−アシル
オキシプレグナ−4−エン−3−オン−20−カル
ブアルデヒドが得られる。代表的な反応例として
挙げられる12α−ヒドロキシプレグナ−4−エン
−3−オン−20−カルブアルデヒドと一般式
()で示されるカルボン酸のクロライドとの反
応はトリエチルアミン、ピリジンなどの第3級ア
ミンの存在下に行なわれる。この反応は溶媒中で
行なうのが好ましく、溶媒として塩化メチレン、
クロロホルム又はこれらとベンゼン、トルエン、
酢酸エチルなどとの混合溶媒が好ましく用いられ
る。この反応は通常室温で行なうが、必要に応じ
て約60℃までの加温下に行なうこともできる。反
応後、反応混合物を希塩酸水、重曹水、水などで
洗滌したのち乾燥し、ついでこれより低沸点物を
留去することにより一般式()で示される12α
−アシルオキシプレグナ−4−エン−3−オン−
20−カルブアルデヒドの粗生成物を得る。この粗
生成物をそのまま次の反応に用いることができ
る。一般式()で示される12α−アシルオキシ
プレグナ−4−エン−3−オン−20−カルブアル
デヒドとピペリジン、ピロリジン、モルホリンな
どの第2級アミンとを反応させることにより一般
式()で示されるエナミンが生成する。第2級
アミンは一般式()で示される12α−アシルオ
キシプレグナ−4−エン−3−オン−20−カルブ
アルデヒドに対して等モル〜2倍モル量用いる。
反応中に副生する水を、ベンゼン、トリエンなど
の水と共沸する溶媒を用いて加熱還流下に反応系
から除去する。この反応は特に触媒を要しない
が、p−トルエンスルホン酸などの触媒の存在下
に反応を行なうこともできる。反応後、反応混合
物から減圧下に低沸点物を留去することにより、
一般式()で示されるエナミンの粗生成物が得
られる。この粗生成物をそのまま次の反応に用い
ることができる。一般式()で示されるエナミ
ンをオゾン酸化又は無水クロム酸、ピリジニウム
クロルクロメート、重クロム酸ナトリウムなどを
用いて酸化することにより、一般式()で示さ
れる12α−アシルオキシプレグナ−4−エン−
3,20−ジオンを得ることができる。なお、無水
クロム酸を用いる酸化反応は通常ピリジン溶媒中
で行なう。この場合、一般式()で示されるエ
ナミンを溶解させたピリジン溶液に無水クロム酸
とピリジンの混合液を徐々に加えるか、又は無水
クロム酸とピリジンの混合液に一般式()で示
されるエナミンを溶解させたピリジン溶液を徐々
に加えることにより反応を行なう。この酸化反応
は氷冷下ないしは室温下に行なわれる。反応後、
反応混合液をベンゼン、トルエンなどで希釈し、
これより固形物を濾過により除去したのち、濾液
に希塩酸水を加え、ついでベンゼン、トルエンな
どで抽出し、抽出液から低沸点物を留去すること
により、一般式()で示される12α−アシルオ
キシプレグナ−4−エン−3,20−ジオンの粗生
成物が得られる。この粗生成物を必要に応じてシ
リカゲルカラムクロマトグラフイーにより精製す
るか、再結晶法により精製することにより高純度
の一般式()で示される12α−アシルオキシプ
レグナ−4−エン−3,20−ジオンを得ることが
できる。一般式()で示される12α−アシルオ
キシプレグナ−4−エン−3,20−ジオンを通常
の加水分解反応に付することにより12α−ヒドロ
キシプレグナ−4−エン−3,20−ジオンが得ら
れる。例えば、この加水分解反応はメタノール、
エタノールなどの溶媒中で水酸化カリウム、水酸
化ナトリウムなどの存在下、室温ないし溶媒の還
流温度で行なわれる。反応後、反応混合物を減圧
下に濃縮し、ついでベンゼン、トルエンなどで希
釈し、水、希塩酸水などで洗滌し、乾燥したの
ち、これより低沸点物を留去することにより12α
−ヒドロキシプレグナ−4−エン−3,20−ジオ
ンの粗生成物が得られる。この粗生成物は例えば
酢酸エチルなどから再結晶することにより精製す
ることができる。
12α−ヒドロキシプレグナ−4−エン−3,20
−ジオンは例えば次の方法によりプレドニゾンに
誘導できる。
〔上記式中、Meはメチル基を表わし、Acはア
セチル基を表わす。〕
すなわち、12α−ヒドロキシプレグナ−4−エ
ン−3,20−ジオンを三酸化クロムで酸化し、さ
らにその酸化生成物を水酸化カリウムの存在下に
二酸化セレンで酸化することによりプレグナ−
4,9(11)−ジエン−3,12,20−トリオンを得
る。このプレグナ−4,9(11)−ジエン−3,12,
20−トリオンを白金の存在下に水素添加すること
により12α−ヒドロキシプレグナ−4,9(11)−ジ
エン−3,20−ジオンとし、ついでこの生成物に
酸性条件下でメタノールを作用させて12α−メト
キシプレグナ−4,9(11)−ジエン−3,20−ジオ
ンを得、この生成物に塩化水素を作用させること
により12−クロロプレグナ−4,9(11)−ジエン−
3,20−ジオンを得る。この12−クロロプレグナ
−4,9(11)−ジエン−3,20−ジオンを炭酸水素
ナトリウムで処理することによりプレグナ−4,
11−ジエン−3,20−ジオンを得、この生成物に
臭素を作用させて11,12−ジブロモプレグナ−4
−エン−3,20−ジオンとし、ついでこれをクロ
ム酸銀及び三酸化クロムの存在下に加水分解する
ことにより12−ブロモプレグナ−4−エン−3,
11,20−トリオンを得、この生成物に金属亜鉛を
作用させてプレグナ−4−エン−3,11,20−ト
リオンとする。このプレグナ−4−エン−3,
11,20−トリオンを無水酢酸で処理することによ
り20−アセトキシ−4,17(20)−ジエン−3,11
−ジオンを得る。この生成物に過テレフタル酸を
作用させることにより20−アセトキシ−17α,
20α−エポキシプレグナ−4−エン−3,11−ジ
オンとし、これを加水分解して17α−ヒドロキシ
プレグナ−4−エン−3,11,20−トリオンを
得、ついでこの生成物にヨードを作用させて17α
−ヒドロキシ−21−インドプレグナ−4−エン−
3,11,20−トリオンを得る。この17α−ヒドロ
キシ−21−インドプレグナ−4−エン−3,11,
20−トリオンをアセトン中で酢酸カリウムで処理
することにより17α−ヒドロキシ−21−アセトキ
シプレグナ−4−エン−3,11,20−トリオンを
得る。ついで、この17α−ヒドロキシ−21−アセ
トキシプレグナ−4−エン−3,11,20−トリオ
ンにその1位に二重結合を形成し得る微生物、例
えば容易に入手できるアルスロバクター・シンプ
レツクスIAM1660(Arthrobacter simplex
IAM1660)菌を通常の発酵方法によつて作用さ
せることにより21−アセトキシ−プレドニゾンを
得、これを加水分解することによりプレドニゾン
とすることができる。
また12α−ヒドロキシプレグナ−4−エン−
3,20−ジオンは次の方法により公知化合物であ
るプレグナ−4,11(12)−ジエン−3,20−ジオン
を経由してハイドロコルチゾン、さらにはプレド
ニゾロンに誘導できる。
(上記式中、R2はアルキル基を表わす。)
すなわち、12α−ヒドロキシプレグナ−4−エ
ン−3,20−ジオンを通常のスルホネート化反応
に付することにより、一般式()で示されるス
ルホネートが得られる。例えば、このスルホネー
ト化反応は、12α−ヒドロキシプレグナ−4−エ
ン−3,20−ジオンをピリジン、ピコリン又はこ
れらとベンゼン、トルエンなどとの混合溶媒に溶
解し、この溶液に一般式()

(式中、R2はアルキル基を表わす。)で示され
るスルホン酸クロライドを該12α−ヒドロキシプ
レグナ−4−エン−3,20−ジオンに対し等モル
〜2倍モル量加え、室温ないしは必要に応じて約
80℃までの加温下に行なわれる。反応後、反応混
合物を希塩酸水などにあけ、ベンゼンなどで抽出
し、抽出液を希塩酸水、重曹水、水などで洗滌
し、乾燥したのち、これより低沸点物を留去する
ことにより一般式()で示されるスルホネート
の粗生成物が得られる。この粗生成物を例えば酢
酸エチルなどから再結晶することにより高純度の
一般式()で示されるスルホネートを得ること
ができる。一般式()で示されるスルホネート
は、これを脱スルホン酸反応に付することによ
り、公知化合物であるプレグナ−4,11(12)−ジエ
ン−3,20−ジオンに誘導される。この脱スルホ
ン酸反応は通常、酢酸カリウム、塩化リチウム、
コリジン、ポタシウムt−ブトキサイドなどの反
応助剤(スルホン酸を捕捉することができる化合
物)の存在下に行なわれる。反応助剤の使用量は
一般式()で示されるスルホネートに対して等
モル〜20倍モル量である。この反応はヘキサメチ
ルホスホルトリアミド、N,N−ジメチルホルム
アミドなどの溶媒中で行なうのが好ましく、通常
約80〜140℃の温度下に加熱して行なわれる。プ
レグナ−4,11(12)−ジエン−3,20−ジオンは常
法によりハイドロコルチゾン、さらにはプレドニ
ゾロンに誘導される。
以下実施例及び参考例によつて本発明をさらに
詳細に説明する。
参考例
シユードモナス・プチダD4014−A357菌株の
取得方法
培地1(組成:デオキシコール酸0.5%、水酸化
ナトリウム0.05%、ペプトン0.5%、酵母エキス
0.5%、塩化ナトリウム0.5%及び寒天1.5%)のス
ラントに生育させたシユードモナス・プチダ
D4014菌株の一白金耳を、予め試験管内に準備し
た培地2(組成:デオキシコール酸2%、水酸化
ナトリウム0.2%、硝酸アンモニウム0.2%、燐酸
2水素カリウム0.1%、燐酸水素2カリウム0.6
%、硫酸マグネシウム・7水和物0.02%及び酵母
エキス0.02%)の10mlに植菌し、30℃で14〜15時
間振盪培養した。この培養液の0.3mlを予め試験
管に準備した培地3(組成:デオキシコール酸0.5
%、水酸化ナトリウム0.05%、グルコース0.1%、
硝酸アンモニウム0.2%、燐酸2水素カリウム0.1
%、燐酸水素2カリウム0.6%、硫酸マグネシウ
ム・7水和物0.02%及び酵母エキス0.02%)の10
mlに加え、30℃で8〜9時間培養した。ついで、
この対数増殖期にある菌体を0.45μのメンブレン
フイルターで無菌的に濾過集菌し、0.1M燐酸塩
緩衝液(PH:7.0)20mlで洗滌後、同じ緩衝液25
mlに懸濁させた。これに終濃度が50μg/mlにな
るようにN−メチル−N′−ニトロ−N−ニトロ
ソグアニジンを添加し、3〜4分放置することに
より突然変異処理を行なつた。突然変異処理を施
した菌体を0.45μのメンブレンフイルターで濾過
集菌し、0.1M燐酸塩緩衝液(PH:7.0)20mlで洗
滌後、同じ緩衝液20mlに懸濁した。得られた菌懸
濁液を滅菌生理食塩水で希釈し、それを培地4
(組成:デオキシコール酸0.5%、水酸化ナトリウ
ム0.05%、硝酸アンモニウム0.2%、燐酸2水素
カリウム0.1%、燐酸水素2カリウム0.6%、硫酸
マグネシウム・7水和物0.02%、酵母エキス0.02
%及び寒天1.5%)の寒天平板上に500〜1000個の
コロニーを出現させるように塗布したのち、30℃
で3〜4日間培養した。出現したコロニー中の極
小コロニーを培地1のスラントに単離したのち、
その一白金耳を予め試験管に準備した培地5(組
成:デオキシコール酸0.2%、水酸化ナトリウム
0.02%、グルコース0.1%、硝酸アンモニウム0.2
%、燐酸2水素カリウム0.1%、燐酸水素2カリ
ウム0.6%、硫酸マグネシウム・7水和物0.02%
及び酵母エキス0.02%)の10mlに植菌し、30℃で
24時間振盪培養した。得られたそれぞれの培養液
中の生産物を薄層クロマトグラフイーにより検定
し、目的とする12α−ヒドロキシプレグナ−4−
エン−3−オン−20−カルブアルデヒドを選択的
に蓄積している一菌株を見い出し、これをシユー
ドモナス・プチダD4014−A357と命名した。
実施例 1
シユードモナス・プチダD4014−A357菌株
(微工研条寄第206号)を次に示す方法で培養し
た。デオキシコール酸0.2g、グルコース0.1g、
硝酸アンモニウム0.2g、燐酸2水素カリウム0.1

により培養中に生じた沈澱物と菌体を集め、水洗
したのち、これにメタノール50mlを加えた。沈澱
物を充分にメタノールに溶解したのち、再度遠心
分離機により上清のメタノール溶液を得た。ロー
タリー・エバポレーターでメタノールを留去する
ことにより、12α−ヒドロキシプレグナ−4−エ
ン−3−オン−20−カルブアルデヒド65mgを得
た。
得られた12α−ヒドロキシプレグナ−4−エン
−3−オン−20−カルブアルデヒドの一部をと
り、これにメタノールを加えて4%溶液とし、こ
の溶液25μをミクロボンダパツクC−18カラム
を備えた高速液体クロマトグラフイー(米国ウオ
ーターズ社製、HLC−GPC−244型)に注入し
た。移動相としてPH4.0に調整した水/メタノー
ルの30/70容量比の混合液を流速1ml/分で流し、
検出を屈折率方式で行なつた。得られた液体クロ
マトグラフにおける各ピークの面積比を積分計
(島津製作所製、島津クロマトパツクC−R1A)
で求め、この面積比から上記の12α−ヒドロキシ
プレグナ−4−エン−3−オン−20−カルブアル
デヒドの純度を求めたところ、98%であつた。
上記で得られた12α−ヒドロキシプレグナ−4
−エン−3−オン−20−カルブアルデヒドの確認
を下記の方法で行なつた。
